The essential oil and fatty acid composition of two provenances of Ruta chalepensis from four organs (leaves, flowers, stems and fruits) was determined. The effect of the plant part on total fatty acid contents, essential oil yields, fatty acid and volatile constituents was significant.Fatty acid profiles varied significantly among the studied provenances and organs. Linolenic acid had the highest amount in leaves of the two provenances. From R. chalepensis, in all organs, the main fatty acids were palmitic (13.10-25.31%), followed by palmitoleic (0-15.72%), stearic (1.03-6.85%), oleic (1.90-24.04%), arachidic (0.11-4.03%), eicosatetraenoic (0.10-5.60%) and behenic (0.47-6.09%) acids. Saturated fatty acids had the highest amounts in growing wild R. chalepensis flowers, and cultivated R. chalepensis stems were characterized by the predominance of polyunsaturated fatty acids. Oil composition of all studied organs has a healthy and nutritionally value. Essential oil yields varied from 0.39% to 2.46% and showed a remarkable variation with plant organs. Thirty-six volatile compounds were identified in different analyzed essential oils; 2-undecanone, 2-nonanol and 2-dodecanone had the highest percentages.  相似文献

The random amplified polymorphic DNA (RAPD) method was used to investigate the genetic variability and population structure of Fusarium culmorum isolated from wheat stem bases. A total of 108 isolates, representing seven geographically distinct populations, was collected from five climatic regions in Tunisia. Pseudo-allelic frequencies were estimated at each of the 25 putative RAPD loci analyzed by scoring for the presence or absence of amplified fragments; 92 haplotypes were found among the 108 strains. The analysis of the population structure did not reveal any trend with regard to geographic origin. Total gene diversity (HT * = 0.318) was mostly attributable to diversity within populations (HS * = 0.308). Analysis of molecular variance confirmed that most of the genetic variability was within populations. Genetic differentiation among populations was low to moderate (GST * ranged from 0 to 0.190 and averaged 0.041 over all loci). Cluster analysis with UPGMA using genetic distances did not reveal any spatial clustering of the isolates collected from the different geographic regions. Based on these results, we conclude that the F. culmorum isolates recovered from different regions in Tunisia might be part of a single population pool.  相似文献

Sixty two rhizospheric and endophytic bacterial strains were evaluated for their biocontrol effect on two aggressive Fusarium culmorum isolates (Fc2 and Fc3). We observed that 35 % and 23 % of the tested strains inhibited the in vitro growth of Fc2 and Fc3 respectively. The observed antagonism was due to inhibition by contact (13–19 % of the strains) or at distance (10–16 % of the strains) for both fungal isolates. Some of the antagonistic bacteria showed the ability to produce diffuse and/or volatile compounds that inhibit the growth, the sporulation and macroconidia germination of F. culmorum. None of the tested antagonistic bacteria showed chitinase activity on synthetic medium. The sequencing of the 16S rDNA genes of some antagonistic bacteria showed that they belong to the genera Bacillus, Pseudomonas and Microbacterium. The double inoculation of durum wheat seeds by the antagonistic bacterial strains (B13, B18, BSE1, BSE3 and B16E) and the two F. culmorum isolates showed that germination and seedling vigor were generally improved in vitro. The percentage of infected seeds was also reduced. In greenhouse trials, the biocontrol effectiveness of F. culmorum was dependant from the virulence of the fungal strain and the specificity of the antagonistic interaction between bacterial and fungal strains. The bacterial strains B18 and B16E reduced F. culmorum infection on durum wheat plants probably due to their antagonistic and plant growth promoting activities and they may be used in a mixture as seed biopriming inoculum for plant growth bio-promoting and Fusarium wheat diseases biocontrol.  相似文献

Fenugreek is an annual leguminous crop grown for hay and grains in Tunisia. It is also considered a valuable rotation crop with cereals. Sclerotinia rot was observed in production fields since 2010. The survey conducted in 2013 revealed that the incidence of diseased plants varied between 5 and 20%. The identification of isolates of Sclerotinia obtained from fenugreek plants with symptoms of stem rot was determined using morphological and molecular criteria. The size, shape and abundance of sclerotia in potato dextrose agar (PDA) cultures were used to classify isolates as S. sclerotiorum or S. trifoliorum. A comparison of colony diameter on PDA after 24, 48 and 72 h at 25 °C, showed that one isolate grew faster (36 mm/day) than the other 10 isolates (14.8 mm/day). There was a significant difference in sclerotial size between the fast and the slow growing isolates, but there was no significant difference in the number of sclerotia produced after 3 weeks on PDA. Two of the slow growing isolates exhibited ascospore dimorphism, whereas the fast growing isolate did not. PCR amplification with the primer pair ITS5/ITS4 produced a fragment of 560 base pairs from the fast growing isolate and 1000 base pairs from all of the slow growing isolates. The ITS sequences of the fast growing isolate had 100% homology with S. sclerotiorum, whereas those of the slow growing isolates had 100% homology with S. trifoliorum. Isolates of both species were pathogenic on fenugreek seedlings in the greenhouse assay and there was no significant difference in the percentage of dead plants two weeks after inoculation between the two species.  相似文献

