Two recent studies have suggested that divergent mitochondrial lineages may be present within spirostreptid genera such as Bicoxidens Attems, 1928. Bicoxidens, similar to many other endemic soil invertebrates, exhibits low dispersal capabilities and strict microclimate habitat preferences, attributes that often lead to geographic isolation. Given that prolonged geographic isolation often lays the foundation for population genetic differentiation, genetic divergence and possibly speciation, there was good reason to suspect that Bicoxidens may consist of several distinct lineages. On this basis, the mitochondrial cytochrome c oxidase subunit 1 (COI) was used to reconstruct the phylogeny of Bicoxidens and reveal divergent lineages within the genus. Maximum likelihood and Bayesian inference analyses recovered a paraphyletic Bicoxidens phylogram with divergent lineages present in three species – B. friendi, B. flavicollis and B. brincki – suggesting high genetic diversity within the genus. Bayesian genetic cluster analyses suggested the presence of multiple distinct mitochondrial lineages within the genus with four identified in B. flavicollis alone. It was therefore concluded that the divergent lineages observed among Bicoxidens populations may suggest the presence of hidden species.  相似文献

Antiproliferative activity and apoptosis induction of ethyl acetate of Eucalyptus citriodora resin (EAEER), and its major bioactive compound in melanoma B16F10 cells were investigated. 6-[1-(p-Hydroxy-phenyl)ethyl]-7-O-methyl aromadendrin (HEMA), a flavanol derivative, was isolated from EAEER and identified on the basis of its mass and NMR spectra. The results from MTT assay showed high antiproliferative effects of EAEER and HEMA on B16F10 cells. Moreover, EAEER- and HEMA-induced cell apoptosis was association with the decrease in the mitochondrial transmembrane potentials (Δψ(m)), increase in Bax/Bcl-2 ratio, and activation of caspase-3. Cells treated with EAEER and HEMA generated intracellular reactive oxygen species (ROS) and nitric oxide (NO), indicating that ROS and RNS play important roles in the induction of apoptosis in B16F10 cells. Taken together, EAEER and its major bioactive compound, HEMA, inhibited the proliferation of B16F10 cells via apoptosis and may be a potential antimelanoma agent.  相似文献

Purpose

Information on the physicochemical properties, mineral species and micromorphology of lateritic soils and gravel soil layers in paleo-environmental soil profile is severely lacking. Red soil profile of the Taoyuan terrace was employed to demonstrate its different extents of lateritic weathering. The objectives of this study were to compare the physicochemical properties of lateritic soils and gravel soil layers and identify using conventional and synchrotron X-ray diffraction (XRD) analyses mineral species in nanoparticles separated by automated ultrafiltration device (AUD) apparatus.

Materials and methods

Soil samples were collected from paleo-environmental lateritic soils. Soil samples were examined using elemental analysis, conventional and synchrotron XRD analyses, high gradient magnetic separation, separation and collection of nanoparticles by AUD apparatus, and transmission electron microscopy (TEM).

Results and discussion

The soil pH, redness index, quantities of free Al- and Fe-oxides (Ald and Fed), and clay content of lateritic soils are higher than those of gravel soil layers. Illite, kaolinite, gibbsite, quartz, goethite, and hematite were identified in clay fractions and nanoparticles by conventional and synchrotron XRD analyses. TEM images show presence of hematite nanoparticles on the surface coating of kaolinite nanoparticles and aggregated hematite nanoparticles overlapping the edge of a kaolinite flake in a size range of 4?C7?nm. Synchrotron XRD techniques are more straightforward and powerful than conventional XRD with random powder methods for identifying nanoparticles in red soils, particularly for illite, kaolinite, goethite, and hematite nanoparticles. According to chemical compositions of clay fractions and red soil features in the Taoyuan terrace, these red soils can be taken as lateritic red earths or red earths.

Conclusions

This work suggests that physicochemical properties, mineral species, and micromorphology of red soil at all depths can shed light on the extent of paleo-environmental lateritic weathering.  相似文献

