The removal of nitrate reductase phosphorylation enhances tolerance to ammonium nitrogen deficiency in rice

Nitrate reductase (NR) is a key enzyme for nitrogen assimilation in plants, and its activity is regulated by posttranslational phosphorylation. To investigate the effects of dephosphorylation of the NIA1 protein on the growth and the physiological and biochemical characteristics of rice under different forms of nitrogen supplies, the phenotypes, nitrogen metabolism and reactive oxygen metabolism were measured in NIA1 phosphorylation site-directed mutant lines (S532D and S532A), an OsNia1 over-expression line (OE) and Kitaake (wild type, WT). Compared with WT and OE, S532D and S532A have stronger nitrogen assimilation capacities. When ammonium nitrate served as the nitrogen source, the plant heights, dry weights of shoots and chlorophyll (Chl) contents of S532D and S532A were lower than those of the WT and OE, whereas hydrogen peroxide (H2O2), malondialdehyde (MDA) and nitrite contents were higher. When potassium nitrate served as the nitrogen source, the plant heights, dry weights of shoots and Chl contents of S532D and S532A were higher than those of the WT and OE, there were no significant differences in the contents of H2O2 and MDA in the leaves of the test materials, and the difference in nitrite contents among different lines decreased. When ammonium sulfate served as the nitrogen source, there were no significant differences in the physiological indexes of the test materials, except NR activity. Compared with ammonium nitrate and ammonium sulfate, the content of NH4+-N in the leaves of each plant was lower when potassium nitrate was used as the nitrogen source. The qPCR results showed that OsGS and OsNGS1 were negatively regulated by downstream metabolites, and OsNrt2.2 was induced by nitrate. In summary, when ammonium nitrate served as the nitrogen source, the weak growth of NIA1 phosphorylation site-directed mutant lines was due to the toxicity caused by the excessive accumulation of nitrite. When potassium nitrate served as the nitrogen source, the assimilation rates of nitrate, nitrite and ammonium salt were accelerated in NIA1 phosphorylation site-directed mutant lines, which could provide more nitrogen nutrition and improve the tolerance of rice to ammonium nitrogen deficiency. These results could provide a possible method to improve the efficiency of nitrogen utilization in rice under low-nitrogen conditions. 相似文献

We report a phenomenal increase in strength, modulus, and fracture strain of ultra high molecular weight polyethylene (UHMWPE) fiber by 103 %, 219 %, and 108 %, respectively through hybridizing this fiber with Nylon 6 as a minor phase and simultaneously reinforcing it with single-walled carbon nanotubes (SWCNTs). Loading of Nylon 6 and SWCNTs into UHMWPE was 20.0 wt% and 2.0 wt%, respectively. Hybridized fibers were processed using a solution spinning method coupled with melt mixing and extrusion. We claim that the enhancement in strain-to-failure of the nanocomposites is due to induced plasticity in the hybridized Nylon 6-UHMWPE polymers. The enhancement in strength and stiffness in the nanocomposites is attributed to the load sharing of the SWCNTs during deformation. Differential scanning calorimetry (DSC), X-ray diffraction (XRD), scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR) studies showed that changes in percent crystallinity, rate of crystallization, crystallite size, alignment of nanotubes, sliding of polymer interfaces and strong adhesion of CNT/polymer blends were responsible for such enhancements. 相似文献

Crop disease has a negative impact on food security. If diverse crop diseases are not identified in time, they can spread and influence the quality, quantity, and production of grain. Severe crop diseases can even result in complete failure of the harvest. Recent developments in deep learning, particularly convolutional neural networks (CNNs), have exhibited impressive performance in both image recognition and classification. In this study, we propose a novel network architecture, namely Mobile-DANet, to identify maize crop diseases. Based on DenseNet, we retained the structure of the transition layers and used the depthwise separable convolution in dense blocks instead of the traditional convolution layers, and then embedded the attention module to learn the importance of interchannel relationship and spatial points for input features. In addition, transfer learning was used in model training. By this means, we improved the accuracy of the model while saving more computational power than deep CNNs. This model achieved an average accuracy of 98.50% on the open maize data set, and even with complicated backdrop conditions, Mobile-DANet realized an average accuracy of 95.86% for identifying maize crop diseases on a local data set. During their ontogeny, a range of fruits develop a semi-transparent white coating on their surface, designated as wax bloom. This wax bloom was examined non-destructively on plums of different colour i.e. blue mature European plum, yellow mirabelle and greengage using a colourimeter and an industrial luster sensor at 15 mm distance from the fruit surface; the results are displayed as ‘luster levels’. European plums contained 175–205 µg epicuticular wax cm?2 fruit surface with linoleic and ursolic acid as predominant fatty acids. The microscopic examination identified the epicuticular fine structure of the fruit surface as platelets viz with natural wax. Polishing with a soft tissue disturbed this fine structure and doubled the luster levels relative to the natural surface from 120–170 to 300–500 arbitrary units. This is related to wax re-distribution and restructuring rather than loss of epicuticular wax. Polishing increased the a value marginally from 2.35 to 2.85, whereas the b values almost tripled from ? 14 to ? 5, representing a more intense blue impression after polishing. This luster or glossiness measurement enables a clear identification of the natural wax coating versus a polished fruit surface irrespective of the respective fruit colour, i.e. blue plum, yellow mirabelle or greengage and hence seems more suitable than colour measurement. It hence seems plausible that fruit with a similar natural wax bloom like grape berry or blueberry etc. might respond similarly and could be used as a hand-hand device or on a sorting line for fast, online and affordable detection of the glossiness of a commodity. 相似文献

