The effect of a novel consensus bacterial 6-phytase variant (PhyG) on apparent ileal digestibility (AID) of amino acids (AA) and phosphorus (P) utilization in young broilers when added to diets with high phytate-P (PP) content without added inorganic phosphate (Pi) and deficient in digestible (dig) AA and metabolizable energy (ME) was investigated. A total of 256 Ross 308 male broilers were assigned to 4 treatments (8 birds/cage, 8 cages/treatment) in a completely randomized design. Treatments comprised a positive control (PC, 2,975 kcal/kg ME, 3.7 g/kg dig P, 2.83 g/kg PP, 8.4 g/kg Ca, 10.6 g/kg dig lysine), a negative control (NC) without added Pi (ME −68 kcal/kg, crude protein −10 g/kg, dig AA −0.1 to −0.4 g/kg, Ca −2.0 g/kg, dig P −2.2 g/kg, Na −0.4 g/kg vs. PC), and NC plus 500 or 1,000 FTU/kg of PhyG. Test diets were corn/soy/rapeseed-meal/rice-bran-based and fed from 5 to 15 d of age. Ileal digesta and tibias were collected on day 15. Excreta was collected during days 12 to 15 to determine P retention. The NC (vs. PC) reduced (P < 0.05) P retention (−10.4% units), tibia ash (−14.3% units), weight gain (−109 g), feed intake (−82 g) and increased FCR (from 1.199 to 1.504), confirming that the NC was extremely deficient in nutrients and energy. Phytase addition to the NC linearly (P < 0.001) improved performance, but did not fully recover it to the level of the PC due to the severe nutrients/energy reduction in NC. Phytase linearly increased P retention (P < 0.001), tibia ash (P < 0.001), AID of dry matter (P < 0.05), nitrogen (P < 0.01), gross energy (P < 0.05), and all 17 individual AA (P < 0.01). At 1,000 FTU/kg, phytase increased (P < 0.05) P retention vs. PC and NC (+14.5 and +24.9% units, respectively) and increased tibia ash vs. NC (+13.8% units), equivalent to PC. The NC decreased AID of Cys, Gly, Thr, and Met vs. PC (P < 0.05). At 1,000 FTU/kg, phytase increased AID of all 17 AA vs. NC (P < 0.01), equivalent to PC. At 1,000 FTU/kg, AID AA responses (above NC) ranged from +4.5% (Met) to +15.0% (Cys), being maximal for essential Thr (+10.4%) and Val (+8.2%) and non-essential Cys (+15.0%) and Gly (+10.4%). The results highlight the efficacy of PhyG at a dose level of 500 to 1,000 FTU/kg in young broilers for improving the ileal digestibility of nitrogen, AA, and energy alongside P retention and tibia ash. The performance data emphasize the need to consider digestible nutrient intake as a response variable in exogenous enzyme studies.  相似文献

This study reports the development of a loop-mediated isothermal amplification procedure (LAMP) for polymerase chain reaction (PCR)-based detection of 'Candidatus Liberibacter solanacearum', the bacterial causal agent of potato zebra chip (ZC) disease. The 16S rDNA gene of 'Ca. Liberibacter solanacearum' was used to design a set of six primers for LAMP PCR detection of the bacterial pathogen in potato plants and the psyllid vector. The advantage of the LAMP method is that it does not require a thermocycler for amplification or agarose gel electrophoresis for resolution. Positive LAMP results can be visualized directly as a precipitate. The LAMP strategy reported here reliably detected 'Ca. Liberibacter solanacearum' and the closely related species 'Ca. Liberibacter asiaticus', the causative agent of huanglongbing disease of citrus, in plant DNA extracts. Although not as sensitive as quantitative real-time PCR, LAMP detection was equivalent to conventional PCR in tests of ZC-infected potato plants from the field. Thus, the LAMP method shows strong promise as a reliable, rapid, and cost-effective method of detecting 'Ca. Liberibacter' pathogens in psyllids and field-grown potato plants and tubers.  相似文献

