The aim of this study was to assess the effect of different supplements on the behavior variables of crossbred dairy cows that were in pasture that comprised two 4?×?4 Latin squares (four periods, four treatments, and four animals). Each experimental period lasted 15 days (10 days to adapt animals to treatments and 5 days for data collection). The animals were supplemented twice a day, with different forage (corn silage and cactus pear) and concentrate sources (soy mea + wheat meal + corn meal + cotton seed together and soy meal as a single constituent of the concentrate). A significant difference (p?<?0.10) was observed for the percentage of time spent consuming the supplement and for idleness, rumination, and bite rate at the time of supplementation. The supplement intake period was greater for the cactus pear-based supplements due to the lower dry matter content. Those based on corn silage resulted in longer rumination periods than those consisting of cactus pear; however, the opposite was observed for supplements based on the cactus, which showed higher percentages of time for idleness. Paratuberculosis is a chronic infection of the intestine of ruminants caused by Mycobacterium avium subsp. paratuberculosis (MAP). Early stage MAP infection can be detected by measuring cell-mediated immune responses using the interferon gamma (IFN-γ) assay. Whole blood samples are cultured overnight with specific MAP antigens followed by quantification of IFN-γ by ELISA. It is recommended that the time interval from sampling to culture does not exceed eight hours but addition of the co-stimulating cytokine interleukin 12 (IL-12) or anti-IL-10 antibodies to culture have been demonstrated to enhance IFN-γ responses of cultures stimulated with Johnin purified protein derivative (PPDj). Here we examined the correlation of IFN-γ production in response to PPDj and 15 recombinant antigens in day-old blood samples from heifers 10-21 months of age from a MAP infected herd with addition of either recombinant bovine IL-12 or anti-bovine IL-10 antibody with IFN-γ production in sample day samples. IFN-γ responses of sample day samples showed high correlation with responses to some antigens in day-old samples with addition of IL-12 or anti-IL-10 antibodies to cultures, indicating that day-old protocols can be applied as an alternative to the conventional IFN-γ protocol. Immunogenicity of the novel antigens was generally low for day-old samples. The most promising antigen using the day-old protocol with addition of IL-12 was latency protein LATP-2 as correlations, immunogenicity and diagnostic specificity collectively was high. The latency protein LATP-1 was the most promising antigen in the day-old protocol with addition of anti-IL-10 antibodies. 相似文献

The aim of this work (from the FP6 project TRACE) was to develop methods based on the use of geochemical markers for the authentication of the geographical origin of cereal samples in Europe (cf. EC regulations 2081/92 and 1898/06). Optimal water quality is considered as being a restriction for marine copepod cultures for live feed. There is a lack of knowledge on the water‐quality conditions in copepod cultures and the effect on copepods. Few studies have investigated the effect of ammonia on copepods, and fewer reports No Observed Effect Concentrations (NOEC) and Lowest Observed Effect Concentrations (LOEC), which provides safety levels before cultures are affected. This study investigates the tolerance of Acartia tonsa nauplii and adults to ammonia, using mortality as the endpoint after 24, 48 and 72 h of exposure. Nauplii were exposed to levels from 0 to 5127 μg NH3 L?1 and adults to levels from 0 to 8481 μg NH3 L?1. Nauplii NOEC was 30 μg NH3 L?1 and LOEC was 81 μg NH3 L?1. Adult NOEC was 477 μg NH3 L?1 and LOEC was 1789 μg NH3 L?1. 50% Lethal Concentrations (LC50) for nauplii of 48 and 72 h was 1257 and 220 μg NH3 L?1. LC50 for adults was 2370 (24 h), 972 (48 h) and 770 (72 h). Combining NOEC with excretion rates of NH4/NH3 a model was developed to calculate densities in batch cultures. We recommend that for batch cultures of A. tonsa, NH3 is kept below NOEC for nauplii and that levels of NH3 together with pH are monitored weekly. 相似文献

