马传染性贫血病毒(EIAV)减毒疫苗是世界首例慢病毒疫苗,但其作用机理尚不明了。我们的前期研究发现EIAV疫苗株EIAVFDDV12编码跨膜蛋白gp45的基因在马体内发生高频率G2230A点突变形成终止子,使该蛋白C末端出现154个氨基酸的截短。为了探讨该截短蛋白对EIAV疫苗株生物学特性的作用,本研究以EIAV弱毒疫苗株感染性克隆基础上,构建了gp45截短型感染性克隆,脂质体转染细胞,增殖传代,获得病毒株EIAVFDDVTM-36,接种到马体内。在接种后167 d内未观察到临床症状,连续注射10 d地塞米松以暂时抑制免疫系统,并用迟发性超敏反应和淋巴细胞增殖实验评价免疫抑制效果。结果显示,截短型跨膜蛋白疫苗株EIAVFDDVTM-36接种组免疫抑制前后病毒载量和中和抗体效价差异不显著,而完整型跨膜蛋白疫苗株感染性克隆EIAVFDDVTM-45接种组4匹马中3匹免疫抑制后病毒载量显著上升,同时组内中和抗体效价明显提高。以上结果表明,马体特异性免疫压力对EIAVFDDVTM-36复制无影响,而对亲本株EIAVFDDVTM-45复制有抑制,显示跨膜蛋白截短型EIAV作为疫苗更为安全。但免疫抑制造成的gp45跨膜蛋白未截短型疫苗株感染性克隆体内病毒载量升高,可以促使机体产生更高的中和抗体效价,提示该疫苗的安全性和有效性在一定程度内呈负相关(相互制约)。因此,安全性和有效性的平衡是慢病毒减毒疫苗研发需注意的重要因素。 相似文献

The probiotics tested in the experiment were isolated from the intestinal of weaning piglets.The isolated probiotics and E.coli K88 were inoculated into the culture of intestinal porcine epithelial cell-1 (IPEC-1).The activity of lactate dehydrogenase (LDH) in the supernatant was measured after incubating for 2.5 h.At the same time, the probiotics and E.coli K88 were co-cultured in vitro, the number of E.coli K88 was counted and the probiotics which could be resistant to the E.coli K88 were selected 2.5 h later.The results showed that the Lactobacillus casei and Bacillus coagulans could significantly reduce LDH activity (P<0.05), decrease the damage of E.coli K88; Bacillus coagulans could inhibit the growth of E.coli K88.At the same time, Bacillus coagulans could resist high temperature, acid and bile salt.The results showed that Bacillus coagulans strains had great potential as the application of probiotics strains.The methods could be used as a model of screen probiotics which could inhibit the growth of E.coli K88 in vitro. 相似文献

植物种群密度及空间分布的研究可以发现植物群落在放牧过程中的生态学现象及规律。为研究荒漠草原无芒隐子草(Cleistogenes songorica)种群在不同放牧强度下的密度差异及空间分布变化,采用随机区组试验设计:对照区(Contral,CK)、轻度(Light grazing,LG)、中度(Moderate grazing,MG)和重度放牧区(Heavy grazing,HG),通过机械取样法收集数据,结合地统计等方法进行分析。结果表明:无芒隐子草密度表现为LG>HG>CK>MG,随放牧强度增大,无芒隐子草的密度先增加,后减少,在重度放牧条件下,密度又增加。无芒隐子草在群落中的绝对密度占比伴随放牧强度增大逐渐增大,占比为HG>MG>CK>LG。无芒隐子草在4个处理区下分型维度D0数值相近,CK,LG,MG的空间结构比C/(C0+C)均>0.75,HG的空间结构比C/(C0+C)为0.100。放牧条件下,无芒隐子草种群空间分布较为均匀,空间异质性减弱,空间分布主要受结构性因素影响。 相似文献

