ABSTRACT:   Stable reproduction is essential for supplying artificially hatched fish to tuna aquaculture. We observed testes maturation in reared Pacific bluefin tuna (PBT) Thunnus orientalis at 2+ years of age. The incidence of males with mature testes was 25.0%, and 40% of the males had developing testes that contain spermatozoa, while oocytes of the same aged females were not mature. These fish were wild-caught at 0+ years old in August 1997 and the gonads were examined in October 1998 and January–February 2000. Therefore, the age at examination in 2000 was estimated to be 2 years and 7–10 months old considering the spawning season of the wild PBT and the size when captured. Histological examination of the matured and developing testes showed that they contained spermatozoa, spermatids, spermatocytes, and spermatogonia. All the spermatozoa were observed to be motile in sea water under light microscopy. From the results of this and previous studies, matured males are probably fertile for at least 5 months a year in Kushimoto. The testes maturation observed at young age in captivity is considered promising to reduce the cost of broodstock maintenance for the juvenile production of PBT, especially if the sperm are cryopreserved.  相似文献

Bacterial community structures were analyzed in water used for rearing fish larvae by fluorescence in situ hybridization. In Experiment 1, red sea bream Pagrus major larvae were reared in two commercial seed production tanks. The survival rate in Tank 1 was higher than in Tank 2, even though phytoplankton, Nannochloropsis sp., was added to both tanks. In Tank 2, γ-proteobacteria became dominant (∼70% of total bacteria) on day 13, there after heavy larval mortalities occurred. In Tank 1, however, α-proteobacteria and the Cytophaga-Flavobacterium cluster were predominant from day − 1 until day 13; no significant mortality was recorded. In Experiment 2, marble goby Oxyeleotris marmoratus larvae were cultured with or without Nannochloropsis sp. At the end of the experiment, larval survival rates in aquaria with Nannochloropsis sp. were significantly (P <0.05) higher than those without. In rearing water without Nannochloropsis sp., γ-proteobacteria increased during rearing. In rearing water with Nannochloropsis sp., α-prote obacteria and the Cytophaga-Flavobacterium cluster were predominant at the beginning of the experiments and the relative abundance of γ-proteobacteria was maintained at a lower level throughout the experiments. The predominance of α-proteobacteria and the Cytophaga-Flavobacterium cluster appears to be a good indicator of successful larval production.  相似文献

Using the full-cycle cultured (FC) Pacific bluefin tuna [body weight 16.3±1.9 kg (pre-fasting group, pre-FG), 14.2±0.9 kg (post-fasting group, post-FG)], changes in the physical/chemical properties of the cephalal parts of dorsal (D) and ventral (V) ordinary muscles (OM) by fasting (6 days) during chilled storage (4°C) were investigated. Condition factors were 26.7 (pre-FG) and 20.3 (post-FG, P < 0.05). Fasting changed the liver color to green. Fasting also decreased the amount of protein and lipid contents of the DOM and VOM of FC tuna. The breaking strength and pH of the DOM and VOM of post-FG tuna were higher (P <0.05) than for pre-FG tuna during storage. In contrast, the glycogen contents of DOM and VOM of post-FG tuna were lower than for pre-FG tuna. The color values (L*, a* and b*) of DOM of post-FG tuna were lower than for pre-FG tuna throughout the storage period. In addition, the metmyoglobin (metMb) content of DOM of post-FG tuna was lower (P <0.05) than that of pre-FG tuna, and the metMb content of VOM of post-FG tuna remained low after fasting. These results indicate that fasting suppresses deterioration (especially meat color) of FC tuna muscles during chilled storage.  相似文献

Fucoxanthin is a marine carotenoid exhibiting several health benefits. The anti-cancer effect of fucoxanthin and its deacetylated metabolite, fucoxanthinol, is well documented. In view of its potent anti-carcinogenic activity, the need to understand the underlying mechanisms has gained prominence. Towards achieving this goal, several researchers have carried out studies in various cell lines and in vivo and have deciphered that fucoxanthin exerts its anti-proliferative and cancer preventing influence via different molecules and pathways including the Bcl-2 proteins, MAPK, NFκB, Caspases, GADD45, and several other molecules that are involved in either cell cycle arrest, apoptosis, or metastasis. Thus, in addition to decreasing the frequency of occurrence and growth of tumours, fucoxanthin has a cytotoxic effect on cancer cells. Some studies show that this effect is selective, i.e., fucoxanthin has the capability to target cancer cells only, leaving normal physiological cells unaffected/less affected. Hence, fucoxanthin and its metabolites show great promise as chemotherapeutic agents in cancer.  相似文献