Norway Spruce is the economically most important tree species in Europe and has been cultivated in plantations on a large-scale at low elevations, far outside its natural range. In the Bohemian Forest, it naturally occurs in pure stands above 1150 m a.s.l. and as a mixed tree species from 650 to 1150 m a.s.l. An understanding of natural distributions and the diversity along temperature gradients at various elevations is important for conservation, pest management, and predictions of future species assemblages by global warming. Here we investigated the species richness of canopy arthropods in spruce trees along a gradient from 300 to 1300 m a.s.l. using flight-interception traps. We analyzed species richness by combining diversity partitioning with a moving window approach after standardizing sample size per plot. Total richness decreased linearly as the elevation increased, which reflected declining temperatures and a declining regional species pool. Phytophages (herbivores excluding xylophages) were the most influenced. Richness did not peak at the transition zones of the three ecological elevation zones, neither for all species, nor for any of the separate functional groups. However, the proportion of both beetle and true bug spruce specialists significantly increased with elevation and actually doubled in richness above 1000 m a.s.l., where spruce is naturally dominating. Our results indicate that even planted spruce trees at lower elevations maintain high levels of species richness. Further climate warming will promote overall species richness, especially of phytophages, at all elevations. However, spruce specialists may be seriously threatened by global warming. 相似文献

Resveratrol is known as a grapevine secondary metabolite with fungicide activity. Its exogenous application on harvested grapes resulted in the reduction of microbial flora growth, and consequently, prolonged shelf life, without affecting the nutritional quality of the fruit. Resveratrol treatment also resulted in being effective on fruit that normally does not accumulate such metabolites as, for example, tomatoes, apples, avocado pears, and peppers. As a result, all treated fruits maintained their post-harvest quality and health longer than the untreated ones. This study demonstrates the potential use of resveratrol as a natural pesticide to reduce post-harvest fungi development on a broad spectrum of fruit types. 相似文献

A reducing solution of 2-mercaptoethanol and its oxidized form 2-hydroxyethyl disulfide, whose variable concentrations set variable disulfide reduction potentials, was applied to progressively reduce the disulfide bonds of proteins extracted from doughs made from Meneba and Robin Hood flour. Several dough proteins had disulfide bonds stronger than those of other dough proteins. A SDS-sedimentation method was applied to monitor the baking of dough into bread. Dough proteins susceptible to heat (baking) were studied by SDS-fractionation, extraction with reducing alcoholic solution, SDS-PAGE, and N-terminal protein sequencing. High or low molecular weight glutenins, α, β, and γ-gliadins, α-amylase inhibitor, and α-amylase trypsin inhibitor were identified among the dough proteins modified by heat (as shown by reduced solubility in aqueous-SDS solution). The heat-induced modification of the gliadins and glutenins might contribute to the coagulation of dough proteins, while the heat-induced modification of the amylase or trypsin inhibitors might contribute to the regulation of endogenous or exogenous amylolytic or proteolytic activities in dough or bread. 相似文献

Xanthohumol (XN) is the major prenylated chalcone of hops and hence an ingredient of beer. Despite many advances in understanding of the pharmacology of XN, one largely unresolved issue is its low bioavailability in the human organism. Also, not much is known about its actual concentrations and pharmacokinetics in liver and intestinal cells. Therefore, the uptake, intracellular distribution, and kinetics of XN were studied in various cell types, namely, hepatocellular carcinoma cells (HuH-7), hepatic stellate cells (HSC), primary cultured hepatocytes, and colorectal adenocarcinoma cells (Caco-2). Fluorescent microscopy allowed for the first time visualization and tracing of the uptake and intracellular distribution of XN. A rapid accumulation of XN concentrations that were up to >60-fold higher than the concentration present in the ambient culture medium was observed. Skatole originates from microbial processing of tryptophan in the large intestine of pigs and accumulates in adipose tissue. Formation may be inhibited by the anti-apoptotic function of butyrate formed out of raw potato starch. Two groups of pigs (each consisting of gilts and barrows) were fed from 30 to 110 kg life weight either a conventional diet (controls; n = 35) or an isocaloric diet containing 300 g of raw potato starch/kg of body weight (RS; n = 34). Skatole concentrations were measured in colon content, blood, and adipose tissue. Odor of cooked meat samples was evaluated by a test panel. RS reduced concentrations in colon content and blood plasma (P < 0.001). Back fat concentrations were decreased significantly from 25 to 1.40 ng/g (barrows; P < 0.001) and from 40 to 9 ng/g (gilts; P < 0.001). Odor rating (scale of 1-5 from very unpleasant to very pleasant) was 3.07 for low skatole concentrations and 2.66 for both medium and high skatole concentrations (P < 0.05). 相似文献
