Background:

Nicotine can activate dopaminergic neurons within the ventral tegmental area (VTA). However, there is no evidence about complete inhibition of VTA on nicotine reinforcement.

Methods:

in the present study, we used conditioned-place preference (CPP) method to study the effect of transient inhibition of left and/or right side of the VTA by lidocaine on nicotine reward properties. Male Wistar rats seven days after recovery from surgery and cannulation were conditioned to nicotine (1.5 mg/kg) in an unbiased designed CPP apparatus. Five min before each nicotine injection in conditioning phase, lidocaine (2%) was administered either uni- or bi-laterally into the VTA (0.5µl/rat).

Results:

results revealed that lidocaine administration into the left but not right side of the VTA reduced nicotine CPP significantly. The reduction was potentiated when lidocaine injected in to both sides of the VTA. In addition, the number of compartment crossing was reduced when lidocaine injected in both side of VTA as well as left side. On the other hand, rearing was reduced when lidocaine injected to the right but not left side of VTA. At last, sniffing was reduced only in the group in which received lidocaine in both side of VTA. Sniffing and rearing increased in the group in which received lidocaine in right side.

Conclusion:

It is concluded that the right and left side of VTA play different role in nicotine-induced activity and reward.  相似文献

Background:

Hyperprolactinemia is a common side effect of antipsychotic drugs that requires further investigation. The current study was designed to evaluate dose-dependent effect of chlorpromazine (CPZ) on hormonal changes and uterine horn histological structure in rats. Moreover, the mammary glands were analyzed to show hyperprolactinemia-induced histological changes.

Methods:

Albino Wistar rats (n = 32) were divided into four groups. The first group was set as a control. In the three drug-treated groups (eight rats in each group), CPZ was administered by a gavage at doses of 3, 10, and 30 mg/kg/day for 28 days. One day after the last administration of the drug, the animals were sacrificed. Histopathological and histomorphometrical analyses of the uterine horns and mammary glands were carried out to evaluate dose-dependent effect of CPZ on histological structure. Serum levels of prolactin (PRL), estradiol, progesterone, follicle-stimulating hormone (FSH), and luteinizing hormone (LH) were also evaluated.

Results:

Remarkable (P < 0.05) elevation was observed in CPZ-administrated animals'' uterine horn endometrium, myometrium, and perimetrium thicknesses, and the mammary glands were observed with galactorrhea features. The serum level of progesterone and PRL significantly (P < 0.05) increased, while the serum concentration of LH, FSH, and estradiol was notably (P < 0.05) decreased depending on administrated CPZ dose. No histological and biological changes were occurred in the control animals.

Conclusion:

The present findings suggest that CPZ-induced disturbances not only depend on PRL level and increased PRL level largely depends on administrated doses of the CPZ. Key Words: Hyperprolactinemia, Uterus, Rats  相似文献

To select and characterize potential probiotic bacteria from the gut microbiota of Siberian sturgeon (Acipenser baerii), 129 strains isolated from the hindgut were screened for antagonistic activity against five fish pathogens. Ten isolates showed antagonism towards three or more pathogens. Nine of these isolates were Gram‐positive, belonging to Lactococcus (seven) and Bacillus (two), and a single strain belonging to the Gram‐negative Citrobacter. These inhibitory isolates were identified using genetic, phentotypic and biochemical traits, and further characterized by in vitro tests assessing the adhesion and growth in mucus and resistance to gastric and intestinal fluids. The candidate probiotics were determined to be non‐pathogenic through an in vivo study. Based on these assays, Lactococcus lactis ssp. lactis STG45 and STG81 showed the broadest inhibitory potential, a high viability in simulated gastrointestinal juice and the highest adhesion capacity to mucus. They were therefore selected as the most promising candidate probiotics. This is the first study screening probiotics among the gut microflora of Siberian sturgeon.  相似文献

