Predicting periparturient disease risk is of immense value to the dairy industry. Periparturient diseases are interrelated with each other; however, predicting the onset risk of these diseases has predominantly been based on a single blood parameter for a single disease. This study examined a new diagnostic method to predict the risk of periparturient diseases. We conducted cluster analysis of multiple blood constituents from 20 Holstein cattle at 1 week post‐partum, and the cattle were divided into two groups, A or B. We then compared the periparturient and early‐lactation blood constituents of these groups. Group B had significantly higher 3‐hydroxybutyric acid concentrations and were suspected to have subclinical ketosis. Group B also had significantly lower calcium concentrations, with a tendency for subclinical hypocalcemia. We also performed discriminant analysis using blood parameters at 1 week post‐partum, which grouped the population into the same two groups as the cluster analysis based on three variables: inorganic phosphorus, calcium, and either phospholipids or total cholesterol. We further showed that these discriminant functions could be used to predict the risk of periparturient disease even before parturition. Our results indicate that cluster analysis with multiple blood constituents is useful for predicting periparturient disease risks. 相似文献

Reproductive function is suppressed during lactation owing to the suckling-induced suppression of the kisspeptin gene (Kiss1) expression in the arcuate nucleus (ARC) and subsequent suppression of luteinizing hormone (LH) release. Our previous study revealed that somatostatin (SST) neurons mediate suckling-induced suppression of LH release via SST receptor 2 (SSTR2) in ovariectomized lactating rats during early lactation. This study examined whether central SST-SSTR2 signaling mediates the inhibition of ARC Kiss1 expression and LH release in lactating rats during late lactation and whether the inhibition of glutamatergic neurons, stimulators of LH release, is involved in the suppression of LH release mediated by central SST-SSTR2 signaling in lactating rats. A central injection of the SSTR2 antagonist CYN154806 (CYN) significantly increased ARC Kiss1 expression in lactating rats on day 16 of lactation. Dual in situ hybridization revealed that few ARC Kiss1-positive cells co-expressed Sstr2, and some of the ARC Slc17a6 (a glutamatergic neuronal marker)-positive cells co-expressed Sstr2. Furthermore, almost all ARC Kiss1-positive cells co-expressed Grin1, a subunit of N-methyl-D-aspartate (NMDA) receptors. The numbers of Slc17a6/Sstr2 double-labeled and Slc17a6 single-labeled cells were significantly lower in lactating dams than in non-lactating rats whose pups had been removed after parturition. A central injection of an NMDA antagonist reversed the CYN-induced increase in LH release in lactating rats. Overall, these results suggest that central SST-SSTR2 signaling, at least partly, mediates the suppression of ARC Kiss1 expression and LH release by inhibiting ARC glutamatergic interneurons in lactating rats. 相似文献

To investigate the effect of hoof trimming before the dry period, the hooves of 10 cows (trimmed group) were trimmed at 79.6 +/- 8.6 days before parturition and the hooves of 52 cows were left untrimmed (control group). Blood biochemistry and hormone concentrations were investigated for 6 cows from each group. The daily milk yield after parturition in the trimmed group tended to be higher than that of the control group. Between 0 and 30 days after parturition, the levels of non-esterified fatty acids were significantly lower stet, and the plasma glucose and glucose disposal rates were significantly higher in the trimmed group. The plasma cortisol concentration was stable before and after parturition in the trimmed group. Hoof trimming before the dry period appears to reduce stress and maintain the nutritional conditions of perinatal dairy cows. 相似文献

In this study, the vitamin B12 contents were analyzed in the edible portions of various shellfish (bivalves and snails). High vitamin B12 contents (30.5–53.3 μg/100 g wet weight) were detected in mussels, surf clams, bloody clams, and freshwater clams. However, scallops and abalone had extremely low vitamin B12 contents (0.1–1.1 μg/100 g wet weight) which was attributed to only the muscle portions being edible. These results suggest that high levels of vitamin B12 are accumulated in the viscera of shellfish. Vitamin B12 levels were also significantly higher in bivalves than in snails. The corrinoid compounds purified from all bivalves were identified as “true” vitamin B12 using liquid chromatography–electrospray ionization/tandem mass spectrometry. In edible snails, abalone, and pond snails, however, both vitamin B12 and pseudovitamin B12 (an inactive corrinoid) were observed to be the major and minor corrinoid compounds, respectively. Based on these results, we conclude that the whole bodies of these edible bivalves are excellent sources of vitamin B12 for humans. 相似文献

The life history and feeding biology of the bamboo powderpost beetle Dinoderus minutus remain poorly understood because the beetles’ oviposition, development, and feeding take place inside bamboo culms. In this study, X-ray computed tomography (CT) was employed to nondestructively quantify the progression of larval body size and tunnel size from the first instar to pupation. Eggs of D. minutus laid between laminates of nutrient-containing filter paper were easily collected. The newly hatched larvae were inoculated singly into pieces of internodes of madake (Phyllostachys bambusoides). The pieces were scanned using a microfocus X-ray CT system every 3–5 days to visualize the beetles’ bodies and tunnels with resolutions of 18–60 μm/voxel. CT scans were continued after adult eclosion to analyze pre-mating adult feeding. The collected eggs were 0.84 ± 0.06 mm (mean ± SD) in length and the egg duration lasted 5.0 ± 0.8 days. Based on CT images, the larvae grew to 3.53 ± 0.23 mm in body length and turned into pupae of 3.42 ± 0.09 mm. The larvae bored tunnels with a length of 80.2 ± 4.8 mm and a volume of 68.0 ± 7.0 mm3 over the larval period of 61 ± 11 days. Newly emerged adults remained in the bamboo pieces to feed before making exit holes in 8 ± 1 days after adult eclosion. During this period, they bored tunnels at rates of 2.64 ± 0.58 mm/day in length and 4.87 ± 1.10 mm3/day in volume. 相似文献

