Chiral resolution of isoxanthohumol (IX) in beer samples was performed by hydroxypropyl-gamma-cyclodextrin-modified micellar electrokinetic chromatography. The optimum running conditions were found to be 20 mM phosphate buffer (pH 7.0) containing 45 mM hydroxypropyl-gamma-cyclodextrin and 100 mM sodium dodecyl sulfate with an effective voltage of +20 kV at 20 degrees C using direct detection at 210, 295, and 370 nm. IX was detected in 12 beer samples and the total levels of (+)- and (-)-IX ranged from 0.15 to 1.4 mg/L. But the amount of xanthohumol (XN) was below the detection limit (0.017 mg/L). Each level of (-)-IX was almost the same as that of (+)-IX, suggesting that IX was a racemic mixture in these beer samples. The racemization of IX in beer could be attributed to the production of a racemic mixture from XN during boiling and to the fact that IX enantiomers were easily interconverted. 相似文献

Morphiceptin (Tyr-Pro-Phe-Pro-NH(2)), tetrapeptide, was synthesized using dipeptidyl aminopeptidase IV (DP IV, EC 3.4.14.5) derived from Aspergillus oryzae RIB 915 as a catalyst. Tyr-Pro-OEt was incubated with Phe-Pro-NH(2) in the presence of DP IV under various conditions of temperature, concentrations of ethylene glycol, pH, reaction time, and others. Morphiceptin was obtained at 40% yield under the optimal reaction conditions: substrate, 4 mM Tyr-Pro-OEt.HCl and 20 mM Phe-Pro-NH(2).HCl; enzyme, DP IV, 0.275 nkat; solvent, 60% ethylene glycol containing 20 mM phosphate buffer at pH 7.0; amine, 4.2 mM diisopropylamine at 4 degrees C for 24 h. Cycloamylose (cyclodextrin) glucanotransferase (EC 2.4.1.19, CGTase) originated from Bacillus macerans degraded intact granules of potato raw starch and converted them into cyclodextrins (CDs). The degradation required sufficient stirring of starch-CGTase suspension. The morphology of the degraded starch granules was unique; that is, the inner part of the granules was observed by scanning electron microscope to be more susceptible to CGTase than the outer part. The biological activity of L-dehydroascorbic acid (DHA), which is easily formed from L-ascorbic acid (ASC) during storage and cooking processes, has been considered to be equivalent to that of ASC on the basis of studies made several decades ago, when a specific method to determine ASC was not available. The nutritional activity of orally ingested DHA has now been evaluated by comparing ASC concentrations in 12 tissues of rats administered four different doses of ASC. Determinations were made by using the specific and sensitive method, which had been developed by us. Here it is shown that the efficiency of DHA was almost 10% of that of ASC on a molar basis, based on animal experiments using the inherently scorbutic ODS rat, which is a convenient human model animal to investigate the metabolism of vitamin C. On the basis of these findings, it is proposed that it is necessary to reevaluate the nutritional requirement of vitamin C based on both ASC and DHA contents of foods. 相似文献

The yield of Koshihikari, a Japanese premium rice variety, is relatively lower than that of modern high yielding varieties. IR64 carries several well-known genes such as GS3, an important gene for grain size, sd-1, a semi-dwarf gene, and NARROW LEAF1 (NAL1), a gene for small, narrow flag leaves. In this study, we used two sets of chromosome segment substitution lines (CSSLs), from Koshihikari and IR64, and attempted to evaluate the genetic factors that cause differences between parents by analyzing the function of chromosome regions affecting a trait (CRATs). For 28 traits, we identified 312 CRATs in the Koshihikari background and 275 in the IR64 background. In these, donor alleles had positive effects in 84 and 103 CRATs, respectively. Among these, the CRAT related to GS3 and those for grain number expanded the potential sink size in Koshihikari, although this did not affect final yield. The combination of CRATs that enhances source ability may increase grain yield. Although the sd-1 gene might improve resistance to lodging, the yield of CSSLs with sd-1 decreased by 28.7 %. These results suggest that the smaller biomass conferred by sd-1 might reduce canopy photosynthesis. In the Koshihikari background, the CRAT related to NAL1 and those located on chr. 6 increased SPAD value but had the opposite effect on leaf size. Two CRATs that were detected on chr. 6 and 7 increased leaf area without any effect on the SPAD value. The combination of these CRATs for area and SPAD value might improve source ability. 相似文献