This study was carried out to show the effects of Sangrovit on growth, blood biochemical parameters, survival and resistance to salinity stress in the Caspian roach. Fish (1.24 ± 0.13 g) were divided into four groups fed diets containing Sangrovit at different levels: 0 (control), 0.05, 0.1 and 0.15% for 45 days. The results showed that there was a significant increase in final weight, weight gain and specific growth rate in those fish fed 0.15% Sangrovit diets compared with the control (P < 0.05). Food conversion rate of fish fed 0.05% Sangrovit diet was significantly (P < 0.05) lower than the control group. Condition factor was not significantly affected by Sangrovit dosage (P > 0.05). The blood glucose and cholesterol levels were significantly lower in treated groups compared with the control (P < 0.05). The highest total protein was obtained in the 0.15% group and it was significantly higher than in all other groups (P < 0.05). Challenges were carried out after 45 days of feeding, to determine the effect of Sangrovit on resistance to salinity stress. Blood samples were obtained four times after challenge (day 1, 3, 5 and 7) for evaluate haematocrit levels. On the first day, haematocrit increased significantly in all groups compared to day 5 and 7 (P < 0.05). The haematocrit levels decreased gradually in all groups from the third day. Before and after stress, no differences were observed for survival rate among the experimental diets (P > 0.05). These results indicate that the inclusion of 0.15% Sangrovit in the diet of the Caspian roach enhances feed efficiency, fish growth and blood biochemical parameters.  相似文献

Evaluating agricultural loss due to flooding is essential for defining an appropriate risk level in flood management projects. This research introduces a method for computing annual damage risk for rice fields with respect to both spatial and temporal pattern of floods, while considering the effect of crop growth stages on the total damage. For finding the spatial pattern of floods, HEC-RAS was employed to derive selected physical parameters of floods of interest and then a physical loss function was used to calculate the unconditional loss over flood prone areas. A time-dependent loss function was introduced for rice to determine the effect of growth on damages derived through the physical loss function. Due to the seasonal nature of agricultural risk, frequency analysis was performed for seasonal discharges for each growth stage of rice. A method was introduced to use the concept of Expected Annual Damage in agricultural fields. A method called “AGEAD” was introduced for evaluating short-term risk for agricultural activities considering their inherent spatial and temporal characteristics. Results showed that damages evaluated by employing AGEAD were completely different from what was obtained by applying a flood of defined return period, i.e. from the results of long-term risk evaluation. While it is common to attribute a unique predicted loss to all flood prone areas, AGEAD application showed that different zones in flooded paddy fields were subjected to different amount of damages from 100 to 0%.  相似文献

This study was carried out to investigate the effect of water‐soluble polysaccharides extract of algae Ulva rigida (WPU) as dietary supplement on growth performance, antioxidant enzyme activity, lysozyme and phenoloxidase activity, and resistance of shrimp (Litopenaeus vannamei) subjected to bacterial infection with Photobacterium damselae. Three replicate groups of shrimp (1.0 g) were fed four diets containing four levels, 0 or control, 0.5, 1 and 1.5 g/kg of WPU for 8 weeks over the growth trial. Thereafter, 30 shrimps from each dietary treatment were infected with bacteria P. damselae to evaluate disease resistance of infected shrimp. The results of this study showed that WPU was effective as a growth promoter for L. vannamei. The best growth rate was observed in shrimp fed 1.5 g/kg of WPU diet. Regarding antioxidant defences, the diets supplemented with three levels of WPU stimulated glutathione peroxidase and catalase activates in experimental shrimps. MDA content of L. vannamei‐fed diet containing WPU 1.5 and WPU 1.0 was lower than WPU 0 and WPU 0. 5 diets. Also, lysozyme and phenoloxidase activities of shrimp receiving WPU at 1.0 and 1.5 level were significantly higher than those fed WPU 0 and WPU 0.5 diets. In addition, using WPU extract in all diets decreased mortality in L. vannamei in a dose‐dependent manner after challenge with P. damselae. These results suggest that incorporation of water‐soluble polysaccharides from green algae U. rigida at 1.5 g/kg doses improves growth and antioxidant activity and enhances the immune responses in shrimp L. vannamei.  相似文献

This study was carried out to evaluation of some nutrients accumulation including Ni (nickel), Fe (iron), Mg (magnesium), Cu (copper), Ca (calcium) as well as Ni translocation factor (TF) in three tomato cultivars grown hydroponically under treatments including L-histidine (His) and calcium chloride (CaCl2) (300 µM) exposed to nickel (II) sulfate (NiSO4) (150 and 300 µM) by ICP-OES (inductively coupled plasma atomic emission spectroscopy) analysis. Tomato cultivars included Cal-J N3, Early Urbana Y and. Results indicated that shoot Ni accumulation increased under Ni stress. Ca and His reduced root, shoot Ni content and Ni TF (translocation factor) under Ni stress in Petoearly CH cultivar. Ni stress also decreased root and shoot Ca, Fe, Mg and Cu while Ca and His improved the nutrients uptake, and inhibited Ni accumulation. Therefore, Ca and His alleviated adverse effects of Ni stress on nutrients in Petoearly CH cultivar. Beside, Ca and His interaction alleviate Ni stress by inhibition of Ni uptake and translocation as well as plant nutrition improvement in the tomato plants.  相似文献

