Breeding progress in black raspberry (Rubus occidentalis L.) has been limited by a lack of genetic diversity in elite germplasm. Black raspberry cultivars have been noted for showing very few phenotypic differences and seedlings from crosses between cultivars for a lack of segregation for important traits. Despite these challenges, little molecular work has been done to explore genetic diversity and relationships in wild and cultivated black raspberry germplasm. Microsatellite, or simple sequence repeat (SSR), markers are highly polymorphic codominant markers useful for studying genetic diversity, population genetics, genetic fingerprinting and other applications. We examined genetic diversity in 148 wild and cultivated black raspberry accessions using 21 polymorphic SSR markers. Black raspberry cultivars clustered tightly and showed higher than expected heterozygosity while that of wild accessions was low. Relationships between wild black raspberry accessions were poorly resolved and regional clusters were mostly absent from our analysis. Our results indicated that wild black raspberry germplasm is a relatively untapped resource available for future breeding.  相似文献

Because dissolved organic matter (DOM) plays an important role is terrestrial C-, N- and P-balances and transport of these three components to aquatic environments, there is a need to include it in models. This paper presents the concept of the newly developed DOM modules implemented in the DAISY model with focus on the quantification of DOM sorption/desorption and microbial-driven DOM turnover. The kinetics of DOM sorption/desorption is described by the deviation of the actual DOM concentration in solution from the equilibrium concentration, Ceq. The Ceq is soil specific and estimated from pedotransfer functions taking into account the soil content of organic matter, Al and Fe oxides. The turnover of several organic matter pools including one DOM pool are described by first-order kinetics.The DOM module was tested at field scale for three soil treatments applied after cultivating grass–clover swards. Suction cups were installed at depths 30, 60 and 90 cm and soil solution was sampled for quantification of dissolved organic C (DOC) and dissolved organic N (DON). In the topsoil, the observed fluctuations in DOC were successfully simulated when the sorption/desorption rate coefficient k was low. In the subsoil, the observed concentrations of DOC were steadier and the best simulations were obtained using a high k. The model shows that DOC and DON concentrations are levelled out in the subsoils due to soil buffering. The steady concentration levels were based on the Ceq for each horizon and the kinetic concept for sorption/desorption of DOC appeared a viable approach. If Ceq was successfully estimated by the pedotransfer function it was possible to simulate the DOC concentration in the subsoil. In spite of difficulties in describing the DOC dynamics of the topsoil, the DOM module simulates the subsoil concentration level of DOC well, and also—but with more uncertainty—the DON concentration level.  相似文献

Inflammatory diseases such as type 2 diabetes (T2D) in humans and mice are under the influence of the composition of the gut microbiota (GM). It was previously demonstrated that treating Lep(ob) mice with antibiotics improved glucose tolerance. However, wild type C57BL/6J mice may also exhibit plasma glucose intolerance reminiscent of human T2D. We hypothesized that antibiotic treatment in C57BL/6 mice would have an impact on glucose tolerance without affecting weight and gut immunology. When compared to mice treated with erythromycin or the controls, treatment for five weeks with ampicillin improved glucose tolerance without significantly affecting the weight or the number of gut mucosal regulatory T cells, tolerogenic dendritic cells or T helper cells type 1. 16S rRNA gene based denaturing gradient gel electrophoresis profiles clearly clustered according to treatment and showed that antibiotic treatment reduced GM diversity. It is concluded that antibiotic treatment changes glucose metabolism as well as the composition of the GM in C57BL/6 mice, and that this does not seem to be correlated to weight development in the mice.  相似文献

  • 1. Anthropogenic‐derived nutrient inputs to coastal environments have increased dramatically worldwide in the latter half of the 20th century and are altering coastal ecosystems. We evaluated the effects of nitrogen loading on changes in macrophyte community structure and the associated fauna of a north temperate estuary. We found that a shift in primary producers from eelgrass to macroalgae in response to increased nutrient loading alters habitat physical and chemical structure and food webs. As nitrogen load increased we found increased macroalgal biomass, decreased eelgrass shoot density and biomass, decreased fish and decapod abundance and biomass, and decreased fish diversity.
  • 2. The central importance of macroalgae in altering eelgrass ecosystem support of higher trophic levels is evident in the response of the ecosystem when this component was manipulated. Removal of macroalgae increased eelgrass abundance and water column and benthic boundary layer O2 concentrations. These changes in the physical and chemical structure of the ecosystem with lower macroalgal biomass resulted in higher fish and decapod abundance and biomass.
  • 3. Both a 15N tracer experiment and the growth of fishes indicated that little of the macroalgal production was immediately transferred to secondary consumers. δ15N values indicated that the most abundant fishes were not using a grazing food web based on macroalgae. Fish tended to grow better and have a greater survivorship in eelgrass compared to macroalgal habitats.
  • 4. Watershed‐derived nutrient loading has caused increased macroalgal biomass and degradation and loss of eelgrass habitat, thus reducing the capacity of estuaries to support nekton.