We evaluated radio frequency (RF) dielectric heating for eradication of pinewood nematodes (PWN) in infested wood. Thirteen temperatures were tested (from ambient to 70 °C) on small wood samples (2.5 × 3.8 × 0.64 cm) to determine the minimum lethal temperature (100 % mortality), which was 56 °C [based on infra-red (IR) thermal images data 55.5–57.4 °C] with a 1 min hold time. We also used thermal probes inside the wood to confirm that temperatures were ≥56 °C. Thirty additional samples were tested bracketing the minimum lethal temperature using 54, 56 and 58 °C with additional replications to produce the minimum sample size equivalent of 100 % mortality of at least 93,616 nematodes to satisfy the Probit 9 efficacy requirement. This minimum lethal temperature was further verified by treating infested large wood blocks (10.2 × 10.2 × 25.4 cm). All samples that met or exceeded the 56 °C lethal temperature for the required 1 min hold time (as measured by probes inserted in the wood and on the wood surface by IR) produced 100 % mortality. The sample size required to show Probit 9 efficacy was also satisfied. This study supports the consideration of RF in addition to microwave (MW) dielectric heating as alternative treatments of wood packaging material for inclusion in ISPM No. 15, provided the treatment delivers the target lethal temperature throughout the profile of the material in industrial scale operations. 相似文献

1. We examined the effects of supplemental dietary vitamin E (Vit E) on semen quality and antioxidative status in male domestic fowls exposed to oxidative stress induced by synthetic glucocorticoid, dexamethasone (DEX) injection. 2. Thirty-six Egyptian local cross males, 42 weeks old, were housed individually in cages in an open-sided building under 16 h light:8 h dark and were provided with commercial feed and water ad libitum. Birds were divided into 4 groups: DEX (4 mg/bird/d), Vit E (200 mg/kg diet), DEX + Vit E (4 mg/bird/d + 200 mg/kg diet, respectively) and control, n = 9. All treatments lasted for 7 continuous days. 3. Oxidative stress induced by injection of DEX (4 mg/bird/d) resulted in decreased sperm count and motility correlated with an increased percentage of dead sperms. Vit E (200 mg/kg diet) enhanced sperm count and viability when supplemented to stress-induced birds, compared to DEX treatment alone. 4. In seminal plasma, low calcium concentration, high lipid peroxidation and reduced activity of glutathione peroxidase were associated with the oxidative stress. Vit E reduced lipid peroxidation in the seminal plasma. 5. In conclusion, excessive supplemental dietary Vit E improved semen quality when cockerels were subjected to stress conditions. It increased both sperm count and motility, reduced the percentage of dead sperm and enhanced the antioxidative status of seminal plasma. 相似文献

Farmers in Pakistan continue to produce maize under various types of risks and adopt several strategies to manage those risks. This study is the first attempt to investigate the factors affecting the concurrent adoption of off-farm income diversification and agricultural credit which the farmers use to manage the risk to maize production. We apply bivariate and multinomial probit approaches to the primary data collected from four districts of Punjab Province in Pakistan. The results show that strong correlations exist between the off-farm diversification and agricultural credit which indicates that the use of one risk management strategy leads to another. The findings demonstrate that education, livestock number, maize farming experience, perceptions of biological risks and risk-averse nature of the growers significantly encourage the adoption of diversification as a risk management tool while farm size inversely affects the adoption of diversification. Similarly, in the adoption equation of credit, maize farming experience, farm size, perceptions of price and biological risks and risk attitude of farmers significantly enhance the chances of adopting agricultural credit to manage farm risks. These findings are important for the relevant stakeholders who seek to offer carefully designed risk minimizing options to the maize farmers. 相似文献

Intensive selection in wheat (Triticum aestivum L.) breeding programs over the past 100 years has led to a genetic bottleneck in modern bread wheat. Novel allelic variation is needed to break the yield plateau, particularly in the face of climate change and rapidly evolving pests and pathogens. Landraces preserved in seed banks likely harbour valuable sources of untapped genetic diversity because they were cultivated for thousands of years under diverse eco-geographical conditions prior to modern breeding. We performed the first genetic characterisation of bread wheat accessions sourced from the N. I. Vavilov Institute of Plant Genetic Resources (VIR) in St Petersburg, Russia. A panel comprising 295 accessions, including landraces, breeding lines and cultivars was subject to single seed descent (SSD) and genotyped using the genotyping-by-sequencing Diversity Arrays Technology platform (DArT-seq); returning a total of 34,311 polymorphic markers (14,228 mapped and 20,083 unmapped). Cluster analysis identified two distinct groups; one comprising mostly breeding lines and cultivars, and the other comprising landraces. Diversity was benchmarked in comparison to a set of standards, which revealed a high degree of genetic similarity among breeding material from Australia and the International Maize and Wheat Improvement Center (CIMMYT). Further, 11,025 markers (1888 mapped and 9137 unmapped) were polymorphic in the diversity panel only, thus representing allelic diversity potentially not present in Australian or CIMMYT germplasm. Open-access to DArT-seq markers and seed for SSD lines will empower researchers, pre-breeders and breeders to rediscover genetic diversity in the VIR collection and accelerate utilisation of novel alleles to improve wheat. 相似文献
