The present study was an investigation of the effect of the contraceptive drug, Nordette, on the stomach of the mouse when administered daily at a recommended therapeutic dose rate of 0.0026 mg kg(-1) for 30 days. Extensive light and electron microscopic changes were noticed. The drug caused enlargement in the all types of cells. The oxyntic cells appeared hypertrophied with irregular cell boundaries, enlarged nuclei and faintly stained cytoplasm. Their cytoplasm contained irregularly distributed mitochondria with dense matrix, decreased rER, obviously increased sER, disorganized intracellular canaliculi and some lysosomes. The peptic cells appeared enlarged and contained hypertrophied rough endoplasmic reticulum and an increase amount of ribosomes and secretory granules. There was an increase in the amount of the secretory granules in the lumen of the gastric gland. The mucus cells at the upper region of the gastric gland were greatly decreased. Smooth muscle fibers showed enlargement and degeneration. The submucosa and lamina propria showed vacuolation. The most pathological effects were restricted to the obvious decrease of the lymphoid cells in the submucosa and lamina propria. Dilatation and congestion of the blood vessels and blood capillaries were noticed. Blood capillaries lined by enlarged endothelial cells containing enlarged heterochromatic nuclei.  相似文献

Three ellagitannins and one disulfated flavonol were isolated from the aerial parts of Reaumuria vermiculata L. Besides that, 16 known compounds were characterized as well. The structures of all compounds were elucidated on the basis of spectroscopic data including 1D and 2D NMR and ESI HR-FTMS. The in vivo antioxidant activity using the oxygen radical absorbance capacity (ORAC) method, of the extract, its column fractions and two of the isolated ellagitannins was accomplished. In addition, a possible cytotoxicity of the extract and two of the new ellagitannins on HaCaT human keratinocytes and the activity of both compounds against the prostate cancer cell line (PC-3) were also assessed, whereby a potent cytotoxicity with IC(50) less than 1μg/ml was determined for both compounds. Besides, the extract exhibited a potential cytotoxic effect against four different solid tumor cell lines, namely liver (Huh-7), colorectal (HCT-116), breast (MCF-7) and prostate (PC-3). The IC(50)s were found to be substantially low (ranged from 1.3±0.15 to 2.4±0.22μg/ml) with relatively low resistance possibility reaching to 0% in the case of Huh-7 cell.  相似文献

The most significant focal points of the embryo transfer technology are as follows: the selection of donors, the response of the selected donor to the superovulation protocol and the obtained number of the transferable embryos. For this purpose, it is suggested that donor selection can be done by anti‐Müllerian hormone (AMH) levels, and embryo production is evaluated. AMH is secreted by the granulosa cells of primordial, pre‐antral and antral follicles below 4 mm in the ovary, independent of FSH. Therefore, the aim of this study was to investigate the relationship between serum AMH levels and the number of corpus luteum (CL), total embryos and transferable embryos that were shaped after a uniform superovulation protocol. For this reason, 48 Simmental cows, which were located at General Directory of Agricultural Enterprises (region, province, etc. instead of the general directorate), were used as donors for the embryo transfer. Blood samples were taken at random, regardless of the stage of animal's sexual cycle. AMH levels were measured by enzyme‐linked fluorescent assay (ELFA) method of the miniVIDAS® (bioMérieux SA) using AMH Bovine Test Kit. According to the statistical analyses of the obtained data, AMH levels were positively correlated with CL and total embryos (p < .05). No significant correlations between AMH and transferable embryos were approved (p > .05). It was also determined that each 200 pg/ml increase in serum AMH level resulted in one increase in CL number. Overall, considering the positive correlation between AMH level and the obtained number of CL and total embryos after a superovulation treatment, it was concluded that measuring blood AMH level prior to any further costly implementation may be an effective method in donor selection.  相似文献