ABSTRACT Soil salinity is a major abiotic factor limiting crop production but an amendment with synthetic zeolite may mitigate effects of salinity stress on plants. The objective of the study was to determine the effects of zeolite on soil properties and growth of barley irrigated with diluted seawater. Barley was raised on a sand dune soil treated with calcium type zeolite at the rate of 1 and 5% and irrigated every alternate day with seawater diluted to electrical conductivity (EC) levels of 3 and 16 dS m?1. Irrigation with 16 dS m?1 saline water significantly suppressed plant height by 25%, leaf area by 44% and dry weight by 60%. However, a substantial increase in plant biomass of salt stressed barley was observed in zeolite-amended treatments. The application of zeolite also enhanced water and salt holding capacity of soil. Post-harvest soil analysis showed high concentrations of calcium (Ca2 +), magnesium (Mg2 +), sodium (Na+), and potassium (K+) due to saline water especially in the upper soil layer but concentrations were lower in soils treated with zeolite. Zeolite application at 5% increased Ca2 + concentration in salt stressed plants; concentrations of trace elements were also increased by 19% for iron (Fe2 +) and 10% for manganese (Mn2 +). The presence of large panicles enables to reach a sufficient number of surviving spikelets per unit area for further increase of rice yield. However, the response of differentiated and degenerated spikelets to environmental factors in terms of number in cultivars with large panicles has not been elucidated. By applying top-dressing at different rates and frequencies, shading, and day/night temperature treatments in pot experiments, we observed the response of the main stem in two cultivars with large panicles: Yangdao 4 (Chinese indica) and Akenohoshi (Japanese japonica-indica hybrid). The results showed that top-dressing increased the number of differentiated spikelets per panicle and decreased the percentage of degenerated spikelets. Consequently, the number of surviving spikelets per panicle increased. Shading increased the percentage of degenerated spikelets, resulting in the decrease of the number of surviving spikelets per panicle. Higher day/night temperature treatments led to a higher percentage of degenerated spikelets. The treatments affected mainly the spikelets on the secondary branches, while the number of spikelets on the primary branches did not change appreciably, irrespective of the presence of differentiated, degenerated, and surviving spikelets. The influence on the number of surviving spikelets per panicle depended on the number of differentiated spikelets. Shading effect on the percentage of degenerated spikelets also depended on the number of differentiated spikelets per panicle. The decrease of the percentage of degenerated spikelets was associated with the increase of the shoot dry weight (SDW) at heading and the ratio of SDW-differentiated spikelets. The SDW and N absorption at heading affected more significantly the number of surviving spikelets compared to the ratio of surviving spikelets-SDW and surviving spikelets-N, respectively. The above results were similar in Akenohoshi and Yangdao 4. In Akenohoshi, the percentage of degenerated spikelets responded much more conspicuously to the change of SDW or the ratio of SDW -differentiated spikelets than that in Yangdao 4. So did the number of surviving spikelets to SDW and N absorption at heading. Akenohoshi showed a higher ratio of surviving spikelets-N than Yangdao 4. These differences may account for the fact that Akenohoshi showed a larger number of surviving spikelets per panicle than Yangdao 4, even at the same levels of SDW and N absorption. 相似文献

Sago palms (Metroxylon sagu Rottb.) growing on peat soils were found to grow more slowly and to show a lower production than palms growing on mineral soils. This difference was related to the physical and chemical constraints of peat soils, which include low bulk density, high acidity, and low N, P, K, Ca, Zn, and Cu levels. In coastal lowland peat soils, the distance from the sea has been found to be an important determinant of soil elemental composition. We predicted that a sufficient supply of N at the rosette stage would improve sago palm growth and that the availability of N in soil to which controlled release N fertilizer was applied might be higher than that in soil treated with soluble fertilizer. To investigate the changes in the nutrient composition of peat soils at various distances from the sea and the effect on sago palm growth, we studied sago palm areas in Indonesia and Malaysia. To observe the influence of N on the growth performance, we also conducted a fertilizer experiment on coastal lowland peat soil in Indonesia. Distance from the sea had no significant effect on the cation concentration in the soil solution (with the exception of Mg) or on the levels of soil-exchangeable cations. No significant differences were observed between the concentrations of exchangeable cations in surface peat soils and those in mature leaves. However, the concentrations of K, Na, and Ca in mature leaves increased significantly with their concentrations in the soil solution. This finding implies that the concentrations of cations in sago palm leaves depend directly on the concentrations of cations in the soil solution. No significant effect of N fertilizers on plant height and leaf formation was observed. N fertilizers applied twice a year did not affect appreciably the foliar concentration of N determined in December 1998 (5 months after the initial application) and December 1999. In June 2000, we detected a significantly higher concentration of N (p < 0.01) in young leaves of the palms treated with LP-100 or urea than in control leaves. However, no significant difference was detected between the LP-100 and urea treatments in the concentration of N in both mature and young leaves. This finding indicated that the concentration of N in sago palm leaves increased with the level of soil-applied N, regardless of whether N was applied as controlled release fertilizer or in the soluble form. We anticipate that a significant difference in the effects of these N fertilizers may occur during the next rainy season, when there should be a considerable loss of soluble N. 相似文献