Gallbladder emptying studies using ultrasonography were performed on ten normal dogs, one normal control dog, three dogs with biliary obstruction and three dogs with nonobstructive hepatobiliary disease. An intravenous bolus of a synthetic cholecystokinin was used to induce emptying. The normal canine gallbladder emptied at least 40% of its volume within one hour of synthetic cholecystokinin (0.04 μg/kg) administration. Maximum response was seen within 5 to 20 minutes. The gallbladder in icteric dogs with nonobstructive hepatobiliary disease (1 cholecystitis, 1 cholangitis, 1 cholangiohepatitis) also emptied at least 40% of its volume within one hour. The obstructed gallbladder (1 chronic pancreatitis, 1 acute pancreatitis, 1 pancreatic mass) emptied less than 20% within one hour. A significant difference (p>0.05) in the maximum percent gallbladder emptying was found between the dogs with biliary obstruction and nonobstructed, icteric dogs. No side effects to the synthetic cholecystokinin were observed.  相似文献

The clinical, radiographic, and sonographic signs in three dogs with splenic torsion are presented. AH dogs exhibited weight loss, anorexia, and lethargy. Splenomegaly was consistent radiographic finding in two dogs and large ill-defined midabdominal mass was seen in one dog. The spleen was easily imaged sonographically in all dogs. Splenic sonography in two dogs showed diffuse, hypoechoic pattern consistent with splenic congestion. Hilar splenic vessels were enlarged on the sonograms of two dogs.  相似文献

In 1989, the Danish Parliament announced a forest policy goal to double the forest area within 60–100 years. One of the objectives of this policy was to improve the recreational possibilities for the urban population. Therefore, focus has been on enhancing public afforestation projects close to urban centres with little nearby forest. We know from previous research that mature local forests possess significant amenity values; the question is whether this is also true for afforestation projects. This study examines the residents’ willingness to pay (WTP) for proximity to urban-fringe afforestation projects, using a hedonic pricing approach to estimate the effect on house prices in the neighbourhood of two afforestation projects. In both cases a significant increase in house prices in the time of afforestation is found; an increase the larger the closer the house is to the new forest. The study also examines the role of property taxes when the rise is proportional to house prices. The increased annual property tax people expect and are willing to pay in addition to the premium on the house value has to be included in the analysis to avoid serious misestimation of the total welfare economic implications of the afforestation.  相似文献

Thirteen strains of mycobacteria isolated from deer and various species of wild birds were analysed by gas chromatography (GG) for cellular fatty acids and by thin-layer chromatography (TLG) for polar lipids. These strains were compared to reference strains of Mycobacterium avium, M. para tuberculosis and M. mal-moense. All the examined strains exhibited a generally similar fatty acid pattern characterized by relatively large amounts of hexadenca-noate (16:0), octadecenoate (18:1), octadecanoate (18:0) and 10-me-thyl-octadecanoate (tuberculostearic acid, 10-Me-18:0). Several additional acids were also generally present but in smaller amounts. By means of small but distinct differences in fatty acid composition, the wild animal isolates could be distinguished from both M. paratuber-culosis and M. malmoense but not from M. avium.The TLG polar lipid patterns on the other hand separated the wild animal isolates into 2 distinct groups of complex and simple polar lipid composition which corresponded to the morphologically smooth and rough types, respectively. The complex patterns of the smooth strains were comparable to those of the M. avium serovars whereas both the rough wild animal isolates and all the M. paratuber-culosis strains showed a simple pattern of polar lipids.Both fatty acid profiles and TLG polar lipid patterns support allocation of the wild animal isolates to the MAIS complex. Moreover, the 2 chemical techniques, particularly the GC procedure, are very useful for a more rapid and precise identification of the slow-growing wild animal mycobacterial isolates which have hitherto been characterized on basis of vague criteria.  相似文献
