Twenty-four wether lambs (BW = 37.5 +/- 0.8 kg) were used in a 64-d randomized complete block design experiment to evaluate the effect of oscillating dietary CP with undegradable intake protein (UIP) on diet digestibility, N retention, and gastrointestinal (GI) organ mass. Four treatments consisted of a 13, 15, or 17% CP diet fed daily or a regimen in which dietary CP was oscillated between 13 and 17% on a 48-h basis (ACP). All diets consisted of 65% bromegrass hay (10.5% CP, 61.9% NDF, 37.2% ADF) and 35% corn-based supplement, and were formulated to contain the same amount of degradable intake protein (9.6% of dry matter), plus additional UIP (from SoyPLUS) to accomplish CP levels above 13%. Beginning on d 52, N balance collections were conducted for 8 d, after which lambs were killed on d 62 and 64 of the trial for measurement of GI organ mass. Because intake was restricted to 3.0% of initial body weight (dry matter basis), dry matter intake did not differ (P > or = 0.67) and no treatment effects (P > or = 0.36) on ADG, feed efficiency, or total tract DM digestibility were observed. Increasing dietary CP from 13 to 17% linearly increased (P = 0.0001) N digestibility, but lambs fed ACP had lower (P = 0.07) total tract N digestibility than those fed 15% CP daily. Although urinary N excretion increased linearly (P = 0.0001) with increasing CP, a linear increase (P = 0.07) was observed in N retention (g/d) with increasing dietary CP. Although the quantity of N retained by lambs fed ACP was not statistically different (g/d, P = 0.19; % of digested N, P = 0.23) from those fed 15% CP daily, N retention in lambs fed ACP was 42% lower than in those fed 15% CP daily (1.8 vs 3.1 g/d, respectively). Increasing CP linearly decreased (P < or = 0.09) weights of the reticulorumen, abomasum, and small intestine, but did not affect (P > or = 0.16) liver or omasum weights. Length of the small intestine was not affected (P > or = 0.45) by treatment, but lambs fed ACP had greater (P = 0.03) small intestine weights than those fed 15% CP daily. Increasing dietary CP linearly decreased (P = 0.03) total GI organ mass, and lambs fed ACP had a greater (P = 0.03) total GI organ mass than those fed 15% CP daily. Oscillating dietary CP may increase the weights of the GI organs, which may subsequently have negative effects on N and energy metabolism in the animal. Likewise, the potential for decreased GI organ mass in response to increased supply of CP with UIP deserves further investigation.  相似文献

The influence of alkylphenol polyoxyethylene non-ionic surfactants on the distribution pattern of MSMA (sodium hydrogen methylarsonate) was studied using scanning electron microscopy and X-ray microanalysis elementmapping techniques. Surfactant concentrations and lipophilic–hydrophilic constitution influenced phytotoxicity and residue patterns. Tanniferous temperate legumes are assumed to possess anthelmintic properties, but it is unclear whether this is the direct result of condensed tannins (CT) or is mediated indirectly via an improved metabolic protein supply. A metabolism experiment was conducted to differentiate between these factors by feeding the CT plant sainfoin (19.7% CP in DM) to lambs infected with the abomasal blood-sucking nematode Hemonchus contortus. A total of 18 infected lambs were fed sainfoin either untreated or treated with polyethylene glycol, a CT-inactivating agent, or a grass-clover mixture (13.2% CP in DM) over 3 wk (n = 6). Six uninfected lambs received the grass-clover mixture as a control. In addition to indicators of infection (fecal egg count, packed-cell volume, abomasal worm burden, and serum protein), nutrient digestibility, the balance of N and selected minerals, ruminal fluid characteristics, and plasma AA levels were determined mostly in the final experimental week. The specific effects of the sainfoin CT, the extra CP with sainfoin, and the infection were statistically evaluated by contrast analysis. The sainfoin CT exerted no beneficial effects on resilience to nematode infection and exerted only minor effects on ruminal ammonia or blood urea concentrations and the excretory pattern of N. Plasma alanine, aspartate, and proline concentrations tended to be greater (P < or = 0.09) because of the sainfoin CT, whereas the other AA remained unaffected. Intake of the mineral supplement was lower (P < 0.001) for lambs fed sainfoin compared with those fed sainfoin treated with polyethylene glycol. Feeding the high-protein sainfoin instead of the grass-clover mixture increased (P < 0.001) N retention and apparent OM digestibility, whereas digestibility of NDF and ADF were decreased (P < 0.001). Feeding sainfoin also decreased (P < or = 0.04) plasma alanine, glycine, isoleucine, and total nonessential AA compared with the grass-clover mixture. Although fecal egg count, worm burden, and packed cell volume were not affected by the greater CP supply associated with sainfoin feeding, serum albumin level was increased (P = 0.008). The lack of effects of sainfoin on resilience to nematode infection might have been the result of the unexpectedly low CT content (3.6% in DM) of the material used. It cannot be excluded that longer term feeding of this batch of sainfoin might have been effective.  相似文献

