Greenhouse and field studies were conducted to investigate response of two rice varieties, Priscilla and Cocodrie, to sub-lethal rates of glyphosate in terms of injury, shikimate accumulation and yield. In the greenhouse, more shikimate accumulated in Cocodrie than Priscilla at comparable glyphosate rates applied to plants at the three-leaf stage. In field studies, glyphosate was applied to both varieties when they were 74-cm tall and in the internode separation growth stage. Visual injury, plant height, and leaf-tissue samples for shikimate analysis were collected at 3, 7, 14, 21 and 28 days after treatment (DAT). Rice yield was also determined. Noticeable visual injury and height reduction to both varieties was observed as early as 7 and 3 DAT in Cocodrie and Priscilla, respectively. Shikimate levels in leaves began to increase in both varieties by 3 DAT in a dose-dependent manner and reached a peak between 7 and 14 DAT. Elevated shikimate levels were still detectable by 28 DAT. Similar levels of shikimate accumulated in both varieties at comparable glyphosate rates. However, glyphosate treatment at comparable rates reduced rice yields more in Cocodrie than in Priscilla. The highest rate of glyphosate reduced yield in Cocodrie by 92% whereas there was only a 60% yield reduction in Priscilla. Shikimate levels in glyphosate-treated rice were strongly correlated to yield reductions across both varieties and appeared to be a better predictor of yield reduction than was visual injury. Visual injury coupled with measured shikimate levels can be used collaboratively to identify glyphosate exposure and estimate subsequent rice yield reductions. 相似文献

BACKGROUND: One of the most studied actions of juvenile hormone (JH) is its ability to modulate ecdysteroid signaling during insect development and metamorphosis. Previous studies in mosquitoes showed that 20‐hydroxyecdysone (20E) regulates vitellogenin synthesis. However, the action of JH and its mimics, e.g. methoprene, on female reproduction of mosquitoes remains unknown. RESULTS: Here, a major malaria vector, Anopheles gambiae Giles, was used as a model insect to study the action of methoprene on female reproduction. Ecdysteroid titers and expression profiles of ecdysone‐regulated genes were determined before and after a blood meal. An ecdysteroid peak was detected at 12 h post blood meal (PBM). The maximum expression of ecdysone‐regulated genes, such as ecdysone receptor (EcR), hormone receptor 3 (HR3) and vitellogenin (Vg) gene, coincided with the ecdysteroid peak. Interestingly, topical application of methoprene at 6 h PBM delayed ovarian development and egg maturation by suppressing the expression of ecdysone‐regulated genes in female mosquitoes. CONCLUSION: The data suggest that ecdysteroid titers are correlated with Vg synthesis, and methoprene affects vitellogenesis by modulating ecdysteroid action in A. gambiae. The genetics of photoperiod sensitivity, flowering date, fruit size, gender, female fertility, and disease resistance were
investigated in progeny between sets of elite F. virginiana selections and F. × ananassa cultivars and selections planted at sites in Michigan, Minnesota and Ontario. Progeny means varied considerably for all the
production traits. Most notable were the large fruit and high fertility observed in crosses with High Falls 22 at all three
sites, and Montreal River 10 in Ontario and Michigan. Fragaria virginiana ssp. virginiana parents yielded progeny with much larger fruit than F. virginianassp. glauca parents. General combining ability was significant for all traits at all locations, while specific combining ability was
significant for only fruit diameter, ovule set and fruit set in Michigan. Overall, the highest number of day-neutral genotypes
were detected in Ontario (mean =44%) compared to Minnesota (31%) and Michigan (26%). In progeny populations of day-neutral
F. × ananassa × short-day F. virginiana almost all fit the 1:1 ratio expected if day-neutrality is regulated by a single dominant gene; however, only a few families
of short-day F. × ananassa ×day-neutral F. virginianacrosses fit a 1:1 ratio. Likewise, in progeny of day-neutral F. virginiana ×day-neutral F × ananassa crosses, only a few of them fit the 3:1 ratio expected if day-neutrality is regulated by a single dominant gene. These data
suggest that it should be relatively easy to useF. virginiana germplasm in strawberry cultivar improvement, and that several different sources of day-neutrality may exist in natural populations.

