ABSTRACT The population structure of Guignardia citricarpa sensu lato (anamorph: Phyllosticta citricarpa), a fungus of which strains pathogenic to citrus are subject to phytosanitary legislation in the European Union and the United States, was investigated. Internal transcribed spacer sequences revealed two phylogenetically distinct groups in G. citricarpa. This distinction was supported by amplified fragment length polymorphism analysis that also supported the exclusion of two isolates that had apparently been misclassified as G. citricarpa. On cherry decoction agar, but not on other media, growth rates of group I isolates were lower than those of group II isolates. Conidial dimensions were similar, but group I isolates formed conidia with barely visible mucoid sheaths, whereas those of group II formed conidia with thick sheaths. Cultures of isolates belonging to group I produced rare infertile perithecia, whereas fertile perithecia were formed by most isolates of group II. Colonies of isolates belonging to group I were less dark than those of group II, with a wider translucent outer zone and a lobate rather than entire margin. On oatmeal agar, exclusively group I isolates formed a yellow pigment. Group I harbored strains from citrus fruits with classical black spot lesions (1 to 10 mm in diameter) usually containing pycnidia. Group II harbored endophytic strains from a wide range of host species, as well as strains from symptomless citrus fruits or fruits with minute spots (<2-mm diameter) without pycnidia. These observations support the historic distinction between slowly growing pathogenic isolates and morphologically similar fast-growing, nonpathogenic isolates of G. citricarpa. The latter proved to belong to G. mangiferae (P. capitalensis), a ubiquitous endophyte of woody plants with numerous probable synonyms including G. endophyllicola, G. psidii, P. anacardiacearum, and P. theacearum. G. mangiferae occurs in the European Union and the United States on many host species including citrus, and does not cause symptoms of citrus black spot, justifying its exclusion from quarantine measures.  相似文献

This study used discriminant analysis to evaluate body measures and sexual dimorphism in Mangalarga Marchador horses. Discriminant analysis is a multivariate method that generates functions to classify animals using a prior criterion. In this study a prior criterion is male or female. The study analyzed 25 linear and 11 angular measures obtained from 25 stallions and 56 mares registered with the Mangalarga Marchador Brazilian Breeders Association. Knee girth, cannon girth, chest width, chest girth, back-loin length, hip width, distance from elbow to knee, hind limb hoof length, head width, shoulder length, and body length were found to be linear measures that provided evidence of sexual dimorphism. Angular measures with evidence of sexual dimorphism were shoulder–humerus, metacarpal–phalanx, coxae–femur, and femur–tibial angles. Stallions were larger than mares for almost all linear measures, except for back-loin length, hip width, and distance from elbow to knee. Shoulder–humerus and coxal–femur angles were larger in mares, while metacarpal–phalanx and femur–tibial angles were larger in stallions. Using linear measures, two mares were classified as stallions and three stallions were classified as mares. For angular measures, two other mares were classified as stallions and three other stallions were classified as mares. The discriminant functions can be useful for the selection of horses and to avoid registration of animals not meeting the phenotypic standards of the breed association.  相似文献

