This paper describes a novel principle for the separation of wheat flour into starch and gluten in a concentrated medium. The process is based on the use of simple shear flow in a cone-and-cone device. The separation takes place in two steps. Initially, local segregation of gluten and starch phases occurs, leading to formation of macroscopically visible gluten patches distributed throughout the dough. This local segregation can be understood by considering the dough as a visco-elastic matrix containing an inert filler (starch). Further shearing leads to aggregation of those patches and migration (large-scale separation) towards the apex of the cone. As a result, the wheat dough is separated into a protein-poor fraction, containing less than 4% protein, and a protein-rich fraction containing almost 50% protein on a dry weight basis. However, under the process conditions used, upon a very long shearing, a redistribution of the aggregated gluten structures in the starch phase was observed, demonstrating a processing limit for the separation performance. Compared to traditional processing, the separation process presented shows opportunities for producing high quality gluten accompanied with significant water savings. Considering the fact that simple shear flow in steady rate is less harmful to gluten quality, such a separation process could benefit gluten quality.  相似文献

The vaccine efficacy of a genetically engineered deletion mutant strain of pseudorabies virus, strain 783, was compared with that of the conventionally attenuated Bartha strain. Strain 783 has deletions in the genes coding for glycoprotein I and thymidine kinase. In experiment 1, which had a 3-month interval between vaccination and challenge exposure, strain 783 protected pigs significantly (P less than 0.05) better against virulent virus challenge exposure than did the Bartha strain. The growth of pigs vaccinated with strain 783 was not arrested, whereas that of pigs vaccinated with the Bartha strain was arrested for 7 days. Of 8 pigs given strain 783, 4 were fully protected against challenge exposure; none of the pigs given strain Bartha was fully protected. In experiment 2, which had a 3-week interval between vaccination and challenge exposure, the growth of pigs vaccinated with strain 783 was arrested for 3.5 days, whereas that of pigs vaccinated with the Bartha strain was arrested for 6 days. In experiment 3, pigs with moderate titer of maternal antibodies were vaccinated twice IM or once intranasally with either strain 783 or Bartha and were challenge-exposed 3 months after vaccination. Pigs given strain 783 twice IM were significantly (P less than 0.05) better protected than were the other pigs. They had growth arrest of only 6 days, compared with 9 days for pigs of other groups, and shed less virus after challenge exposure. Results of this study indicate that the vaccine based on the deletion mutant strain 783 is more efficacious than is the Bartha strain of pseudorabies virus.  相似文献

Seed tubers that are planted soon after their harvest give low yields because of dormancy and low growth vigor. In the research reported in this paper, we investigated the advancing effect of a haulm application of gibberellic acid (750 g GA/ha) 6 days before haulm pulling and its interaction with storage temperature regimes on the growth vigor of immaturely harvested seed tubers of three cultivars. The effects on tuber yield were also examined in one experiment. The storage regimes were: 18 C continuously, hot pre-treatments of different duration (different periods at 28 C and subsequently 18 C) and a cold pre-treatment (20 days at 2 C and subsequently 18 C). Both a foliar spray with GA and storage at 28 C enhanced physiological aging of the tubers and greatly advanced the growth vigor, without negative effects on the morphology of the plants. There was a clear interaction between the GA treatment and the storage temperature regime. At early planting, the effect of the treatments on tuber yield were small for Diamant (short dormancy), but strongly positive for Désirée and Draga (long dormancy).  相似文献

In wheat, the ability to store and remobilise large amounts of stem water soluble carbohydrates (WSC) to grain constitutes a desirable trait to incorporate into germplasm targeted to regions with frequent terminal drought. The main aim of this paper was to examine the relationships between WSC storage, grain number and grain weight across several environments. A small set of recombinant inbred lines (2–4) contrasting in stem WSC were grown in six field trials where water availability, sowing date and/or N level were manipulated, with line yields ranging from 400 to 850 g m−2 across experiments. Biomass, N and WSC concentration (WSCc, mg g−1 dry weight) and amount (WSCa, g m−2) were monitored. A resource-oriented area-based model [Fischer, R.A., 1984. Growth and yield of wheat. Lettuce, spinach and chicory are generally considered the main leafy vegetables, while a fourth group denoted by ‘minor leafy vegetables’ includes, amongst others, rocket salad, lamb’s lettuce, asparagus, artichoke and rhubarb. Except in the case of lettuce, central crop databases of leafy vegetables were lacking until recently. Here we report on the update of the international Lactuca database and the development of three new central crop databases for each of the other leafy vegetable crop groups. Requests for passport data of accessions available to the user community were addressed to all known European collection holders and to the main collection holders located outside Europe. Altogether, passport data of 17,530 accessions from a total of 129 collections were collected. The four separate databases were made available on line via a common entry page accessible at http://documents.plant.wur.nl/cgn/pgr/LVintro/. Based on a literature study, an analysis of the gene pool structure of the crops was performed and an inventory was made of the distribution areas of the species involved. The results of these surveys were related to the contents of the newly established databases in order to identify the main collection gaps. Priorities are presented for future germplasm acquisition aimed at improving the coverage of the crop gene pools in ex situ collections.  相似文献