Aspects of husbandry, productivity parameters, diseases, marketing and farmer attitudes in small-scale broiler farms in Botswana were studied using a questionnaire-based survey. The number of broilers per production cycle in small-scale operations ranged from 100 to 2000, with an average of 640. On average, small-scale producers had 4-5 production cycles per annum with an interval of 2-3 weeks between batches. The average marketing age was 48.3 days at a live weight of 1.79 kg and a carcase weight of 1.42 kg. The average feed conversion ratio was estimated to be 2.72 (kg feed/kg live weight) and the average mortality was 9.15%. Overall, the performance indices of broilers under small-scale farm conditions were much poorer than those obtained elsewhere under improved management conditions, but these lower performance levels are suggestive of the potential that exists for further improvement. The survey also identified several areas of concern that constrain the realization of the full potential of this sector. Factors contributing to the poor performance under small-scale farm conditions are complex, but feed supply and quality, chick quality and farmer training appear to be areas for immediate improvement.  相似文献

ABSTRACT

1. The influence of barley inclusion level and supplementation of a multi-component non-starch polysaccharide degrading enzyme on performance and nutrient utilisation in broilers was investigated. Normal-starch hulled barley was evaluated with five levels of inclusion (0, 141, 283, 424 and 565 g/kg) in a wheat-based diet and two levels of enzyme supplementation (0 and 150 g/tonne of feed; a 5 × 2 factorial arrangement of 10 dietary treatments). All diets were equivalent in metabolisable energy and digestible amino acid contents. A total of 400, one-d old male broilers (five cages/treatment; eight birds/cage) were used in the experiment.

2. Regardless of enzyme supplementation, weight gain (WG) increased up to 283 g/kg of barley and was reduced afterwards (P < 0.01). Increasing levels of barley resulted in greater (P < 0.001) gain per feed (G/F). Enzyme addition increased WG (P < 0.05) and G/F (P < 0.001) at each barley inclusion level.

3. Birds fed diets with 0 and 565 g/kg barley showed the lowest and highest (P < 0.001to 0.05) digestibility for all nutrients measured, respectively. Digestibility of all nutrients was improved by enzyme supplementation at each barley inclusion level (P < 0.05). The nitrogen-corrected apparent metabolisable energy improved with increasing inclusion of barley (P < 0.001) and supplemental enzyme (P < 0.01). Increasing inclusion of barley increased the relative weight of gizzard (P < 0.001) and reduced jejunal digesta viscosity (P < 0.001). Supplemental enzyme (P < 0.001) reduced digesta viscosity.

4. The optimum inclusion level of barley, with respect to growth performance, was 283 g/kg of diet. Increasing barley inclusion improved nutrient and energy utilisation, possibly through lowered digesta viscosity and better function of the gizzard. Feed efficiency and nutrient and energy utilisation can benefit from carbohydrase supplementation in barley-based diets, regardless of barley inclusion level.  相似文献

The nutrient and antinutrient components of tubers from seven cultivars ofDioscorea alata were determined. The average crude protein content ofD. alata tubers was 7.4%. Starch (75.6–84.3%) was the predominant fraction of the tuber dry matter. Significant differences in crude protein and starch contents were observed among cultivars. Vitamin C content of the yam tubers ranged from 13.0 to 24.7 mg/100 g fresh weight. The results showed yams to be reasonably good sources of minerals. Phytic acid contents of the yams were low, with values ranging from 58.6 to 198.0 mg/100 g dry matter. Total oxalate levels in yam tubers were found to be in the range of 486–781 mg/100 g dry matter, but may not constitute a nutritional concern since 50–75% of the oxalates were in the water-soluble form. The overall results are suggestive of the nutritional superiority of yams compared to other tropical root crops.  相似文献

A method is described for determining heats of heterolysis, DeltaH(het), from the heats of reaction of resonancestabilized carbenium ions with carbanions in solution. These results can be converted into heats of homolysis, DeltaH(homo), for the same bonds by combining DeltaH(het) values with electron transfer energies, deltaG(ET) (deltaG(ET) approximately deltaH(ET)), obtained from redox potentials of the reacting ions. The results provide a comprehensive tabulation of DeltaH(het), DeltaH(homo), and DeltaG(ET) values for an extended series of organic compounds and are examined in terms of their interrelationships and a number of related properties of the ions and radicals.  相似文献