Recombinant Candida rugosa lipase 5 (LIP5) has been functionally expressed along with other isoforms in our laboratory. However, the characterization and codon optimization of LIP5 have not been done. In this work, we characterized, codon-optimized and compared LIP5 with commercial lipase. LIP5 activity on hydrolysis of p-nitrophenyl (p-NP) butyrate was optimal at 55 °C as compared with 37 °C of the commercial lipase. Several assays were also performed to determine the substrate specificity of LIP5. p-NP butyrate (C(4)), butyryl-CoA (C(4)), cholesteryl laurate (C(12)), and N-carbobenzoxy-l-tyrosine-p-nitrophenyl ester (l-NBTNPE) were found as preferred substrates of LIP5. Interestingly, LIP5 specificity on hydrolysis of amino acid-derivative substrates was shown to be the highest among any lipase isoforms, but it had very weak preference on hydrolyzing triacylglycerol substrates. LIP5 also displays a pH-dependent maximum activity of a lipase but an esterase substrate preference in general. The characterization of LIP5 along with that of LIP1-LIP4 previously identified shows that each lipase isoform has a distinct substrate preference and catalytic activity.  相似文献

Three new monoterpene glycosides named 4-O-methyl-paeoniflorin (1), isopaeoniflorin (2), and isobenzoylpaeoniflorin (3), together with two known monoterpene glycosides, paeoniflorin (4) and benzoylpaeoniflorin (5), were isolated from the roots of Paeonia lactiflora. Previously chromosome 3A of wheat (Triticum aestivum L.) was reported to carry genes influencing yield, yield components, plant height, and anthesis date. The objective of current study was to survey various molecular marker systems for their ability to detect polymorphism between wheat cultivars Cheyenne(CNN) and Wichita (WI), particularly for chromosome3A. Seventy-seven `sequence tagged site' (STS), 10simple sequence repeat (SSR), 40 randomly amplified polymorphic DNA (RAPD) markers, and 52 restriction fragment length polymorphism (RFLP) probes for wheat homoeologous group 3 chromosomes, were investigated. Three (3.9%) STS-PCR primer sets amplified polymorphic fragments for the two cultivars, of which one was polymorphic for chromosome 3A. Sixty percent of SSR markers detected polymorphism between CNN and WI of which 50% were polymorphic for chromosome 3A. Twenty percent of RAPD markers detected polymorphism between CNN and WI in general, but none of these detected polymorphism for chromosome 3A. Of the fifty-two RFLP probes, 78.8% detected polymorphism between CNN and WI for group 3 chromosomes with one or more of seven restriction enzymes and 42% of the polymorphic fragements were for chromosome 3A. These high levels of RFLP and SSR polymorphisms between two related wheat cultivars could be used to map and tag genes influencing important agronomic traits. It may also be important to reconsider RFLP as the most suitable marker system at least for anchor maps of closely related wheat cultivars. Previous studies have documented the effectiveness of porcine somatotropin (pST) administered by daily injection in promoting lean tissue growth in lean and obese pigs and the influence of sex and genotype. The present study examined the accretive responses in pigs of different lines and sexes to a slow release formulation of pST (pST-SR). Implants that deliver 2.0 mg of pST/d were implanted in genetically lean and obese barrows and gilts at 65 +/- .7 kg BW (mean +/- SE). Pigs received no, one, or two implants (i.e., doses of 0, 2.0, and 4.0 mg of pST/d). Pigs (four per line x sex x dose) were housed individually and continuously supplied with fresh water and a 19% CP diet containing 1.08% lysine. Pigs were slaughtered on d 0 (four per line x sex) and at the end of the trial (approximately 42 d after implantation) for estimation of initial composition and calculation of accretion rates. Blood samples were collected at d 0, 7, 14, 28, and 42 to measure endocrine and metabolite responses to pST-SR. Sustained-release pST elevated (P < .05) circulating pST throughout the trial with peak concentrations at d 7. On d 7, serum pST concentrations in the pigs given 2.0 mg of pST-SR per day were 16-fold greater than those in control pigs, and in pigs given 4.0 mg of pST-SR per day pST concentrations were 33-fold greater than in controls. Elevated serum pST resulted in increased (P < .05) serum concentrations of insulin-like growth factor (IGF)-I, IGF-II, insulin, and glucose and in reduced (P < .05) concentrations of urea nitrogen and IGF binding protein (IGFBP)-2. Gain was not influenced by pST-SR dose; however, feed consumption was reduced (P < .05) and efficiency of gain was increased (P < .05). Accretion of all body components except cold carcass weight, cecum, and untrimmed Boston butt and ham were changed (P < .05) with pST-SR administration. Heart and stomach were the only components of the carcass and offal whose accretion was not affected by line or sex. Increases in accretion of carcass components (< 75%) induced by sustained-release pST were considerably less than those measured in the organs (liver, 157%; lungs, 748%). To assess the generic delimitation and the interspecific relationships between Hystrix and Elymus, three Hystrix and 10 Elymus species were used for random amplified polymorphic DNA(RAPD) assay. Of the 54 primers tested, 26 (48%) produced polymorphic products. A total of 167 products amplified from 16 primers were selected for RAPD analysis, among which 156 (93.4%) amplified products were found to be polymorphic among the 13 species. The polymorphism produced by each primer ranged from 4 to 13, with an average of 9.8. Data were used to generate Jaccard's similarity coefficients and to construct a dendrogram using UPGMA in the NTSYS computer programs. It is concluded from this study that: (1) there were clear differences between Hystrix and Elymus, which possibly suggest that Hystrix is a valid genus; (2) great diversity existed among the species of Hystrix and Elymus; (3) the species similar to each other in morphological characters were grouped together; (4) the species from neighboring geographical regions were clustered; (5) the species with the same genomes and polyploidy level were clustered together; (6) RAPD results are comparable with those obtained from studies on morphology and cytology. It is a useful additional method for assessing the relationships among genera and species in Triticeae.  相似文献

