Aphanomyces euteiches causes severe root rot of peas. Resistance is limited in commercial pea cultivars. Real-time fluorescent PCR assay specific for A. euteiches was used to study the relationship between disease severity and pathogen DNA content in infected peas. Five pea genotypes ranging in levels of resistance were inoculated with five isolates of A. euteiches. Plants were visually rated for disease development and the amount of pathogen DNA in roots was determined using the PCR assay. The susceptible genotypes Genie, DSP and Bolero tended to have significantly more disease and more pathogen DNA than the resistant genotypes 90-2079 and PI 180693. PI 180693 consistently had less disease, while 90-2079 had the lowest amount of pathogen DNA. The Spearman correlation between pathogen DNA quantity and disease development was positive and significant (P < 0.05) for three isolates, but was not significant for two other isolates. This suggests that the real-time PCR assay may have limited application as a selection tool for resistance in pea to A. euteiches. Its utility as a selection tool would be dependent on the correlation between disease development and pathogen DNA content for a given pathogen isolate. The accuracy and specificity of the real-time PCR assay suggests considerable application for the assay in the study of mechanisms of disease resistance and the study of microbial population dynamics in plants.  相似文献

BACKGROUND: Interpretation of serial urine protein:creatinine (UPC) values is confounded by a lack of data regarding random biologic variation of UPC values in dogs with stable glomerular proteinuria. HYPOTHESIS: That there is minimal day-to-day variability in the UPC of dogs with unchanging proteinuria and the number of measurements needed to reliably estimate UPC varies with the magnitude of proteinuria. ANIMALS: Forty-eight heterozygous (carrier) female dogs with X-linked hereditary nephropathy (XLHN) causing stable proteinuria. METHODS: Urine samples were obtained daily by cystocentesis for 3 consecutive days on 183 occasions (549 samples). The UPC was measured for each sample with a single dry-film chemistry auto-analyzer. Data were analyzed retrospectively by a power of the mean model because the variance of UPC values within the 3-day evaluation periods increased as the magnitude of proteinuria increased. RESULTS: To demonstrate a significant difference (P < .05) between serial values in these proteinuric dogs, the UPC must change by at least 35% at high UPC values (near 12) and 80% at low UPC values (near 0.5). One measurement is adequate to reliably estimate the UPC when UPC <4, but 2-5 determinations are necessary at higher UPC values. CONCLUSIONS AND CLINICAL IMPORTANCE: These guidelines for interpretation of serial UPC values in female dogs with XLHN may also be helpful for interpretation of UPC values in dogs with other glomerulopathies.  相似文献

Background

Data on the movement behavior of translocated wild pigs is needed to develop appropriate response strategies for containing and eliminating new source populations following translocation events. We conducted experimental trials to compare the home range establishment and space-use metrics, including the number of days and distance traveled before becoming range residents, for wild pigs translocated with their social group and individually.

Results

We found wild pigs translocated with their social group made less extensive movements away from the release location and established a stable home range ~5 days faster than those translocated individually. We also examined how habitat quality impacted the home range sizes of translocated wild pigs and found wild pigs maintained larger ranges in areas with higher proportion of low-quality habitat.

