The long-term (9 years) effect of pig slurry applications vs mineral fertilization on denitrifying activity, N2O production and soil organic carbon (C) (extractable C, microbial biomass C and total organic C) was compared at three soil depths of adjacent plots. The denitrifying activities were measured on undisturbed soil cores and on sieved soil samples with acetylene method to estimate denitrification rates under field or potential conditions. Pig slurry applications had a moderate impact on the C pools. Total organic C was increased by +6.5% and microbial biomass C by ≥25%. The potential denitrifying activity on soil suspension was stimulated (×1.8, P<0.05) 12 days after the last slurry application. This stimulation was still apparent, but not significant, 10 months later and, according to both methods of denitrifying activity measurement (r 2=0.916, P<0.01 on sieved soil; r 2=0.845, P<0.001 on soil cores), was associated with an increase in microbial biomass C above a threshold of about 105 mg kg−1. The effect of pig slurry on denitrification and N2O reduction rates was detected on the surface layer (0–20 cm) only. However, no pig slurry effect could be detected on soil cores at field conditions or after NO3 enrichments at 20°C. Although the potential denitrifying activity in sieved soil samples was stimulated, the N2O production was lower (P<0.03) in the plot fertilized with pig slurry, indicating a lower N2O/(N2O + N2) ratio of the released gases. The pig-slurry-fertilized plot also showed a higher N2O reduction activity, which is coherent with the lower N2O production in anaerobiosis.  相似文献

A laboratory procedure for the analysis of the oxygen-18/oxygen-16 isotope ratios of ethanol derived from sugars and fruit juices by pyrolysis-isotope ratio mass spectrometry (IRMS) has been applied to the study of isotopic fractionation induced by the isotope effects of fermentation and distillation. For both processes, an experimental model has been established to describe and explain the observed fractionation phenomena. It is shown that reproducible results can be obtained when appropriate analytical conditions are used. Moreover, the ability of ethanol to act as a reliable indicator of the (18)O/(16)O ratio of sugars in orange juice (and therefore to be used as an internal reference for detecting water addition) is demonstrated both in theory and in practice.  相似文献

Two-dimensional electrophoresis was used to investigate sarcoplasmic protein expression in pig Semimembranosus muscles sampled 20 min after slaughter. Two groups (light and dark) of 12 animals were selected from 1000 pigs, based on meat L values measured 36 h postmortem. Twenty-two proteins or fragments (p < 0.05) were differentially expressed. Muscles leading to darker meat had a more oxidative metabolism, indicated by more abundant mitochondrial enzymes of the respiratory chain, hemoglobin, and chaperone or regulator proteins (HSP27, alphaB-crystallin, and glucose-regulated protein 58 kDa). Conversely, enzymes of glycolysis were overexpressed in the lighter group. Such samples were also characterized by higher levels of glutathione S-transferase omega, which can activate the RyR calcium channels, and higher levels of cyclophilin D. This protein pattern is likely to have severe implications on postmortem metabolism, namely, acceleration of ATP depletion and pH fall and subsequent enhanced protein denaturation, well-known to induce discoloration.  相似文献

The determination of the occurrence and level of cocoa shells in cocoa products and chocolate is an important analytical issue. The recent European Union directive on cocoa and chocolate products (2000/36/EC) has not retained the former limit of a maximum amount of 5% of cocoa shells in cocoa nibs (based on fat-free dry matter), previously authorized for the elaboration of cocoa products such as cocoa mass. In the present study, we report a reliable gas-liquid chromatography procedure suitable for the determination of the occurrence of cocoa shells in cocoa products by detection of fatty acid tryptamides (FATs). The precision of the method was evaluated by analyzing nine different samples (cocoa liquors with different ranges of shells) six times (replicate repeatability). The variations of the robust coefficient of variation of the repeatability demonstrated that FAT(C22), FAT(C24), and total FATs are good markers for the detection of shells in cocoa products. The trueness of the method was evaluated by determining the FAT content in two spiked matrices (cocoa liquors and cocoa shells) at different levels (from 1 to 50 mg/100 g). A good relation was found between the results obtained and the spiking (recovery varied between 90 and 130%), and the linearity range was established between 1 and 50 mg/100 g in cocoa products. For total FAT contents of cocoa liquor containing 5% shells, the measurement uncertainty allows us to conclude that FAT is equal to 4.01 +/- 0.8 mg/100 g. This validated method is perfectly suitable to determine shell contents in cocoa products using FAT(C22), FAT(C24), and total FATs as markers. The results also confirmed that cocoa shells contain FAT(C24) and FAT(C22) in a constant ratio of nearly 2:1.  相似文献

