The incidence of disease caused by Pasteurella sp. in farmed lumpsuckers in Norway has been steadily increasing in recent years, causing significant economic losses and fish welfare issues. The disease affects all life stages, both in hatcheries and after release into salmon cages. Therefore, it is important to establish robust challenge models, to be used for vaccine development. Exposure experiments via intramuscular and intraperitoneal injection underlined the high virulence of the bacteria, whereas the cohabitation and bath models allowed the chronic symptoms of the disease to be studied more accurately. Skin lesions and haemorrhage at the base of fins were observed in the more acute cases of the disease. Symptoms including white spots over the skin, especially around the eyes, characterized the chronic cases. The latter were most prominent from the bath challenge model. Histopathology indicated a systemic pattern of disease, whereas qPCR analysis from head kidney showed that bacteria may be present in survivor fish at the end of the challenges. In all the challenge models investigated, Pasteurella sp. was re‐isolated from the fish, thus fulfilling Koch's postulates. These findings highlight the importance of screening of lumpsuckers prior to transfer to minimize the risks of carrying over asymptomatic carriers.  相似文献

A mesocosm experiment was established to evaluate the effect of two organic wastes: fermented sugar beet residue (SBR) and urban waste compost on the stimulation of plant growth, phytoaccumulation of heavy metals (HM) and soil biological quality and their possible use in phytostabilization tasks with native (Piptatherum miliaceum, Retama sphaerocarpa, Bituminaria bituminosa, Coronilla juncea and Anthyllis cytisoides) and non-native (Lolium perenne) plants in a heavy metal-contaminated semiarid soil. Except R. sphaerocarpa, SBR increased the contents of shoot N, P and K and shoot biomass of all plants. The percentage of mycorrhizal colonization was not affected by the organic amendments. The highest increase in dehydrogenase and β-glucosidase activities was recorded in SBR-amended P. miliaceum. SBR decreased toxic levels of HM in shoot of P. miliaceum, mainly decreasing Fe and Pb uptake to plants. This study pointed out that the SBR was the most effective amendment for enhancing the plant performance and for improving soil quality. The combination of SBR and P. miliaceum can be regarded the most effective strategy for being employed in phytostabilization projects of this contaminated site.  相似文献

β-Asarone content in Acorus calamus is a paramount issue because it limits the usage of plant for medicinal purpose. In the present study A. calamus L. accessions based on RAPD marker, ploidy level and β-asarone content were characterized and correlated on the basis of β-asarone content/ploidy level. Of the 40 random primers used, 6 primers generated polymorphism. Genetic relatedness among accessions evaluated by a similarity matrix based on Dice's coefficient ranged from 0.72 to 0.97. A phenetic dendrogram based on UPGMA analysis grouped accessions into two clusters. A. calamus L. accessions were found to be triploid and tetraploid and their β-asarone content was found in two ranges 6.92–8.0% and 73–88%. The study clustered the accessions as per their ploidy level, β-asarone content and geographical locations. This study would have extensive application in quality control of raw materials.  相似文献

A field experiment was performed in eastern Kenya to estimate N2 fixation by Sesbania sesban over an 18-month period using the 15N dilution method. The influence of three reference species, Senna spectabilis, Eucalyptus saligna and Grevillea robusta, on the estimates of N2 fixation was also assessed. Percentage Ndfa (nitrogen derived from the atmosphere) was calculated based on foliar atom excess (FAE), above-ground atom excess (AAE) or whole tree atom excess (WAE) data. The differences in atom% 15N excess values between species and plant parts are presented and discussed. We recommend the use of several reference species for estimating %Ndfa and that the different results obtained should be carefully considered in relation to the issues being addressed. In this study, Senna was the most suitable of the three reference species because its N uptake pattern and phenology were very similar to those of Sesbania. When well established, the amount of N fixed by Sesbania accounts for more than 80% of its total N content, according to FAE-based estimates. We estimated the Ndfa by Sesbania after 18 months to between 500 and 600 kg ha−1 , depending on whether FAE, AAE or WAE data were used and on the choice of reference species. The substantial accumulation of N in planted Sesbania highlighted its potential to increase the sustainability of crop production on N-limited soils. We consider the 15N dilution method to be appropriate for quantifying N2 fixation in improved fallows in studies, similar to this one, of young trees with high N2-fixing ability.  相似文献