Somatic hybridization is a biotechnology tool that can be used in citrus breeding programs to produce somatic hybrids with the complete genetic combination of both parents. The goal of this work was to test the reaction of citrus somatic hybrids that may be useful as rootstocks to trunk and root infections caused by Phytophthora nicotianae van Breda de Haan (P. parasitica Dastur) and to citrus tristeza virus (CTV). The somatic hybrids evaluated were ‘Caipira’ sweet orange (Citrus sinensis L. Osbeck) + ‘Rangpur’ lime (C. limonia Osbeck), ‘Caipira’ sweet orange + ‘Cleopatra’ mandarin (C. reshni hort. ex Tanaka), ‘Caipira’ sweet orange + ‘Volkamer’ lemon (C. volkameriana V. Ten. & Pasq.), ‘Caipira’ sweet orange + rough lemon (C. jambhiri Lush.), ‘Cleopatra’ mandarin + ‘Volkamer’ lemon, ‘Cleopatra’ mandarin + sour orange (C. aurantium L.), ‘Rangpur’ lime + ‘Sunki’ mandarin (C. sunki (Hayata) hort. ex Tanaka), ‘Ruby Blood’ sweet orange (C. sinensis L. Osbeck) + ‘Volkamer’ lemon, ‘Rohde Red’ sweet orange (C. sinensis L. Osbeck) + ‘Volkamer’ lemon, and ‘Valencia’ sweet orange + Fortunella obovata hort. ex Tanaka. For P. nicotianae trunk and root infection assays, plants of the somatic hybrids, obtained from 9-month semi-hardwood cuttings, were evaluated and compared with diploid citrus rootstock cultivars after mycelia inoculation in the trunk or spore infestation in the substrate, respectively. ‘Cleopatra’ mandarin + sour orange, ‘Rangpur’ lime + ‘Sunki’ mandarin, ‘Cleopatra’ mandarin + ‘Volkamer’ lemon, ‘Ruby Blood’ sweet orange + ‘Volkamer’ lemon, ‘Rohde Red’ sweet orange + ‘Volkamer’ lemon, and ‘Caipira’ sweet orange + ‘Volkamer’ lemon had less trunk rot occurrence, whereas the somatic hybrids ‘Cleopatra’ mandarin + ‘Volkamer’ lemon, ‘Cleopatra’ mandarin + sour orange, ‘Caipira’ sweet orange + ‘Volkamer’ lemon, and ‘Caipira’ sweet orange + ‘Rangpur’ lime were tolerant to root rot. For CTV assays, plants of the somatic hybrids along with tolerant and intolerant rootstocks were budded with a mild strain CTV-infected or healthy ‘Valencia’ sweet orange budwood. Differences in average scion shoot length indicated that the hybrids ‘Cleopatra’ mandarin + sour orange and ‘Valencia’ sweet orange + Fortunella obovata were intolerant to CTV.  相似文献

The caprine arthrite encephalite (CAE) is a disease that affects especially dairy goat. The virus shows compartmentalization features, that allows it to hide at certain times during the course of the disease, making it difficult to control. The present study was conducted to identify the major seminal plasma protein profile of goats infected by CAE and its associations with seroconversion using Western blotting. Two groups containing five males each, were used in this experiment. The first group was composed by seropositive animals and the control by seronegative confirmed by Western blotting and PCR. The semen was collected through artificial vagina and after that, two-dimensional electrophoresis and MALDI-TOF MS were used. Seventy-five spots were identified in the goat seminal plasma gels, equivalent to 13 different proteins with more expression. The similar proteins found in both groups and related to reproduction were spermadhesin Z13-like, bodhesin and bodhesin-2, Lipocalin, protein PDC-109-like, and albumin. In infected goats, proteases such as arisulfatase A have been identified, whose function probably is related to metabolism control of sulfatides, involved to virus control. The other ones were bifunctional ATP-dependent dihydroxyacetone kinase/FAD-AMP lyase, cathepsin F isoform X1, disintegrin and metalloproteinase domain-containing protein 2-like isoform X1, clusterin, carbonic anhydrase 2, electron transfer flavoprotein subunit beta, and epididymal secretory glutathione peroxidase. The results of this study show the reaction of the innate immune system against chronic infection of goats by CAE.  相似文献

