Five native Sargassaceae species from Brittany (France) living in rockpools were surveyed over time to investigate photoprotective strategies according to their tidal position. We gave evidences for the existence of a species distribution between pools along the shore, with the most dense and smallest individuals in the highest pools. Pigment contents were higher in lower pools, suggesting a photo-adaptive process by which the decreasing light irradiance toward the low shore was compensated by a high production of pigments to ensure efficient photosynthesis. Conversely, no xanthophyll cycle-related photoprotective mechanism was highlighted because high levels of zeaxanthin rarely occurred in the upper shore. Phlorotannins were not involved in photoprotection either; only some lower-shore species exhibited a seasonal trend in phlorotannin levels. The structural complexity of phlorotannins appears more to be a taxonomic than an ecological feature: Ericaria produced simple phloroglucinol while Cystoseira and Gongolaria species exhibited polymers. Consequently, tide pools could be considered as light-protected areas on the intertidal zone, in comparison with the exposed emerged substrata where photoprotective mechanisms are essential.  相似文献

Exopolysaccharides play an important role in the pathogenicity of Ralstonia solanacearum. We compared in vitro and in planta exopolysaccharide production of the pathogenic strain AW1 with that of three related mutant strains impaired in both their exopolysaccharide production and aggressiveness on tomato. The distinction between the two hexosamine-rich exopolysaccharides, namely the N-acetyl-glucosaminorhamnan and the major N-acetyl-galactosamine-containing acidic polymer was emphasized. The major acidic polymer was identified specifically by electron microscopy using glutaraldehyde/ruthenium red/uranyl acetate staining, by immunofluorescence using specific monoclonal antibodies and correlated to an appropriate biochemical analysis. The two mutant strains AW1-1 and AW-19A were totally devoid of any production of the major exopolysaccharide in vitro or in planta whatever the technique used. Infection and vascular colonization of tomato roots by the pathogenic strain were also compared to those of the mutant strains by light microscopy. Pathogenicity on tomato was assessed by root infection without any artificial injury. Light microscopy showed that the two mutant strains AW1-1 and AW-19A were poorly infective and unable to invade xylem vessels, while they induced defence mechanisms in root tissues and appeared aggregated or degenerated within cortical infection pockets. These two mutant strains were non-pathogenic or weakly aggressive, respectively. In contrast, the pathogenic strain AW1 and the hypoaggressive AW1-41 strains, which produce large amounts of the major acidic exopolysaccharide in planta, were both infective and invasive, and tomato root tissues exhibited only limited defence reactions. Thus, the major acidic exopolysaccharide produced by Ralstonia solanacearum is involved in root infection and vascular colonization, though its precise role is still unknown.  相似文献

Crop residues are among the main inputs that allow the organic carbon (C) and nutrients to be maintained in agricultural soil. It is an important management strategy that can improve soil fertility and enhance agricultural productivity. This work aims to evaluate the extent of the changes that may occur in the soil heterotrophic microbial communities involved in organic matter decomposition and C and nitrogen (N) mineralization after the addition of crop residues. Soil microcosm experiments were performed at 28°C for 90 days with the addition of three crop residues with contrasting biochemical qualities: pea (Pisum sativum L.), rapeseed (Brassica napus L.), and wheat (Triticum aestivum L.). Enzyme activities, C and N mineralization, and bacterial and fungal biomasses were monitored, along with the bacterial and fungal community composition, by the high-throughput sequencing of 16S rRNA and ITS genes. The addition of crop residues caused decreases in β-glucosidase and arylamidase activities and simultaneous enhancement of the C mineralization and net N immobilization, which were linked to changes in the soil microbial communities. The addition of crop residues decreased the bacterial and fungal biomasses 90 days after treatment and there were shifts in bacterial and fungal diversity at the phyla, order, and genera levels. Some specific orders and genera were dependent on crop residue type. For example, Chloroflexales, Inquilinus, Rubricoccus, Clitocybe, and Verticillium were identified in soils with pea residues; whereas Thermoanaerobacterales, Thermacetogenum, and Hypoxylon were enriched in soils with rapeseed residues, and Halanaerobiales, Rubrobacter, and Volutella were only present in soils with wheat residues. The findings of this study suggest that soil C and N dynamics in the presence of the crop residues were driven by the selection of specific bacterial and fungal decomposers linked to the biochemical qualities of the crop residues. If crop residue decomposition processes showed specific bacterial and fungal operational taxonomic unit (OTU) signatures, this study also suggests a strong functional redundancy that exists among soil microbial communities.  相似文献

Two distinct approaches were used to characterise spray-drift during the application of atrazine and alachlor to a maize crop. The first consisted in determining the quantities which did not reach their target. A first experiment was carried in 2001 to improve the sampling method. A second experiment in 2002 showed that losses represented 46 and 38% for atrazine and alachlor, respectively. The second approach was to follow the spatiotemporal evolution of the cloud formed during application. The concentrations observed near the application zone during spraying reached 4.5 microg m(-3) for atrazine and 8.5 microg m(-3) for alachlor. With alachlor these concentrations decreased rapidly when increasing distance from the plot or time following treatment, whereas in the case of atrazine they stabilised rapidly (between 0.5 and 0.3 microg m(-3)) both in space and in time. Deposits around the plot were light and slightly higher for alachlor (from 20 to 130 microg m(-2)). Alachlor was more rapidly diluted in space than atrazine, reflecting a differentiated evolution of physical form during the process. Alachlor, being more volatile than atrazine, is quickly transferred to the gaseous phase which was more rapidly dispersed than aerosols.  相似文献