Table grapes (Vitis vinifera L. cv. Crimson Seedless) were coated with Aloe vera gel according to our developed patent (SP Patent P200302937) and then stored for 35 days at 1 degrees C, and the subsequent shelf life (SL) was monitored at 20 degrees C. Uncoated clusters showed a rapid loss of functional compounds, such as total phenolics and ascorbic acid. These changes were accompanied by reduction of the total antioxidant activity (TAA) and increases in total anthocyanins, showing an accelerated ripening process. On the contrary, table grapes coated with Aloe vera gel significantly delayed the above changes, such as the retention of ascorbic acid during cold storage or SL. Consequently, Aloe vera gel coating, a simple and noncontaminating treatment, maintained the functional properties during postharvest storage of table grapes.  相似文献

Purpose

The objective of this work was to evaluate the effectiveness of a plant bioassay (Phytotoxkit®) for screening ecotoxicological risks in sediments affected by mining activities.

Materials and methods

A total of 42 sediment samples affected by mining activities were studied, including 39 sediment samples from the Sierra Minera, Spain, an area affected by old extraction procedures, and three sediments from an area affected by opencast mining. These three samples were then mixed with limestone filler at 10, 20 and 30 %, providing nine stabilised samples. The total and soluble metal(loid) content (As, Cd, Cu, Fe, Pb and Zn) was determined in all samples, and the Phytotoxkit® bioassay was applied to determine the ecotoxicological effect of this procedure.

Results and discussion

The stabilised material had a neutral pH and low soluble metal(loid) concentration, similar to that of samples in which a natural attenuation process had taken place because of mixing with surrounding carbonate-rich materials. An ecotoxicological survey identified the low toxicity levels of the stabilised samples.

Conclusions

The applied bioassay is a good tool for screening metal(loid) contamination in areas affected by mining activities, since it provides information on both natural and simulated attenuation processes. The mixing of sediments with limestone filler could be applied to the remediation of zones affected by mining activities, because the toxicological effect on the tested organisms in the stabilised sediments was reduced significantly and the metal(loid) content was diminished.  相似文献

Purpose

Knowledge of sediment sources is a prerequisite for sustainable management practices and may furthermore improve our understanding of water and sediment fluxes. Investigations have shown that a number of characteristic soil properties can be used as “fingerprints” to trace back the sources of river sediments. Spectral properties have recently been successfully used as such characteristics in fingerprinting studies. Despite being less labour-intensive than geochemical analyses, for example, spectroscopy allows measurements of small amounts of sediment material (>60 mg), thus enabling inexpensive analyses even of intra-event variability. The focus of this study is on the examination of spectral properties of fluvial sediment samples to detect changes in source contributions, both between and within individual flood events.

Materials and methods

Sediment samples from the following three different origins were collected in the Isábena catchment (445 km2) in the central Spanish Pyrenees: (1) soil samples from the main potential source areas, (2) stored fine sediment from the channel bed once each season in 2011 and (3) suspended sediment samples during four flood events in autumn 2011 and spring 2012 at the catchment outlet as well as at several subcatchment outlets. All samples were dried and measured for spectral properties in the laboratory using an ASD spectroradiometer. Colour parameters and physically based features (e.g. organic carbon, iron oxide and clay content) were calculated from the spectra. Principal component analyses (PCA) were applied to all three types of samples to determine natural clustering of samples, and a mixing model was applied to determine source contributions.

