Germination ability, equilibrium relative humidity (eRH), and moisture content of ‘control’ seed samples representing 183 rice accessions stored in the active (2–4 °C) and base (?10 °C until 1993, then ?20 °C) collections of the T. T. Chang Genetic Resources Center were determined after storage for 20.5–30.5 years. Germination of seeds that had been stored in the base collection was generally high (>70 %), whereas germination was more variable for seeds stored in the active collection. Samples with lower viability after storage in the active collection were likely to have lower viability after storage in the base collection. There were significant differences in the moisture content-eRH relationship of the seeds depending on whether the seeds had been stored in the active or base collection. Based on re-test data for regular seed samples regenerated in 1979–1980 and stored in the active collection for up to 31 years, estimates of the time for ability to germinate to fall to 50 % (p 50) ranged from 54 to 997 years. For the same seed samples stored in the base collection for approximately 31 years, ability to germinate has been maintained and germination increased due to improved procedures. The ability to germinate of base collection samples was also generally higher than that of ‘safety duplicate’ samples of the same seed lots that had been sent to the National Center for Genetic Resources Preservation, USA in 1981 and stored at ?18 °C. This may have been due to uptake of moisture either during processing for dispatch or as a consequence of poor packaging material. The results are discussed in relation to long-term seed storage and genebank management. 相似文献

Barley is one of the world??s most important crops providing food and related products for millions of people. Diseases continue to pose a serious threat to barley production, despite the use of fungicides and resistant varieties, highlighting the impact of fungicide resistance and the breakdown of host plant resistance on the efficacy of control measures. This paper reviews progress towards an integrated approach for disease management in barley in which new methods may be combined with existing measures to improve the efficacy of control in the long-term. Advances have been made in genetic mapping of resistance (R) genes and in identifying novel sources of genes in wild barley populations and land races. Marker assisted selection techniques are being used to pyramid R genes to increase the durability of resistance. Elicitors to induce host resistance used in combination with fungicides can provide effective disease control in the field and could delay the evolution of fungicide insensitivity. Traits that may contribute to disease tolerance and escape have been identified and the extent of genetic variation within barley germplasm is being determined. Tools are being developed to integrate the above methods via an assessment of the risk of economic injury occurring from disease to guide decisions on the requirement for fungicide treatment. Barriers exist to the adoption of integrated management approaches from growers and end-users further down the supply chain (e.g. acceptance of variety mixtures) and policy incentives from government may be required for it to be taken up in practice. 相似文献

This study reports the occurrence of Toxoplasma gondii in the brain of three striped dolphins (Stenella ceoruleoalba) found stranded on the Ligurian Sea coast of Italy between 2007 and 2008. These animals showed a severe, subacute to chronic, non-purulent, multifocal meningo-encephalitis, with the cerebral parenchyma of two dolphins harbouring protozoan cysts and zoites immunohistochemically linked to T. gondii. Molecular, phylogenetic and mutation scanning analyses showed the occurrence of Type II and of an atypical Type II T. gondii isolates in one and two dolphins, respectively. In spite of the different molecular patterns characterizing the above T. gondii genotypes, the brain lesions observed in the three animals showed common microscopic features, with no remarkable differences among them. The role of T. gondii in causing the meningo-encephalitis is herein discussed. 相似文献

OBJECTIVES: To describe a technique for collecting cancellous bone graft from the proximal humerus in horses. STUDY DESIGN: Prospective evaluation of an experimental bone graft collection technique. ANIMAL POPULATION: Eight horses, 3-15 years, weighing 495-605 kg. METHODS: Horses were anesthetized and positioned in lateral recumbency. The lateral aspect of the proximal humerus was exposed by a 7-10-cm incision extending distally from the greater humeral tubercle, followed by sharp dissection through the omotransversarius muscle and between the infraspinatus and deltoideus muscles. A 12-mm cortical defect was incrementally created in the lateral proximal humerus. Human bone graft harvesting equipment (Acumed, Beaverton, OR) was drilled through this defect to collect a core of cancellous bone. In five horses additional cancellous bone was then collected with conventional instruments. Bone samples were weighed and histologically examined. Horses were monitored and graded for quality of anesthetic recovery, incisional complications, and postoperative lameness. RESULTS: Total mean (+/-SD) surgical time for harvesting bone with the Acumed system and traditional techniques (n=5) was 38+/-6 minutes (range, 32-47 minutes). Mean cancellous bone weight collected with the Acumed system was 3.6+/-0.8 g (range, 2.0-4.6 g), and cancellous bone collected conventionally was 25.6+/-7.5 g (range, 16.8-34.2 g). Minimal incisional complications or postoperative lameness were observed. Mortality was 12.5%; one horse fractured the operated humerus during anesthetic recovery. CONCLUSION: The Acumed system provided limited cancellous bone when used with the technique described. However, the quantity of cancellous bone collected with traditional harvesting instruments was comparable to other sites used in horses. The procedure was associated with minimal postoperative incisional complications or lameness, but because one horse suffered a catastrophic humeral fracture further research is required to assess the effects of this procedure on humeral breaking strength. CLINICAL RELEVANCE: Based on the risk of catastrophic fracture, this technique cannot be recommended for use in clinical cases, especially if an unassisted recovery from general anesthesia is planned. 相似文献

