In mammals, thyrotropin, or thyroid-stimulating hormone (TSH), assay is used for the diagnosis of primary hypothyroidism. Hypothyroidism is the most common type of thyroid disorder in cattle. The aim of this study was to develop and validate, under physiologic and pathologic conditions, a radioimmunoassay (RIA) for bovine TSH (bTSH). Double RIA was performed with purified bTSH and specific bovine antiserum. Laboratory validation included research of minimal detection limit, accuracy, and reproducibility. The physiologic validation included a thyrotropin-releasing hormone (TRH) challenge performed on euthyroid cows and a follow-up of bTSH concentration over a 24-hour period. Furthermore, bTSH concentration was assayed in a large population of healthy dairy and beef cows to define reference interval. The pathologic validation was made by assaying bTSH and thyroid hormones on healthy and goitrous newborn calves. The minimum detection limit (MDL) for bTSH assay was 1.3 microU/ml. The recovery was 101% to 106%. The intra- and interassay coefficients of variation (CVs) ranged from 5% to 11% and 11% to 15%, respectively. The RIA covered the whole range of physiologic bTSH values, as shown by bTSH values induced by TRH-challenge. A pulsatile secretion of bTSH was observed, accompanied by a diurnal variation with lower night values than day values. Reference intervals of bTSH ranged from 1.3 to 13.0 microU/ml for beef and dairy breeds. Finally, bTSH easily discriminated goitrous newborn calves from healthy ones, leading to the definition of a cutoff value of 35 microU/ml. The bTSH assay positively reacted to physiologic and pathologic conditions. The accuracy and precision of the RIA were satisfying.  相似文献

Subjective microscopic sperm motility results have recently been demonstrated to differ between Holstein-Friesian (HF) and Belgian Blue (BB) bulls. However, such assessments are rather imprecise. In the present study, sperm motility was assessed objectively by means of the Hamilton Thorne CEROS version 12.2c computer-assisted sperm motility analyser (CASA), and differences between the BB and HF breed could also be demonstrated. Higher percentages of both totally (p < 0.0001) and progressively (p < 0.0001) motile spermatozoa were encountered in the HF breed compared with the BB breed. Furthermore, a lower kinetic efficiency of the BB spermatozoa, evidenced by a lower beat cross-frequency (p = 0.0007) combined with a higher lateral head displacement (p = 0.0015), was the basis for the lower velocity of BB sperm cells. Additionally, BB spermatozoa move less straight forward, resulting in a lower straightness (p < 0.0001). No sperm motility differences were observed between age groups within the BB breed. The breed differences were observed in the examined bull populations residing at AI centres, in Belgium for the BB bulls and in the Netherlands for the HF bulls. However, these bull populations are selected for fertility. A similar pattern was observed in an unselected bull population of both breeds, although these differences were mostly non-significant for the different CASA parameters. Nevertheless, these data suggest that a genetic component might be responsible for the observed sperm motility breed differences.  相似文献

In dairy cattle, many studies have reported quantitative trait loci (QTL) on the centromeric end of chromosome 14 that affect milk production traits. One of the candidate genes in this QTL region – thyroglobulin (TG) – was previously found to be significantly associated with marbling in beef cattle. Thus, based on QTL studies in dairy cattle and because of possible effects of this gene on fat metabolism, we investigated the association of TG with milk yield and composition in Holstein dairy cattle. A total of 1279 bulls from the Cooperative Dairy DNA Repository Holstein population were genotyped for a single nucleotide polymorphism in TG used previously in beef cattle studies. Analysis of 29 sire families showed no significant association between TG variants and milk production traits. Within‐sire family analysis suggests that TG is neither the responsible gene nor a genetic marker in association with milk production traits.  相似文献

Six mature Arabian geldings were used in a two-period crossover study to investigate the effects of cellulase supplementation on fiber digestion. Horses were randomly assigned to either a control (CO; n = 3) or a cellulase (CE; n = 3) treatment for the first period and then treatments were switched for period 2. Each period consisted of a 10-day diet adaptation followed by a 3-day total fecal collection. The enzyme mixture contained 40,000 cellulase units/g and was fed at a rate of 3 g/day split evenly between two feedings. During the diet adaptation period, horses had ad libitum access to timothy hay and were also fed 165 g whole oats as a carrier for the supplement. When eating the CO treatment, horses consumed 16% more hay than when on the CE treatment (P = .004). Fecal output also tended to be greater when horses consumed the CO treatment as compared with CE treatment (P = .07). No differences were found between treatments for fecal percent dry matter (DM%), fecal neutral detergent fiber (NDF), fecal acid detergent fiber (ADF), fecal nitrogen (N), or fecal gross energy (GE). There was a trend for horses consuming the CO treatment to digest more NDF than when consuming the CE treatment (34.6% ± 1.5 vs 31% ± 1.5; P = .07). Horses also digested a greater %ADF, %N, and Mcal of energy when consuming the CO treatment than when consuming the CE treatment (P < .05). Cellulase addition to a hay-based horse diet decreased digestion of fiber components.  相似文献