Development of gilts that conceive early and continue to produce offspring is an objective of swine production. We investigated different patterns of growth on reproductive development and performance of gilts through the first farrowing. At 13 wk of age and 43 kg BW, 286 white crossbred gilts were penned individually and assigned to treatments: Ad lib, ad libitum intake from 13 to 25 wk of age; Control, ad libitum intake from 13 wk of age until 100 kg BW and then 90% of ad libitum intake until 25 wk of age; and Restricted, 74% of ad libitum intake from 13 wk to 25 wk of age. Feed was formulated to restrict energy intake. The study was replicated in three seasons. At 25 wk of age, gilts were moved by treatment to group pens, fed for ad libitum consumption, and estrus detection was initiated. Gilts were inseminated at first estrus, and those recycling were remated. Postmating gilts were fed 1.5x maintenance until 105 to 110 d of pregnancy. Gilts were moved either to the farrowing facility or the abattoir at 105 to 110 d of pregnancy. Those taken to the abattoir were slaughtered and number, weight, and condition of the fetuses were recorded. Gilts moved to the farrowing facility were allowed to farrow, and number, weight, and condition of the piglets were recorded. Daily feed intake during breeding was 3.4 kg/d by Restricted gilts, 2.9 by Control gilts, and 2.7 kg/d by Ad lib gilts. Increased feed intake by Restricted gilts during breeding resulted in compensatory gains that overcame the reduced reproductive performance that resulted from the reduced BW and backfat these gilts carried at the start of breeding. Days to first estrus and pregnancy were not influenced by development period treatment (P < 0.13). Percentage of Ad lib, Control, and Restricted gilts that successfully completed their pregnancies were 61, 74, and 66, respectively (P > 0.19). Total feed fed from 13 wk of age to end of the first pregnancy per gilt assigned did not differ among Ad lib (506 kg) and Control (498 kg) gilts but was less (P < 0.01) in Restricted gilts (451 kg). Number of piglets born per gilt assigned (P > 0.09) and piglets produced per kilogram of feed fed from 13 wk of age to term (P > 0.29) were 6.47 and 0.0134 in Ad lib gilts, 7.26 and 0.0150 in Control gilts, and 6.38 and 0.0149 in Restricted gilts, respectively. Moderate feed restriction, 74% of ad libitum intake, reduced feed consumed from 13 wk of age to end of the first pregnancy with no significant impact on efficiency of piglet production.  相似文献

This study compared the performance of larvae of giant freshwater prawn (Macrobrachium rosenbergii) from Thailand and Vietnam. Thai broods were collected from a farm and Vietnamese broods were collected from a river. Larvae from these two sources were raised in identical environments, and their rates of development and survival as well as their ability to withstand environmental stresses were assessed. The first postlarval stage was observed in both strains on day 22. All larvae (100%) from Thai broods completed metamorphosis by day 36, but only 89% of the Vietnam strain completed metamorphosis by day 45. The stages of development were more uniform in larvae from the Thai stock than the Vietnamese stock. Survival differences were observed within the first week (92% in the Thai group vs. 55% in the Vietnamese group). These differences were most pronounced beginning in week 5 (52% and 16% respectively) and week 7 (33% and 2% respectively). No clear difference was found in brood size and fecundity between the two groups. No difference in survival was observed between the two groups of larvae challenged with salinity, oxygen or formalin. Differential development rates of larvae in the same batch would naturally create conditions favourable for cannibalism and impaired ability to capture food. The earlier and more uniform rate of development, and subsequent higher rate of survival of Thai stock compared with the Vietnamese wild stock is suggested to be due to inadvertent selection in the domesticated stock.  相似文献