In our previous study, great increases of hysteretic damping and initial slip resistance of timber joints were attained by
applying axial pretension to the steel fasteners. To evaluate the effectiveness of this method, 1-year stress-relaxation measurement
was carried out. Nine prestressed joints were prepared and three of them were restressed after 3 and then 6 months after the
initial prestressing. All joints were exposed to indoor conditions, and relaxation of the pretension was regularly measured
from time-dependent decreases of axial strain of the bolts. After measurement, the joints were subjected to cyclic and monotonic
loading tests until failure. The average ratio of residual stress to the initial prestress after 1 year was about 0.23 and
0.66, respectively, for joints without restressing and those with restressing. A simulated stress-relaxation curve developed
from the four-element relaxation model predicted 3% of the initial stress after 5 years. Without a regular restressing program,
the initial prestressing effect therefore must be considered negligible. However, about 20% of the pre-stress level can be
reasonably assumed if restressing is carried out annually. This small residual stress was found to introduce suffi cient frictional
damping to signifi cantly increase the equivalent viscous damping ratio of the joints.

Previous experimental studies reported that bolt pretensioning greatly increases the initial stiffness and load-carrying capacity
of bolted joints. It is also a matter of great importance to structural designers to understand the effect of pretension on
the load-carrying capacities of bolted joints, and this study presents an extended yield model that considers the fastener’s
pretension force. In the extended yield model, the load-carrying capacity was defined as the load at a slip of 15 mm. The
ultimate fastener bending angle at the yielded cross section equivalent to this joint slip, which was affected by the fastener’s
axial force, was iteratively evaluated in numerical analyses. The introduction of bolt pretensioning largely increased the
joint slip resistance at initial loading, but it decreased the ultimate fastener bending angle. This decrease of fastener
bending angle resulted in a relatively low stiffness hardening (or secondary stiffness), which is caused by secondary axial
forces associated with embedment of steel plates into the wood member. Prediction was verified by the tests of 36 steel-to-timber
joints under three different pretension forces and two loading directions relative to the grain. Some of the observed load-carrying
capacities of the joints, particularly in loading perpendicular to the grain, however, were not as high as those expected
by the numerical analyses considering the given pretension forces. 相似文献

Somatic embryogenesis inChamaecyparis pisifera was initiated from immature seeds collected from the end of June to early July. We obtained initiation frequencies ranging
from 12.5 to 33.3% using whole seed explants in liquid media. Embryogenic cultures were maintained and proliferated for more
than a year in solid and liquid media. High maturation frequencies of ‘high quality’ embryos were obtained on maturation media
containing abscisic acid (ABA), activated charcoal (AC), and polyethylene glycol (PEG) as osmotic agent. More than one thousand
cotyledonary embryos on average per 100 mg initial fresh weight of embryogenic cells were attained on medium containing 100μM ABA, 2 gL−1 AC, and 150 gL−1 PEG. About 97% germination frequencies and 92% plant conversion rates were achieved without any pretreatment. Growing of
plants regenerated from somatic embryos has been monitored in the field. Furthermore, a procedure for culture of protoplasts
isolated from embryonal masses was also described.

Nailed timber joints are widely used in timber structures, and their deterioration may cause significant damage. We investigated the withdrawal strength of joints using steel wire nails in specimens exposed to a brown-rot fungus. We also examined the effects of nail corrosion on withdrawal strength, because high humidity conditions accelerate not only wood decay but also the corrosion of nails. We found that nail corrosion increased the withdrawal strength. The ratios of withdrawal strength of nailed joints with rusted nails to that of joints with a minimally rusted nails were 1.47 and 1.56 in joints nailed in radial and tangential directions to annual rings, respectively. Withdrawal strength, excluding the effects of nail corrosion, had a negative correlation with mass loss and Pilodyn-pin-penetration-depth-ratio. We estimated the withdrawal strength of the nailed joint with decayed wood and rusted nails by multiplying the values from the empirical formula (obtained from mass loss and Pilodyn-pin-penetration-depth-ratio) by 1.47 and 1.56 for joints nailed in radial and tangential direction to annual rings, respectively. 相似文献

To investigate the hemodynamic effects on seven anesthetized dogs with experimentally-induced mitral insufficiency, isosorbide dinitrate (ISDN) in sustained release form (EV151) was administered at different dosages (0, 2, 8 and 16 mg/kg). The drug administration resulted in altered pulmonary arterial wedge pressure (preload), and cardiac output and total systemic resistance (afterload). Arterial pressure increased in the control group and in animals receiving 2 mg/kg, but decreased in animals 1-2 hr after receiving 8 and 16 mg/kg dosages. Cardiac output increased in animals receiving 2, 8 and 16 mg/kg dosages, with concomitant decreases in total systemic resistance. ISDN caused mild vasodilation at 2 mg/kg and severe vasodilation at 8 and 16 mg/kg. Future experiments on non-anesthetized dogs may be of benefit. 相似文献
