绿豆抗豆象基因PCR标记的构建与应用 总被引:5,自引:1,他引:5
采用PCR分子标记技术,对16个绿豆品种(系)进行了遗传分析。在选用的56个随机引物中,发现抗豆象品种与感豆象品种间有一定差异。根据聚类分析结果将它们分成抗豆象野生种(TC1966)、抗豆象栽培种(V2709)、抗豆象杂交后代(VC3890A2/TC1966-23)和混合类型4个大组。以绿豆抗豆象和感豆象品种及抗豆象品种×感豆象品种组合的F2群体为试验材料,利用BSA法,对抗(感)豆象品种池和一个组合F2的抗(感)豆象池进行了鉴定,获得一个共显性标记。经F2分析,在抗豆象个体中扩增出约1.79 kb的特异片段或2个特异片段(1.79 kb/1.03 kb);在感豆象个体中仅扩增出约1.03 kb的特异片段。初步认为此标记与TC1966的抗豆象基因位点紧密联锁,可用于绿豆抗豆象种质鉴定和遗传育种的分子标记辅助选择。 相似文献

OBJECTIVE: To determine the percentage of broodmares and foals that shed Clostridium perfringens in their feces and classify the genotypes of those isolates. DESIGN: Prospective cross-sectional study. ANIMALS: 128 broodmares and their foals on 6 equine premises. PROCEDURES: Anaerobic and aerobic bacteriologic cultures were performed on feces collected 3 times from broodmares and foals. All isolates of C. perfringens were genotyped. RESULTS: Clostridium perfringens was isolated from the feces of 90% of 3-day-old foals and 64% of foals at 8 to 12 hours of age. A lower percentage of broodmares and 1- to 2-month-old foals shed C. perfringens in their feces, compared with neonatal foals. Among samples with positive results, C. perfringens type A was the most common genotype identified (85%); C. perfringens type A with the beta2 toxin gene was identified in 12% of samples, C. perfringens type A with the enterotoxin gene was identified in 2.1% of samples, and C. perfringens type C was identified in < 1% of samples. CONCLUSIONS AND CLINICAL RELEVANCE: Clostridium perfringens was identified from the feces of all but 6 foals by 3 days of age and is likely part of the normal microflora of neonatal foals. Most isolates from broodmares and foals are C. perfringens type A; thus, the clinical relevance of culture results alone is questionable. Clostridium perfringens type C, which has been associated with neonatal enterocolitis, is rarely found in the feces of horses. 相似文献

When we look at our hands, we immediately know that they are part of our own body. This feeling of ownership of our limbs is a fundamental aspect of self-consciousness. We have studied the neuronal counterparts of this experience. A perceptual illusion was used to manipulate feelings of ownership of a rubber hand presented in front of healthy subjects while brain activity was measured by functional magnetic resonance imaging. The neural activity in the premotor cortex reflected the feeling of ownership of the hand. This suggests that multisensory integration in the premotor cortex provides a mechanism for bodily self-attribution. 相似文献

OBJECTIVES: To clone and sequence the cDNA for feline preproparathyroid hormone (preproPTH) and to compare that sequence with other known parathyroid hormone (PTH) sequences. SAMPLE POPULATION: Parathyroid glands from 1 healthy cat. PROCEDURES: A cDNA library was constructed in lambda phage from feline parathyroid gland mRNA and screened with a radiolabeled canine PTH probe. Positive clones were sequenced, and nucleic acid and deduced amino acid sequences were analyzed and compared with known preproPTH and PTH sequences. RESULTS: Screening of approximately 2 X 10(5) recombinant plaques revealed 3 that hybridized with the canine PTH probe; 2 clones comprised the complete sequence for feline preproPTH. Feline preproPTH cDNA consisted of a 63-base pair (bp) 5'-untranslated region (UTR), a 348-bp coding region, and a 326-bp 3'-UTR. The coding region encoded a 115-amino acid peptide. Mature feline PTH consisted of 84 amino acids. Amino acid sequence analysis revealed that feline PTH was > 83% identical to canine, bovine, swine, equine, human, and macaque PTH and 69, 71, and 44% identical to mouse, rat, and chicken PTH, respectively. Within the region responsible for hormonal activity (amino acids 1 to 34), feline PTH was > 79% identical to other mammalian PTH sequences and 64% identical to the chicken sequence. CONCLUSIONS AND CLINICAL RELEVANCE: The amino acid sequence of PTH is conserved among mammalian species. Knowledge of the cDNA sequence for feline PTH may be useful to investigate disturbances of calcium metabolism and alterations in PTH expression in cats. 相似文献

