As Europe ages, the proportion of people who work will decline unless older individuals remain in the labor force. Such reform could be part of a more general redistribution of work. If a greater share of the population worked, then the average number of hours worked per week could be reduced. This could particularly help younger people and increase Europe's low birth rates. The challenges facing Germany, Europe's most populous country, are highlighted, but statistics are also given for five other European countries and, for comparison, the United States. Polyphenols are secondary plant compounds showing anticarcinogenic effects both in vitro and in animal experiments and may thus reduce the risk of colorectal cancer in man. The identification of polyphenol metabolites formed via their passage through the small intestine of healthy ileostomy subjects after apple juice consumption is presented. Identification and quantification of polyphenols and their metabolites were performed using HPLC-DAD as well as HPLC-ESI-MS/MS. Total procyanidin content (TPA) was measured, and additionally the mean degree of polymerization (DPm) of the procyanidins was determined in the apple juice and ileostomy effluents. As products of polyphenol metabolism, D-(-)-quinic acid and methyl esters of caffeic acid and p-coumaric acid are liberated from the corresponding hydroxycinnamic acid esters. 1-Caffeoylquinic acid and 3-caffeoylquinic acid were determined as products of isomerization. Phloretin 2'-O-glucoside (phloridzin) and phloretin 2'-O-xyloglucoside were metabolized into the corresponding aglycons phloretin and phloretin 2'-O-glucuronide and all were found in the ileostomy effluent. Ninety percent of the consumed procyanidins were recovered in the ileostomy effluent and therefore would reach the colon under physiologic circumstances. The DP m was reduced (DP m of apple juice=5.7) and varied depending on the time point of excretion. The gastrointestinal passage seems to play an important role in the colonic availability of apple polyphenols. 相似文献

Principal component analysis (PCA) was applied to the chromatographic and spectroscopic data of authentic Mexican tequilas (n = 14) and commercially available samples purchased in Mexico and Germany (n = 24). The scores scatter plot of the first two principal components (PC) of the anions chloride, nitrate, sulfate, acetate, and oxalate accounting for 78% of the variability allowed a classification between tequilas bottled in Mexico and overseas; however, no discrimination between tequila categories was possible. Mexican products had a significantly (p = 0.0014) lower inorganic anion concentration (range = 1.5-5.1 mg/L; mean = 2.5 mg/L) than the products bottled in the importing countries (range = 3.3-62.6 mg/L; mean = 26.3 mg/L). FTIR allowed a rapid screening of density and ethanol as well as the volatile compounds methanol, ethyl acetate, propanol-1, isobutanol, and 2-/3-methyl-1-butanol using partial least-squares regression (precisions = 5.3-29.3%). Using PCA of the volatile compounds, a differentiation between tequila derived from "100% agave" (Agave tequilana Weber var. azul, Agavaceae) and tequila produced with other fermentable sugars ("mixed"tequila) was possible. The first two PCs describe 89% of the total variability of the data. Methanol and isobutanol influenced the variability in PC1, which led to discrimination. The concentrations of methanol and isobutanol were significantly higher (methanol, p = 0.004; isobutanol, p = 0.005) in the 100% agave (methanol, 297.9 +/- 49.5; isobutanol, 251.3 +/- 34.9) than in the mixed tequilas (methanol, 197.8 +/- 118.5; isobutanol, 151.4 +/- 52.8). 相似文献

The delta15N(AIR) and delta2H(VSMOW) data for several alkylpyrazines formed during the roasting process of coffee are reported. Samples of commercially available roasted (n = 9) as well as self-roasted (n = 8) coffee beans (Coffea arabica L. and Coffea canephora var. robusta) of different origins were investigated. By use of extracts prepared by simultaneous distillation extraction (SDE) and subsequently fractionated by liquid chromatography on silica gel, on-line capillary gas chromatography-isotope ratio mass spectrometry was employed in the combustion (C) and pyrolysis (P) modes (HRGC-C/P-IRMS) to determine the delta15N(AIR) and delta2H(VSMOW) values, respectively. In addition to the constituents of coffee beans, data for commercial synthetic alkylpyrazines and substances declared to be "natural" were determined. The delta15N(AIR) data for coffee alkylpyrazines under study-2-ethyl-5-methylpyrazine (1) and 2-ethyl-6-methylpyrazine (2) (measured as sum 1/2), 2-ethyl-3-methylpyrazine (3), 2-methylpyrazine (4), 2,5-dimethylpyrazine (5) and 2,6-dimethylpyrazine (6) (measured as sum 5/6), and 2,3-dimethylpyrazine (7), as well as 2,3,5-trimethylpyrazine (8)-varied in the range from +8.3 to -10.2 per thousand, thus revealing their biogeneration from amino acids (delta15N(AIR) ranging from +8 per thousand to -10 per thousand). The delta2H(VSMOW) values were determined in the range from -5 per thousand to -127 per thousand. Owing to the analytical differentiation observed between coffee alkylpyrazines and synthetic/"natural" samples of 3, 4, and 7, authenticity assessment of coffee-flavored products seems to be promising, provided that extended data will be available in the future. A screening study with 25 common European wild plant species were performed over three consecutive growing seasons to investigate the effects of ozone on plant growth, reproductive development, and resource allocation. Species were grown from seedling stage until the flowering stage or seed maturity, respectively, in open-top chambers in different ozone-enriched atmospheres at environmentally-relevant concentrations. Ozone treatments covered a range of concentrations from 20 to 55 ppb ozone (seasonal 8 h daily mean). The experiments revealed significant differences between species with respect to the sensitivity of different end points toward ozone exposure. Ozone caused a significant reduction in leaf biomass of more than 20% in six species, and a significant increase in leaf biomass in three species. The relative ozone sensitivities of the species in terms of leaf biomass were different from those inferred from total shoot biomass or seed production, indicating that ozone alters resource allocation patterns in wild plants but there was considerable variation between species in effects on the allocation to leaves, stems, flowers/fruits and seeds. Germinability of seeds was affected by ozone such that germination rate was up to 30% lower in ozone-treated plants compared to control plants. Based on the genotypes screened and by combining different sensitivity criteria (vegetative growth, reproductive growth, exposure-growth response relationships) Malva sylvestris must be regarded as the most sensitive species in this study. 相似文献