Conclusion

Collectively, our findings suggest translocations of invasive wild pigs have a greater probability of establishing a viable population near the release site when habitat quality is high and when released with members of their social unit compared to individuals moved independent of their social group or to low-quality habitat. However, all wild pigs translocated in our study made extensive movements from their release location, highlighting the potential for single translocation events of either individuals or groups to have far-reaching consequences within a much broader landscape beyond the location where they are released. These results highlight the challenges associated with containing populations in areas where illegal introduction of wild pigs occurs, and the need for rapid response once releases are identified. © 2023 The Authors. Fasciolosis is an endemic zoonotic parasitic disease with significant impacts on human health and both animal health and production. Early post-infection impacts on the host remain unclear. The objective of this study was to determine the changes, if any, to levels of endotoxin in cattle plasma in response to early-stage infection with Fasciola hepatica. Thirty-six (36) commercial bred cattle were experimentally infected with approximately 400 viable metacercariae. Plasma lipopolysaccharide (endotoxin) levels were examined on 24 occasions from 0 h before infection to 336 h after infection using the Limulus Amoebocyte Lysate chromogenic end point assay and compared with that of six (6) uninfected control animals. Peak lipopolysaccharide levels in infected animals were reached at 52 h after infection and returned to pre-infection levels at time 144 h after infection. Infected animals had significantly elevated lipopolysaccharide levels between 24 and 120 h after infection when compared to uninfected animals. The mean change in endotoxin units (EU)/mL over time after infection was statistically significant in infected animals. Elevations of lipopolysaccharide occurred in all infected animals suggesting a possible repeatable and titratable endotoxemia conducive to therapeutic agent model development.  相似文献

The acute and chronic toxicities of three benzoylphenyl ureas, diflubenzuron, hexaflumuron and teflubenzuron were assessed under laboratory conditions against two-day-old second (II) instar Desert Locust, Schistocerca gregaria (ForskÅl) nymphs. Following exposure by ingestion of a single precise dose applied to short pieces of spring barley, nymphs were monitored for two moults until the fourth (IV) instar. Analysis of acute response data gave three significantly different LD50 statistics (P < 0·05), 68·0, 26·6 and 0·71 μg per nymph respectively for diflubenzuron, hexaflumuron and teflubenzuron. The probit regression slopes also differed significantly, indicating distinct tolerance distributions for the three compounds, the narrowest response being to diflubenzuron and the widest range of response being to teflubenzuron. The timing of death was found to vary between the compounds; most nymphs died during the first moult following treatment with either hexaflumuron or teflubenzuron. However, the majority of nymphs that died after exposure to diflubenzuron did so after completing the first moult after treatment, but before the second. The mean development times of nymphs during the II and especially third (III) instars were significantly longer (P < 0·05) than those of the controls following exposure to diflubenzuron and hexaflumuron. Teflubenzuron had no significant effect (P < 0·05) on the duration of the II instar. The potential of the three compounds to control S. gregaria populations in the field is discussed with particular reference to the timing and nature of acute and chronic responses.  相似文献

Commercial fertilizer (particularly nitrogen) costs account for a substantial portion of the total production costs of cellulosic biomass and can be a major obstacle to biofuel production. In a series of greenhouse studies, we evaluated the feasibility of co-applying Gibberellins (GA) and reduced nitrogen (N) rates to produce a bioenergy crop less expensively. In a preliminary study, we determined the minimum combined application rates of GA and N required for efficient biomass (sweet sorghum, Sorghum bicolor) production. Co-application of 75 kg ha?1 (one-half of the recommended N rate for sorghum) and a modest GA rate of 3 g ha?1 optimized dry matter yield (DMY) and N and phosphorus (P) uptake efficiencies, resulting in a reduction of N and P leaching. Organic nutrient sources such as manures and biosolids can be substituted for commercial N fertilizers (and incidentally supply P) to further reduce the cost of nutrient supply for biomass production. Based on the results of the preliminary study, we conducted a second greenhouse study using sweet sorghum as a test bioenergy crop. We co-applied organic sources of N (manure and biosolids) at 75 and 150 kg PAN ha?1 (representing 50 and 100% N rate respectively) with 3 g GA ha?1. In each batch of experiment, the crop was grown for 8 wk on Immokalee fine sand of minimal native fertility. After harvest, sufficient water was applied to soil in each pot to yield ~1.5 L (~0.75 pore volume) of leachate, and analyzed for total N and soluble reactive P (SRP). The reduced (50%) N application rate, together with GA, optimized biomass production. Application of GA at 3 g ha?1, and the organic sources of N at 50% of the recommended N rate, decreased nutrient cost of producing the bioenergy biomass by ~$375 ha?1 (>90% of total nutrient cost), and could reduce offsite N and P losses from vulnerable soils.  相似文献
