Abstract. Nitrogen (N) loss by leaching poses great challenges for N availability to crops as well as nitrate pollution of groundwater. Few studies address this issue with respect to the role of the subsoil in the deep and highly weathered savanna soils of the tropics, which exhibit different adsorption and drainage patterns to soils in temperate environments. In an Anionic Acrustox of the Brazilian savanna, the Cerrado, dynamics and budgets of applied N were studied in organic and inorganic soil pools of two maize (Zea mays L.) – soybean (Glycine max (L.) Merr.) rotations using 15N tracing. Labelled ammonium sulphate was applied at 10 kg N ha?1 (with 10 atom%15N excess) to both maize and soybean at the beginning of the cropping season. Amounts and isotopic composition of N were determined in above‐ground biomass, soil, adsorbed mineral N, and in soil solution at 0.15, 0.3, 0.8, 1.2 and 2 m depths using suction lysimeters throughout one cropping season. The applied ammonium was rapidly nitrified or immobilized in soil organic matter, and recovery of applied ammonium in soil 2 weeks after application was negligible. Large amounts of nitrate were adsorbed in the subsoil (150–300 kg NO3?‐N ha?1 per 2 m) matching total N uptake by the crops (130–400 kg N ha?1). Throughout one cropping season, more applied N (49–77%; determined by 15N tracers) was immobilized in soil organic matter than was present as adsorbed nitrate (2–3%). Most of the applied N (71–96% of 15N recovery) was found in the subsoil at 0.15–2 m depth. This coincided with an increase with depth of dissolved organic N as a proportion of total dissolved N (39–63%). Hydrophilic organic N was the dominant fraction of dissolved organic N and was, together with nitrate, the most important carrier for applied N. Most of this N (>80%) was leached from the topsoil (0–0.15 m) during the first 30 days after application. Subsoil N retention as both adsorbed inorganic N, and especially soil organic N, was found to be of great importance in determining N losses, soil N depletion and the potential of nitrate contamination of groundwater.  相似文献

Morphological, geochemical and mineralogical studies were carried out in a representative soil catena of the low‐elevation plateaux of the upper Amazon Basin to interpret the steps and mechanisms involved in the podzolization of low‐activity clay soils. The soils are derived from Palaeozoic sandstones. They consist of Hydromorphic Podzols under tree savannah in the depressions of the plateaux and predominantly of Acrisols covered by evergreen forest elsewhere. Incipient podzolization in the uppermost Acrisols is related to the formation of organic‐rich A and Bhs horizons slightly depleted in fine‐size particles by both mechanical particle transfer and weathering. Weathering of secondary minerals by organic acids and formation of organo‐metallic complexes act simultaneously over short distances. Their vertical transfer is limited. Selective dissolution of aluminous goethite, then gibbsite and finally kaolinite favour the preferential cheluviation of first Fe and secondly Al. The relatively small amount of organo‐metallic complexes produced is related to the quartzitic parent materials, and the predominance of Al over Fe in the spodic horizons is due to the importance of gibbsite in these low‐activity clay soils. Morphologically well‐expressed podzols occur in strongly iron‐depleted topsoils of the depression. Mechanical transfer and weathering of gibbsite and kaolinite by organic acids is enhanced and leads to residual accumulation of sands. Organo‐metallic complexes are translocated in strongly permeable sandy horizons and impregnate at depth the macro‐voids of embedded soil and saprolite materials to form the spodic Bs and 2BCs horizons. Mechanical transfer of black particulate organic compounds devoid of metals has occurred later within the sandy horizons of the podzols. Their vertical transfer has formed well‐differentiated A and Bh horizons. Their lateral removal by groundwater favours the development of an albic E horizon. In an open and waterlogged environment, the general trend is therefore towards the removal of all the metals that have initially accumulated as a response to the ferralitization process and have temporarily been sequestrated in organic complexes in previous stages of soil podzolization.  相似文献

To determine the distribution of genes that encode enterotoxins A, B and C, 36 strains of Staphylococcus aureus isolated from goat mastitis and 64 isolated from bovine mastitis were analyzed by Multiplex PCR. Of the total strains studied, 37 (37%) were detected to have some of the SEs genes. From the bovine mastitis strains, 4 (6.3%) co-amplified the sea and seb genes and 2 (3.1%) were positive for the sec gene. From the goat mastitis strains, 31 (86%) tested positive to the Multiplex, and the sec gene was detected in all of them. The production of SE was detected in all strains harboring the corresponding gene. The results demonstrated that S. aureus isolated from goat mastitis had a higher enterotoxigenic potential than those isolated from bovine mastitis. Additionally, the presence of the sec gene in the majority of goat mastitis strains suggests a possible involvement of SEC in goat mastitis pathogenesis.  相似文献