Genetic diversity and relationships of 11 species and cultivars belonging to different Angiosperms families were examined using sodium dodecyl sulfate seed protein markers. The protein was resolved into 36 bands (for soybean), 41 (for quinoa), 35 (for buckwheat), and 28 to 39 bands of Amaranth species, respectively. All species and cultivars can be distinguished from each other. Soybean, quinoa, and buckwheat species had a characteristic protein pattern showing a high degree of polymorphism. The protein patterns of soybean were considerably different from other species. Amaranth species had similar seed protein electrophoretic profile. The similarity coefficients calculated on the basis of presence and absence of bands ranged from 0.08 to 0.97. Following the UPGMA algorithm of similarity coefficients, the examined species and varieties could be clustered into two similarity groups. Our results did not confirm the Tachtadzjan hypothesis that Polygonales (e.g., buckwheat) and Caryophyllales (e.g., quinoa and amaranth) are closely related. Our data rather indicate occurrence of significant genetic distance (similarity coefficients 0.05-0.10). Also, it is doubtful that amaranth and quinoa species are also closely related (similarity coefficients varied from 0.16 to 0.25). It seems that soybean, quinoa, buckwheat, and amaranth (as a genus) can be considered as phylogenetic distant taxa. Differences and similarities in the secondary structure were observed by circular dichroism spectra. This study investigated the underlying mechanisms of action for blood lipid lowering effects of citrus flavonoids and their methoxylated analogues (n = 19; dose range: 0-100 μM) in HepG2 cells. Cholesterol (CH) and triglyceride (TG) syntheses were assessed by measuring the incorporation of (14)C-acetate and (14)C-glycerol, respectively, whereas apoB secretion was determined by ELISA. Results show that two polymethoxylated citrus flavonoids (PMFs), tangeretin and nobiletin, potently inhibited apoB secretion (IC(50) = 13 and 29 μM, respectively) and modestly inhibited CH synthesis (IC(50) = 49 and 68 μM) and TG synthesis (IC(50) = 14 and 73 μM), without effecting LDL-receptor activity. Other PMFs (e.g., sinensetin) and non-PMFs (e.g., hesperetin and naringenin) had only weak effects on CH and TG syntheses and apoB secretion (IC(50) > 100 μM). The structure-activity analysis indicated that a fully methoxylated A-ring of the flavonoid structure was associated with a potent inhibitory activity on hepatic apoB secretion. In conclusion, this study using HepG2 cells indicates that citrus flavonoids with a fully methoxylated A-ring may lower blood CH and TG concentrations primarily by suppressing hepatic apoB secretion as a main underlying mode of action. 相似文献

It was the aim of the present study to determine the mobile nutrient content in leaves and pods of oilseed rape in relation to leaf age and growth stage and to develop an instrument to estimate the possible significance for interpretation of plant analytical data. From older leaves an increasing amount of sulfur (S), potassium (K), magnesium (Mg), calcium (Ca), and phosphorus (P) was leached, while no N was extracted after 24 h of leaching. In younger leaves the potential for nutrient leaching was below 10% for all investigated nutrients while in older leaves this value increased to 58% for S, 28% for P, 21% for Mg, 18% for Ca and 16% for K. Generally the potential for nutrient leaching from leaves and pods increased with growth stage. Until BBCH 83 nutrient leaching from pods was very low with less than 5% for the investigated elements, except S, but increased with further ripening drastically. 相似文献
