Adzuki bean is an important legume crop originating in temperate regions, with photoperiod in sensitivity being a key factor in its latitudinal adaptation. The Flowering Date1 (FD1) gene has a large effect on the photoperiodic response of flowering time, but the molecular basis for the effect of this locus is undetermined. The present study delimited the FD1 locus to a 17.1 kb sequence, containing a single gene, an E1 ortholog (VaE1). A comparison between Vigna angularis ‘Shumari’ (photoperiod insensitive) and ‘Acc2265’ (photoperiod sensitive) identified 29 insertions/deletions and 178 SNPs upstream of VaE1 in the FD1 locus. VaE1 expression in ‘Acc2265’ was greater under long-day than short-day conditions, whereas VaE1 expression in ‘Shumari’ was lower regardless of day length. These findings suggested that responsible gene of FD1 is a VaE1, which acts as a floral repressor by being upregulated in response to long-day conditions. The inability to upregulate VaE1 under long-day conditions was linked to its ability to flower under these conditions. These results provide greater understanding of the molecular control of a flowering date and clues enabling the breeding of adzuki bean at higher latitudes. 相似文献

In this study, we cryopreserved pig spermatozoa using carboxylated poly-L-lysine (CPLL) as the cryoprotectant to determine its efficacy. Pig spermatozoa were placed in a freezing extender containing 3% (v/v) glycerol and different CPLL concentrations. The motility indices of the spermatozoa cryopreserved with 0.25% (v/v) CPLL at 6 (59.3), 9 (53.7), and 12 (26.2) h after thawing were significantly higher (P < 0.01 or P < 0.05) than those of the spermatozoa cryopreserved without CPLL (53.7, 40.1, and 17.5 at 6, 9, and 12 h after thawing, respectively). The concentration of CPLL in the freezing extender did not affect the ability of frozen-thawed spermatozoa to fertilize oocytes in vitro. However, the blastocyst formation rate of embryos derived from spermatozoa cryopreserved with 0.25% CPLL (24.6%) was significantly higher (P < 0.01) than that of embryos derived from spermatozoa cryopreserved without CPLL (11.2%). The conception rate of the sows inseminated with spermatozoa cryopreserved with 0.25% CPLL (72.2%) was not significantly different from that of the sows inseminated with spermatozoa stored at 17°C (81.3%). However, the mean number of total piglets born to the former (10.0) was significantly lower (P < 0.05) than that of total piglets born to the latter (13.4). The results showed that CPLL in the freezing extender maintained the motility of frozen-thawed pig spermatozoa and improved the in vitro development of embryos produced by in vitro fertilization. In addition, we have demonstrated that piglets could be obtained with artificial insemination using spermatozoa cryopreserved with CPLL. 相似文献

suwari gels, set at various temperatures and times, and acid-induced kamaboko gel, which was prepared by soaking suwari gel in 5% acetic acid for 20 h, were studied to evaluate the mechanical properties that are affected by setting conditions.
Unset squid meat paste did not form a gel when soaked in acetic acid. The breaking strength of both suwari gel and acid-induced kamaboko gel showed a tendency to increase with setting temperature and time. SDS-PAGE analysis of suwari gel and acid-induced kamaboko gel, which were set at various temperatures and times, showed that myosin heavy chain (MHC)
was observed at 30°C only for the first hour. The intensity of the MHC band at 30°C gradually decreased with setting time,
while the intensity of the polymer band gradually increased with setting time. These results suggest that the protein-protein
bonds in suwari gel affect the final texture of acid-induced kamaboko gel. Based on the analysis of the mechanical properties, and in consideration
of the fact that the purpose of this experiment was to reduce energy usage, the best setting condition was determined to be

ABSTRACT: Parvalbumin, a calcium-binding sarcoplasmic protein of approximately 12 kDa, represents the cross-reactive, major allergen in fish. In consideration of the fact that parvalbumin is contained at high levels not only in fish muscle but also in frog muscle, the present study was undertaken to clarify whether fish-allergic patients react to two parvalbumins (α- and β-parvalbumins) purified from the bullfrog Rana catesbeiana , which is sometimes consumed as a delicacy in Japan. In enzyme-linked immunosorbent assays (ELISA), sera from 12 of the 14 patients tested reacted equally to both parvalbumins purified from the Pacific mackerel Scomber japonicus and the bigeye tuna Thunnus obesus . Of the 12 sera positive to fish parvalbumins, eight sera also reacted to α- and β-parvalbumins of the bullfrog with different spectra: one serum reacted strongly to α-parvalbumin, six sera reacted strongly to β-parvalbumin and one serum reacted equally to both α- and β-parvalbumins. In addition, inhibition ELISA experiments revealed cross-reactivity between fish and bullfrog parvalbumins. Based on these results, it is proposed that fish-allergic patients should avoid the consumption of frog meat unless they are accurately diagnosed as lacking immunoglobulin E against frog. 相似文献

