The effect of sodium taurocholate (S. T.) on the contractile response of rat-phrenic nerve diaphragm, frog's musculus rectus abdominis and frog's musculus gastrocnemius sciatic nerve preparation was studied. Moreover, interaction of S. T. with neuromuscular blockers are carried out, too. S. T. was found to have a dose-dependent reduction in the contractile response of the tested preparation. On the other hand, the inhibitory effect induced by S. T. did not block or alter the stimulatory effect of either acetylcholine or prostigmine. Trials were made to estimate the potency of S. T. in a comparison with other skeletal muscle relaxant drugs. In this respect S. T. exhibited a more potent effect than gallamine. In contrast, other skeletal muscle relaxants (dtubocurarine, atracurium, pancuronium and succinylcholine) were highly potent. There was also a marked synergistic effect between S. The immunopotentiating activity of a new delivery system was investigated comparatively to Alhydrogel adjuvant, as an antiviral inactivated vaccine after one injection. The efficiency of the new formulation (BioMed) was evaluated with an inactivated porcine strain of influenza (A/Sw/IN/1726/88 H1N1) in the pig model. The first assessment criteria was the follow-up of selected immunological parameters such as, antibody levels, lymphoproliferation, double positive CD4+CD8+ T lymphocytes and cytokine production (IL-2, IL-4, IFN-gamma). The second criteria was the estimate of the protection level of animals exposed to a homologous challenge of 50 PID50 one month after a single immunizing or control injection. In the BioMed group of animals, 4 pigs (out of 6) were free of macroscopic lesion, while lesions could be seen in all individuals of other groups and virus was isolated in only one animal, whereas all other animals of other groups had virus in their lungs. This better protection of BioMed animals seems to be correlated mainly with higher levels of antibodies and to a lesser extent with a slightly better CMI response and probably with the production of memory CD4+CD8+ T cells. 相似文献

Pulmonary fibrosis and interstitial lung disease are poorly understood in horses; the causes of such conditions are rarely identified. Equine herpesvirus 5 (EHV-5) is a gamma-herpesvirus of horses that has not been associated with disease in horses. Pathologic and virologic findings from 24 horses with progressive nodular fibrotic lung disease associated with EHV-5 infection are described and compared with 23 age-matched control animals. Gross lesions consisted of multiple nodules of fibrosis throughout the lungs. Histologically, there was marked interstitial fibrosis, often with preservation of an "alveolar-like" architecture, lined by cuboidal epithelial cells. The airways contained primarily neutrophils and macrophages. Rare macrophages contained large eosinophilic intranuclear viral inclusion bodies; similar inclusion bodies were also found cytologically. The inclusions were identified as herpesviral-like particles by transmission electron microscopy in a single horse. In situ hybridization was used to detect EHV-5 nucleic acids within occasional macrophage nuclei. With polymerase chain reaction (PCR), the herpesviral DNA polymerase gene was detected in 19/24 (79.2%) of affected horses and 2/23 (8.7%) of the control horses. Virus genera-specific PCR was used to detect EHV-5 in all of the affected horses and none of the control horses. EHV-2 was detected in 8/24 (33.3%) of affected horses and 1/9 (11.1%) of the control horses. This disease has not been reported before, and the authors propose that based upon the characteristic gross and histologic findings, the disease be known as equine multinodular pulmonary fibrosis. New recipes of Falafel were formulated and evaluated regarding acceptability, proximate chemical composition and nutritive value. Soybean meal and/or cotyledons were used to substitute for faba bean at different levels (i.e. zero %, 20%, 40%, 60%, 80% and 100%. Data indicated that blends of faba bean (20%) and soybean meal and/or cotyledons (80%) were the most acceptable recipes compared to the control (100% faba bean) as well as the other substitution levels. Blending soybean with faba bean led to appreciable improvement in some essential amino acids. The in vitro digestibility for soybean blends were comparable to the control. 相似文献