Maize rough dwarf virus (MRDV) is the most important disease affecting maize growth in Iran. In the present study, the putative resistance capacity of 35 maize inbred lines against the causal agents of MRDVwas studied under natural field infection in 2010 and 2011. The viral transmitter was a plant hopper known as Laodelphax steriatellus. Early sowing provided suitable temperatures for vector propagation and transmission and therefore natural infection under field conditions. Sowing single rows of SC704 as vector spreader between every five rows of inbred lines caused adequate vector propagation and viral transmission. This shows that viral infection was sufficient and possible resistance was linked to viral resistance, not failure, in vector transmission. In principal component biplot, plant height vector had wide angles with the vectors for disease severity index and MRDV incidence that showed the negative association of plant height with MRDV disease. The percentage of natural infection to MRDV in susceptible control(SC704) was about 64%. The genotypes A679, K3547/3, K3545/6, K3653/2, and B84 were more susceptible to MRDV. Only the line K3640/3 had low disease incidence and severity and it can be considered as resistant for possible use in genetic studies. Results showed that an early sowing of maize in temperate regions increases the chance of transmitting viruses via their vector.  相似文献

The effect of Mentha longifolia (L.) leaf hydroalcoholic extract (MLE) was examined on rat ileal smooth muscle contractions. Last portion of ileum from male adult Wistar rat was mounted in an organ bath containing Tyrode solution. The tissue was contracted by carbachol (CCh, 10 microM), KCl (60 mM) and BaC12 (4 mM) and then MLE (0.0625-1 mg mL(-1)) was added to the bath cumulatively. The effect of MLE on KCl-induced contraction was examined after tissue incubation with propranolol (1 microM), naloxone (1 microM) and N omega-nitro-L-arginine methyl ester (L-NAME, 100 microM). The effect of MLE on CaCl2-induced ileal contraction in Ca(2+)-free with high potassium Tyrode solution was also evaluated. The role of potassium channels was examined by ileum incubation (5 mim) with tetraethylammonium (TEA, 1 mM). The results showed that KCl-, CCh and BaCl2-induced ileal contractions were inhibited (p < 0.001) by cumulative concentrations of MLE with the same potency. In addition, MLE (0.25-1 mg mL(-1)) inhibited (p < 0.01) ileal contractions induced by CaCl2 (0.45-2.7 mM) in a concentration-related manner. The antispasmodic effect of MLE was affected neither by propranolol, L-NAME nor by naloxone. The MLE concentration-response curve was shifted to the right (p < 0.05) by tissue incubation with TEA. From results it may be suggested that Mentha longifolia hydroalcoholic leaf extract induces its spasmolytic activity mainly through disturbance in calcium mobilization and partly by potassium channels activation. Present results show that Mentha longifolia leaf extract exerts relaxant effects on intestinal smooth muscle, consistent with the traditional use of the plant to treat gastrointestinal disorders such as diarrhea and colic.  相似文献

森林生态服务直接或间接地造福人类。本文利用模拟模型和地理信息系统作为工具,分析生态因子对生态系统服务功能的作用,评估了伊朗西部扎格罗斯森林对Bazoft河流域水源保护的经济价值。应用HEC-HMS模型中的曲线数字法对降雨量-径流进行了模拟。该模型需要输入土地覆被、土壤、短期降雨和流出数据。用观测数据并分阶段校对对拟合模型的功效进行了修订。测定了森林在4种设定的土地覆盖方案中保水性和减少地表径流量的作用以及土地用途改变对该区降雨量-径流行为的影响。结果表明:方案一假定该流域全被森林覆盖,总的流出量将最小,降雨初期损失将增加。用重置成本法估算了森林水文功能之保水性经济价值。评估结果表明,每公顷Bazoft河流域的森林可以蓄存84.8立方米水,年产值0.5 US$/m3。因此,每公顷BazoR河流域的森林保水性经济评价值是43US$。说明覆被森林有利于该流域经济发展,该研究有助于决策者选择适当的、经济可行的发展策略。  相似文献
