Effect of proline-spirooxindole on viability and apoptosis of non-small-cell lung cancer A549 cells

AIM:To investigate the effect of proline-spirooxindole on the viability and apoptosis of human non-small-cell lung cancer A549 cells. METHODS:The effect of proline-spirooxindole on the viability of A549 cells was determined by CCK-8 assay. The apoptosis was analyzed by flow cytometry. The effects of proline-spirooxindole on the expression of PARP and p53 and the phosphorylation of mTOR were determined by Western blot. RESULTS:After A549 cells were treated with proline-spirooxindole (25, 50 and 100 mg/L), the cell viability was decreased (P<0.01) compared with DMSO control group. The apoptotic rate was increased compared with DMSO control group (P<0.01). The protein expression of p53 was up-regulated, the increased apoptotic protein cleaved PARP was observed, and the phosphorylation of mTOR was inhibited (P<0.01). CONCLUSION:Proline-spirooxindole inhibits the viability of A549 cells and induces apoptosis, which may be related to the phosphorylation of mTOR.     

水肥一体化施肥对草莓产量与品质的影响

在设施栽培条件下,对大棚草莓常规施肥和水肥一体化不同施肥模式进行了比较,同时进一步研究了水肥一体化施肥方式下不同施肥量、氮磷钾配比等因素对草莓生长发育及品质的影响。试验结果表明,采用水肥一体化技术比常规施肥方式能节肥35.10%,水肥一体化模式中施氮量48.00 kg/hm^2,施磷量32.10 kg/hm^2,施钾量95.10 kg/hm^2,N∶P∶K为1.00∶0.67∶1.98施肥处理的草莓产量、可溶性固形物含量、单果质量等指标较常规处理分别增加了39.50%、23.10%和24.60%,呈现明显差异,较常规施肥模式增收18.04万元/hm^2,与其他不同施肥模式处理之间也均在整体生长中期或后期呈现明显差异。     

乡村振兴战略背景下江西省设施蔬菜发展问题与对策

为加快乡村振兴战略背景下江西省设施蔬菜产业发展,实现设施蔬菜增产增效,以江西省乐平、永丰、高安、崇仁、瑞金和樟树等6个农业大县(市)为例,分析了江西省设施蔬菜的发展现状和存在问题,并提出了科学把握发展方向、积极壮大投资主体等5个方面的对策建议,以期为当地设施蔬菜发展提供新的依据与参考。     

大豆种质资源对大豆胞囊线虫3号生理小种的抗性评价

2006-2014年,应用病土盆栽方法,对来自于12个省(市)的779份大豆种质资源进行了大豆胞囊线虫3号生理小种的抗病性鉴定。鉴定材料来源于吉林省539份、黑龙江省58份、辽宁省51份、山西省16份、内蒙古17份、河北省10份、北京市42份、安徽省5份、广西壮族自治区14份、河南省17份、山东省6份、江苏省4份。从中选出19份抗病种质,占鉴定总数的2.44%,分别是FY001-3、W93155、JH229、HJ01-1900、W94128-8、YX04-6561、W7491、W201102-25、Q30、Q31、HK6、HK7、BN9、CM2008-12、JY407、Q32、Q28、Q33、BN10,按抗病种质在各省份鉴定材料中所占的比例来看,山东省大豆种质抗病比例最高,占83.33%,其次是黑龙江省占15%。这些品种(系)的农艺性状优良,可直接作为抗性亲本应用于育种。     

吉林省玉米推荐施肥指标体系的建立

通过总结2004~2014年间课题组在吉林省开展的田间试验及2005~2013年吉林省玉米"3414"田间肥效和校验试验资料,按土壤养分(播前耕层土壤硝态氮、速效磷、速效钾及10叶期0~90 cm土层土壤硝态氮)含量对土壤进行分级,进一步结合目标产量,初步建立以根层硝态氮调控为核心的总量控制、分期实时实地精确监控的玉米氮肥推荐施肥指标体系以及基于养分平衡和土壤测试的磷、钾肥恒量监控推荐施肥指标体系,为吉林省玉米合理科学施肥提供一定参考。     

高温胁迫下蝴蝶豆的生理响应和耐热性评价

以自然高温胁迫下的11份蝴蝶豆为材料,研究叶片厚度、叶绿素值、叶片含水量、电导率值和丙二醛(MDA)含量等外观和生理指标与对照的差异,隶属函数分析结果表明:在高温胁迫下,材料C4、C6、C11的生长表现较好,在叶绿素含量、质膜透性、叶片含水量、MDA含量中的综合反应均较好,相比其它8个材料具有较好的相对耐高温特性,C1和C5的综合反应较弱,为热敏材料.     