The present study was carried out using juveniles of silver pomfret, Pampus argenteus, produced in the hatchery by stripping eggs from spawners collected from the wild during 2004 and 2005 culture period. The experiments carried out during 2004 investigated the efficiency of using salmon feed mixed with cyclopeeze, salmon feed mixed with shrimp meat, salmon feed alone, and shrimp meat alone in the diet using 1 m3 capacity round fiberglass tanks in three replicates for each treatment with flow‐through seawater. The results showed that inclusion of shrimp meat in the diet either with salmon feed or alone give significantly high (P < 0.01) weight gain of up to 1.10 ± 0.06 g/fish/d and significantly lower (P < 0.001) feed conversion ratio compared to that of feeds without shrimp meat. The experiments carried out during 2005 investigated the use of commercially available feeds with feed additives. The results showed that feeding with “Gemma” feed with 54.0% crude protein and 19.0% crude fat or salmon feed with 41.4% crude protein and 23.9% crude fat give significantly high (P < 0.01) growth rates compared to that of pompano feed with 43.0% crude protein and 6.0% crude fat. The results of these investigations although show high growth rate of the fish compared to that of previous observations; there is yet an urgent need to formulate a suitable feed for this fish species for commercial ventures. 相似文献

This study was conducted to determine the effects of dietary highly unsaturated fatty acids (HUFA) on flame angelfish (Centropyge loriculus) reproduction, and egg and larval quality. In the experiment, formulated diets containing 1.8, 2.9 or 3.6% n‐3 HUFA were fed to flame angelfish broodstock (n = 4) for 5 months. Mean fecundity (daily egg production), egg fertilization rates and embryo viability were used as indicators of egg quality. In addition, mean egg diameter, oil globule diameter, per cent hatch, larval size at hatch, per cent survival to yolk exhaustion and larval size at yolk exhaustion were recorded for each treatment. Flame angelfish fed the diet containing 3.6% n‐3 HUFA exhibited significantly increased fecundity, fertilization rates and embryo viability than fish that were fed the other two formulated diets. Egg diameter, egg oil globule diameter, larval size at hatch, larval survival to yolk exhaustion and larval size at yolk exhaustion from the 3.6% n‐3 HUFA broodstock treatment group were not significantly different from those derived from the Control broodstock treatment. These data revealed that flame angelfish egg quality could respond rapidly (within weeks) to maternal dietary changes. Results from this study further support that dietary HUFA composition can significantly affect broodstock reproductive performance as well as subsequent performance of eggs and larvae. 相似文献

Nets in traditional Porphyra mariculture are seeded with conchospores derived from the conchocelis phase, and spend a nursery period in culture tanks or calm coastal waters until they reach several centimeters in length. Some species of Porphyra can regenerate the foliose phase directly through asexual reproduction, which suggests that the time, infrastructure, and costs associated with conchocelis culture might be avoided by seeding nets with asexual spores. Here, we present work from a short-term mariculture study using nets seeded with asexual spores (neutral spores) of a native Maine species of Porphyra. Porphyra umbilicalis (L.) Kützing was selected for this proof of concept research because of its reproductive biology, abundance across seasons in Maine, and evidence of its promise as a mariculture crop. We studied the maturation, release, and germination of the neutral spores to develop an appropriate seeding protocol for nets, followed by development of a nursery raceway to provide an easily manipulated environment for the seeded nets. Neutral spores were produced throughout the year on the central Maine coast; however, there was a temporal variability in the number and survival of released neutral spores, depending upon thallus position in the intertidal zone. Small thalli were strictly vegetative, but most thalli reproduced by neutral spores; sexual reproduction was absent. Neutral spores germinated quickly at 10 and 15 °C, but germination was delayed at 5 °C. Unlike some algal zygotes and spores, neutral spores of P. umbilicalis required light to germinate; however, irradiances of 25 and 100 μmol photons m− 2 s− 1 were equally sufficient for germination. Rafts of seeded nets were deployed in Cobscook Bay, Maine, at two distances from salmon aquaculture pens and at a control site on a nearby, fallow aquaculture site (no salmon). There was no difference in nitrogen content of harvested thalli; however, both the density and the surface area of harvested thalli were different among the sites.