A cross-sectional study evaluating potential risk factors for odontoclastic resorptive lesions (ORL) in feline teeth was conducted. Owners of 32 cats with ORL and 27 cats without ORL were interviewed regarding their respective cat's demographic characteristics, diet, and medical and dental histories. Four factors were identified as significantly associated with ORL using unconditional logistic regression. A history of dental disease (gingivitis, calculus, or periodontal disease; odds ratio [OR], 4.5); city residence (OR, 4.4); and being an exclusively indoor cat (OR, 4.5) were associated with an increased risk for ORL. Consumption of commercial treats (OR, 0.3) appeared protective for ORL.  相似文献

The objective of this study was to examine the association of adenosine monophosphate (AMP)-activated protein kinase (AMPK) with glycogen content in bovine muscle and their links with intramuscular fat (IMF) and muscle fiber type composition. Five steers with high intramuscular fat (High IMF, IMF content is 5.71 +/- 0.36%) and five steers with low intramuscular fat (Low IMF, IMF content is 2.09 +/- 0.19%) in the longissimus thoracis muscle (LM) were selected for immunoblotting, glycogen, and myofiber type composition analyses. The glycogen content was higher in Low IMF muscle than in High IMF muscle (1.07 +/- 0.07 versus 0.85 +/- 0.08 g/100 g muscle, P < 0.05). Phosphorylation of the AMPK alpha subunit at Thr 172, which is correlated with its activity, was lower (P < 0.05) in High IMF compared to Low IMF. In agreement with the lower AMPK phosphorylation in High IMF muscle, the phosphorylation of acetyl-CoA carboxylase (ACC) was also lower (P < 0.05) in High IMF muscle than in Low IMF muscle. Glycogen synthase kinase 3 (GSK3) down-regulates glycogen synthesis through phosphorylation of glycogen synthase. The phosphorylation of GSK3 in High IMF was lower (P < 0.05) than that in Low IMF, which should down-regulate glycogen synthase activity and reduce the glycogen content in High IMF beef. Type IIB myosin isoform was absent in beef muscle. No noticeable difference in myosin isoform composition was observed between Low and High IMF muscle. In summary, High IMF cattle had lower LM glycogen levels than low IMF cattle, and AMPK activity was less in High IMF than in Low IMF cattle. The difference in glycogen content between Low and High IMF muscle was not correlated with muscle fiber composition. This data shows that LM lipid and glycogen metabolisms are affected by AMPK activity. Thus, AMPK may be a molecular target to alter IMF and glycogen levels in beef muscle.  相似文献

Infectious diarrhea in piglets can be caused by TGE, EVD and rota virus. The present report shows another viral agent (para-rota virus) to be present. In addition the report describes light and electronmicroscopic findings of the enteric mucosa caused by the agents mentioned. The degree and distribution of villous atrophy, one of the main histological findings, of infections with TGE, EVD, rota and para-rota virus is compared. A correlation of the results obtained with fluorescent antibody staining and ultrastructural diagnosis of agents was done. An application of the latter seems advisable in case of negative FA staining results.  相似文献

Objective: To determine endogenous serum insulin concentration in dogs with diabetic ketoacidosis (DKA), and to compare it to endogenous serum insulin concentration in diabetic dogs with ketonuria but no acidosis (KDM), diabetic dogs with uncomplicated diabetes mellitus (DM) that did not have ketonuria or acidosis, and dogs with non‐pancreatic disease (NP). Design: Prospective study. Setting: Veterinary Hospital of the University of Pennsylvania. Animals: Forty‐four client‐owned dogs; 20 dogs with newly diagnosed diabetes mellitus (7 dogs with DKA, 6 dogs with KDM, and 7 dogs with DM) and 24 dogs with non‐pancreatic disease. Interventions: Blood and urine samples were obtained at the time of admission to the hospital. Measurements and main results: Signalment, clinical signs, physical examination findings, and concurrent disease were recorded for all dogs. Blood glucose concentration, venous blood pH, venous blood HCO3? concentration, urinalysis, and endogenous serum insulin concentration were determined in all dogs. Dogs with DKA have significantly decreased endogenous serum insulin concentrations compared to dogs with DM (P = 0.03) and dogs with non‐pancreatic disease (P = 0.0002), but not compared to dogs with KDM (P = 0.2). Five of 7 dogs with DKA had detectable endogenous serum insulin concentrations, and 2 of these dogs had endogenous serum insulin concentration within the normal range. Conclusions: Diabetic dogs with ketoacidosis have significantly decreased endogenous serum insulin concentration compared to dogs with uncomplicated diabetes mellitus. However, most dogs with DKA have detectable endogenous serum insulin concentrations, and some dogs with DKA have endogenous serum insulin concentrations within the normal range.  相似文献

Thirty-three pigs in three groups of nineteen, ten, and four pigs were infected with three different African swine fever (ASF) virus isolates, respectively. All virus isolates were attenuated to varying degrees by passaging in cell cultures, and they retained sufficiently low virulence to produce subacute and chronic infections in pigs. Sera collected at various intervals were tested for antibody activity by the immunoelectroosmophoresis, agar gel diffusion precipitin, and complement-fixation tests using a modified Kolmer technique. Results clearly indicated that the immunoelectroosmophoresis test is a rapid (30 minute) and accurate method with extreme sensitivity and superior to the complement-fixation and agar gel diffusion precipitin tests in detecting antibody against ASF virus. Possible use of this method in detecting ASF virus infection is suggested.  相似文献