为了研究不同猪种背最长肌中氨基酸沉积规律,试验采用高效液相色谱检测了不同体重梯度(90、110和130 kg)的川藏黑猪和杜长大(DLY)猪背最长肌中氨基酸含量。结果表明,谷氨酸在两猪种肌肉中含量最高,含硫氨基酸总量在90和130 kg体重梯度时表现为川藏黑猪显著高于DLY猪(P〈0.05),110 kg体重梯度时极显著高于DLY猪(P〈0.01),而同一猪种不同体重梯度间各种氨基酸组成差异均不显著(P〉0.05),鲜味氨基酸占总氨基酸比例(FAA/TAA)在90 kg体重梯度表现为川藏黑猪极显著高于DLY猪(P〈0.01),130 kg体重梯度时显著高于DLY猪(P〈0.05)。本研究说明川藏黑猪肌肉中与风味相关的氨基酸含量高于DLY猪,我们认为这也是川藏黑猪的肌肉风味比DLY猪更浓郁的重要原因之一。我们的研究初步揭示了不同猪种背最长肌中氨基酸的沉积规律及构成差异,为进一步研究猪肉风味形成机理提供数据。 相似文献

AIM:To investigate the antitumor activity and mechanism of Paecilomyces tenuipes polysaccharide(PTPS).METHODS:PTPS-I was obtained by water extraction and alcohol precipitation, and purified by DEAE-cellulose and Sephadex G-100 chromatography. Human erythroleukemia cell line K562, laryngocarcinoma cell line Hep2 and hepatic carcinoma cell line SMMC-7721were co-cultured with PTPS-I or the conditioned medium which prepared with PTPS-I-stimulated human mononuclear cells (PTPS-I-MNC-CM), and the proliferation of tumor cells was determined. The cell counting kit-8 (CCK-8) was used to determine the proliferation of MNCs. The FQ-RT-PCR was applied to investigate the expression of TNF-α and IL-6 mRNA in MNCs. RESULTS:PTPS-I-MNC-CM inhibited the proliferation of K562, Hep2 and SMMC-7721 cells in vitro (P<0.01). Cytotoxicity of PTPS-I against K562, Hep2 and SMMC-7721 cells was not observed (P<0.01). PTPS-I stimulated the proliferation of MNCs (P<0.01) and significantly enhanced the expression of TNF-α and IL-6 mRNA in MNCs (P<0.05, P<0.01). CONCLUSION:The results suggest that PTPS-I is an immunomodulator and its antitumoral activity is through the immunomodulatory mechanism rather than the direct cytotoxicity against tumor cells. 相似文献

According to the multiple alignments identified major histocompatibility complex Ⅰ (MHC Ⅰ) gene conserved sequence registered in GenBank from the family ducks (Anatidae) anser waterfowl, a pairs of specific primers for the fragments of MHCⅠgene of goose F1 from fast-growth lines were designed and synthesized by Primer Premier 5.0. Using the genome DNA of goose F1 from fast-growth lines, the target gene fragment was obtained by PCR. To conduct sequencing of the fragments of MHCⅠgene of goose F1 from fast-growth lines and make sequence alignment and analysis of protein structure and function by bioinformatics, and research the characteristics of MHCⅠgene of goose F1 and the physicochemical properties of the protein. Bioinformatics was analyzed the nucleic acid data, deduced amino acid sequence and phylogenetic trees. The result of sequence analysis showed that the fragments of MHCⅠgene of goose F1 from fast-growth lines was 1036 bp in length, which coded 96 amino acids polyprotein. The homology were 93% and 83% with MHC Ⅰ gene and coding sequence of Wulong goose in NCBI respectively. There were 72 different bases sequence and 16 amino acids change. There also was higher homology with other poultry, and existed genetic relationship of Siji goose > chickens > ducks.The homology segment sequences corresponding to the fragments of MHCⅠ gene of goose F1 coded 96 amino acids protein, which molecular weight, PI, positively or negatively charged amino acid, estimated half-life, instability index, aliphatic index and average hydrophobicity were 11.342 ku, 5.32, 14, 17, 2.8 h, 34.92, 42.81, -1.066, respectively, and appeared 9 B cell epitopes, but contained no signal peptide. These results indicated that the protein for hydrophilic non-secreted proteins, had the high immunogenicity. In addition, The protein structure study indicated that alpha-helix, beta-sheet, beta-turn and random coil were 31.25%,16.67%, 14.58% and 37.50%, respectively. There existed amino terminal domain and carboxyl terminal domain in the tertiary structure. Therefore, MHC gene had significant difference between species and populations of individuals by the pathogen pressure in environment, and there were the interaction between polymorphism of MHC molecules and the diversity of antigenic peptide. MHC determined the differences of individual susceptibility to disease, and could be treated as a candidate gene for disease resistance. 相似文献