Malassezia pachydermatis is usually associated with otitis and dermatitis in dogs but it can also cause diseases in other species, including humans. In a human neonatal intensive care unit, M. pachydermatis was isolated from an infant's ocular discharge. Therefore, the aim of this study was to ascertain the presence of Malassezia spp. and its possible consequences in dogs' eyes. This research included 19 dogs with unilateral or bilateral corneal ulcers and 60 healthy dogs. A total of 158 clinical specimens from both the groups were obtained from the conjunctival sac of each eye by a calibrated platinum loop. The samples were placed on Dixon and blood agar, incubated at 35 degrees C, and examined daily for 15 days. Then, the strains were subcultured on Sabouraud agar. Of 22 clinical specimens collected from the eyes with corneal ulcers, five cultures (23%) were positive for M. pachydermatis. Of 16 samples collected from the contralateral healthy eye, cultures were positive in three samples (19%). Three animals had unilateral corneal ulcer and positive cultures for M. pachydermatis in both the eyes. Two dogs had unilateral corneal ulcer and positive cultures for M. pachydermatis in the same eye. However, from the 120 samples of 60 healthy dogs, only four clinical specimens (3%) had positive cultures for M. pachydermatis. The findings of M. pachydermatis, in a considerable percentage of clinical specimens from dogs with corneal ulcer, suggest its possible role at least as an aggravating factor in the pathophysiology of this disease.  相似文献

In this work, the gene and cDNA of the sea bass CD8alpha have been isolated and characterized. The coding sequence has an ORF of 666 bp. It retains the Ig motif that interacts with MHC and the two cysteines responsible for an intra-chain disulfide bridge. The hinge region contains the two essential cysteines involved in dimerization. The transmembrane region is well conserved in all analysed sequences. Similar to other teleosts, the cytoplasmic region lacks the consensus p56(lck) motif common in higher vertebrates. Analysis of the expression pattern using RT-PCR shows the highest expression in the thymus. Like in the human gene, the sea bass CD8alpha genomic structure is organized into six exons, which roughly correspond to separate functional domains of the protein. Southern blotting shows that CD8alpha exists as a single copy gene. Together, these results support the concept that the basic structure of CD8alpha has been maintained through evolution.  相似文献

The essential oil of Achillea ligustica from Corsica was investigated by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). A total of 82 compounds representing 94.0% of the oil were tentatively identified. The main constituents were the camphane derivatives, representing >30% (camphor, 21.3%; borneol, 6.2%; bornyl acetate, 3.5%) of the whole oil, and santolina alcohol (19.3%). The enantiomeric distribution of 8 chiral constituents was determined by GC-MS using two enantioselective stationary phases (DIME-beta-CD and Lipodex-E). Racemic santolina alcohol, required for optimization of the enantioselective GC conditions, was prepared by an original two-step synthesis from 2,5-dimethylhexa-2,4-diene. The whole essential oil was tested for its antibacterial activity against a wide range of bacteria using a paper disk method. The results show a promising activity against Streptomyces species.  相似文献

Common bacterial blight (CBB) caused by Xanthomonas axonopodis pv. phaseoli (Xap) is the main bacterial disease of snap bean. The present work aimed to select snap bean genotypes that are resistant to CBB based on three components of resistance: area under the disease progress curve, latent period, and lesion diameter on pods (DL). A completely randomized two-factor factorial design with six replications was used to evaluate leaves and pods of 14 snap bean and three common bean genotypes (PI 207262, BAC-6 and A-794) with high resistance to CBB. Two Xap isolates, 1394-98 and 775-90, were used to inoculate leaves and pods. Data were analyzed using nonparametric statistics. Significant differences were observed among the evaluated genotypes for all of the components of resistance, except for DL. The snap bean UENF 1482 demonstrated good performance in two disease resistance components. For this reason, this genotype can be recommended for use in breeding programs that aim to generate snap bean genotypes resistant to Xanthomonas axonopodis pv. phaseoli.  相似文献

Sheath blight (SB), caused by Rhizoctonia solani, is a serious disease of cultivated rice (Oryza sativa L.) for which genetic resistance is in demand by breeders. With the goal of resistance (SBR)-QTL discovery in U. S. japonica breeding material, 197 doubled-haploid lines from a cross between MCR10277 (resistant) and Cocodrie (susceptible) were evaluated in field and greenhouse assays with U. S. and Colombian pathogen isolates and genotyped at 111 microsatellite marker loci. Four SBR QTLs from MCR10277 were identified, together accounting for 47% of field genetic variation. In all trials the strongest effect was provided by a chromosome-9 QTL, qsbr_9.1, but some QTLs differed for U. S. and Colombian R. solani isolates. SBR QTLs coincided with only two of several height or heading-time QTLs, suggesting that the relationship between these developmental traits and SBR is not simple. For the U. S. isolates, a microchamber greenhouse assay revealed the same QTLs as did field inoculation.  相似文献