High moisture corn (HMC) can undergo fermentation during storage that may improve its nutritional value for pigs. Stored HMC used in this study contained (n = 4) 75% dry matter (DM), and, on a DM basis, 63.8% starch, 0.5% mono and disaccharides, 9.9% protein, 19 mM acetic acid, 18 mM lactic acid (LA), 1.25 × 107 cfu g− 1 of LA producing bacteria, 2.88 g kg− 1 total phosphorus (P), and 1.26 g kg− 1 soluble P. The soluble P content was higher than in freshly harvested HMC (0.27 g kg− 1 DM) and indicates P release during storage. Studies were aimed at altering the nutritional value of stored HMC by controlled fermentation or steeping. Changes in LA content were not affected (P > 0.05) by addition of exogenous Lactobacillus and Bacillus bacteria. Steeping of HMC with phytase (Ronozyme) was examined at four inclusion levels (0, 500, 750, 1000 FTU kg− 1) and at either 21 °C or 37 °C. At 37 °C added phytase released virtually all phytate P within 6 h irrespective of the level (levels increased by 1.44 g P kg− 1 DM); at 21 °C, the increase was maximized at 1.2 g P kg− 1 DM after 24 h. The feeding value of HMC for pigs can be altered by steeping with exogenous phytase and does not appear to require microbial inoculants.  相似文献

In a previous study, a reduced efficiency of ileal digestible threonine (THR) use for body protein deposition was observed in growing pigs when pectin was included in the diet. This response was not due to increased physical endogenous ileal THR loss. Our aim was to explore the contribution of diet-induced increases in protein synthesis in the colon, especially mucins, to dietary THR requirements. Twelve barrows (21 kg mean BW) were fed either a cornstarch–soybean meal-based diet (Control) or Control with 12% pectin (Pectin). Pigs were given intravenously 1.5 mmol/kg BW of L-1-13C valine (40 mol%) to measure fractional and absolute synthesis rates (FSR, ASR, respectively) of mucosal and whole intestinal protein in the jejunum and colon. Dietary pectin inclusion increased plasma levels of glucose, isoleucine and glutamine (P < 0.05) but had no effect on insulin or urea nitrogen (P > 0.10). There were no differences in FSR and ASR of whole intestinal protein in jejunum and colon (P > 0.10). The FSR of mucosal proteins in colon, not in jejunum, was increased with dietary pectin supplementation (P < 0.05). Assuming mucosal protein mass is constant, these results imply that the higher protein synthesis in colon mucosa contributes to the reduced THR efficiency observed in pectin-supplemented diet.  相似文献

Previous experiments have shown that mixed or conjoint grazing, when two or more animal species are grazed together, can lead to improved performance of one or more of the species and a higher total output per unit area. Sequential grazing systems, when different animal species graze an area in succession, also have the potential to improve productivity. This experiment directly compared these two approaches for integrating the grazing of cattle and sheep when pastured on improved permanent pasture. Four treatments were compared: 1) sheep only from May to October (S/S); 2) cattle only from May to July followed by sheep only from August to October (C/S); 3) cattle and sheep from May to July, sheep only for the rest of the growing season (C + S/S); and 4) cattle and sheep from May to October (C + S/C + S). Each treatment was replicated three times. From May until weaning at the end of July the plots were grazed by steers and ewes and lambs, and from weaning until October by steers and lambs. Sward heights were maintained at 6 cm using a “put and take” stocking system. Animal performance and sward composition data were collected during three growing seasons, 2001–2003. During the pre-weaning period lambs grazing plots which were grazed only by sheep had significantly poorer growth rates than those grazing plots where there was mixed grazing. Lambs grazing the sheep only plots also had lower liveweight gain during the post-weaning period, with the highest growth rates being recorded on the C + S/C + S treatment. Carrying capacity and total liveweight gain per unit area post-weaning were also consistently higher for the C + S/C + S treatment, while carrying capacity pre-weaning was highest for the C/S treatment. Measurements of clover content in the grazed horizon and botanical separation of material from quadrat cuts indicated that even under relatively controlled conditions choice of grazing system can influence sward composition. However, there was no clear link between any single sward change and the growth of the animals, indicating that the improved performance of cattle and sheep under mixed grazing is the result of a combination of factors, rather than a response to a particular sward parameter.  相似文献

The aim of the study was to measure the effect of creep feeding during lactation on net absorption in the small intestine at 4 days after weaning. Intermittent suckling was used to increase creep feed intake during lactation. Creep feed containing chromic oxide was provided. Based on the colour of the faeces, piglets were classified as ‘eaters’ or ‘non-eaters’, respectively. At day 4 after weaning, an in vivo small intestine segment perfusion test was performed at 5 sites along the small intestine in 24 piglets (12 eaters and 12 non-eaters). At both sides of each intestinal segment a tube was fitted to perfuse and drain fluid in order to assess net absorption. Net absorption was higher in eaters than in non-eaters (P < 0.001). Net absorption varied greatly between and within piglets and was highest in the caudal segments of the small intestine (P < 0.001). These data suggest that creep feeding could be a useful tool in the prevention of post-weaning diarrhoea.  相似文献

Field tests of a prototype microwave‐based weed killer machine were conducted on Abutilon theophrasti, Panicum miliaceum, lucerne and oilseed rape pure stands. The approach can be considered a thermal weed control method, the microwave radiation causing dielectric heating of plant tissue water that eventually kills the plant. The method could overcome the limitations of other thermal methods, such as fire risk with flaming or the heavy loads required for hot water treatments. Species were effectively controlled by microwave irradiation, but their sensitivity and the evolution of damage symptoms over time differed. Lucerne showed no sigmoidal response and was the least affected by the treatment, while a log‐logistic curve expressed the dose–response relationships of the other species quite well. The estimated microwave dose for a 90% dry weight reduction ranged from 1015 kJ m?2 in A. theophrasti to 3433 kJ m?2 in P. miliaceum. Energy cost evaluation indicated that increased efficiency is required for this technique to compete with other thermal methods. Microwave efficiency could be increased by a flux configuration that minimizes soil penetration and maximizes absorption by plants, which, in turn, depends on plant growth form.  相似文献
