新农村形势下“红色村镇”建设规划的探索——以湖南省浏阳市大围山镇为例

“红色村镇”的建设规划是社会主义新农村建设的工作重点之一,本文以湖南省浏阳市大围山镇为例,通过分析大围山镇的现状条件、发展优势和制约条件,对以大围山镇为代表的湖南“红色村镇”建设规划进行了深入的研究,并提出了建设规划的具体措施。     

重组多子小瓜虫抑动抗原ISCOMs制备

提取表达于四膜虫(Tetrahymena thermophila)细胞膜表面的多子小瓜虫抑动抗原.通过Western Blotting证实重组多子小瓜虫抑动抗原约为35.7 kD,其免疫原性与多子小瓜虫抑动抗原相似.以Quil-A作为佐剂,采用离心、超声波和透析等方法制备小瓜虫抑动抗原免疫刺激复合物(Iag-ISCOMs).电镜观察可见Iag-ISCOMs经磷钨酸负染后呈直径30～40 nm的笼格状结构.免疫和攻毒试验证实Iag-ISCOMs对鳗鲡具有一定保护能力,攻毒后存活率达71.4％,对照组存活率为43.9％,结果表明,本实验成功制备了具有一定免疫原性的Iag-ISCOMs.     

Morphometric and molecular characterisation of Cyprinus carpio × Carassius auratus hybrids

Among members of the Cyprinidae family, hybridisation is more widespread than in any other group of freshwater fish. Therefore, the aim of this work has been to use two different approaches, biometric and genetic, to assess differences between common carp (Cyprinus carpio) × goldfish (Carassius auratus) hybrids and their parents. Twenty-three length measurements of 10 fish from each group have been performed. Statistical analyses of results were conducted using Levene’s, ANOVA, NIR and Kruskal–Wallis tests. Genetic analysis has been conducted by means of amplified fragment length polymorphism (AFLP) technique. The AFLP allowed us to distinguish groups of fish and showed that hybrids had more in common with C. auratus than with C. carpio.     

百合花粉管粘附与定向生长相关基因SCA的克隆及基因多样性分析

以亚洲百合‘Tresor’、麝香百合‘White heaven’和东方百合‘Caruso’为试材,首次在百合基因组中扩增到SCA基因全长序列,在亚洲百合和东方百合中分别获得4个SCA基因拷贝,在麝香百合中获得3个SCA基因拷贝,各拷贝之间具有72.97%～99.68%相似性。从百合基因组中克隆到的SCA基因均含有两个外显子和一个内含子,推导编码113～115个氨基酸残基的前体蛋白(其中成熟蛋白均含91个氨基酸残基)。SCA蛋白具有典型的LTP蛋白N端信号肽,保守的8个半胱氨酸残基和2个五肽模序,并且氨基酸序列之间的相似性高于基因序列相似性,为87.91%～98.90%。亚洲百合、麝香百合、东方百合SCA氨基酸序列比对发现在某些氨基酸位点存在着明显种系间差异。在系统进化树中,三个杂种系的11个SCA基因的氨基酸序列与单子叶植物LTP亲缘关系较近,与双子叶植物较远,这与植物分类学中亲缘关系的远近相一致。     

WTO环境下竹笋生产加工产业发展思考

根据我国种子和竹笋资源状况，分析我国竹笋生产，加工和销售过程中的问题，阐述在WTO市场背景下，应对《卫生和植物检疫措施（SPS）协定》和国际食品法典委员会制定的标准和规范着重关注，重视农药残留量的检测；推动竹笋生产和加工中有机农产品的开放。     

Effect of Rabbit Age on Sperm Chromatin Structure

The aim of this study was to determine the relationship between the age of male rabbits and the sperm chromatin structure. The studies involved the semen of New Zealand White rabbits between 5 and 28 months of age. A flow cytometry and sperm chromatin structure assay (SCSA) method was used to determine chromatin structure. The results of cytometric chromatin structure assay suggested a relatively high stability of sperm chromatin in the rabbit. Between 6 and 16 months of age, the mean percentage of sperm with damaged chromatin was the lowest and ranged from 1.7 to 2.4%. Decreased sperm chromatin stability was found in ejaculates taken from male rabbits less than 5 months and more than 20 months of age.     

Identification of perivascular and stromal mesenchymal stem/progenitor cells in porcine endometrium

Mammalian uterus contains a population of mesenchymal stem/progenitor cells that likely contribute to endometrial regeneration during each reproductive cycle. In human and mouse, they reside in perivascular, epithelial and stromal compartments of the endometrial functionalis and basalis. Here, we aimed to identify tissue resident cells expressing mesenchymal stem cell markers CD29, CD44, CD90, CD105, CD140b and CD146 in the porcine endometrium. We used single immunofluorescence and Western blotting. Each of these markers was detected in small cells surrounding endometrial blood vessels. CD105 and CD146 were also expressed in single stromal cells. A few stromal and perivascular cells showed the presence of pluripotency marker Oct4 in the cytoplasm, but not in the nucleus, which may imply they are not truly pluripotent. Endometrial cell cultures were examined for the expression of CD29, CD44, CD90, CD105 and CD140b proteins and tested in wound‐healing assay and culture model of chemotaxis. In conclusion, our results demonstrate perivascular location of prospective mesenchymal stem/progenitor cells in the porcine endometrium and may suggest that stromal CD105⁺ and CD146⁺ cells represent more mature precursors originating from their perivascular ancestors.     

Breeding of naked oats having high-quality grain

Ecologically plastic naked oat Bekas, which combines high productivity (up to 5.63 t/ha) and grain quality (protein content up to 19.7%; fat content up to 6.7%; starch content up to 54.61%; test weight up to 712 g/L; kernel yield 95–98%; grain yield from sheaf up to 42.3%), was created using selective sampling under different natural conditions: the Northeast Agricultural Research Institute and Falenki Breeding Station (Kirov region), as well as Samara Agricultural Research Institute (Samara region). The grain is large—1000 grain mass is 26.9–29.1 g, grain size is 7–9 × 2.5–3.5 mm, and 2–6 grains are in a spikelet. The new variety is resistant to falling out, to helminthosporium leaf blotch and to crown rust in natural infectious background; it is tolerant to fruit fly.     

CFD 在油气储运工程领域的应用

当前，CFD 商用软件日渐增多及成熟，CFD 技术已成为油气储运工程领域非常重要的科研开发工具。对 CFD 技术在油气储运工程领域的应用现状进行梳理，介绍了 CFD 技术的基本原理、工作流程及其显示和输出的计算结果内容；分述了CFD 技术在管道不同运行方式、管道设备和附件、管道安全运行 3 个方面的应用及取得的成果。以全球第一个通过 ISO9001质量认证的 CFD 商用软件 CFX 对缩径直管段中气固两相管流流场的数值模拟为例，分析了气固两相管流流场特征以及颗粒的冲蚀情况，全面展示了 CFX 软件的模拟流程，对于 CFD 技术进一步有效应用于油气储运工程领域具有重要的现实意义。