Objective— To describe diagnostic findings, surgical technique, and outcome in dogs with thoracic spinal canal stenosis and vertebral instability secondary to congenital vertebral anomalies.
Study Design— Retrospective clinical study.
Animals— Dogs (n=9) with thoracic spinal canal stenosis.
Methods— Medical records (1995–1996; 2000–2006) of 9 dogs with a myelographic diagnosis of spinal canal stenosis and/or vertebral instability secondary to congenital vertebral anomaly that were surgically managed by vertebral stabilization with or without laminectomy were reviewed. Data on pre- and postoperative neurologic status, diagnostic findings, surgical techniques, and outcomes were retrieved. Follow-up evaluations were performed at 1, 2, and 6 months. Long-term outcome was assessed by means of clinical examination or owner telephone interviews.
Results— Spinal cord compression was confirmed by myelography, and in 2 dogs, dynamic compression by stress myelography. Eight dogs regained the ability to ambulate postoperatively. One dog with a partial recovery regained voluntary movement but did not become ambulatory.
Conclusions— Spinal cord injury secondary to congenital vertebral anomaly may have a good outcome when treated by vertebral stabilization with or without laminectomy. Adequate stabilization of the vertebrae and improved neurologic outcome were achieved in most dogs.
Clinical Relevance— Vertebral stabilization using positively threaded profile pins and polymethylmethacrylate with or without laminectomy is an effective treatment for spinal canal stenosis and vertebral instability secondary to congenital thoracic vertebral anomalies. 相似文献

In 2007–2008, a canine distemper virus (CDV) epidemic occurred among wild animals in
Wakayama Prefecture, Japan, and many mammals, including the wild boar and deer, were
infected. In this study, CDV prevalence among wild animals was surveyed before and after
the epidemic. At first, an enzyme-linked immunosorbent assay (ELISA) with horseradish
peroxidase-conjugated protein A/G was established to detect CDV antibodies in many
mammalian species. This established ELISA was available for testing dogs, raccoons and
raccoon dogs as well as virus-neutralization test. Next, a serological survey of wild
mammalians was conducted, and it was indicated that many wild mammalians, particularly
raccoons, were infected with CDV during the epidemic, but few were infected before and
after the epidemic. On the other hand, many raccoon dogs died during the epidemic, but CDV
remained prevalent in the remaining population, and a small epidemic occurred in raccoon
dogs in 2012–2013. These results indicated that the epidemic of 2007–2008 may have been

A 6-month-old female beagle dog, assigned to the low-dose group in a toxicity study, was evaluated for compound toxicity, and spontaneous hyperadrenocorticism was suspected. The animal had an externally apparent distended abdomen on clinical examination upon arrival. Pre-dose clinical pathology showed slightly higher erythroid parameters and stress leukogram on hematology; plasma biochemistry showed higher total protein, gamma-glutamyl transferase, total cholesterol, and triglyceride levels than the reference data. On necropsy, a prominent increase in adipose tissues of the subcutis and abdomen and increased weight of the adrenal gland and liver were observed. Histopathology revealed diffuse hyperplasia of adrenocortical cells in the zona fasciculata and reticularis, cortical atrophy of the thymus, and abundant glycogen accumulation in the hepatocytes. These findings were incidental and not test-substance-related. Electron microscopy of the adrenocortical cells in the zona fasciculata revealed decreased typical translucent lipid droplets, increased electron-dense lipid droplets, and abundant smooth endoplasmic reticulum and lysosomes. Additionally, increased numbers of various sizes and forms of mitochondria with tubular, vesicular, or lamellar cristae compared to that of normal animals were observed. These ultrastructural characteristics of the adrenocortical cells suggested hyperfunction. The pre-dose plasma cortisol levels were slightly higher than those of other females assigned to the toxicity study, while plasma adrenocorticotropic hormone levels were within the normal range. These findings indicate that hyperadrenocorticism is a possible cause of the systemic changes in this case. 相似文献

Synthetic hexaploid wheat is an effective genetic resource for transferring agronomically important genes from Aegilops tauschii to common wheat. Wide variation in grain size and shape, one of the main targets for wheat breeding, has been observed among Ae. tauschii accessions. To identify the quantitative trait loci (QTLs) responsible for grain size and shape variation in the wheat D genome under a hexaploid genetic background, six parameters related to grain size and shape were measured using SmartGrain digital image software and QTL analysis was conducted using four F2 mapping populations of wheat synthetic hexaploids. In total, 18 QTLs for the six parameters were found on five of the seven D-genome chromosomes. The identified QTLs significantly contributed to the variation in grain size and shape among the synthetic wheat lines, implying that the D-genome QTLs might be at least partly functional in hexaploid wheat. Thus, synthetic wheat lines with diverse D genomes from Ae. tauschii are useful resources for the identification of agronomically important loci that function in hexaploid wheat. 相似文献

Lysosomes are acidic and highly dynamic organelles that are essential for macromolecule
degradation and many other cellular functions. However, little is known about lysosomal
function during early embryogenesis. Here, we found that the number of lysosomes increased
after fertilization. Lysosomes were abundant during mouse preimplantation development
until the morula stage, but their numbers decreased slightly in blastocysts. Consistently,
the protein expression level of mature cathepsins B and D was high from the one-cell to
morula stages but low in the blastocyst stage. One-cell embryos injected with siRNAs
targeted to both lysosome-associated membrane protein 1 and 2 (LAMP1 and LAMP2) were
developmentally arrested at the two-cell stage. Pharmacological inhibition of lysosomes
also caused developmental retardation, resulting in accumulation of lipofuscin. Our
findings highlight the functional changes in lysosomes in mouse preimplantation