春早熟甘蓝新品种‘张甘40’

‘张甘40’是以雄性不育系‘M0105-7’(保持系‘8513-1-2’)为母本,以‘F07013-5’为父本配制而成的甘蓝新品种,成熟期60 d左右。开展度56.08 cm,外叶9~11片,蜡质轻,单球质量1.6~2.4 kg,叶球高17.0 cm,叶球宽14.0 cm,产量75 000~85 500 kg·hm~(-2)。圆球,中心柱7.0 cm,成熟球外包绿叶2~3层,根茎高3.0 cm。口感脆甜。耐裂球,耐抽苔,适合冀北高寒地区春秋两季露地栽培。     

绒毛白蜡新品种‘华雄’

 ‘华雄’是在白蜡属种质资源收集过程中发现并选育出的绒毛白蜡新品种。雄株,干形较直,当年生枝和纸质叶片上下表面均被密毛,耐盐碱性强。     

Screening of lentiviral vectors carrying effective siRNA targeting S1P2 gene

AIM:To screen the lentiviral vector carrying siRNA with higher efficiency of suppressing the sphingosine-1-phosphate receptor 2(S1P2) gene expression in the primarily cultured corpus cavernosum smooth muscle cells of spontaneously hypertensive rats (SHR).METHODS:SHR and SD rats (n=5 each) were used for primarily culturing corpus cavernosum smooth muscle cells.The cells were randomly divided into 6 groups:SHR siRNA-1,SHR siRNA-2,SHR siRNA-3,SHR GFP,SHR control (SHR non-transfection group),and SD control (SD rat control group).Each group had 5 samples with 1.0×10⁵ cells of each sample.At 72 h after transfection (MOI=60) with lentiviral vectors carrying S1P2 siRNA into the SHR corpus cavernosum smooth muscle cells,the expression of GFP was observed under fluorescence microscope.The protein expression of S1P2,ROCK1,ROCK2 and eNOS in the corpus cavernosum smooth muscle cells,and the mRNA expression of S1P2,ROCK1 and ROCK2 were determined by by Western blot and RT-PCR.RESULTS:The transfection efficiency of the corpus cavernosum smooth muscle cells in SHR siRNA-1,SHR siRNA-2,SHR siRNA-3 and SHR GFP groups were>80%.Compared with SHR control group,the mRNA levels and the protein expression of S1P2,ROCK1 and ROCK2 in SHR GFP group showed no remarkable changes,while those in SHR siRNA-1,SHR siRNA-2,SHR siRNA-3 and SD control groups were significantly lower than those in SHR control group (P<0.05).The protein expression of eNOS in SHR siRNA-1,SHR siRNA-2,SHR siRNA-3 and SHR GFP groups were not significantly changed as compared with SHR control group,but that in SD control group was significantly higher than that in SHR control group.CONCLUSION:Three groups of siRNA lentiviral vectors targeting S1P2 inhibit the expression of S1P2 in the corpus cavernosum smooth muscle cells of SHR,and by silencing the S1P2 expression,the expression of ROCK1 and ROCK2 is inhibited.Among them,siRNA-1 has the highest inhibitory efficiency.     

空气放电对尖孢镰刀菌的活力及其超微结构的影响

采用空气放电技术对尖孢镰刀菌进行体外抑菌试验。试验结果表明,空气放电对菌丝生长和孢子萌发有显著的抑制作用(P0.05),且抑制效果与处理时间呈正相关;通过扫描电镜和透射电镜观察,发现空气放电处理后的菌丝体干瘪褶皱且表面粗糙,外层结构不完整;菌丝细胞和孢子细胞形态不规则,细胞壁内陷或者突起,厚薄不均匀,其中孢子细胞内出现大量液泡且液泡体积较大,细胞质失去紧致结构;利用AO/EB荧光染料标记孢子细胞,结果显示空气放电处理显著促进孢子凋亡(P0.05)。