Au cours du printemps 2002, une grave maladie s'est manifestée dans une culture de tomate (cv. Collibri) de primeur sous abris-serres dans une exploitation située à Chott Mariem (120 km au sud de Tunis). Les plantes attaquées montrent les symptômes caractéristiques d'une maladie vasculaire. L'isolement de l'agent pathogène à partir des vaisseaux à différents niveaux de la tige des plantes malades a révélé systématiquement la présence de Verticillium alboatrum . La virulence de ce nouveau pathotype de V. alboatrum a été vérifiée sur quatre cultivars de tomate portant le gène de résistance Ve et comparée à un témoin sensible. Ce nouveau pathotype a montré une virulence accrue aussi bien chez les cultivars résistants que chez le témoin sensible entraînant une réduction de la croissance de 18%à 34%, le dessèchement des feuilles et le brunissement des vaisseaux. Ces résultats confirment la présence pour la première fois en Tunisie d'un nouveau pathotype de V. alboatrum pouvant attaquer les cultivars de tomate pourvus du gène de résistance Ve .  相似文献

L'analyse fongique des semences de pastèque a révélé leur contamination par différentes espèces de Fusarium. En se basant sur des critères morphologiques et des analyses de I'ADNr, quatre espèces de Fusarium ont été identifiées. II s'agit de F. oxysporum, F. solani, F. moniliforme et F. equiseti. L'amplification de I'ADN ribosomique par deux amorces universelles suivie de digestion enzymatique a permis de distinguer entre les différentes espèces de Fusarium isolées à partir des semences de pastèque. La pathogénicité des isolats de chaque espèce de Fusarium a été testée sur des plants de pastèque sensibles (cv. Sugar Baby). Sur la base de ces tests d'inoculation, les isolats pathogènes de F. oxysporum et F. solani ont été séparés des non pathogènes. Tous les isolats de F. moniliforme et F. equiseti testés se sont révélées non pathogènes sur pastèque.  相似文献

We evaluated annual productivity and carbon fluxes over the Fontainebleau forest, a large heterogeneous forest region of 17,000 ha, in terms of species composition, canopy structure, stand age, soil type and water and mineral resources. The model is a physiological process-based forest ecosystem model coupled with an allocation model and a soil model. The simulations were done stand by stand, i.e., 2992 forest management units of simulation. Some input parameters that are spatially variable and to which the model is sensitive were calculated for each stand from forest inventory attributes, a network of 8800 soil pits, satellite data and field measurements. These parameters are: (1) vegetation attributes: species, age, height, maximal leaf area index of the year, aboveground biomass and foliar nitrogen content; and (2) soil attributes: available soil water capacity, soil depth and soil carbon content. Main outputs of the simulations are wood production and carbon fluxes on a daily to yearly basis. Results showed that the forest is a carbon sink, with a net ecosystem exchange of 371 g C m(-2) year(-1). Net primary productivity is estimated at 630 g C m(-2) year(-1) over the entire forest. Reasonably good agreement was found between simulated trunk relative growth rate (2.74%) and regional production estimated from the National Forest Inventory (IFN) (2.52%), as well as between simulated and measured annual wood production at the forest scale (about 71,000 and 68,000 m(3) year(-1), respectively). The chemical composition of essential oil isolated by hydrodistillation from the leaves of Tunisian Eucalyptus erythrocorys Linn. was analyzed by gas chromatography (GC) and gas chromatography–mass spectrometry (GC/MS). Fifteen compounds were identified, representing 97 % of total oil, which was found to be rich in oxygenated monoterpenes (66.7 %), particularly in 1,8-cineole (54.8 %). The yield and the physico-chemical properties of oil were determined. The study of antifungal activity revealed that E. erythrocorys essential oil significantly inhibited the growth of five plant pathogenic fungi especially Bipolaris sorikiniana and Botritys cinerea. Moreover, herbicidal properties of the oil, tested on Sinapis arvensis L. and Phalaris canariensis L. indicated that the E. erythrocorys oil completely inhibited the seed germination at high concentration (1.5 μl/ml), while at low doses (0.25, 0.5, 0.75, 1 μl/ml) the oil acted by decreasing and delaying the germination and inhibiting the seedling growth of all tested weeds.  相似文献
