The objective of this study was to describe the population pharmacokinetics (PK) of mosapride under fasting and fed conditions. A single 5‐mg oral dose of mosapride was administered to fasted (n = 15) and fed (n = 12) beagle dogs. Plasma concentrations of mosapride were subsequently measured by liquid chromatography–tandem mass spectrometry. Data were analyzed using modeling approaches with the NONMEM 7.2 software. A one‐compartment open PK model utilizing model event time (MTIME) with first‐order absorption and first‐order elimination was found to be more appropriate than all other PK models tested. The absorption rate constants of mosapride were significantly decreased under fed conditions, compared to fasting conditions. The observed bootstrap medians of PK parameters were generally consistent with the corresponding population mean estimates. Furthermore, with the exception of some mosapride concentrations, most of observed data fell into the range of the 5th and 95th percentiles of the simulated values. Overall, the final model was able to describe the observed mosapride concentrations reasonably well. These findings suggest that food intake affects both the rate and extent of absorption of mosapride and that the pharmacological effect of mosapride can differ significantly depending on food intake. 相似文献

Root-lesion nematodes of the genus Pratylenchus are migratory endoparasites with worldwide economic impact on several important crops including potato, where certain species like P. penetrans, P. neglectus, and P. scribneri reduce the yield and quality of potato tubers. Morphological identification of Pratylenchus spp. is challenging, and recent advancements in molecular techniques provide robust and rapid diagnostics to differentiate species without the need of specialist skills. However, the fact that molecular diagnostics are not available for all Pratylenchus species means that there are limitations in worldwide application. In general, root-lesion nematodes are difficult to manage once introduced into agricultural land and damage can be related to pathogenicity and population densities. In addition, root-lesion nematodes interact with fungi such as Verticillium dahliae, resulting in disease complexes that enhance the damage inflicted on the potato crop. Management interventions are often focused on limiting nematode reproduction before planting crops and include the application of nematicides, and cultural practices such as crop rotation, cover crops, biofumigation, and biological control. Understanding the limitations of the available crop protection strategies is important and there are many gaps for further study. This review discusses the status of the diagnosis, distribution, pathogenicity, and management of the main species of root-lesion nematodes, reported to infect potatoes worldwide, and highlights areas for potential future research. 相似文献

REASONS FOR PERFORMING STUDY: Overload injuries in sport horses commonly occur; shoeing techniques are believed to be important in prevention of these injuries, but there is a paucity of scientific information identifying the potential connection. OBJECTIVES: To test a horseshoe with a modified rolled toe designed to ease the process of breakover and decrease loading of lesion-prone structures of the distal limb. METHODS: Twenty clinically sound Warmblood horses trotted over a track containing a pressure/force measuring system and 6 infrared cameras. The horses were measured with 2 types of shoes, standard flat shoes and shoes with a rolled toe. The shoeing procedure was randomised and horses had 2 days between measurements to adapt to the shoes. RESULTS: Limb placement and timing characteristics, e.g. breakover duration, did not change significantly. There was an improvement in the ease of movement to roll over the toe in the shoes with a rolled toe, due mainly to a smoother hoof-unrollment pattern. The peak indicative moment decreased substantially at the onset of breakover in the shoe with the rolled toe. CONCLUSIONS: With a rolled toe the process of hoof-unrollment is smoother, which improves the coordination of this process, and lowers peak loading of the distal limb during breakover. POTENTIAL RELEVANCE: This study stresses the importance of proper shoeing in sound horses, showing that shoe modifications can optimise the loading characteristics of the distal limb and therefore might be a means to prevent sport horses from overload injuries. 相似文献

In March 2011, witches’ brooms comprising many small shoots were observed on desert rose plants, Adenium obesum, in PyinOoLwin, Myanmar. The causal agent of the symptomatic leaves was diagnosed as a phytoplasma by polymerase chain reaction
(PCR) analysis. Sequence analysis of the PCR product (1.8 kbp) showed the closest phylogenic relationships with members of
the peanut witches’ broom phytoplasma group. Additionally, phylogenetic analyses revealed the phytoplasma is a member of ‘Candidatus Phytoplasma aurantifolia’. This is the first report of desert rose plant as a new host for ‘Ca. P. aurantifolia’. 相似文献

Equine canker is a chronic, hyperplastic, exudative pododermatitis affecting one or more feet. Although many causes and treatments have been suggested, the cause of the disease is still unknown and most probably multifactorial. Local treatments include radical surgical debridement of the diseased hoof tissue and application of caustic substances, antibiotics, and pressure bandaging. Nevertheless, the number of recurrences is high (45%). This article presents a 3-year-old New Forest pony-cross mare in which all horny structures (frogs, coronets, spurs, chestnuts) of all feet were affected. Bacteriological and fungal cultures of the frogs were found negative for the pathogens tested. Papilloma virus was not found. Clinical findings raised the hypothesis that the non-specific hyperplastic inflammation of these horn-like structures might have been caused by an (auto-)immune reaction. On the basis of the clinical findings, the pony was treated with surgical debridement of the frogs of a diagonal pair of feet and oral administration of prednisolone (1 mg/kg sid). The frogs, coronets, spurs, and chestnuts of all four feet healed completely within 8 weeks, thus making an (auto-)immune reaction more likely. In conclusion, this case report raised the hypothesis that an aspecific, hyperplastic inflammation of all four feet ('equine canker') and other horny structures may be caused by an (auto-)immune reaction, and that corticosteroids (prednisolone 1 mg/kg sid per os) are effective as treatment. 相似文献