A selective and fast method for the quantitative determination of 2,4,6-trichloroanisole (TCA) and 2,4,6-tribromoanisole (TBA) in wine was developed. Microextraction in packed syringe (MEPS) was optimized for the extraction and preconcentration of the analytes using extremely small volume samples (0.1-1 mL). For GC-EI-MS, the limit of detection (LOD) for red and white wine was in the range 0.17-0.49 microg L(-1) for TCA and TBA. In addition to GC-EI-MS both GC-NCI-MS and GC-HRMS were used to further improve both selectivity and sensitivity. The lowest LODs were achieved using GC-HRMS in the EI mode. In red and white wine samples the LODs were between 0.22-0.75 ng L(-1) for TCA and TBA. The reproducibility and linearity for the GC-HRMS method was good, with RSD-values of 4-10% for spiked red wine samples at 1 ng L(-1) and linearity with R (2) > 0.962 over a concentration range of 1 to 100 ng L(-1).  相似文献

Juvenile, 0.87 g Colossoma macropomum (Cuvier) were submitted to different ad libitum feeding regimes. Feeding frequency ranged from one to five meals per day and intervals between meals from 3 h to 24 h. Higher feeding frequencies resulted in higher feed uptake, a higher growth rate and a decreased feed utilization efficiency. Feed consumption was lower at 0700 h than at 1900 h. The amount of feed from different meals persisting in the digestive tract was determined with chromic-oxide-marked feed. This method revealed that with ad libitum feeding, about 21% of the feed remained uneaten. Feed losses did not differ significantly between feeding regimes. Feed accumulated in the digestive tract in the morning and early afternoon. In the late afternoon most feed rests disappeared from the digestive tract and feed consumption increased. It is suggested that feed uptake and growth of C. macropomum can be improved by extending the feeding period after 1900 h.  相似文献

Home gardens in southern Ethiopia are regarded as efficient farming systems, allowing interactions and synergies between crop, tree and livestock components. However, these age-old traditional home gardens are evolving rapidly in response to changes in both the socio-economic and biophysical environment. Altered cropping patterns, farm size and component interactions may affect the systems’ sustainability. Home gardens exhibit a huge diversity in farms and farming systems, which needs to be understood in order to design interventions for improvement. Dynamics of home gardens were studied over two-decades (1991–2013) based on a survey of 240 farm households and focus group discussions. Farms were grouped into five types: Khat-based, Enset-cereal-vegetable, Enset-based, Enset-coffee and Enset-livestock. Farm trajectories revealed a shift from food-oriented Enset-based and Enset-livestock systems to (1) cash crop oriented khat-based systems, and (2) combined food and cash crop oriented Enset-cereal-vegetable systems. In densely populated, market proximate areas a major trend was expansion of khat, from 6 to 35% of the area share per farm, while the combined area share of enset and coffee decreased from 45 to 25%. Concurrently, the cattle herd size fell from 5.8 TLU to 3.9 TLU per household. In medium populated, less accessible areas the trend was consolidation of combined production of food and cash crops. Enset and coffee together maintained a share of over 45%. Easy transport and marketing of the perishable cash-generating khat compared with traditional crops favoured its cultivation among smallholders located close to markets. The insights in home garden change in response to increasing population pressure, decreasing farm size and market development may help to design interventions to increase system sustainability.  相似文献

Bark beetles are notorious pests of natural and planted forests causing extensive damage. These insects depend on dead or weakened trees but can switch to healthy trees during an outbreak as mass-attacks allow the beetle to overwhelm tree defences. Climatic events like windstorms are known to favour bark beetle outbreaks because they create a large number of breeding sites, i.e., weakened trees and for this reason, windthrown timber is generally preventively harvested and removed. In December 1999, the southwest of France was struck by a devastating windstorm that felled more that 27 million m3of timber. This event offered the opportunity to study large-scale spatial pattern of trees attacked by the bark beetle Ips sexdentatus and its relationship with the spatial location of pine logs that were temporally stored in piles along stand edges during the post-storm process of fallen tree removal. The study was undertaken in a pure maritime pine forest of 1300 ha in 2001 and 2002. We developed a landscape approach based on a GIS and a complete inventory of attacked trees. During this study more than 70% of the investigated stands had at least one tree attacked by I. sexdentatus  . Spatial aggregation prevailed in stands with n≥15n15 attacked trees. Patches of attacked trees were identified using a kernel estimation procedure coupled with randomization tests. Attacked trees formed patches of 500–700 m2 on average which displayed a clumped spatial distribution. Log piles stemming from the sanitation removals were mainly distributed along the large access roads and showed an aggregated spatial pattern as well. The spatial relationship between patches of attacked trees and log pile storage areas was analyzed by means of the Ripley’s statistic that revealed a strong association at the scale of the studied forest. Our results indicated that bark beetle attacks were facilitated in the vicinity of areas where pine logs were stored. The spatial extent of this relationship was >1000 m. Similar results were obtained in 2001 and 2002 despite differences in the number and spatial distribution of attacked trees. The presence of a strong “facilitation effect” suggests that log piles should be removed quickly in order to prevent outbreaks of bark beetles.  相似文献

Summary When Asiatic lilies are forced during the winter period, additional light is needed to prevent flower bud abscission and flower bud blasting. Growth room experiments showed considerable differences among eight lily cultivars in their response to low light. Among these cultivars, Connecticut King and Enchantment appeared to be the most sensitive to low light conditions while Uncle Sam and Scout were the least sensitive.Analysis of an incomplete diallel cross between nine cultivars showed significant differences in general combining ability (GCA) for flower bud abortion, number of buds, forcing time, plant height and leaf scorch under winter greenhouse conditions. Enchantment had the highest positive GCA for bud abortion, indicating a high sensitivity to low light conditions. Uncle Sam had the lowest GCA. Pirate and Scout appeared to have a high GCA for sensitivity to leaf scorch. Connecticut King was found to be a probable source of cytoplasmic male sterility. The data suggest that male sterility is associated with better response to low light conditions.  相似文献
