Almond (Prunus dulcis) is one of the well known stone fruit species grown for its unripe fruits and delicious seeds in Turkey. In the Trakya region, however, some prevailing virus infections have reduced almond yields and quality. In ten districts of Trakya, 260 leaf samples were collected from affected almond trees in June 2010. DAS–ELISA assays and RT-PCR tests were employed for the identification of viruses. As a result of these detection studies, five of the 260 leaf samples gathered from symptomatic almond trees had Plum pox virus (PPV), 81 of them had Prunus necrotic ringspot virus (PNRSV), and 11 samples contained Prune dwarf virus (PDV). Only four out of 260 samples had a mixture of these viruses. Partial nucleotide sequences of five almond isolates of PPV were determined and compared with 17 other PPV isolates in databases. Computer analysis of obtained and published nucleotide sequences showed identity ranged from 75.72% to 96.87%. Of the five PPV almond isolates obtained, however, there was a close nucleotide identity of 95.82–96.61% to Turkish isolates. Phylogenetic analysis of nucleotides and amino acids showed that five PPV isolates of almond from the Trakya Region of Turkey were clustered in the same subgroup with PPV-T Turkish isolates in GenBank. Therefore we can consider almond isolates of PPV as PPV-T strain, like the two other isolates from apricot trees in Turkey.  相似文献

ABSTRACT

The diffusion properties of sodium chloride in rainbow trout muscle during dry salting at 10°C were investigated. The average initial dry matter content of the samples increased from 27.80 ± 0.10 to 36.01 ± 0.16%, and the average salt content increased to 17.98 ± 0.38 g NaCl/100 g dry matter at the end of the 10-h dry salting process. The analytical solution of Fick’s second law considering one-dimensional diffusion through an infinite slab was used to calculate effective salt diffusion coefficients (Deff). The Deff values in rainbow trout fillets decreased with increasing salting time and ranged from 6.64 × 10?10 to 16.45 × 10?10 m2/s.  相似文献

Genetic transformation is a useful technique to complement conventional breeding in crop improvement. Although carrot has been a model organism for in vitro embryogenesis study, genetic transformation of carrot is still lengthy and labor intensive. An efficient transformation and detection system is desirable. Direct infection of Agrobacterium to carrot calli has provided an easy way for carrot genetic transformation. To improve the efficiency of antibiotic selection in this method, we report the combined use of an improved green-fluorescent protein, referred to as smGFP, to establish a versatile selection method for carrot callus transformation system. By combining antibiotic selection with the bright fluorescence observed in the callus tissue, we were able to easily identify stable transformants in early stage of the transformation process. In addition to the GFP expression of the callus cells, the transgenic nature of callus cells was confirmed with Southern and Western analysis. We found we can link the simplicity of carrot-callus-cell transformation, early detection of stable transformants with antibiotic selection, visualization of GFP fluorescence, and molecular analysis (Southern and Western) of callus tissue (non-photosynthetic tissue) to provide a more efficient way in identifying stable transformants at early stage of carrot transformation.  相似文献

The effects of plant growth promoting bacteria (PGPB) on the fruit yield, growth and nutrient element content of strawberry cv. Fern were investigated under organic growing conditions between 2006 and 2008. The experimental plot was a completely randomized design with 3 replicates. Three PGPB strains (Pseudomonas BA-8, Bacillus OSU-142 and Bacillus M-3) were used alone or in combination as bio-fertilizer agent in the experiment. Data through 3 years showed that the use of PGPB significantly increased fruit yield, plant growth and leaf P and Zn contents. Root inoculation of M3 and floral and foliar spraying of OSU-142 and BA-8 bacteria stimulated plant growth resulting in significant yield increases. M3 + BA-8, BA-8 + OSU-142, M3, M3 + OSU-142 and BA-8 applications increased cumulative yield by 33.2%, 18.4%, 18.2%, 15.3% and 10.5%, respectively. Number of fruits per plant significantly increased by the applications of M3 + BA-8 (91.73) and M3 (81.58) compared with the control (68.66). In addition, P and Zn contents of strawberry leaves with bacterial inoculation significantly increased under organic growing conditions. Available P contents in soil were increased from 0.35 kg P2O5/da at the beginning of the study to 2.00, 1.97 and 1.82 kg P2O5/da by M3 + OSU-142, M3 + BA-8 and M3 + BA-8 + OSU-142 applications, respectively. Overall, the results of this study suggest that root inoculation of Bacillus M3 alone or in combination with spraying Bacillus OSU-142 or Pseudomonas BA-8 have the potential to increase the yield, growth and nutrition content of strawberry plant under organic growing conditions.  相似文献

Forest structure changes continuously by natural and anthropogenic effects. Because the level of goods and services provided by forest ecosystems are related to this structure, some attributes have to be controlled while they are being managed. In this paper we describe the long-term temporal changes in land area and landscape metrics related to different land uses of a managed forest in Turkey. The study was carried out for the Daday Forest Planning Unit located in the west Black Sea region of northern Turkey. The total area is 16,813 ha and besides wood production, it is managed for erosion control, public health, aesthetics, and recreation. Stand type maps that were constructed in 1970, 1989, 1999, and 2010 were used in this analysis. Transition matrixes that illustrate area changes among cover types and temporal changes on some landscape metrics were obtained using Geographic Information Systems. Stands were separated into small patches, and thus the number of patches increased nearly two-fold between 1970 and 2010. The total forest edge increased and through the associated fragmentation, the amount of core forest area decreased at the landscape scale. Landscape metrics were applied to digitized versions of historical maps to assess how forest area changed. Human use of the land has changed, forest management practices have evolved, and these along with natural forest growth have contributed to interesting changes in landscape character.  相似文献