Ariidae catfish species are numerically abundant along the coast of Brazil. Their benthic habits and broad spectrum diets make them good potential candidates for sub-lethal biomonitoring studies. In this study, we assess the levels of various metal contaminants (Pb, Cd, Hg, Cu and Zn) in the muscle tissues of two Ariidae species, Cathorops spixii and Genidens genidens, from three sites in S?o Paulo State, Southeast Brazil: two polluted sites in the Santos-S?o Vicente estuary and a relatively unpolluted site in the Cananéia estuary. The Zn levels observed in the polluted areas in the Santos-S?o Vicente estuary were similar to those obtained for Ariidae from the reference site in Cananéia. The concentrations of Hg and Cu in the muscle tissue of both fish species were higher in individuals from the Santos-S?o Vicente than those from the Cananéia estuary. Both Ariidae species were observed to accumulate the Cu, Zn and Hg in their tissues; however C. spixii showed a more stable response suggesting its potential utility as a bioindicator species. The Zn and Cu concentrations probably reflect normal levels without a significant influence of anthropogenic contamination. The levels of Cd and Pb in muscle tissue of C. spixii and G. genidens were relatively low, but the PCA indicated the presence of the levels of these metals in the reference area (thereby supporting the need for monitoring in Cananéia estuary). The detection of Hg in fish from Santos-S?o Vicente and from the unpolluted site (Cananéia) is of particular concern as scientists frequently use this area as a reference site for biomonitoring studies on the Southwest coast of Brazil.  相似文献

Corynebacterium pseudotuberculosis is the etiological agent of caseous lymphadenitis (CLA), a common disease in small ruminant populations throughout the world. Once established, this disease is difficult to eradicate because drug therapy is not effective and because the clinical detection of infected animals is of limited efficiency. We reviewed the microbiological, biochemical and taxonomic features of C. pseudotuberculosis, general aspects of infection, the main virulence determinants and currently available commercial vaccines. We also examined the current molecular strategies for the study of virulence in C. pseudotuberculosis, including the latest research on the identification of novel virulence factors and genes, which will help us to better understand the biology of this microorganism. This knowledge may also contribute to the development of improved CLA vaccines, including subunit and DNA-based types, as well as to improve the diagnosis, treatment and control of this disease.  相似文献

Metal-ion contamination (Cd, Cu, Pb, and Zn) on sunflower (Helianthus annuus L.) growth and total leaf protein expression were studied in the present work. The height, mass production, and metal distribution (Ca, K, Fe, Mg, Na, and P) in all plant fractions (roots, stems, and leaves) were evaluated. Sunflowers plants contaminated with four metal ions decreases height and mass by 35% and 40%, respectively, compared to control. Significant differences of total protein composition were noted after SDS-PAGE separation. Sunflower proteomics were more affected when 500 mg L(-1) of metal ion was added as contaminant of both zinc and mixed ions solution. In these cases, proteins having a molar mass of 14.5, 34.5, and 54.0 kDa were present at a lower level and alterations in enzymatic activities (SOD and GR) were found. Sunflowers plants contaminated with zinc and the mixed ions solution showed some degree of oxidative stress.  相似文献

The presence of carbonyl compounds (CCs) in wines has sparked the interest of researchers in several countries. The quantification of some of these compounds has been used as a parameter of quality for many fermented beverages. Although present in minute quantities (except for acetaldehyde), they have a strong olfactory impact. In addition, the CCs found in wines have a strong affinity for bisulfite and can form stable adducts, which will also interfere in the characteristics of aroma. The greatest challenge, however, is to predict which CCs have the strongest affinity for S(IV) and what conditions favor this interaction. To better understand the reaction of CC-bisulfite adduct formation (HASA), this study has evaluated the profile of 22 CCs in a "synthetic wine" containing bisulfite and in 10 real samples of different wines from the S?o Francisco Valley, northeastern Brazil. On the basis of principal component analysis (PCA) and dissociation constants, the results revealed that aliphatic aldehydes form adducts with S(IV), whereas ketones, cyclic aldehydes, and trans-alkenes interact weakly and are found predominantly in their free form. These results revealed also that pH 10 and 11 were defined as the most appropriate for CC-SO 2 adduct dissociation, and the total CCs were quantified reliably.  相似文献

The seroprevalence of caprine arthritis–encephalitis virus (CAEV) in the Cariri Region of Paraíba State, Brazil, was determined in 60 goat herds using the agar gel immunodiffusion test. The overall seroprevalence was 8.2%, with seropositivity in 21/60 (35%) herds and 13/15 (86.6%) municipalities. Bucks had a significantly higher frequency of infection (28.3%) than does (5.9%), and bucks that originated in other states had a significantly higher frequency of infection (76.5%) than those from Paraíba State (9.3%).  相似文献
