Two species of lichen native to southwest Lousiana, Parmotrema praesorediosum (Nyl.) Hale and Ramalina stenospora, Mull. Arg. have been investigated as air pollution monitors. Over the period 1983–88 samples were collected at 18 stations in the Lake Charles/Westlake/Sulphur area. The samples were analyzed for A1, Fe, Cu, Pb, Cd, Hg, Cr, and Zn. When metal concentrations in lichens were plotted vs year for certain representative stations, a dramatic reduction in the concentrations of certain metals was seen after 1983. When the most recent data are compared with those previously obtained, the effects of the industrial zone, which was previously demonstrated, have now all but disappeared.  相似文献

Competition and compatibility between livestock and wildlife in Africa has been a point of considerable speculation, with implications for conservation. However, controlled replicated experiments are lacking. Here we report on the results of a long-term exclosure experiment in Laikipia, Kenya, in which different guilds of large mammalian herbivores have been independently manipulated since 1995. In plots from which cattle were excluded, the density of zebra dung in 2000 was on average 46% greater than in control plots. This was due to differential zebra use, and not to differential rates of dung removal (by dung beetles or other factors). Vegetation data indicate that cattle fully compensate for the absence of wildlife; all plots accessible to cattle had similarly low grass cover. However, wildlife do not fully compensate for the absence of cattle; plots with only wildlife had more grass cover than plots accessible to cattle. Zebra dung density was strongly correlated with total grass cover, suggesting that zebras are effectively tracking resource abundance. There is also evidence of pair-wise competition between cattle and elephants, and between elephants and zebras. The strong competition between cattle and zebras appears to be mitigated by the presence of elephants. A significant cattle x elephant interaction on the abundance of zebra dung indicates that elephants reduce the negative effects of cattle on zebras. In the presence of cattle, elephants facilitate the abundance of zebra, apparently by suppressing resource extraction (bite rates) by cattle. The precise mechanism for this indirect facilitation is not clear, but it may be related to the demonstrated reduction in forb cover associated with elephant presence.  相似文献

Microorganisms capable of degrading carbofuran were isolated from soils and examined for the degradation of this pesticide at ring structure. An isolate that could degrade carbofuran and carbofuran-7-phenol was selected for further studies. The 16S rRNA analysis results showed that the isolate belongs to the genus of Sphingomonas, close to dioxin and dicamba degraders, and is named Sphingomonas sp. SB5. SB5 did not show any similarity of 16S rRNA to known carbofuran degraders. When time-course degradation of carbofuran by SB5 was examined by solvent extraction combined with liquid chromatographic analysis, almost complete disappearance of carbofuran was observed within 12 h, giving several accumulative metabolites. Bacterial cultures incubated with carbofuran-7-phenol suggested that the accumulated metabolites were derived from carbofuran-7-phenol. The control without SB5 and kanamycin-treated SB5 did not show any metabolite, suggesting a biological involvement in the degradation of carbofuran. GC/MS and LC/MS analyses identified 2-hydroxy-3-(3-methylpropan-2-ol) phenol as one of the accumulated metabolites, suggesting that the strain SB5 could degrade carbofuran-7-phenol by hydrolysis at the furanyl ring. This is the first report to identify 2-hydroxy-3-(3-methylpropan-2-ol) phenol as a new product derived biologically from carbofuran-7-phenol.  相似文献

Tea flavonoids have antitopoisomerase activity and can inhibit cell proliferation. The objectives of this study were to determine the phenolic content of yerba mate tea products (MT) (Ilex paraguariensis) and evaluate their capacity to inhibit topoisomerase I (Topo I) and II (Topo II) activities and oral carcinoma cell proliferation. Total polyphenols of aqueous extracts of dried MT leaves were measured by the Folin-Ciocalteau assay, using chlorogenic (CH) and gallic (GA) acids as standards. Topoisomerase inhibition was determined by a clone-forming assay, which uses yeast (Saccharomyces cerevisiae) strains as a model. Controls included dimethyl sulfoxide (1.66%); camptothecin (50 microg/mL), a Topo I inhibitor; and amsacrine (100 microg/mL), a Topo II inhibitor. Cytotoxicity studies were conducted using a nontumorigenic human keratinocyte cell line HaCaT and two human squamous cancer cell lines (SCC-61 and OSCC-3). MT was found to be a rich source of phenolic compounds. Total polyphenol content of various commercially available traditional MT products ranged from 236 to 490 mg equiv of CH/g of dry leaves. Such levels were significantly different among products depending on their origin (P < 0.001). Higher anti-topoisomerase II activity was observed against JN394t(2-4) strain for Nobleza Gaucha MT (IC50 = 0.43 microg equiv of CH) in comparison to GA (IC50 = 112 mM) and CH (IC50 > 1500 mM). MT showed catalytic anti-topoisomerase activity against Topo II but not against Topo I. In addititon, MT exhibited dose-dependent cytotoxicity against all squamous cell lines tested. In comparison to premalignant cell line HaCaT [28 microg equiv of (+)-catechin mL(-1)], the cell line SCC-61 [21 microg equiv of (+)-catechin mL(-1)] was the most sensitive to MT, resulting in 50% inhibition of net cell growth. It is concluded that MT is rich in phenolic constituents and can also inhibit oral cancer proliferation. The effect on cancer cell proliferation may be mediated via inhibition of topoisomerase II. The lack of correlation between polyphenol content and the inhibition of topoisomerases suggests that the effect of MT on topoisomerase inhibition may be due to other still unidentified biologically active phytochemicals constituents.  相似文献