Some reports have shown that for single species the correlation between modulus of elasticity (MOE) and modulus of rupture (MOR) in bending is quite high. Tropical timbers consist of hundreds of species that are difficult to identify. This report deals with the mechanical stress grading of tropical timber regardless of species. Nine timber species or groups of species with a total number of 1094 pieces measuring 60 × 120 × 3000 mm, were tested in static bending. The MOE was measured flat wise, while MOR was tested edge wise. Statistical analysis of linear regression with a dummy model and analysis of covariance were used to analyze the role of MOE and the effect of species on prediction of MOR. The analysis showed that using MOE as a single predictor caused under/overestimation for one or more species and/or groups of species. The accuracy of prediction would be increased with species identification. An allowable stress and reference resistance for species and/or groups of species were provided to compare with the prediction of strength through timber grading. The timber strength class for species and/or groups of species was also established to support the application of mechanical timber grading.  相似文献

Flavonoids are dietary components and the most ubiquitous phenolic compounds found in nature, showing a range of pharmacological activities including antiviral action. This study describes the antiviral screening of 60 different flavones and flavonols against human rotavirus (Wa-1 strain) as well as their cytotoxicity in MA104 cells. Cytotoxicity was investigated by cell morphology assessment and antirotavirus activity by cytopathic effect inhibition. Results were expressed as CC50 and IC50, respectively, in order to calculate the selectivity index (SI = CC50/IC50) of each compound. Pacific Island communities are experiencing significant societal changes as a result of rural urban, inter‐island migration and migration to New Zealand. These relocations have significant implications for children's relationship to place. However, virtually nothing is known about children's sense of place in Samoa or places and activities that are important to them. This exploratory study worked with eight children aged 5–12 years growing up in an urban village in Samoa drawing on ‘Talanoa’ as research methodology and method by employing photo‐elicitation. The study provides a first snapshot of Samoan children's sense of place growing up in an urban village.  相似文献

Structured lipids (SLs) containing palmitic and oleic acids were synthesized by transesterification of tripalmitin with either oleic acid or methyl oleate as acyl donor. This SL with palmitic acid at the sn-2 position and oleic acid at sn-1,3 positions is similar in structure to human milk fat triacylglycerol. LIP1, an isoform of Candida rugosa lipase (CRL), was used as biocatalyst. The effects of reaction temperature, substrate molar ratio, and time on incorporation of oleic acid were investigated. Reaction time and temperature were set at 6, 12, and 24 h, and 35, 45, and 55 degrees C, respectively. Substrate molar ratio was varied from 1:1 to 1:4. The highest incorporation of oleic acid (37.7%) was at 45 degrees C with methyl oleate as acyl donor. Oleic acid resulted in slightly lesser (26.3%) incorporation. Generally, higher percentage incorporation of oleic acid was observed with methyl oleate (transesterification) than with oleic acid (acidolysis). In both cases percentage incorporation increased with reaction time. Incorporation decreased with increase in temperature above 45 degrees C. Initially, oleic acid incorporation increased with increase in substrate molar ratio up to 1:3. LIP1 was also compared with Lipozyme RM IM as biocatalysts. The tested reaction parameters were selected on the basis of maximum incorporation of C18:1 obtained during optimization of LIP1 reaction conditions. Reaction temperature was maintained at 45, 55, and 65 degrees C. Lipozyme RM IM gave highest oleic acid incorporation (49.4%) at 65 degrees C with methyl oleate as acyl donor. Statistically significant (P < 0.05) differences were observed for both enzymes. SL prepared using Lipozyme RM IM may be more suitable for possible use in human milk fat substitutes.  相似文献

Crispness of bread is rapidly lost because of water migration inside the crumb towards the crust. How crumb properties determine this process independent of crust properties has not been examined before. Therefore, the aim of this study was to analyze and explain the influence of crumb morphology on the overall crispness retention. Crispness retention was determined by analyzing the acoustic emission of breads differing in either crust or crumb morphology. When crumb morphology is coarse with a lower number of large connections between the air cells, the effective diffusion coefficient is reduced. This effective diffusion coefficient of crumb, which equals approximately half the value of air, was estimated using X-ray micro-computer tomography images of crumb pieces. If the crumb has a lower effective diffusion coefficient, bread with similar crust properties has significantly longer crispness retention. Despite this, our data show that variations in properties of crust, which has 30 times higher permeability than crumb, have a larger impact on crispness retention than variations in properties of crumb.  相似文献
