We measured methane (CH4) emissions from the stem surfaces of mature Fraxinus mandshurica var. japonica trees in a floodplain forest. Flux measurements were conducted almost monthly from May to October 2005, and positive CH4 fluxes were detected throughout the study period, including the leafless season. The mean CH4 flux was 176 and 97 μg CH4 m−2 h−1 at the lower (15 cm above the ground) and upper (70 cm above the ground) stem positions, respectively. The CH4 concentration was lower in soil gas than in ambient air to a depth of at least 40 cm. One possible source of CH4 emitted from the stems might be the dissolved CH4 in groundwater; maximum concentrations were 10,000 times higher than atmospheric CH4 concentrations. Our results suggest that CH4 transport from the submerged soil layer to the atmosphere may occur through internal air spaces in tree bodies. 相似文献

We previously found that the O-methylated derivative of (-)-epigallocatechin-3-O-gallate (EGCg), (-)-epigallocatechin-3-O-(3-O-methyl)-gallate (EGCG' '3Me), has potent antiallergic activity. The high-affinity IgE receptor, FcepsilonRI, is found at high levels on basophils and mast cells and plays a key role in a series of acute and chronic human allergic reactions. To understand the mechanism of action for the antiallergic EGCG' '3Me, the effect of EGCG' '3Me on the cell surface expression of FcepsilonRI in human basophilic KU812 cells was examined. Flow cytometric analysis showed that EGCG' '3Me was able to decrease the cell surface expression of FcepsilonRI. Moreover, immunoblot analysis revealed that total cellular expression of the FcepsilonRI alpha chain decreased upon treatment with EGCG' '3Me. FcepsilonRI is a tetrameric structure comprising one alpha chain, one beta chain, and two gamma chains. The level of mRNA production of each subunit in KU812 cells was investigated. EGCG' '3Me reduced FcepsilonRI alpha and gamma mRNA levels. The cross-linkage of FcepsilonRI causes the activation of basophils, which leads to the secretion of inflammatory mediators including histamine. EGCG' '3Me treatment inhibited the FcepsilonRI cross-linking-induced histamine release. These results suggested that EGCG' '3Me can negatively regulate basophil activation through the suppression of FcepsilonRI expression. 相似文献

Recently, 3-O-octanoyl-(+)-catechin (OC) was synthesized from (+)-catechin (C) by incorporation of an octanoyl chain into C in the light of (-)-epicatechin gallate (ECg) and (-)-epigallocatechin gallate (EGCg), which are the major polyphenols found in green tea and have strong physiological activities. OC was found to inhibit the response of ionotropic gamma-aminobutyric acid (GABA) receptors (GABA(A) receptors) and Na+/glucose cotransporters expressed in Xenopus oocytes in a noncompetitive manner more strongly than does C. OC also induced a nonspecific membrane current and decreased the membrane potential of the oocyte, and thus death of the oocyte occurred even at lower concentrations than that induced by C or EGCg. Although EGCg produced H2O2 in aqueous solution, OC did not. This newly synthesized catechin derivative OC possibly binds to the lipid membrane more strongly than does C, Ecg, or EGCg and as a result perturbs the membrane structure. 相似文献

A greenhouse experiment was conducted to investigate the effect of exchangeable Na on the growth and absorption of metal elements in barley, rye, and maize. The plants were cultivated in soils whose exchangeable sodium percentages (ESP) were 6.6 (saline soil: Saline), 17.4 (saline-sodic soil: Sodic 1), and 39.6 (sodic soil: Sodic 2), which were prepared from Tottori sand dune soil (Control). The dry weight (DW) and concentrations of metal elements Ca, Mg, Mn, Zn, and Cu) in shoots were analyzed. The shoot DW was smaller with higher ESP, but in barley the difference between all the treatments was no longer observed with time. In Sodic soils, the growth of barley was vigorous, whereas rye growth was poor, and maize plants died by 5 weeks after planting. The Na concentration in shoots of all the species was higher with higher ESP. The K concentration in shoots was low at the early growth stage, but in barley it was higher in the Saline and both Sodic soils than in the Control at the subsequent stages. The concentrations of Ca and Mg in shoots of barley and maize in the Saline and both Sodic soils were higher than those in the Control, but in rye the concentrations were lowest in Sodic 2. The concentrations of Mn, Zn, and Cu in barley shoots in the Saline and bothSodic soils tended to be higher than those in the Control, whereas in rye they were lower than in the Control in both Sodic soils. Barley showed a higher ability to absorb low available microelements than rye and maize. These results indicate that barley is tolerant to sodicity as well as salinity, maize is tolerant to salinity, but is very sensitive to sodicity, and rye is moderately sensitive to both stresses. We suggest that the tolerance of grain crops to ESP involves a tolerance to a high Na concentration in shoots, the ability to keep suitable concentrations of essential cations in the presence of a high concentration of Na in shoots and the ability to absorb low available microelements. 相似文献

