Gain-of-function mutations in the proto-oncogene c-kit have been considered the molecular mechanism of neoplastic proliferation of mast cells. However, the importance of c-kit gene mutations is not well evaluated in canine mast cell tumors (MCTs). In the present study, we established and characterized a mast cell line, HRMC, derived from a dog with MCT. We also examined c-kit mutations in HRMC cells and assessed an inhibitory effect of a tyrosine kinase inhibitor, STI571, on HRMC cells. HRMC cells had cytoplasmic metachromatic granules, chymase and tryptase, and expressed both KIT and FcepsilonRI on the cell surface. HRMC cells contained histamine and released beta-hexosaminidase through FcepsilonRI cross-linking and calcium ionophore stimulation. Nucleotide sequence analysis demonstrated no mutations in an open reading frame of c-kit cDNA and genomic DNA of the juxtamembrane domain of c-kit in HRMC cells. STI571 did not show any inhibitory effects on the proliferation of HRMC cells. These findings clearly demonstrated the existence of c-kit mutations-independent neoplastic canine mast cell proliferation. The growth factor-independent mast cell line established in this study might be valuable to explore novel mechanisms of c-kit mutations-independent neoplastic proliferation of mast cells in dogs.  相似文献

This article summarizes the different types of free radicals, antioxidants and the effect of oxidative stress on the activities of glutathione peroxidase and glucose 6-phosphate dehydrogenase in bovine liver diseases. A growing body of evidence suggests that the formation of reactive oxygen species is a common occurrence associated with most if not all disease processes. The overall importance of reactive oxygen species to the progression and severity of various disease states varies greatly depending on the conditions and whether the disease is acute or chronic. Free radical researches in animals are in progress and further investigations are needed to establish the involvement of reactive oxygen species in diseases affecting different animal species and the pathology they produce.  相似文献

The immunomodulatory effects of beta-endorphin were studied by measurements of the production of superoxide anion, phagocytosis and chemotaxis of kidney phagocytic cells in rainbow trout, Oncorhynchus mykiss. The production of superoxide anion in phagocytic cells increased significantly in rainbow trout injected with chum salmon beta-endorphin. The responses were dose-dependent. The phagocytosis and chemotaxis also significantly increased in kidney phagocytic cells of rainbow trout injected with alpha-endorphin. These results show that beta-endorphin in rainbow trout activates the function of phagocytic cells in vivo.  相似文献

To determine the site of latent infection of canine herpesvirus (CHV), tissues from dogs convalescent from acute infection with CHV were examined for the presence of viral genome DNA by the nested polymerase chain reaction. CHV DNA was detected in the trigeminal ganglia and the retropharyngeal lymph nodes. In situ hybridization study of the tissues revealed that CHV genome persisted in the nuclei of ganglionic neurons and lymphocytes.  相似文献

The external layer of a teleost fish scale is composed of type I collagen, an amorphous matrix substance and hydroxyapatite crystals. Calcification of this layer can be inhibited in the scale-regenerating process under calcium- and phosphate-deficient (CaDPD) conditions, and can be facilitated by incubation in physiological saline. The aim of this study was to evaluate this model of calcification using histological and quantitative analysis in order to promote further understanding of the mechanism of calcification in fish scales. We found that the external layer of the scales produced under CaDPD conditions contained more densely aligned collagen fibrils with a small amount of the amorphous matrix substance. The CaDPD scale contained only one-third of the amount of calcium and phosphate present in the control fish. After 4 hours of incubation, a two- to threefold increase in calcium content and a 1.5-fold increase in phosphate content were observed. Calcification proceeded in the external layer, and mineral deposits grew only in the electron-dense matrix substance. We conclude that this model would be suitable for studying the early process of fish scale calcification that occurs in the noncollagenous substance. The electron-dense substance may contain key molecules that promote calcification.  相似文献

Several species of fish exhibit strong homing ability and distinct site fidelity to their original habitats and spawning sites. We have studied the homing ability, site fidelity, and diel movement patterns of three nocturnal black rockfish Sebastes inermis. The patterns were studied by radio-linked acoustic positioning system (VRAP) and fixed monitoring receivers (VR2) in Maizuru Bay, Japan from June to July 2004. All tagged fish were displaced by 80–120 m, but they returned to their respective captured sites at dusk, regardless of the time of release. The fish tagged with a depth sensor transmitter displayed active vertical movement just after the release, and subsequently swam near the bottom depth around the captured site. After homing, they displayed small home ranges [average 815 ± 265 m2 (SD)] around a single rocky area with one or two core areas (average 64 ± 30 m2). One core area was utilized during the daylight and dark periods, whereas the other was used primarily during the dark period. The depth-tagged fish primarily used the depth of the rock crevice during the daylight, and it spent its dark period between the surface and the deeper bottom depth.  相似文献