Late blight caused by the oomycete Phytophthora infestans is a disease of potato and tomato of worldwide relevance and is widespread throughout Europe and the Mediterranean region. While pathogen populations in northern Europe have been sampled and characterized for many years, the genetic structure of populations from southern Europe, including Italy, has been less studied. Between 2018 and 2019, we collected 91 samples of P. infestans from potato and tomato crops in Italy, Algeria, and Tunisia on FTA cards and genotyped them using 12-plex microsatellites. These samples were compared to genotypes of P. infestans previously collected within the framework of the EuroBlight network and from published sources. Four clonal lineages were identified: 13_A2 (Blue 13), 2_A1, 23_A1, and 36_A2. Two other isolates collected could not be matched to any currently known clonal lineage. The 13_A2 and 36_A2 lineages were found exclusively in southern Italy and Algeria, while 2_A1 was only found in Algeria. This is the first report of the 36_A2 lineage in Italy. Two isolates from Solanum nigrum were 13_A2, suggesting this weed host could be a reservoir of inoculum. The 23_A1 lineage was found widely on infected tomato crops in Italy and is the same as the lineage US-23 that is widespread in North America. Differences in genotypes across the country suggests that there may be different sources of introduction into Italy, possibly via infected seed tubers from other countries in Europe, tubers for consumption from North Africa, or tomatoes.  相似文献

Abstract

A laboratory experiment was carried out to evaluate the impact of the nanoparticles of water treatment residuals (nWTRs) and salt stress on germination and growth parameters of cucumber seedling. The interaction between three nWTRs treatments (0, 500, and 1000 mgL?1) nWTRs and five saline solution (fresh water: sea water) treatments had 0.70, 2, 3, 6, and 11?dSm?1 were studied. The results revealed that increasing salinity levels significantly reduced the percentage of germination (GP) for the primed seeds treated in fresh water and nWTRs. The GP reduction was higher in seeds primed in fresh water compared to which primed in nWTRs. Salt stress negatively affected radicle length of cucumber seedling for both priming treatments. However, this impact was more pronounced for the primed seeds treated in fresh water than which treated in nWTRs at high salinity stress. Priming in nWTRs significantly decreased the root radius of cucumber seedlings, and the1000 mgL?1 priming treatment obtained a lowest value of radicle radius. Increasing salt concentration in culture medium reduced total biomass of cucumber seedling, however for the primed seeds treated in nWTRs, the total biomass was increased in comparison with which treated in fresh water. Salt tolerance and vigor indices were significantly (p?<?0.01) affected by salinity levels, nWTRs treatments and their interaction. It can be concluded that nWTRs are ameliorating materials for plant growth under salt stress conditions.  相似文献

This study presents a combination of dispersive liquid-liquid-solidified floating organic drop microextraction (DLLSFODM) and slotted quartz tube (SQT) with conventional flame atomic absorption spectrometry (FAAS) to improve the sensitivity for cadmium determination. A ligand namely 2-(4-methylphenyl)-1H-imidazo-[4,5-f]-[1,10]-phenanthroline which has not been used in trace analyte determination was used to form a cadmium complex. Stepwise optimization of parameters affecting complex formation (pH, ligand, and buffer solution) and extraction (extraction and dispersive solvents, salt effect and mixing) was done to maximize cadmium absorbance. The slotted quartz tube was fitted onto the flame burner and optimized to increase residence time of atoms in the flame. Instrumental parameters such as sample and fuel flow rate were also optimized to further enhance the absorbance signal for cadmium. Using optimal parameters and values, the limits of detection and quantification were determined to be 0.81 and 2.69 μg L?1, respectively. Low percent relative standard deviations (<?6.0%) indicated good precision for both extraction and instrumental measurements. Recovery tests were used to determine the accuracy of the method and the recovery results obtained were between 88 and 113%.

