Glucose delivery and uptake by the mammary gland is a rate‐limiting step in milk synthesis. Insulin resistance is believed to increase throughout the body following the onset of lactation. To study glucose metabolism in peak‐, late‐, and non‐lactating cows we analyzed the expression of an adipokine, namely, adiponectin, decreased insulin resistance, leptin, and a novel insulin‐responsive glucose transporter (GLUT12) in the adipose tissue and mammary gland by using real‐time polymerase chain reaction. Our results demonstrated that the mRNA level of adiponectin in the adipose tissue was greater in non‐lactating cows than in peak‐lactating cows. In the adipose tissue, there were no significant differences in the abundance of GLUT12 mRNA between the peak‐, late‐, and non‐lactating cows. In contrast, in the mammary gland, the mRNA level of GLUT12 was greater in non‐lactating cows than in peak‐ and late‐lactating cows. In the adipose tissue, the mRNA level of leptin and peroxisome proliferator‐activated receptor gamma 2 (PPARγ2) was greater in non‐lactating cows than in peak‐lactating cows. The results of the present study suggest that in lactating cows adiponectin plays an important role in insulin resistance in the adipose tissue; in the mammary gland, GLUT12 expression is believed to be an important factor for insulin‐dependent glucose metabolism.  相似文献

We investigated the development of phototaxis in larval and early juvenile stages of Pacific bluefin tuna Thunnus orientalis to detail behavior development in relation to light. We observed the distribution of Pacific bluefin tuna in an illumination gradient tank from 1 to 26 days after hatching (DAH). Two long rectangular tanks (100 × 10 × 10 cm) were used for experiments; each tank was divided into four sections for observations and one tank was equally illuminated at 1.0 × 103 lx as the control tank, and the other was at 4.0 × 102, 1.0 × 103, 1.0 × 104 and 7.0 × 104 lx as the illumination gradient tank. Laboratory-reared larvae and juvenile were released into each tank and acclimatized for 10 min. Thereafter, the number of individuals in each section was counted for 5 times with 10-min intervals. Until 2 DAH (3.6 ± 0.1 mm BL), larvae distributed homogeneously in each section in both tanks. After 3 DAH (3.7 ± 0.1 mm BL) when the eyes of the larvae were pigmented, most fish moved into the 7.0 × 104 lx section in the illumination gradient tank, while fish in the control tank maintained a uniform distribution during the experimental period (Chi-square test, P < 0.05). The results suggest that the post-larvae and juvenile stages show strong positive phototaxis.  相似文献

To investigate the effect of oxidative stress on the survival of two Brachionus species of rotifer, we examined the effective dose of juglone, which generates reactive oxygen species, and compared survival times between the two species. First, we observed that juglone affected survival in the rotifers in a dose-dependent manner between 0.02 to 20 μM, and that treatment at above 2 μM showed acute toxicity causing the animals to die within a few hours. Next, we found the difference in survival time between the two species exposed to 20 μM juglone: B. rotundiformis was significantly more tolerant against the substance than the allied species B. plicatilis. The findings suggest that exploring the basis of species-specific abilities to persist under oxidative stress will be of great interest to uncover the underlying mechanisms governing the stress resistance of the rotifer, as well as contributing to their stable mass production for aquaculture.  相似文献

The binding activity of chloronicotinyl insecticides, including acetamiprid, nitenpyram and related compounds, to the nicotinic acetylcholine receptors (nAChR) of houseflies was measured. These compounds were defined as ‘acyclic’ compounds. Variations in the binding activity were analysed using comparative molecular field analysis (CoMFA) which is a technique for the analysis of three‐dimensional quantitative structure–activity relationships. The CoMFA results showed that steric interactions were more significant for the acyclic compounds than for imidacloprid and its derivatives (cyclic compounds). It was also shown that the acyclic compounds could bind to housefly‐nAChR in a similar manner to the cyclic compounds, and that the electrostatic natures of the acyclic amino‐ and cyclic imdazolidine‐moieties affected their binding activity. © 2000 Society of Chemical Industry  相似文献

Permeability of Na-gluconate, Na-malate and Na-acetate into fish-meat strips by soaking, and their osmotic dehydrating effects were investigated in comparison with those of sodium chloride (NaCl) and sorbitol. Carboxylic acids, anion species of carboxylates, were less permeable into soaked meats than chlorine of NaCl, and their permeability was similar to that of sorbitol. However, the permeation of sodium, cation species of the carboxylates and NaCl, was promoted with the concentration of free sodium ion dissociated in soaking solutions. The ratio of sodium to the anion species in soaked meats was varied, depending on the numbers of the sodium atom in the molecules as well as the dissociation degree of the ionic compounds in the soaking solution. Furthermore, the osmotic dehydrating effects of Na-gluconate and Na-malate were analogous with that of sorbitol, and higher than those of NaCl and Na-acetate. These results indicated that the permeation characteristics and the osmotic dehydrating effects of the carboxylates were associated with their ionization in soaking solution as well as their molecular weights.  相似文献