Results and discussion

We found that fine sediment stored in the river bed seems to be mainly influenced by grain size and seasonal variability, while sampling location—and thus the effect of individual tributaries or subcatchments—seem to be of minor importance. Suspended sediment sources were found to vary between, as well as within, flood events; although badlands were always the major source. Forests and grasslands contributed little (<10 %), and other sources (not further determinable) contributed up to 40 %. The analyses further suggested that sediment sources differ among the subcatchments and that subcatchments comprising relatively large proportions of badlands contributed most to the four flood events analyzed.

Conclusions

Spectral fingerprints provide a rapid and cost-efficient alternative to conventional fingerprint properties. However, a combination of spectral and conventional fingerprint properties could potentially permit discrimination of a larger number of source types.  相似文献

The in vitro antimicrobial activity of commercial coffee extracts and chemical compounds was investigated on nine strains of enterobacteria. The antimicrobial activity investigated by the disc diffusion method was observed in both the extracts and tested chemical compounds. Even though pH, color, and the contents of trigonelline, caffeine, and chlorogenic acids differed significantly among the coffee extracts, no significant differences were observed in their antimicrobial activity. Caffeic acid and trigonelline showed similar inhibitory effect against the growth of the microorganisms. Caffeine, chlorogenic acid, and protocatechuic acid showed particularly strong effect against Serratia marcescens and Enterobacter cloacae. The IC(50) and IC(90) for the compounds determined by the microtiter plate method indicated that trigonelline, caffeine, and protocatechuic acids are potential natural antimicrobial agents against Salmonella enterica. The concentrations of caffeine found in coffee extracts are enough to warrant 50% of the antimicrobial effect against S. enterica, which is relevant to human safety.  相似文献

A comparative study was made of the evolution and modification of various carotenoids and vitamin A in untreated orange juice, pasteurized orange juice (90 degrees C, 20 s), and orange juice processed with high-intensity pulsed electric fields (HIPEF) (30 kV/cm, 100 micros), during 7 weeks of storage at 2 and 10 degrees C. The concentration of total carotenoids in the untreated juice decreased by 12.6% when the juice was pasteurized, whereas the decrease was only 6.7% when the juice was treated with HIPEF. Vitamin A was greatest in the untreated orange juice, followed by orange juice treated with HIPEF (decrease of 7.52%) and, last, pasteurized orange juice (decrease of 15.62%). The decrease in the concentrations of total carotenoids and vitamin A during storage in refrigeration was greater in the untreated orange juice and the pasteurized juice than in the juice treated with HIPEF. During storage at 10 degrees C, auroxanthin formed in the untreated juice and in the juice treated with HIPEF. This carotenoid is a degradation product of violaxanthin. The concentration of antheraxanthin decreased during storage, and it was converted into mutatoxanthin, except in the untreated and pasteurized orange juices stored at 2 degrees C.  相似文献

A study of the algal pigments of the biomass from aerobic treatment ponds was made with the aim of supporting other procedures to identify the predominant species that grow in the experimental condition assayed. Chlorophillic and carotenoids pigments were separated and purified by TLC for later identification and quantification by HPLC. A significant predominance of fucoxanthin and diadinoxanthin which are specific of Diatoms were found, as well as very low proportions of other non-specific pigments from other minor growing species. The absence of chlorophyll ‘b’ and a low level of ‘c’ owing to a possible coexistence of Diatom and Cyanophyceae in the culture, must be emphasized. In the experimental conditions assayed the addition of silicates to the waste water promotes the growth of Diatoms specifically Fragilaria pinnata, and also induces the formation of a secondary microflora of Cyanophyceae type with bentonic characteristics, mainly Phormidium fragile, the principal characteristics of which, are a ready tendency to sedimentation and a large size, facilitating their separation by filtration or decantation.  相似文献
