A search for double‐stranded RNA (dsRNA) was conducted among 204 European isolates of the pathogenic fungus Heterobasidion annosum. Nucleic acids were extracted and purified by cellulose CF‐11 chromatography or lithium chloride precipitation. dsRNA was present in eight of the isolates and was confirmed by nuclease digestion. The dsRNA elements ranged between 1.8 and 2.4 kbp and were found in two H. annosum intersterility groups, S and P. Partial amino acid sequence information from one dsRNA element showed significant homology to RNA‐dependent RNA polymerases from several fungal partitiviruses. This is the first report of the presence of dsRNAs in H. annosum. Possible implications of dsRNA for the biology of the fungus and the potential for biological control are discussed. 相似文献
Seeds of Juniperus procera collected from five provenances across its geographic range in Ethiopia were subjected to cold-moist stratification at 5°C
or 10°C for 6–12 weeks. The effect of aqueous smoke solution in overcoming the light requirement for germination, and the
potential of visible (VIS) and near infrared reflectance (NIR) spectroscopy for sorting sound and insect-damaged seeds were
also investigated. Highly significant differences in germination were detected among provenances (P < 0.0001) and stratification periods (P < 0.0001), but not between temperature regimes (P=0.111). Seeds from the south and southeast distribution ranges had higher percentage germination after 6 weeks of stratification
than seeds collected from north, northwest and central ranges of distribution. The smoke treatment did not affect germination
regardless of whether the seeds were exposed to light. Exposure to light increased germination capacity three fold. Sound
and damaged seeds were distinguished with 90% accuracy using VIS + NIR spectroscopy. It can be concluded that dormancy in
juniper seeds varies with provenances, and cold stratification for 6 weeks alleviates dormancy in some seed lots. Tentatively,
smoke treatment seems ineffective in overcoming photo-dormancy in juniper seeds. VIS + NIR spectroscopy has demonstrated a
great potential for sorting damaged seeds, thereby upgrading seed lot purity. 相似文献
The aim of this study was to determine changes in crystallinity and crystalline unit cell type of heat-treated Scots pine
(Pinus sylvestris L.) and Uludağ fir (Abies nordmanniana stev. subsp. bornmuelleriana Mattf.) wood samples by means of FT-IR spectroscopic method. Heat treatment was applied on the test samples in an oven at
three different temperatures (120, 150, and 180°C) and for two different periods of time (6 and 10 h) under atmospheric pressure.
It was designated that crystallinity of both Scots pine and Uludağ fir wood samples increased during heat treatment depending
on the duration. However, monoclinic structure in crystalline unit cells of Scots pine and Uludağ fir wood samples converted
to triclinic structure when heat treated. It was estimated that monoclinic structure was dominant in the crystalline unit
cell. It was established that the crystalline structure of Scots pine wood samples was more affected by heat treatment than

The mechanical behavior of steamed spruce wood changes dramatically with compression along the grain, the change being much more moderate perpendicular to the grain. The stiffness decrement due to increased temperature is greatest in the tangential material direction. The stiffness decrement due to compression is greatest along the grain. Compression to 80% compressive strain at 131°C inverts the order of the material directions regarding stiffness, the stiffness being the least along the grain. Plastic strain due to compression is greater at higher temperatures. The compression-induced decrement of stiffness along the grain is greater at higher temperatures, but the off-axis decrement of stiffness is less at higher temperatures. 相似文献
Training of Standardbred race horses aims to improve cardiovascular and metabolic functions but studies on the effects of different training strategies from breaking till racing are lacking. Sixteen horses with the goal to race as 3-year-olds were studied from breaking (1-year-olds) to December as 3-year-olds. Horses were allocated to either a control (C) or reduced (R) training program from 2 years of age. The aim was to evaluate the effect of reducing the distance of high intensity exercise by 30% with respect to velocity at lactate concentration 4 mmol/l (VLa4), blood lactate and cardiovascular response. All training sessions were documented and heart rate (HR) was recorded. A standardized exercise test of 1,600 m was performed 10 times and a VLa4 test was performed five times.
Results
C horses initially exercised for a longer time with a HR >180 beats per minute compared to R horses (P < 0.05) but after 6–9 months, time with HR >180 bpm decreased in C and were similar in the two groups (P > 0.05). Over the 2-year period, recovery HR after the 1,600 m-test decreased in both groups but was within 2 months lower in C than in R (P < 0.05). C horses also had lower resting HR as 3-year-olds (P < 0.01) than R horses. In C, post exercise hematocrit was higher than in R (P < 0.05). There was a tendency (P < 0.1) towards a larger aortic diameter in C as 3-year-olds (C: 1.75 ± 0.05, R: 1.70 ± 0.05 cm/100 kg BW). Left ventricle diameter and blood volume (in December as 2-year-olds) did not differ between groups. There were no differences between groups in post exercise blood lactate concentration or in VLa4. Both groups were equally successful in reaching the goal of participation in races.
