The growth inhibitory effects of recombinant canine interferon alpha (IFN-alpha), beta (IFN-beta) and gamma (IFN-gamma) were examined on Madin-Darby canine kidney cells infected with Neospora caninum tachyzoites. The parasite growth was inhibited by all IFNs in a dose-dependent manner. IFN-gamma inhibited the parasite growth with greater efficacy than IFN-alpha or IFN-beta. Moreover, the effect of IFNs on N. caninum growth associated with the suppression of the host cell viability. The present study indicates IFN-alpha and -beta, besides IFN-gamma, play a crucial role for N. caninum growth in host cells.  相似文献

The dendritic cell (DC) network is a specialized system for presenting antigen to naive or quiescent T cells, and consequently plays a central role in the induction of T cell and B cell immunity in vivo. Despite considerable achievements in the last ten years, in our understanding of how DC induce and regulate immune responses, much remains to be learned about this complex system of cells. The history and current status of DC termed "directors of the immune system orchestra" is reviewed. The present understanding of DC cell biology, function and use, taking into account their complexity is discussed.  相似文献

Babesia equi (EMA-1) and Babesia caballi (BC48) gene fragments were amplified by polymerase chain reaction (PCR), in blood samples, and partially fed-females and egg and larval progenies of Dermacentor nuttalli, collected from horses in Altanbulag, Tuv Province, Mongolia. While Babesia parasite DNA was detected in some horse blood samples during the first PCR, all positive cases in partially fed-female ticks, eggs and larvae were confirmed by nested PCR. Present study reinforces earlier similar findings in unfed D. nuttalli ticks collected from an open space vegetation in Bayanonjuul, Tuv Province in Central Mongolia, pointing to the most likely important role of D. nuttalli in the transmission of equine babesiosis in Mongolia. The detection of parasite DNA in eggs and larval progenies is likewise suggestive of transovarial parasite transmission in this tick species.  相似文献

We reported previously that doxorubicin, an anticancer agent that has an anthracycline structure, alters Ca2+ releasing and uptake mechanisms in the sarcoplasmic reticulum of myocardial cells. These effects of doxorubicin are apparently related to its cardiotoxicity. Mitoxantrone is a similar anticancer agent with an anthracenedion structure that has been shown to be significantly less cardiotoxic. In the present study, the effects of mitoxantrone on the functions of the sarcoplasmic reticulum were examined in isolated muscle preparations obtained from the guinea-pig heart. In electrically-stimulated left atrial muscle preparations, incubation in vitro for 4 hr with 30 or 100 microM mitoxantrone significantly prolonged the time to the peak of twitch tension, markedly increased the developed tension observed at lower stimulation frequencies, thereby attenuating the slope of positive force-frequency relationships, and increased the postrest contraction observed after a 60-sec quiescent period. In myocytes isolated from ventricular muscles, 30 microM mitoxantrone increased the peak and the size of intracellular Ca2+ concentrations ([Ca2+] i), and prolonged the time to peak [Ca2+]i. In skinned muscle fiber preparations obtained from the left ventricular muscle, 30 muM mitoxantrone significantly increased the caffeine-induced contraction without affecting the Ca2+ sensitivity of contractile proteins. These results suggest that mitoxantrone enhances Ca2+ release from the sarcoplasmic reticulum in isolated atrial muscle preparations obtained from the guinea-pig heart. Apparent enhancement of the sarcoplasmic reticulum functions, in contrast to anthracyclines that has been shown to suppress these functions, seems to explain the relative lack of marked cardiotoxicity of mitoxantrone.  相似文献

Dietary supplement of bioflavonoid rutin and calorie restriction were examined to investigate possible reduction in oxidative DNA damage, protein oxidation, and lipid peroxidation in animals. Rats were fed ad libitum 20% casein semipurified diet or a diet that was supplemented by 2.5% water soluble rutin derivative, 4(G)-alpha-glucopyranosylrutin (G-rutin) for 18 days. Two other groups of rats were fed the respective diets at 60% of the mean food intake of the ad libitum fed animals for the same period. Urinary excretion of thymine glycol and thymidine glycol, indices of DNA base damage in the whole body, were significantly low in the G-rutin-supplemented groups. The protein carbonyl contents, a measure of protein oxidation, were significantly low in the liver of G-rutin-supplemented groups and calorie-restricted groups. The data indicate that G-rutin provides an antioxidant defense in rodents against free radical-caused oxidative damage of DNA and proteins.  相似文献

