AIM: To explore the effects and mechanism of eleutheroside (ETS) B or E on the proliferation of HBZY-1 cells treated with high glucose. METHODS: The HBZY-1 cells were cultured under high glucose condition. The 4th generation of HBZY-1 cells was used for determining the optimal cell density, which was consistent with the growth regulation curve of the cells. The cells were divided into 6 groups: low glucose (LG) group, high glucose (HG) group, high glucose plus ETS-B/E (low dose, medium dose and high dose) groups, and high glucose plus losartan (LTG) group. After all cells were treated with the corresponding drugs at 24 h, 48 h and 72 h, the inhibitory rate of the proliferation was measured, and the expression of TGF-β1 and PPARγ was detected by immunocytochemistry and Western blotting. RESULTS: The best cell density was 2 000 cells/well, which was complied with the basic rules of the cell growth, and high glucose significantly promoted the HBZY-1 cell proliferation. At each time point, the inhibitory effects of ETS-B/E were significantly different between HG group and LTG group on the proliferation of the HBZY-1 cells (P<0.05). The expression of TGF-β1 was significantly inhibited, and the expression of PPARγ was significantly promoted by ETS-B/E (P<0.05). ETS-E showed stronger effect than ETS-B (P<0.05) in a concentration- and time-dependent manner. CONCLUSION: ETS-B/E significantly inhibits the proliferation of HBZY-1 cells under high glucose condition by decreasing TGF-β1 expression and promoting PPARγ expression. 相似文献

本试验旨在研究骆驼乳清蛋白(CWP)对热应激(HS)大鼠肝损伤、氧化应激及肝功能的保护作用。选取6周龄SD大鼠36只,适应性饲养2周后随机分为6组:正常对照组饲喂基础日粮;CWP对照组添加400 mg·kg-1的CWP;HS组除饲喂基础日粮外,每天进行2 h HS处理,连续8 d;3个CWP干预组分别于每次HS前灌服100、200和400 mg·kg-1的CWP。试验结束后,取大鼠肝组织,HE染色观察肝组织病理学变化,同时检测肝功能生物标志物、氧化应激标志物水平及抗氧化酶活性。结果表明:CWP降低了HS大鼠血清谷草转氨酶(AST)和谷丙转氨酶(ALT)活性,400 mg·kg-1的CWP干预效果最好(P<0.01);400 mg·kg-1CWP干预组有效降低了HS诱导的大鼠肝组织病理学改变; CWP降低了大鼠肝活性氧(ROS)及丙二醛(MDA)水平,增加了超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性及谷胱甘肽(GSH)水平,400 mg·kg-1的CWP干预效果最好(P<0.01)。结果提示,CWP干预能够以剂量依赖性方式降低大鼠肝氧化应激,增加抗氧化系统防御能力,从而缓解HS所致大鼠肝损伤。 相似文献

试验旨在探索羊源D型多杀性巴氏杆菌入侵宿主脾脏组织中引发的免疫应答途径。首先利用羊源D型多杀性巴氏杆菌(HN01菌株)感染小鼠,建立羊源D型多杀性巴氏杆菌感染的动物模型;之后利用转录组测序技术获得感染小鼠与正常小鼠的脾脏转录组数据,并使用COG、KOG、eggNOG、GO、KEGG数据库对测序结果中的差异表达基因(differentially expressed genes,DEGs)进行功能注释与分析,同时对于显著富集到关键免疫通路的差异表达基因使用STRING软件和KEGG mapper进行蛋白互作分析,筛选出核心通路中起关键作用的基因;最后选取关键的10个基因进行实时荧光定量RT-PCR验证。转录组分析结果显示,与正常组相比,感染组中筛选出3 380个差异表达基因(P<0.01,log2|FoldChange|≥0.5),其中1 691个基因上调,1 689个下调。基因功能富集分析结果表明,感染组脾脏中的差异表达基因主要发挥信号转导的功能,其主要参与的生物途径包括细胞因子与细胞因子受体互作通路、趋化因子信号通路、HIF-1信号通路、TNF信号通路。蛋白互作分析筛选出约28个核心差异表达基因,结合实时荧光定量RT-PCR验证后,其中9个基因的表达结果与测序一致,分别是C3、Cd4、Cxcl13、Lck、Gnai1、Grap2、IL-6、Cxcr6及Serping1基因。本研究初步证明了脾脏在抵抗多杀性巴氏杆菌入侵中参与了一系列免疫应答反应,为进一步研究羊源D型多杀性巴氏杆菌与宿主之间相互作用的分子机制奠定理论基础。 相似文献

以杜仲转录组数据为试材,采用CodonW和SPSS软件相结合的方法,研究了杜仲基因密码子组成和长度对基因表达量的影响,以期为将来改造外源基因及利用基因工程技术改良杜仲提供理论基础。结果表明:杜仲转录组中基因的表达量与G3s、C3s、G+C和GC3s含量均呈极显著正相关(P0.01),与A3s含量、T3s含量、单条基因总密码子数(L_aa)和有效密码子数ENC均呈极显著负相关(P0.01),与Aro含量呈显著负相关(P0.05),表明高表达基因的密码子使用偏好性较强,且偏好于使用以G/C结尾的密码子;ENC与C3s、G3s、G+C和GC3含量均呈极显著负相关(P0.01),与A3s、T3s、Aro含量呈极显著正相关(P0.01);确定了15个杜仲最优密码子,分别为AAC、AAG、ACC、AGG、AUC、CAC、CAG、CCG、CUC、CUG、GCC、GGC、GUC、UAC和UCC。果实和叶片特异表达基因在氨基酸Leu、Ile、Ser、Pro、Thr、Asn、Glu和Cys密码子的使用上存在差异。 相似文献

In TT&C system,unified spread spectrum framework and adaptive antenna digital beamforming technologies are used to track, telemetry and command objects. Constant modulus array (CMA) and adaptive signal canceller estimate the direction of arrival (DOA) of every signal and use the optimum weighted vector to form the beams. This paper analyzes the structure and line current waveform of 3-phase AC voltage regulation circuit. As an application of 3-phase AC voltage regulation circuit, a high-frequency welding device's simulation model and harmonic analysis of line current are presented. Results of simulation are compared to field measurements. The two are in a good agreement, so this simulation model is correct. Two single-tuning filters and one second-order HPF are designed according to the device's harmonic condition. Simulation results of the circuit with filters indicate decrease of the harmonics in current of power supply, and the waveform is close to sinusoidal wave, which prove correctness of the solution to harmonics pollution. 相似文献

This paper introduces the development of the electronic throttle control system (ETCS) , the structure and the principle of the system are also involved. After analyzing the nonlinearity problem existing in the mechanical structure of the system, a mathematical model of the ETCS is built up. Then three controllers (PID controller, Fuzzy controller, sliding mode controller) were designed and simulated in the matlab simulink.