A feeding trial was conducted to investigate the effects of the replacement of dietary fish meal and fish oil with oilseed meals (soybean or canola) and canola oil on growth, nutrient utilization, body composition, diet digestibility and hematological parameters of rainbow trout Oncorhynchus mykiss. Seven diets were used; the control diet (designated FM) contained fish meal and fish oil as the main protein and lipid sources. For the experimental diets, 40% of fish meal protein was substituted with soybean meal, canola meal or a soybean/canola meal mixture, and these diets (designated SM, CM and SCM, respectively) contained fish oil as the lipid source. Three additional diets (SM?+?CO, CM?+?CO and SCM?+?CO) were formulated with the same vegetable protein meals but with fish oil replaced by canola oil. Fish were fed twice daily to apparent satiation for 11?weeks. The growth of fish fed the CM?+?CO diet was significantly lower than that of fish fed the FM, SCM, SM?+?CO and SCM?+?CO diets. The feed conversion ratio (FCR) was significantly better in fish fed the FM and SCM?+?CO diets than in fish fed the CM and CM?+?CO diets. Furthermore, feed intake was significantly lower for fish fed the CM?+?CO diet than in fish fed the SCM diet, and lipid digestibility of the CM?+?CO diet was significantly lower than that of all other diets. No significant differences of body composition were observed. Circulating leukocyte levels, leukocyte ratios and serum lysozyme activity remained unaffected by dietary treatment. However, it was observed that fish fed the CM?+?CO diet displayed hematocrit levels significantly lower (P?<?0.05) than that of fish fed the other diets. The results indicate that when diets contain either fish oil or canola oil, canola meal and soybean meal can be incorporated into rainbow trout feeds at a combined 32% inclusion level (replacing 40% of fish meal protein) without inducing significant negative effects on growth, nutrient utilization or health.  相似文献

Pretreatment with oil of sea turtle Caretta caretta protected tomato plants against bacterial canker caused by Clavibacter michiganensis subsp. michiganensis (Cmm). The turtle oil was ineffective in inhibiting Cmm in an agar diffusion test, suggesting a mechanism of induced resistance. Under controlled conditions in the greenhouse, turtle oil lowered the disease index and had reduced the growth of bacteria up to 50.4% by 7 days after inoculation. Applying turtle oil to the foliage of tomato plants increased per-oxidase and lipoxygenase activities. Gas chromatography analyses of turtle oil indicated the presence of polyunsaturated fatty acids such as eicosapentaenoic acid and docosahexaenoic acid. These fatty acids may lead to the higher activity of the enzymes and probably induced plant resistance against the pathogen.  相似文献

: Five dogs, four small mixed breed and a Doberman Pinscher, presented in our clinic with hemivertebra. Complete physical, radiological and neurological examinations were done and the spinal deformities were characterized in accord with the Nasca classification used in human medicine. Two dogs had multiple hemivertebrae (round, oval or wedge-shaped: Type 3) in the thoracic region; one dog had an individual surplus half vertebral body (Type 1) plus a wedge-shaped hemivertebra (Type 2b) in the lumbar region; one dog had multiple hemivertebrae which were fused on one side (Type 4a) in the thoracic region; and one dog had a wedge-shaped hemivertebra (Type 2a) in the cervical region.  相似文献

From 2002 to 2006, plant growth promoting effects of Bacillus M3, Bacillus OSU-142 and Microbacterium FS01 were tested alone or in combination on apple (Malus domestica L.) cv. Granny Smith in terms of yield, growth and nutrient composition of leaves in the province of Malatya, Turkey. The presence of M3 and/or OSU-142 and/or FS01 combinations stimulated plant growth and resulted in significant yield increases in Granny Smith. Root inoculation of PGPR strains significantly increased cumulative yield (26.0–88.0%), fruit weight (13.9–25.5%), shoot length (16.4–29.6%) and shoot diameter (15.9–18.4%) compared with the control. In addition, all nutrient element contents (N, P, K, Mg, Ca, Fe, Mn and Zn) investigated in Granny Smith, except Mg was significantly affected by bacterial applications compared with the control. The highest N content (3.38%) was obtained from OSU-142 + FS01 application. Phosphorus contents of leaves of Granny Smith increased from 0.29% in the control to 0.42, 0.37, 0.36, 0.36 and 0.35% by M3, M3 + FS01, M3 + OSU-142, OSU-142 and M3 + OSU-142 + FS01 application, respectively. All bacterial applications significantly increased Ca content of leaves. The highest K, Fe, Mn, Cu and Zn content of leaves were obtained from OSU-142, M3, FS01, M3 and M3 + FS01 applications, respectively. The results of this study suggest that Bacillus M3 and/or OSU-142 and/or Microbacterium FS01 in combination have the potential to increase the yield, growth and nutrition of apple trees.  相似文献