Arbor Day, or Week, is a well-established greening initiative worldwide, focusing on tree planting and the benefits thereof. Frequently most effort is targeted at institutions such as schools and community groups. Yet there has been limited assessment of the success of Arbor Day, or Week, activities both at the schools, or the wider dissemination in neighbouring communities and suburbs. The objective of this study was to determine the influence of school-based Arbor Week activities on learners’ home-based practices regarding tree planting and urban forestry. Learners from three schools in Grahamstown South Africa, with good Arbor Week participation, were assessed on their tree planting knowledge. The learners’ parents were also interviewed to determine whether the information obtained by the learners at school was taken home. A control group consisting of people with no children or very young children was also assessed. This study found that Arbor Week activities were, for the most part, successfully taught in the case study schools, and most of the learners stated that their knowledge of trees and their benefits came from their school activities. However, many learners could not remember the activities in which their schools partook the previous year. Whilst most learners were aware of the importance of trees, few had encouraged tree planting at home. However, over one-fifth of adults claimed that their knowledge of the benefits of trees was as a consequence of Arbor Day activities when they were young. Numerous constraints to tree planting were provided by learners and both the adult groups, the two most frequent being limited space and falling leaves making their yards untidy. External organisations should become more actively involved and provide much needed support systems if greening initiatives are to reach the wider community. 相似文献

Forest ecosystems are increasingly expected to produce multiple goods and services, such as timber, biodiversity, water flows, and sequestered carbon. While many of these are not mutually exclusive, they cannot all be simultaneously maximised so that management compromise is inevitable. We used a 42-year dataset from a naturally regenerating floodplain forest of the river red gum (Eucalyptus camaldulensis) to investigate the effects of pre-commercial thinning on long-term patterns in habitat quality, forest structure and rates of carbon storage (i.e. standing aboveground carbon). Estimates of habitat quality were based on the density of hollow-bearing trees because hollows are ecologically important to many species of vertebrates and invertebrates in these forests. Thinning improved habitat value by producing 20 (±8) hollow-bearing trees per ha after 42 years, while the unthinned treatment produced none. Unthinned (highest density) stands were dominated by many slender trees, mostly <25 cm in diameter, whereas thinned stands produced negatively skewed size distributions with higher median and maximum stem diameters. Moderately thinned stands (560 trees ha−1) had the highest aboveground carbon storage rate (4.1 t C year−1) and the highest aboveground carbon stocks (200.2 ± 9.6 t C ha−1) after 42 years, while the unthinned treatment had the lowest carbon storage rate (1.6 t C year−1) and an intermediate level of aboveground standing carbon (165.1 ± 31.1 t C ha−1). Our results highlight the importance of early stand density as a determinant of long-term forest structure, habitat quality and carbon storage rates. We recommend that thinning be considered as one component of a broader strategy for enhancing the structure, habitat value and aboveground carbon storage of developing floodplain forests. 相似文献

This paper details predictive models of tree-level hollow incidence for State forests in central and eastern Victoria. Models are based on the hollows component of the Statewide Forest Resource Inventory (SFRI), the most comprehensive database of hollow incidence in Australia. A two-stage methodology was used at the individual tree-level that in its first-stage generated a statement of the probability of hollows presence, and in its second-stage estimated the size of hollows. The hierarchical nature of SFRI data prompted the search for statistical methodology capable of explicitly modelling a complicated error structure. Subsequently, generalised linear mixed models (GLMMs) were used to estimate first-stage, tree-level models in the presence of spatial and nested dependence. The developed models were statistically and biologically plausible and performed well when validated using independent data. By overcoming the limitations facing previous research associated with insufficient data and inappropriate statistical methodology viable predictive models were developed. Tree-level models will help us understand patterns of hollow incidence, can be used for predictive purposes when individual-tree information is available, and can improve habitat tree retention guidelines. 相似文献

Brewers’ spent grain (BSG), one of the co-products of the brewing industry, has been mainly used as cattle feed. Spent grain was shown to contain a number of potentially high-value components such as feruloylated arabinoxylan and protein, as confirmed by microscopy and chemical analysis. A significant quantity of starch was also identified, a polysaccharide generally considered to be removed through the malting and mashing steps of brewing. As part of a study to increase the exploitation of spent grain, five separate fractions were prepared through combined milling and vibratory sieving and characterised in terms of chemical composition (polysaccharide composition and linkage; phenolic composition) and by fluorescence microscopy. Material retained on sieve mesh plates of 500, 250 and 150 μm consisted mainly of arabinoxylan-rich palea and lemma, while material passing through 106 and 55 μm sieves was fine, crumb-like material enriched in protein and starch. Lignin was present in all fractions, and originated from the fragmented palea and lemma. The results are discussed in relation to the potential for whole BSG exploitation. 相似文献