Small cardamom (Elettaria cardamomum Maton) is extensively cultivated in the Western Ghats of South India either as a monocrop under the forest trees or as an intercrop along with arecanut and coffee plantations. Colletotrichum species responsible for severe outbreaks of anthracnose on small cardamom in South India are reported. Small cardamom anthracnose, popularly known as “Chenthal”, manifests itself on the foliage as yellowish lesions, which later coalesce to form large blighted areas. In advanced stages, the affected leaves dry up giving a burnt appearance to the plant. Twenty-five isolates of Colletotrichum were isolated from leaves of small cardamom in Karnataka, Kerala and Tamil Nadu states of India. The isolates were characterized through morphological studies and multilocus phylogenetic analysis (ITS, ACT, CHS-1, GAPDH, TUB2, CYLH3, GS and ApMat gene regions) to test whether different species are present and identified: C. karstii (2 isolates), C. gloeosporioides (1), C. siamense (7), C. syzygicola (6), Colletotrichum sp (5), and C. guajavae (4), as the cause of anthracnose on small cardamom for the first time. Pathogenicity of the six species was confirmed. To our knowledge, this is the first detailed study of Colletotrichum species which cause anthracnose diseases on small cardamom.  相似文献

Asiatic cotton (Gossypium arboreum) is mostly grown in the rainfed regions of India. However, little is known about the effects of nutrient‐management practices on plant growth and fruiting pattern of Asiatic cotton. Therefore, plant growth and fruiting pattern under four nutrient‐management treatments, N, NPK, FYM (10 Mg ha–1), and INM (integrated nutrient management: a combination of NPK and FYM) were quantified during 2000–01 to 2002–03 (years 16 to 18 of a long‐term field experiment). Plants of the INM and FYM treatments were taller (68.4–149.5 cm) and had more main stem nodes per plant (30.5–44.5) as compared to N and NPK treatments. In treatment N, the shortest plants (50.9–83.6 cm) and the least number of fruiting structures were produced. Plants of the INM and FYM treatments accumulated more squares and bolls. Maximum boll production was 10–19 days earlier with the manure‐amended than the N and NPK treatments. Treatment N had the lowest seed cotton yield (639–790 kg ha–1), because of small boll size (1.48–1.73 g) and few open bolls. Seed cotton yield followed the trend: NPK (815–1278 kg ha–1) < INM (776–1551 kg ha–1) < FYM (902–1593 kg ha–1). Water stress and nutrient deficiencies (P and Zn in the N and Zn in the NPK treatments) as a consequence of nutrient depletion over the years may have decreased seed cotton yields in treatments that received mineral fertilizer alone in comparison with manure‐amended treatments. On a long‐term basis, FYM application should therefore form an integral part of nutrient recommendation.  相似文献

Accurate knowledge of the actual nutritional value of individual feed ingredients and complete diets is critical for efficient and sustainable animal production. For this reason, feed evaluation has always been in the forefront of nutritional research. Feed evaluation for poultry involves several approaches that include chemical analysis, table values, prediction equations, near-infrared reflectance spectroscopy, in vivo data and in vitro digestion techniques. Among these, the use of animals (in vivo) is the most valuable to gain information on nutrient utilization and is more predictive of bird performance. However, in vivo methods are expensive, laborious and time-consuming. It is therefore important to establish in vitro methods that are reliable, rapid and practical to assess the nutritional quality of feed ingredients or complete diets. Accuracy of the technique is crucial, as poor prediction will have a negative impact on bird performance and, increase feed cost and environmental issues. In this review, the relevance and importance of feed evaluation in poultry nutrition will be highlighted and the various approaches to evaluate the feed value of feed ingredients or complete diets will be discussed. Trends in and practical limitations encountered in feed evaluation science, with emphasis on in vitro digestion techniques, will be discussed.  相似文献