Twenty-day old seedlings were exposed to different concentrations ranging from 0–1.5 mM of chlorpyrifos through foliar spray in the field condition. The seedlings were uprooted for analyses and observed at the preflowering (five days after treatment, DAT), flowering (10 DAT) and post-flowering (20 DAT) stages for various morphological parameters such as plant height, number of branches, leaves per plant, total leaf area, plant biomass and photosynthetic pigments viz. Chlorophyll (Chl) a, Chl b, Total Chl, and Carotenoid (Car) content. Yield attributing characters like number of pods plant?1, number of seeds pod?1 and weight of 100 seeds were analyzed from both control and treated plant after harvest stage. All the growth parameters, pigments’ activity and yield parameters increased at 0.3 mM insecticidal treatment, when compared with control. Further increases in insecticide level had a negative impact upon all studied parameters.  相似文献

In this study, the effect of serum paraoxonase-1 (PON-1) activity on superovulation response and embryo yield was evaluated. The study material comprised 50 Holstein cows aged 3–4 years on postpartum day 90–120 with a body condition score of 3–3.25. A progesterone-based estrus synchronization protocol was initially administered to the selected donors. For this purpose, progesterone source was inserted intravaginally (day 0) and gonadotropin-releasing hormone injection was performed (day 6). Seven days after the insertion of progesterone device, follicle-stimulating hormone injections (total dose of 500 µg in decreasing doses for 4 days) were administered for superovulation. On the morning of the ninth day, prostaglandin (PG) F2α was administered, and the progesterone device was removed from the vagina in the evening on the same day. Two days after PGF2α administration, fixed-time artificial insemination was performed in the morning and in the evening. On the day of artificial insemination, blood samples were taken from the donors to determine the serum PON-1 activity. Uterine flushing was performed seven days after insemination. The results revealed that the serum PON-1 activity (mean ± SD, 562.71 ± 140.23 U/l) of the cows that responded to superovulation (donors with total corpus luteum count of ≥3 in both ovaries) was higher than those (389.91 ± 80.51 U/l) that did not (P<0.05). On the day of insemination, a positive correlation was determined between serum PON-1 activity and the counts of total corpus luteum (r=0.398), total oocyte/embryo (r=0.468), transferable embryo (r=0.453), and Code I embryos (r=0.315, P<0.05). Unlike the Code I embryos, there was no significant correlation between serum PON-1 activity and the number of Code III embryos. Moreover, no significant difference in the number of Code III embryos between the two PON-1 groups was observed. However, embryo yield and quality were found to have increased with increased PON-1 activity. Therefore, it was concluded that serum PON-1 activity may be associated with superovulation response, embryo yield and quality in donor cows.  相似文献

Aqueous and alcoholic extracts, volatile oil and four flavonoids, xanthomicrol (1), kumatokenin (2), jaceidin (3) and 3, 3'-di-O-methylquercetin (4), isolated from Varthemia iphionoides were investigated for their in vitro anti-platelet activity. Aqueous extract and compounds (1), (2) and (4) showed anti-platelet activity. Volatile oil and the alcoholic extract did not exhibit any anti-platelet activity. Structures of the isolated compounds (1)-(4) were determined by spectroscopic methods (UV, IR, MS, (1)H-NMR).  相似文献

The Russian wheat aphid (RWA), Diuraphis noxia (Kurdjumov), is an important pest of small‐grain cereals, particularly wheat, worldwide. The most efficient strategy against the RWA is to identify sources of resistance and to introduce them into susceptible wheat genotypes. This study was conducted to determine the mode of inheritance of the RWA resistance found in ICARDA accession IG 100695, to identify wheat microsatellite markers closely linked to the gene and to map the chromosomal location of the gene. Simple sequence repeat (SSR) marker scores were identified in a mapping population of 190 F2 individuals and compared, while phenotypic screening for resistance was performed in F2 : 3 families derived from a cross between ‘Basribey’ (susceptible) and IG 100695 (resistant). Phenotypic segregation of leaf chlorosis and rolling displayed the effect of a single dominant gene, temporarily denoted Dn100695, in IG 100695. Dn100695 was mapped on the short arm of chromosome 7D with four linked SSR markers, Xgwm44, Xcfd14, Xcfd46 and Xbarc126. Dn100695 and linked SSR markers may be useful for improving resistance for RWA in wheat breeding.  相似文献