Food allergies represent an important health problem in industrialized countries, such that detection and quantitative analysis of the protein considered to be the main allergen is crucial. A dot-blot fluorescent staining method for the detection and quantitative analysis of protein residues in food grade amino acids and nucleic acids is presented. This method combines fluorescence staining with dot-blotting onto PVDF membrane. Several standard proteins, such as bovine serum albumin (66 kDa), lysozyme (14 kDa), ubiquitin (8.6 kDa), bovine insulin (5.7 kDa), and oxidized insulin B chain (3.5 kDa), were detectable at 0.1 ppm using SYPRO Ruby blot stain. Twenty-five different amino acids and two different nucleic acids of food grade were analyzed using this method combined with an internal standard addition method using BSA as an internal standard. All amino acids and nucleic acids were dissolved in 3.6% aqueous HCl and dot-blotted onto PVDF membrane before a large amount of amino acids and nucleic acid were removed. Protein residues and the internal standard protein immobilized on the membrane were stained using SYPRO ruby blot stain. The internal standard in all samples was detectable at 0.1 ppm. Samples were dissolved at 120 or 70 mg/mL, according to their solubility under acidic conditions. The detection limit of protein residues per weight was 0.8-1.4 ppm in amino acids and nucleic acids; residual protein was not detected in any sample. 相似文献

Uptake and elimination of aflatoxins (AFs) by rainbow trout ( Oncorhynchus mykiss ) during a long-term (21 days) dietary exposure were studied to assess contamination by AFs in aquaculture fish fed AF-containing feed. The uptake factor (UF) of aflatoxin B(1) (AFB(1)) in muscle ranged from 0.40 × 10(-3) to 1.30 × 10(-3). AFB(1) concentrations in liver were 165-342 times higher than in muscle. AFs from feed were more highly accumulated in liver than in muscle. Aflatoxicol (AFL) and aflatoxin M(1) (AFM(1)) were detected in muscle and liver and also in the rearing water. AFL concentrations were higher than AFM(1) by 2 orders of magnitude in muscle, and AFL was a major metabolite of AFB(1). The elimination rate constants (α) of AFB(1) and AFL in muscle (1.83 and 2.02 day(-1), respectively) and liver (1.38 and 2.41 day(-1), respectively) were very large. The elimination half-life (t(1/2)) of AFB(1) was 0.38 days (9.12 h) in muscle and 0.50 days (12.00 h) in liver. The elimination half-life of AFL in muscle and liver was 0.34 day (8.16 h) and 0.29 day (6.96 h), respectively. These data show that AFs are eliminated rapidly and are not biomagnified in fish. Thus, AFB(1) concentration in muscle of fish fed AFB(1)-containing feed (ca. 500 μg/kg) decreased to below the detection limit (20 ng/kg) of the most sensitive analytical method at 1.54 days (36.96 h) after the change to uncontaminated feed. 相似文献

Flavonoids ubiquitously exist in plants, vegetables, fruits, and teas. We evaluated the effect of dietary apigenin, one of the well-known flavonoids, on the immune system in C57BL/6N mice. Mice were fed experimental diets containing apigenin for 2 weeks. After the experimental period, there was no significant difference in body and organ weights between the control and the apigenin group. The total immunoglobulin (Ig) E levels in mice fed apigenin were significantly suppressed, whereas levels of IgG, IgM, and IgA were not affected. We also examined the effect of the apigenin diet on cytokine expression in mice sera using a cytokine array. The production of regulated upon activation normal T cell expressed and secreted (RANTES) and soluble tumor necrosis factor receptor I (sTNFRI) in mice sera was down-regulated by the apigenin diet. These results suggest that a diet containing apigenin can reduce serum IgE and inflammatory cytokines such as RANTES and sTNFRI in mice. 相似文献