To halt biodiversity loss in the humid tropics of developing countries, it is crucial to understand the roles and effects of human-modified landscapes with fragmented forest remnants in maintaining biodiversity while fulfilling the demands of local communities and reducing poverty. To implement appropriate landscape planning for conserving biodiversity and ecosystem functioning, appropriate information is required about parameters of habitat suitability among various anthropogenic habitats with a range of distances to forests and vegetation characteristics, but such information is limited. We examined differences in avian communities between a remnant forest and four types of man-made forest (two mature plantations and two agroforests) in a forest–agricultural landscape of West Java, and we analyzed the effects of both local and landscape factors on various types of species richness in this landscape. The results from non-metric multidimensional scaling revealed avifauna in the two types of agroforest was clustered separately from that in the remnant forest, mainly because drastic declines in the abundance of forest specialists (including IUCN red-listed species) and their replacement with open-habitat generalists. The mixed-tree agroforests were colonized by 30 % of forest specialists and forest-edge species found in the remnant forest, and maintained the highest richness of species endemic to Indonesia among man-made forests, implying that some forest specialists and endemics might have adapted to ancient landscape heterogeneity. High proportion of insectivorous birds was found in the remnant forest (more than 50 %) and drastically decline in man-made forests, although the species richness of insectivores did not decline significantly in broad-leaved plantations. We concluded that protection of remnant forests should be prioritized to conserve forest bird diversity. However, as different environmental factors affected the richness values of different ecological groups, appropriate landscape design and habitat management could improve functional diversity in forest–agricultural landscapes in the tropics.  相似文献

Information regarding the distributional patterns of top predators is undoubtedly important for the monitoring and management of marine ecosystems. Nevertheless, previous knowledge on the distributional patterns of cetacean species in relation to physical oceanography is usually limited to a small fraction of each species' range because of their vast distribution and infrequency of encountering many species. We carried out comprehensive analyzes on the distributions of small odontocetes covering almost the entire North Pacific Ocean. Using data from over 300 000 nautical miles (≈ 555 600 km) of dedicated sighting surveys and 7000 school sightings of 14 small odontocete species, which have accumulated for approximately a quarter of a century, we describe interspecific habitat differences in small odontocetes with respect to physical oceanography. Multivariate analyzes were used to discriminate the 14 species into four ecological groups: (i) Subtropical Domain species; (ii) Subarctic Boundary species; (iii) Transitional Domain species; and (iv) Subarctic Domain species. By comparing environmental variables in their habitats with variables indicative of water mass and frontal structures, the four species groups were further divided into several subgroups. Groups at higher latitudes comprised fewer species than those at lower latitudes, while species diversity was maximal at lower and intermediate latitudes. Our findings indicate that large‐scale oceanographic structures are important determinants for the present patterns in biogeography and species diversity of small odontocetes.  相似文献

This study investigated the optimal timing of day to promote initial swimbladder inflation (ISI) for improved Pacific bluefin tuna (PBT), Thunnus orientalis, larval survival. Larval swimbladder inflation frequency was compared based on three experiments using different time schemes of surface film removal (SFR) from 3 to 9 days post hatch (dph). SFR was conducted from 05:00 to 19:00 hours (light period: S.5–19), 19:00 to 05:00 hours (dark period: S.19–5), 08:00 to 19:00 hours (S.8–19) and the entire day (S.24) in Experiment 1; from 08:00 to 19:00 hours (S.8–19‐E2), 08:00 to 13:00 hours (S.8–13), 13:00 to 19:00 hours (S.13–19) in Experiment 2; and from 13:00 to 16:00 hours (S.13–16), 16:00 to 19:00 hours (S.16–19), 18:00–19:00 hours (S.18–19) in Experiment 3. The swimbladder inflation frequency at the experiment termination (9 dph) was significantly higher (< 0.001) in S.24 (91.1 ± 5.7%), S.5–19 (92.2 ± 5.1%) and S.8–19 (93.3 ± 3.4%) than in S.19–5 (11.1 ± 5.1%) in Experiment 1, and remarkably higher in S.8–19‐E2 (81.7%) and S.13–19 (88.3%) than in S.8–13 (0.0%) in Experiment 2, and significantly higher (< 0.001) in S.16–19 (84.4 ± 5.1%) and S.18–19 (70.0 ± 12.0%) than in S.13–16 (7.8 ± 3.9%) in Experiment 3. These results suggest that the optimal timing to promote larval ISI by SFR is a few hours before the end of light period (16:00–19:00 hours) from 3 to 9 dph.  相似文献