Summary To provide a truly genotype-independent transformation system, it is necessary to be able to transform a wide range of potato genotypes. The ability to regenerate shoots in vitro was determined for 34 potato varieties using tuber disc explants. Following a culture regime used extensively in previous studies with the variety Desiree, half of the varieties could be regenerated from tuber discs and half could not. From a sample of varieties that could be regenerated from tuber discs, all but one variety gave transgenic plants. Twelve varieties were evaluated for the capacity to regenerate shoots from leaf and internode explants excised from in vitro grown plants. All of the varieties tested regenerated adventitious shoots. Leaf and internode explants from 5 varieties were subsequently used for transformation, and transgenic plants were produced from two potato varieties that did not give transgenic plants from tuber disc explants. Some varieties could not be transformed by either method, and will require modification of the in vitro regeneration and transformation system to be successful.Abbreviations 2,4-D

Agricultural sustainability considers the effects of farm activities on social, economic, and environmental conditions at local and regional scales. Adoption of more sustainable agricultural practices entails defining sustainability, developing easily measured indicators of sustainability, moving toward integrated agricultural systems, and offering incentives or imposing regulations to affect farmer behavior. Landscape ecology is an informative discipline in considering sustainability because it provides theory and methods for dealing with spatial heterogeneity, scaling, integration, and complexity. To move toward more sustainable agriculture, we propose adopting a systems perspective, recognizing spatial heterogeneity, integrating landscape-design principles and addressing the influences of context, such as the particular products and their distribution, policy background, stakeholder values, location, temporal influences, spatial scale, and baseline conditions. Topics that need further attention at local and regional scales include (1) protocols for quantifying material and energy flows; (2) standard specifications for management practices and corresponding effects; (3) incentives and disincentives for enhancing economic, environmental, and social conditions (including financial, regulatory and other behavioral motivations); (4) integrated landscape planning and management; (5) monitoring and assessment; (6) effects of societal demand; and (7) integrative policies for promoting agricultural sustainability. 相似文献

Black bears hibernate for 5 to 7 months a year and, during this time, do not eat, drink, urinate, or defecate. We measured metabolic rate and body temperature in hibernating black bears and found that they suppress metabolism to 25% of basal rates while regulating body temperature from 30° to 36°C, in multiday cycles. Heart rates were reduced from 55 to as few as 9 beats per minute, with profound sinus arrhythmia. After returning to normal body temperature and emerging from dens, bears maintained a reduced metabolic rate for up to 3 weeks. The pronounced reduction and delayed recovery of metabolic rate in hibernating bears suggest that the majority of metabolic suppression during hibernation is independent of lowered body temperature. 相似文献

The salmonid orthomyxovirus infectious salmon anaemia virus (ISAV) causes disease of varying severity in farmed Atlantic salmon, Salmo salar L. Field observations suggest that host factors, the environment and differences between ISAV strains attribute to the large variation in disease progression. Variation in host mortality and dissemination of ISAV isolates with high and low virulence (based on a previously published injection challenge) were investigated using immersion challenge. Virus dissemination was determined using real‐time PCR and immunohistochemistry in several organs, including blood. Surprisingly, the low virulent virus (LVI) replicated and produced nucleoprotein at earlier time points post‐infection compared to the virus of high virulence (HVI). This was particularly noticeable in the gills as indicated by different viral load profiles. However, the HVI reached a higher maximum viral load in all tested organs and full blood. This was associated with a higher mortality of 100% as compared to 20% in the LVI group by day 23 post‐infection. Immersion challenge represented a more natural infection method and suggested that specific entry routes into the fish may be of key importance between ISAV strains. The results suggest that a difference in virulence is important for variations in virus dissemination and pathogenesis (disease development). 相似文献

Summary The major geneH1 confers resistance in potatoes to the golden potato cyst nematode (Globodera rostochiensis). No direct effect of the gene on resistance to the white potato cyst nematode (G. pallida) was found, but factors increasing resistance toG. pallida were detected and it is possible that these are linked to theH1 gene.