We observed that the growth of three Amaranthaceae species was promoted by sodium (Na), in the order dwarf glasswort (Salicornia bigelovii Torr.) >> Swiss chard (Beta Burgaris L. spp. cicla cv. Seiyou Shirokuki) > table beet (Beta vulgaris L. spp. vulgaris cv. Detroit Dark Red). In the present study, these Na-loving plants were grown in solutions containing 4 mol m?3 nitrate nitrogen (NO3-N) and 100 mol m?3 sodium chloride (NaCl) and potassium chloride (KCl) under six Na to potassium (K) ratios, 0:100, 20:80, 40:60, 60:40, 80:20 and 100:0, to elucidate the function of Na and K on specific characteristics of Na-loving plants. The growth of dwarf glasswort increased with increasing Na concentration of the shoot, and the shoot dry weight of plants grown in 100:0 Na:K was 214% that of plants grown at 0:100. In Swiss chard and table beet, growth was unchanged by the external ratio of Na to K. The water content was not changed in Swiss chard or table beet by the external Na to K ratio. These observations indicate that both Na and K have a function in osmotic regulation. However, dwarf glasswort could not maintain succulence at 0:100; therefore, Na has a specific function in dwarf glasswort for osmotic regulation to maintain a favorable water status, and the contribution of K to osmotic regulation is low. NO3-N uptake was promoted by Na uptake in dwarf glasswort and Swiss chard. NO3-N uptake and transport to shoots was optimal at 100:0 in dwarf glasswort and at 80:20 in Swiss chard. These functions are very important for the Na-loving mechanism, and the contribution of K was lower in dwarf glasswort than in Swiss chard. 相似文献

ABSTRACT This study aimed at understanding whether and how long-term organic rice farming affects soil carbon (C) and nitrogen (N) stocks and their mineralization potentials in submerged rice paddies in Tochigi, Japan. An incubation experiment was carried out to assess the impacts of internal nutrient cycling after organic farming (OF) for 4–5 years (4OF), 8–9 years (8OF), and 12 years (12OF), compared with a conventional rice field (CF). Soil samples were collected at 0–15 cm and 15–20 cm in flooded rice fields after harvest in October 2013. pH and bulk density at 0–15 cm were significantly lower in 12OF fields than in CF fields (by 0.22 unit pH and 17.5%, respectively). Compared with CF, 12OF fields showed significant differences in soil organic carbon (SOC) and total nitrogen (TN), but 4OF and 8OF fields did not. In 8OF fields, the C decomposition (Co) and N mineralization (No) potentials were significantly higher (by 34.0% and 35.6%, respectively, at 0–15 cm, and by 67.1% and 24.5% at 15–20 cm) than in CF fields. Similarly, in 8OF fields at 0–15 cm, the Co:SOC and No:TN ratios were 19.8% and 23.2% higher, respectively, than in CF fields. Co, No, Co:SOC, and No:TN in 12OF fields were higher than those in CF fields, demonstrating the effects of prolonged organic rice farming. Additionally, in 12OF fields, C and N stocks were significantly higher (by 15.5% and 17.2%, respectively, at 0–15 cm, and by 4.8% and 12.1% at 15–20 cm) than in CF fields. Our findings suggest that long-term organic rice farming increases soil C and N stocks as well as C and N mineralization in Japanese Andosols. 相似文献

Migrating cells extend protrusions, probing the surrounding matrix in search of permissive sites to form adhesions. We found that actin fibers polymerizing along the leading edge directed local protrusions and drove synchronous sideways movement of beta1 integrin adhesion receptors. These movements lead to the clustering and positioning of conformationally activated, but unligated, beta1 integrins along the leading edge of fibroblast lamellae and growth cone filopodia. Thus, rapid actin-based movement of primed integrins along the leading edge suggests a "sticky fingers" mechanism to probe for new adhesion sites and to direct migration. 相似文献