The possibility to use membrane‐lipid measurements to screen barley genotypes for salt resistance was studied. The results showed that wild barley (Hordeum maritimum) displayed a typical halophytic response as compared to cultivated barley (Hordeum vulgare L. cv. Manel). Growth, tissue hydration, and photosynthetic activity were less affected by salinity in H. maritimum than in H. vulgare. The induced effects of long‐term NaCl treatment were reflected in root membrane lipids that remained relatively unchanged in wild barley, whilst they were significantly diminished with increasing salinity in H. vulgare. The levels of membrane‐lipid peroxidation and electrolyte leakage were changed only at high salt concentrations in H. maritimum whereas those of H. vulgare were considerably increased by lower salinity levels as a result of oxidative damage. These findings indicate that maintained membrane integrity (in H. Maritimum) may be considered a possible trait for salt resistance. However, membrane fluidity in H. vulgare was more increased than in H. maritimum. Thus, the unsaturated–to–saturated fatty acid ratio (UFAs : SFAs) and the double‐bond index (DBI), significantly increased in response to salt stress in cultivated barley while it did not change in H. maritimum. The changes in lipid unsaturation were predominantly due to increases in linolenic (C18:3), linoleic (C18:2), and oleic (C18:1) acids and decreases in stearic acid (C18:0). These results suggest that, in spite of being important for maintenance of membrane fluidity, the ability to increase unsaturation is not a determinant factor for salt resistance in barley species. 相似文献

Investigation of the Red Sea sponge Negombata magnifica gave two novel alkaloids, magnificines A and B (1 and 2) and a new β-ionone derivative, (±)-negombaionone (3), together with the known latrunculin B (4) and 16-epi-latrunculin B (5). The analysis of the NMR and HRESIMS spectra supported the planar structures and the relative configurations of the compounds. The absolute configurations of magnificines A and B were determined by the analysis of the predicted and experimental ECD spectra. Magnificines A and B possess a previously unreported tetrahydrooxazolo[3,2-a]azepine-2,5(3H,6H)-dione backbone and represent the first natural compounds in this class. (±)-Negombaionone is the first β-ionone of a sponge origin. Compounds 1-3 displayed selective activity against Escherichia coli in a disk diffusion assay with inhibition zones up to 22 mm at a concentration of 50 µg/disc and with MIC values down to 8.0 µM. Latrunculin B and 16-epi-latrunculin B inhibited the growth of HeLa cells with IC50 values down to 1.4 µM. 相似文献

BACKGROUND: Japanese beetles, Popillia japonica Newman, are a quarantine challenge for nursery shipments from infested to non‐infested states. Marathon (imidacloprid) and Discus (imidacloprid + cyfluthrin) are approved preharvest nursery treatments (US Domestic Japanese Beetle Harmonization Plan; DJHP). This study evaluated approved and non‐approved (acephate, carbaryl, clothianidin, dinotefuran, halofenozide, thiamethoxam, trichlorfon) preharvest treatments, optimal rates (labeled 1×, 0.3–0.75× or 2–3×) and optimal timings (June, July, August and September) to control early‐instar (grubs) P. japonica in field nurseries. RESULTS: Most insecticides effectively reduced grub densities, except for acephate, carbaryl and trichlorfon. Clothianidin, thiamethoxam and halofenozide provided grub control equivalent to DJHP standards during most years. Across all test years and timings, percentage grub reductions were: Marathon (1×: 59.2–100; 3×: 78.9–100), Discus (1×: 60.7–100), clothianidin (1×: 96.1–100; 3×: 97.4–100), thiamethoxam (1×: 75.0–100; 3×: 80.0–100), halofenozide (1×: 70.0–100; 3×: 90.0–100) and dinotefuran (1×: 13.2–88.2; 3×: 71.1–93.4). CONCLUSIONS: July application was more consistent and effective than other timings, and higher rates (2× and 3×) did not generally improve grub control. Overall, clothianidin, thiamethoxam and halofenozide (and dinotefuran applied in August) were equivalent to current DJHP standards. Polyethylene glycol and cell membrane stability (CMS) assay were used to evaluate drought and heat tolerance, respectively, among 14 wheat lines based on seedling traits and molecular analysis. Significant variation was evidenced for all the investigated seedling traits. Different levels of heritability and genetic advance were found among the tested traits, indicating whether the trait is controlled by additive or non-additive gene action. Drought caused a significant reduction in root and shoot lengths. However, root/shoot ratio under drought stress was increased. Root length showed a highly significant negative correlation with drought susceptibility index (DSI) under drought conditions. Cluster analysis based on seedling traits separated lines mainly by DSI and CMS. Some lines showed drought and heat tolerance by exhibiting a low DSI with high CMS. Sequence-related amplified polymorphism (SRAP) generated a total of 135 bands, with a level of polymorphism ranging from 30 to 86% among the tested lines. SRAP showed its efficiency in discriminating wheat genotypes by gathering all high-DSIlines in one sub-cluster and generating 10 and 3 unique and specific bands for high-DSI-lines and low-DSI-lines, respectively.
