The effects of temperature on growth and production of Lipophilic Toxins (LT) by a monoclonal culture of Dinophysis caudata was studied. The cell density of D. caudata increased significantly with increasing temperature, and was the highest under 27, 30, and 32.5 °C. Temperature affected the average specific growth rate (µ) during the exponential growth phase (EG), which increased from 15 °C to 30 °C, and then decreased at 32.5 °C. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) revealed that this strain of D. caudata produced only pectenotoxin-2 (PTX-2) whose concentration increased significantly with incubation period, except at 32.5 °C. It was significantly different between temperatures ≤18 °C, ≥21 °C, and 32.5 °C. The cellular toxin production (CTP, pg·cell−1·day−1) showed variation with growth phase and temperature, except at 32.5 °C. The average net toxin production (Rtox) was not affected by temperature. During EG, the average specific toxin production rate (µtox) increased significantly with increase in temperature, reaching a peak of 0.66 ± 0.01 day−1 at 30 °C, and then decreased. Over the entire growth span, µtox was significantly correlated to µ, and this correlation was most significant at 27 and 30 °C. During EG, µtox was affected by both temperature and growth. This study shows that temperature affects growth and toxin production of this strain of D. caudata during EG. In addition, a positive correlation was found between toxin production and growth.  相似文献

Starch and protein separated from oat were chemically modified using cross‐linking and acetylation protocols for starch, and deamidation and succinylation for protein isolate. Cross‐linking decreased swelling power of starch, whereas syneresis increased, but cross‐linking does not have a significant effect on gelatinization temperature. Acetylation increased swelling power, but gelatinization temperature and syneresis diminished. Deamidation and succinylation increased nitrogen solubility index, emulsion activity, foaming capacity, and water and oil binding capacity. Emulsion stability did not change with deamidation and it diminished with succinylation, while foaming stability decreased with both treatments. Acetylated starch and two types of modified proteins were substituted for 5, 10, 15, and 20% of oat flour to bake cake samples and then physical properties of the cakes were measured. Acetylated starch increased batter viscosity, cake volume, and whiteness of cake crust. Increased level of deamidated protein produced cakes with lower batter viscosity, higher volume, and darker color (increase in redness). Application of higher levels of succinylated protein led to higher batter viscosity and lightness, and lower cake volume. Therefore, substitution of deamidated protein and acetylated starch can improve cake properties.  相似文献

In recent years, common bean (Phaseolus vulgaris) and cowpea (Vigna unguiculata) plants in the north of Iran have exhibited symptoms resembling Cercospora leaf spot (CLS) disease. This study was initiated to elucidate the taxonomy and pathogenicity of cercosporoid taxa associated with leaf spot diseases of these two legume crops in Iran. A total of 138 samples with CLS symptoms were collected from cultivated common bean and cowpea species in northern Iran and subjected to microscopic examination, resulting in identification of 98 Cercospora and 59 Pseudocercospora samples. A six-locus phylogenetic analysis (ITS, actA, tef1, gapdh, his3, and cmdA) coupled with examination of the morphology of 42 representative isolates from these samples confirmed that several cercosporoid fungi occur on common bean and cowpea in Iran. Five Cercospora species (C. iranica, C. cf. flagellaris, Cercospora sp. G, Cercospora sp. T, and C. vignigena) and two Pseudocercospora species (P. griseola f. griseola and P. cf. cruenta) were found; of these, C. cf. flagellaris was the dominant species, occurring on both common bean and cowpea. Pathogenicity tests confirmed that all seven species could infect leaves of common bean and/or cowpea. This is the first report of C. iranica, Cercospora sp. G, and Cercospora sp. T associated with common bean and/or cowpea in the world. In addition, C. vignigena was recorded for the first time in Iran. Results achieved in this study will assist strategies for the management of CLS disease of common bean and cowpea.  相似文献

In order to study the effect of plant growth promoting rhizobacteria (PGPR), Azolla compost and Azolla biochar on some soil quality indicators and rosemary growth, a greenhouse experiment was conducted in a completely randomized design with six replications. Treatments consisted of T1 (control), T2 (1% Azolla compost), T3 (1% Azolla biochar), T4 (PGPR (P. fluorescens), T5 (1% compost + PGPR) and T6 (1% biochar + PGPR). Rosemary growth parameters and nutrients concentration increased in all treatments compared to control. Treatments increased soil nutrient concentrations, soil microbial respiration (SMR) and microbial biomass C (MBC) but decreased soil metabolic quotient (qCO2) compared to control treatment. A significant enhancement in rosemary growth occurred due to the improved soil quality as a result of organic fertilizers application, particularly by co-application of P. fluorescens and compost or biochar of Azolla.  相似文献

Water shortage is a critical issue worldwide, and it may adversely impact non-food landscape plants. Thus, the impact of two levels of evapotranspiration-based (ETc) water stresses and two biostimulants consisting of s-abscisic acid (s-ABA) and glycine betaine (GB), and their combined applications on perennial ryegrass (Lolium perenne) under climatic and soil conditions of the Intermountain West, USA, were studied. Clippings with 50% ETc had higher percentage dry weight (DW) but lower fresh weight (FW) and chlorophyll index (CI) than those with 75% ETc. The performance rating of plots with 75% ETc was significantly higher (better) than those receiving the 50% ETc treatment. Clippings with the 75% ETc treatment had higher concentrations of nitrogen (N), nitrate, phosphorous (P), sulfur (S), potassium (K), sodium (Na), and copper (Cu) than those with the 50% ETc treatment. Considering all mineral nutrient values, CIs, and performance ratings, we conclude that the application of 75% ETc is sufficient for maintaining a healthy turfgrass with a satisfactory appearance, while we can save 25% water as compared to the application of water at 100%. The application of biostimulants had no effect on clipping or root FWs, DWs, or percentage DWs. Clippings from GB-treated turfgrass had significantly higher N than those from all other biostimulants or non-treated control. Additionally, clippings from the plots with the GB treatment had significantly higher S, K, and Cl but lower Zn. Clippings from the s-ABA-treated turfgrass also had significantly higher K than those in non-treated control. Average performance ratings in s-ABA and GB, and s-ABA and GB were significantly higher than those in non-treated control, underscoring the values of these biostimulants in the reduction of drought stress.  相似文献