Conclusions
Horses subjected to a reduced distance of high intensity training from the age of 2 showed an attenuated heart rate response, but were able to maintain the same VLa4 and race participation as horses subjected to longer training distances. 相似文献
The Swedish control program for salmonella includes restrictions and on-farm control measures when salmonella is detected in a herd. Required control measures are subsidised by the government. This provides an opportunity to study costs for on-farm salmonella control. The aim of this study was to describe the costs for on-farm salmonella control in Swedish cattle herds and to investigate the effects of herd factors on these costs in dairy herds.
Results
During the 15 years studied there had been a total of 124 restriction periods in 118 cattle herds; 89 dairy herds, 28 specialised fattening herds and three suckler herds. The average costs per herd for on-farm salmonella control was 4.60 million SEK with a median of 1.06 million SEK corresponding to approximately 490 000 and 110 000 EUR. The range was 0.01 to 41 million SEK corresponding to 1080 EUR to 4.44 million EUR per farm. The costs cover measures required in herd-specific control plans, generally measures improving herd hygiene. A mixed linear model was used to investigate associations between herd factors and costs for on-farm salmonella control in dairy herds. Herd size and length of the restriction period were both significantly associated with costs for on-farm control of salmonella with larger herds and longer periods of restrictions leading to higher costs. Serotype detected and administrative changes in the Swedish Board of Agriculture aiming at reducing costs were not associated with costs for on-farm salmonella control.
Conclusions
On-farm control of salmonella in Swedish cattle herds incurred high costs but the costs also varied largely between herds. Larger herds and longer restriction periods increased the costs for on-farm control of salmonella in Swedish dairy herds. Two trials were performed to assess the meat fatty acid profile of goat kids from different genotypes, production systems and sex. In the first trial, genotype effect was determined in 24 suckling male kids from Turkish Saanen, Maltese and Gokceada breeds. In the second trial, male and female Gokceada Goat kids were used to compare the effect of extensive and semi‐intensive production systems on fatty acid composition of meat. Significant genotype effect was observed in the percentages of myristic acid (C14:0), palmitic acid (C16:0), oleic acid (C18:1 n‐9), linolenic acid (C18:3 n‐3), arachidonic acid (C20:4 n‐6) and docosahexaenoic acid (C22:6 n‐3), despite no differences on the ratios of polyunsaturated fatty acids to saturated fatty acids (PUFA/SFA) and n‐6/n‐3 (P > 0.05). The effect of production system had also significant effects on fatty acids, but sex only influenced significantly stearic acid (C18:0), C18:1 n‐9 and C18:3 n‐3 fatty acids and total PUFA level and PUFA/SFA ratio. This study confirms that dairy breeds are prone to produce higher levels of unsaturated fatty acids in their muscle. Meanwhile, meat from Gokceada goat kids, which is one of the indigenous breeds in Turkey, had similar PUFA/SFA and n‐6/n‐3 ratios to Turkish Saanen and Maltase. 相似文献
This paper describes a sensitive isocratic HPLC/ECD method developed for the determination of rosmarinic acid (RA) in plant material, animal feed, and pig plasma. The plasma sample preparation only includes protein precipitation and adjustment of the pH. The applicability of the method was tested on plasma samples of pigs that were exposed to a 91-day oral intake of RA via feed enriched by aerial parts of Prunella vulgaris. The plasma was directly analyzed using the method described as well as after enzymatic hydrolysis. When no hydrolysis step was included, RA and caffeic acid (CA) were quantified in the plasma. In hydrolyzed plasma samples, several other metabolites were determined, including dihydrocaffeic, ferulic, and dihydroferulic acid. The dual-channel coulometric detection employed, as an alternative to mass spectrometry, offers good selectivity and sensitivity owing to the electrochemical properties of the phenolic constituents. 相似文献

Silvopastoral systems integrate the complexity of forestry and forage-livestock management in an economically and ecologically sustainable way. However, these systems have undergone intensive pressure over the past decades caused by changes in land uses, as well as, in local socioeconomic priorities that suppressed traditional practices such as livestock grazing. These changes have caused land cover spatiotemporal diversifications whose effects were investigated in a traditional oak silvopastoral system at western Greece. In order to investigate the effect of human activities over the study area, the spatiotemporal land cover changes in conjunction with human and animal population changes and the environmental factors altitude, soil depth, inclination and rock type were analyzed. Also, forest-crown density changes were examined as a complimentary factor for the investigation of human intervention in the study area. Data about land-cover changes were derived from aerial orthorectified photographs of years 1945 and 2008, complimented by maps of environmental factors. Data digitization was implemented by the use of ArcGis. Based on the data analysis it derives that land cover changes are mainly attributed to direct human intervention. Environmental factors affect the ecosystem in two ways; first by affecting the location, type and intensity of human activities in the area, for example, soil depth determines where land is more productive and so predetermines the location, type and intensity of agricultural activities. Secondly, in the areas without or with minimal human intervention, environmental factors affect directly the land cover in conjunction with natural species necessities and their antagonistic capabilities.