OBJECTIVE : To describe immunolocalization of TGF-beta receptors (RI and RII) in normal equine skin and in thoracic or limb wounds, healing normally or with exuberant granulation tissue (EGT). STUDY DESIGN : Group A: six wounds on one metacarpus and one midthoracic area. Group B: six wounds on both metacarpi, one of which was bandaged to stimulate EGT. Immunohistochemistry was used to detect RI and RII expression in wound margins. ANIMALS : Eight horses, randomly assigned to one of two study groups. METHODS : Neutralizing polyclonal anti-rabbit RI and RII antibodies were used to detect spatial expression of RI and RII in biopsies obtained before wounding, at 12 and 24 hours, and 5, 10 and 14 days after wounding. RESULTS : RI and RII were co-localized in both unwounded and wounded skin. There were no differences in cell types staining positively between tissues obtained from the limb and the thorax, or from normally healing limb wounds and limb wounds with EGT, at any time. Because of increased cellularity within EGT, staining intensity of limb wounds with 'proud flesh' was greater than limb wounds healing normally, and thoracic wounds, during the proliferative phase of repair. CONCLUSIONS : Strong expression of RI and RII, particularly in limb wounds with EGT, suggested that signalling for stimulation of matrix proteins is in place to contribute to scarring. CLINICAL RELEVANCE : This information may help determine the appropriate time for using receptor antagonists to prevent scarring of limb wounds of horses.  相似文献

The ability of the nematophagous fungus Duddingtonia flagrans to reduce the number of infective nematode larvae in coproculture was investigated in goats using different doses of chlamydospores (0, 1.25 x 10(5), 2.5 x 10(5), 5 x 10(5) chlamydospores/kg BW/day) given by oral administration or by voluntary consumption in feed during natural or experimental infections with nematodes. The kinetics of excretion of D. flagrans chlamydospores in the faeces was also determined using a dose of 5 x 10(5) chlamydospores/kg BW/day for five days. For all the trials, the faecal nematode egg outputs were determined by a modified McMaster method and standard coprocultures were set up (14 days, 25 degrees C) to determine the number of larvae emerging from culture in fungus treated and control faeces. When chlamydospores were orally administered, the number of larvae were reduced by 50 to 97% when compared to control cultures. No difference in the level of larval emergence from the culture was seen for experimental or natural infections at the different chlamydospore dose rates. In contrast, when chlamydospores were distributed in the feed, a dose-dependent relationship was observed 10 days after the start of administration, the larval development being 2.0%, 14.0% and 86.9% for 5 x 10(5), 2.5 x 10(5) and 0 spores/kg BW/day, respectively. In addition, the kinetic study showed that the larval emergence from coproculture in the fungus group was statistically lower than in the control group from the second day of administration of the chlamydospores and remained lower until the second day after the last administration (p < 0.05). The results indicate that, for goats in farm conditions, a minimum daily dose of 5 x 10(5) chlamydospores/kg BW must be used to ensure a high treatment efficacy and that daily administration is preferable for maintenance of efficacy over time.  相似文献

OBJECTIVE: To develop an epiduroscopic technique for use in standing cattle and describe the endoscopically visible anatomic structures of the epidural space in the sacrococcygeal area. ANIMALS: 6 healthy nonlactating, nonpregnant cows (mean +/- SD age, 60 +/- 18.5 months; mean weight, 599.7 +/- 63.87 kg) and 3 bovine cadavers. PROCEDURES: Cadavers were used to allow familiarization with the equipment and refinement of the technique. Following these experiences, procedures were performed in live animals. Each cow was restrained in a stock. After sedation with xylazine (0.03 mg/kg, IV), 2% lidocaine hydrochloride (0.25 mg/kg) was injected epidurally in the first intercoccygeal or the sacrococcygeal intervertebral space. By use of an introducer set (guidewire and dilation trocar and shaft), a flexible endoscope (length, 75 cm; diameter, 2.3 mm) was inserted through the dilation shaft into the epidural space. To obtain an optimal view, small amounts of air were insufflated into the epidural space through the working channel of the endoscope via a syringe with special filter. RESULTS: Anatomic structures of the epidural space that were viewed by means of the endoscopic procedure included blood vessels, connective tissue, fat, nerves, and the spinal dura mater. No adverse events were detected during epiduroscopy, and it was tolerated well by all 6 cows. CONCLUSIONS AND CLINICAL RELEVANCE: In ruminants, epidural structures can be viewed via endoscopy. Such epiduroscopic procedures may be useful in anatomic studies as well as for the diagnosis of disease or therapeutic interventions in ruminants.  相似文献

Medical records of 42 cows that underwent both thoracic radiographic and postmortem examinations within a period of 7 days were reviewed to develop an evaluation grid to interpret bovine thoracic radiographs and to determine the sensitivity and the specificity of thoracic radiographs for detection of thoracic lesions, based on postmortem examination. Most cows (64%) had clinical signs of respiratory disease, whereas 19% showed signs of cardiac problems. The sensitivity and specificity of radiographs for identifying cows with thoracic lesions were 94% and 50%, respectively. In this study, with a prevalence of thoracic lesions of 86%, the positive- and negative-predictive values were 92% and 57%, respectively. This study provides an evaluation grid that allows standardization of the reading of bovine thoracic radiographs and the identification of most thoracic lesions. Bovine thoracic radiographs are useful in detecting thoracic lesions in cows.  相似文献