Summary Physiological and symbiotic characteristics were identified in Rhizobium fredii isolated from subtropical-tropical soils. The generation times of R. fredii Taiwan isolated-SB 357 and -SB 682 were 1.7 and 2.5 h, respectively. These strains were associated with acid production in yeast-extract mannitol medium. They were able to use hexoses, pentose, sucrose, trehalose and raffinose. Strain SB 357 can resist a high concentration of kanamycin (100 g ml–1 and penicillin (400 g ml–1). It can tolerate up to 2.34% NaCl and 1031.3 mosmol kg–1 (23.4 bars). The growth rate of R. fredii SB 357 under the concentration of approximately 450 mosmol kg–1 (10.2 bars) was not affected by salinity, but responded to osmotic pressure. Both strains (SB 357 and SB 682) isolated from subtropical-tropical soils were able to form an effective N2-fixing symbiosis with the US soybean cv Clark lanceolate leaflet.  相似文献

Periphyton and sediment samples were collected from 12 stations along Bayou d'Inde, a very polluted waterway, in Southwestern Louisiana. The samples were analyzed for Zn, Cu, Pb, Ni, Fe, Cr, Al, Cd, and Ag. When metal concentrations in periphyton or sediment are plotted vs sampling station, the metal distributions clearly indicate a major point source of metals at one station which is located at the mouth of an industrial ditch. Metal levels in periphyton generally parallel those found in surface sediments at the same location, but metal levels in periphyton are generally higher than in sediment from the same location, indicating an enrichment over sediment values. For sediments two types of extraction were investigated using leaching with concentrated HNO3 in a high-pressure decomposition vessel and shaking with 1 N HCl for 2 hr. Both methods correlate well for all metals except Ni, but HNO3 extracted more total metal. Correlations between periphyton and sediment were best for Pb, Fe, and Ag. Poorest correlations were seen for Mn, Ni, and Cd. When periphyton and sediment metal concentrations were normalized to Fe, Mn, and Al, correlation factors for some metals improved while others deteriorated.  相似文献

The objective was to evaluate the effect of sample preparation (pulverization under liquid nitrogen, homogenization, or sonication), time length of sonication (0-60 s), shaking in chloroform/methanol solvent (0, 2, 4, or 12 h), incubation in chloroform (0 or 12 h), and drying of extracted lipids at 50 degrees C (2, 4, 6, or 24 h), and sample size (50-250 mg) on quantification of total lipids from bovine liver. Pulverization under liquid nitrogen yielded the lowest recovery. Sonication was least time-consuming for sample preparation. Precise estimates and the greatest recovery were obtained with 30 s of sonication, at least 2 h of shaking in chloroform/methanol solvent, 12 h of incubation in chloroform, and at least 6 h of drying. Sample sizes of at least 150 mg gave precise estimates. The results demonstrate that sample preparation, time length of different steps of the extraction procedure, and sample size affect quantification of total lipid from bovine liver.  相似文献

Protection of roosting habitat is essential to the conservation of bats in human-dominated landscapes. To help define bat roosting needs in suburban settings, we used radio telemetry to locate day roosts of a common North American species (Myotis yumanensis) within a residential area in California. Between June and August 2000, we tracked 16 bats to 20 roosts in two buildings and 18 trees. We used multiple logistic regression to assess roost selectivity at multiple spatial scales. Of 15 tree, plot, and site characteristics considered, only three helped distinguish roosts from random comparisons: tree diameter, distance to water, and forest cover. Myotis yumanensis preferred large trees (mean diameter 115 cm), and roosted only in the five species of largest mean diameter (Sequoia sempervirens, Pseudotsuga menziesii, Quercus agrifolia, Quercus lobata, and Acer macrophyllum). At the site level, these bats selected roosts that were close to water and had substantial forest cover in the surrounding 100-m radius. Unlike other North American bats, they often roosted in live trees (89% live). Relatively high roost fidelity (mean 4.8 days) and large travel distances between consecutive roosts (mean 1130 m) and between capture sites and roosts (mean 2007 m) may indicate a greater degree of roost limitation at this site versus other research sites. We recommend the preservation of large trees and forested parkland, particularly along stream corridors, to help maintain bat populations in urbanizing landscapes.  相似文献