Juvenile Pacific bluefin tuna (Thunnus orientalis; PBT) often experience high mortality during ship transportation. This study investigated whether the addition of colours or patterns to the walls of tanks affected survival rate. In the first experiment, three colours and lattice patterns were tested: dark blue single‐colour, red single‐colour, and red–blue lattice pattern. Fish in all tanks exhibited abnormal behaviours when sunlight entered the tanks between 0800 and 1000 hours, but mortality only increased in the single‐coloured tanks as a result of collision with the tank walls. In the second experiment, four colours and patterns were tested: dark blue single‐colour, red–blue lattice pattern, red–blue lattice pattern with shade sheet and red–green lattice pattern with shade sheet. Again, we visually observed that fish in all treatment groups exhibited abnormal behaviour when sunlight entered the tanks, but there were no collision deaths in the lattice‐patterned tanks and survival in this group was significantly higher than in the single‐coloured tanks. Thus, the use of a high‐contrast colour pattern can prevent mass death of juvenile PBT during ship transportation.  相似文献

This study investigated the influence of initial swimbladder inflation (ISI) failure on survival of Pacific bluefin tuna (PBT), Thunnus orientalis, larvae to prevent mass mortality due to sinking death. In Experiment 1, swimbladder inflation frequency and survival within an ISI promoted (PS) group, for which surface film of rearing water was removed, and a group without ISI promotion (NPS) were compared in 20 and 30 kL tanks. The PS group demonstrated significantly higher swimbladder inflation frequency and increased survival than the NPS group within 20 kL tanks at 9 days post hatching (dph). Similar tendencies were observed within 30 kL tanks. In Experiment 2, larval distribution and swimbladder inflation frequency in the night‐time were examined through larval sampling within the upper and middle layers of the water column and tank bottom within 1.6 and 30 kL tanks at 5 dph. Larvae at tank bottom had higher distributional density and significantly lower swimbladder inflation frequency than those distributed in the upper and middle layers within 1.6 kL tanks. Similar tendencies were observed within 30 kL tanks. Results of this study suggest the improvement of larval survival due to prevention of sinking death through ISI promotion in mass‐scale PBT larviculture.  相似文献

Nematodes of the family Heligmonellidae (Heligmosomoidea; Trichostrongylina) reside in the digestive tracts of rodents and lagomorphs. Although this family contains large numbers of genera and species, genetic information on the Heligmonellidae is very limited. We collected and isolated adult worms of three species in Japan that belong to the family Heligmonellidae, namely Heligmonoides speciosus (Konno, 1963) Durette-Desset, 1970 (Hs) from Apodemus argenteus, Orientostrongylus ezoensis Tada, 1975 (Oe) from Rattus norvegicus and Lagostrongylus leporis (Schulz, 1931) (Ll) from Pentalagus furnessi, and sequenced the entire internal transcribed spacer regions, ITS-1 and ITS-2 of ribosomal DNA. ITS-1 of Hs, Oe and Ll was 426, 468 and 449 bp in length, and had a G+C content of about 41, 41 and 37 %, respectively. ITS-2 of Hs, Oe and Ll was 297, 319 and 276 bp in length and had a G+C content of about 38, 40 and 28%, respectively. The data of Hs, Oe and Ll were compared with those of two other known species within the family Heligmonellidae, Calorinensis minutus (Dujardin, 1845) (Cm) and Nippostrogylus brasiliensis (Travassos, 1914) (Nb), and with those of two species of Heligmosomidae (Heligmosomoidea), Heligmosomoides polygyrus bakeri and Ohbayashinema erbaevae. Phylogenetic analysis placed Hs, Oe and Ll in the same clade with Cm and Nb, forming a Heligmonellidae branch in both ITS-1 and ITS-2, separate from the Heligmosomoidea branch. These results demonstrated that the ITS-1 and ITS-2 sequences are useful for differentiating the Heligmonellidae nematode species. This study is the first to describe the ITS-1 and ITS-2 sequences of Hs, Oe and Ll.  相似文献