Group A saponins are the principal factors underlying the undesirable bitter and astringent tastes in soybean food products. Therefore, the genetic reduction or elimination of the undesirable tastes is of great significance in soybean taste breeding. Using thin‐layer chromatography and liquid chromatography–mass spectrometry, we identified a novel mutant lacking group A saponins in a wild soybean (Glycine soja Sieb. & Zucc.), reflecting the failed arabinose binding of arabinosyltransferase to the C‐22 hydroxyl group, consequently leading to a lack of group A saponins and the concomitance expression of two new components A‐αg and KA‐αg in seeds. The C‐3 glycosylated soyasapogenol A (SS‐A), A‐αg, is a steady precursor compound in biosynthesis of Aa and Ab types in the seeds of soybean plants and exhibits stable inheritance. In this study, we observed a critical step necessary for arabinose binding to the C‐22 hydroxyl group in the biosynthetic pathway of group A saponins and identified an important germplasm accession for the genetic improvement in the tastes of soybean milk and processed soybean foods.  相似文献

Dietary fiber has high functional value in relation to gut flora. We searched for a high-lysine mutant of the most popular rice cultivar in Japan, ‘Koshihikari’, as a route to a higher dietary fiber content like a success case in new barley cultivar, ‘Beau-fiber’. We found several promising high-lysine mutants with high dietary fiber content. One of these, ‘WFE5’, has three times the dietary fiber content in white rice. Two rounds of backcrossing to Koshihikari produced a near-isogenic line with a high fiber content. The line’s agronomic traits were close to those of Koshihikari except for yield and eating quality. As these two traits are critical, we discuss how to improve them.  相似文献

The mid-gut gland of scallop Patinopecten yessoensis has been discarded in scallop processing factories as a fishery waste and various attempts have been made to turn the waste into valuable resources. In the present study, we tried to use mid-gut gland drips from scallop as a source of β-1,3-glucanase. The mid-gut gland drips were collected in a local fishery factory in Yubetsu-cho, Hokkaido Prefecture. β-1,3-Glucanase was purified from the mid-gut gland drips by ammonium sulfate fractionation followed by successive chromatography on Toyopearl Phenyl-650M and Toyopearl DEAE-650M. The scallop β-1,3-glucanase, named PyLam38 in the present study, showed a molecular mass of approximately 38 kDa by sodium dodecylsulfate-polyacrylamide gel electrophoresis, and hydrolyzed laminarin, a β-1,3-glucan from Laminaria sp., producing laminaribiose and glucose with an optimal pH and temperature of 6.0 and 45°C, respectively. PyLam38 exhibited high transglycosylation activity toward various accepter substrates such as monosaccharides, alcohols and xylooligosaccharides. Thus, PyLam38 was found to be useful for the production of various novel heterooligosaccharides consisting of laminarioligosaccharides and various accepters.  相似文献

Information about potential risks of iron nanomaterials is still limited, while a wide variety of applications are expected. We recently reported acute phase responses of male and female Fischer 344 rats after a single intratracheal spray instillation of Fe3O4 nanoparticles (magnetite), clearly showing dose-dependent pulmonary inflammatory changes (Tada et al., J Toxicol Pathol 25, 233–239, 2012). The present study assessed long-term responses of male and female Fischer 344 rats to multiple administrations of magnetite. Ten-week-old male and female Fischer 344 rats (n=20/group) were exposed to a total of 13 quadweekly intermittent intratracheal spray instillations of magnetite during the experimental period of 52 weeks, at doses of 0, 0.2 (low), 1.0 (medium) and 5.0 (high-dose) mg/kg body weight per administration. Absolute and relative lung weights of the high-dose group were significantly higher than those of the control group. Macroscopically, slight enlargement and scattered black patches were recognized in the lungs and the lung-associated lymph nodes of the high-dose group. Histopathologically, infiltration of macrophages phagocytosing magnetite (all dose groups) and of chronic inflammatory cells (medium- and high-dose males and high-dose females), alveolar bronchiolization and granuloma (high-dose group) were observed. In addition, alveolar hyperplasias were observed in some rats of the high-dose group, and cytoplasmic overexpression of β-catenin protein was immunohistochemically found in such lesions. The present results clearly show that instilled magnetite causes chronic inflammatory responses in the lung. These responses occur in a dose-dependent manner without apparent differences among sexes  相似文献