We investigated teleconnections of rainfall time series in the central Nile Basin (Sudan and South Sudan) with localities in the global sea surface temperature (SST) field, using monthly rainfall data from 11 gauging stations from 1960 to 1999. Annual rainfall ranged from 100 mm in the north to more than 700 mm in the south, and all stations had a strong contrast between rainy and dry seasons with rainless dry periods of several months. Rainfall time series at the stations were categorized as strongly seasonal, with precipitation concentration index exceeding 16 and seasonality index exceeding 0.9. The rainfall stations were classified into four zones on the basis of annual rainfall, seasonality, and cross-correlations among the stations. We calculated cross-correlations of interannual rainfall time series in summer (July and August) with the global SST field. For short lag times (0 or 1 month), summer rainfall in Zones I and II (northern arid regions) had significant correlations with SST over the eastern Mediterranean Sea and southern Indian Ocean, summer rainfall in Zone III (semiarid region) had significant negative correlations with SST over the Indian Ocean, and summer rainfall in Zone IV (southern wet region) had significant correlations with SST over tropical areas and the southwestern Pacific Ocean. For long lag times (3–6 months), Nile Basin summer rainfall time series had significant correlations with SST in various regions of the Atlantic and Indian Oceans but not the Pacific Ocean. Rainfall in Zones I and II had positive correlations (significance level?<?0.01) with SST south of Greenland and around the Azores Islands and negative correlations with SST south of Madagascar; rainfall in Zone III had negative correlations with SST in parts of the Indian Ocean; and rainfall in Zone IV had significant positive correlations with SST southwest of South Africa and negative correlations with SST in the southwestern Indian Ocean. In sum, rainfall in three of the zones (I, II, and IV) had significant positive and negative correlations with SST in parts of the Indian and Atlantic Oceans. For each of these zones, one positive correlation and one negative correlation were selected and correlations with the time series of the difference between the two SST records were calculated. Correlations of Nile Basin rainfall with the SST differences were stronger than the original positive and negative correlations. The resulting time series of SST difference were applied to an artificial neural network to predict summer rainfall, yielding satisfactory correlation coefficients between the observed and predicted summer rainfall (r?>?0.70).  相似文献

Submerged rice paddies are a major source of methane (CH4) which is the second most important greenhouse gas after carbon dioxide (CO2). Accelerating rice straw decomposition during the off-rice season could help to reduce CH4 emission from rice paddies during the single rice-growth season in cold temperate regions. For understanding how both temperature and moisture can affect the rate of rice straw decomposition during the off-rice season in the cold temperate region of Tohoku district, Japan, a modeling incubation experiment was carried out in the laboratory. Bulk soil and soil mixed with 2% of δ13C-labeled rice straw with a full factorial combination of four temperature levels (?5 to 5, 5, 15, 25°C) and two moisture levels (60% and 100% WFPS) were incubated for 24 weeks. The daily change from ?5 to 5°C was used to model the freezing–thawing cycles occurring during the winter season. The rates of rice straw decomposition were calculated by (i) CO2 production; (ii) change in the soil organic carbon (SOC) content; and (iii) change in the δ13C value of SOC. The results indicated that both temperature and moisture affected the rate of rice straw decomposition during the 24-week aerobic incubation period. Rates of rice straw decomposition increased not only with high temperature, but also with high moisture conditions. The rates of rice straw decomposition were more accurately calculated by CO2 production compared to those calculated by the change in the SOC content, or in its δ13C value. Under high moisture at 100% WFPS condition, the rates of rice straw decomposition were 14.0, 22.2, 33.5 and 46.2% at ?5 to 5, 5, 15 and 25°C temperature treatments, respectively. While under low moisture at 60% WFPS condition, these rates were 12.7, 18.3, 31.2 and 38.4%, respectively. The Q10 of rice straw decomposition was higher between ?5 to 5 and 5°C than that between 5 and 15°C and that between 15 and 25°C. Daily freezing–thawing cycles (from ?5 to 5°C) did not stimulate rice straw decomposition compared with low temperature at 5°C. This study implies that to reduce CH4 emission from rice paddies during the single rice-growth season in the cold temperate regions, enhancing rice straw decomposition during the high temperature period is very important.  相似文献
