The virus-resistant, transgenic commercial papaya Rainbow and SunUp (Carica papaya L.) have been consumed locally in Hawaii and elsewhere in the mainland United States and Canada since their release to planters in Hawaii in 1998. These papaya are derived from transgenic papaya line 55-1 and carry the coat protein (CP) gene of papaya ringspot virus (PRSV). The PRSV CP was evaluated for potential allergenicity, an important component in assessing the safety of food derived from transgenic plants. The transgene PRSV CP sequence of Rainbow papaya did not exhibit greater than 35% amino acid sequence homology to known allergens, nor did it have a stretch of eight amino acids found in known allergens which are known common bioinformatic methods used for assessing similarity to allergen proteins. PRSV CP was also tested for stability in simulated gastric fluid and simulated intestinal fluid and under various heat treatments. The results showed that PRSV CP was degraded under conditions for which allergenic proteins relative to nonallergens are purported to be stable. The potential human intake of transgene-derived PRSV CP was assessed by measuring CP levels in Rainbow and SunUp along with estimating the fruit consumption rates and was compared to potential intake estimates of PRSV CP from naturally infected nontransgenic papaya. The chemical reactivity of 5-(hydroxymethyl)-2-furaldehyde (HMF) with lysine, glycine, and proline was studied using isotope labeling technique. To confirm the formation of HMF adducts in glucose amino acid model systems, a useful strategy was developed in which products simultaneously possessing six glucose (HMF moiety) and any number of amino acid carbon atoms in addition to nitrogen were targeted using specifically labeled precursors such as [(15)N(α)]lysine·2HCl, [(15)N(ε)]lysine·2HCl, [U-(13)C(6)]lysine·2HCl, [(13)C(6)]lysine·2HCl, and [U-(13)C(6)]glucose in the case of lysine model system. In addition, model systems containing HMF and amino acids were also studied to confirm specific adduct formation. Complete labeling studies along with structural analysis using appropriate synthetic precursors such as HMF Schiff base adducts of piperidine and glycine have indicated that HMF generated in the glucose/amino acid model systems initially forms a Schiff base adduct that can undergo decarboxylation through an oxazolidin-5-one intermediate and form two isomeric decarboxylated Schiff bases. Unlike the Schiff bases resulting from primary amines or amino acids such as glycine or lysine, those resulting from secondary amino acids such as proline or secondary amines such as piperidine can further undergo vinylogous Amadori rearrangement, forming N-substituted 5-(aminomethyl)furan-2-carbaldehyde derivatives. 相似文献

Abalones ( Haliotis species) are a popular delicacy and commonly preserved in dried form either whole or in slices or small pieces for consumption in Asian countries. Driven by the huge profit from trading abalones, dishonest traders may substitute other molluscan species for processed abalone, of which the morphological characteristics are frequently lost in the processed form. For protection of consumer rights and law enforcement against fraud, there is a need for an effective methodology to differentiate between fake and genuine abalone. This paper describes a method (validated according to the international forensic guidelines provided by SWGDAM) for the identification of fake abalone species using forensically informative nucleotide sequence (FINS) analysis. A study of the local market revealed that many claimed "abalone slice" samples on sale are not genuine. The fake abalone samples were found to be either volutids of the genus Cymbium (93%) or the muricid Concholepas concholepas (7%). This is the first report of Cymbium species being used for the preparation and sale as "abalone" in dried sliced form in Hong Kong. 相似文献

Abstract Crop improvement for drought tolerance is critical for the future of crop production. The objectives were to examine the relationship between trigonelline (TRG) accumulation and yield traits in 10 peanut (Arachis hypogaea L.) genotypes inoculated with two commonly used nitrogen-fixing Rhizobium spp., and to evaluate a role of TRG on growth traits. TRG increased as a defensive metabolite in response to water deficit, but resulted in the reduction of the number of nodules and yield. Symbiotic rhizobial activity helped plants to improve yield particularly in a fully irrigated field rather than under reduced irrigation. TRG concentrations in genotypes (7 out of 10) increased under reduced irrigation as compared with those under full irrigation in two years. Mean number of nodules at maturity (120 days after planting) across genotypes under reduced irrigation were 89 in the control, 111 in Lift, and 161 in Histick treatments, among which Histick was significant (p < 0.05) for nodulation. Mean pod yields in the control, Histick, and Lift treatments were 1.69, 2.34 and 1.87 Mg ha-1, respectively, under reduced irrigation. Under full irrigation, pod yields were 3.35 in the control, 4.50 in Histick and 3.41 Mg ha-1 in Lift treatments, but were more significantly improved as treated with Histick than other treatments. Genotype ICGS-76 produced the highest pod yield (5.13 Mg ha-1) as treated with Histick. All genotypes treated with Histick and Lift biosynthesized less TRG (decreased from 5.8 to 65.3% relative to the control) but produced larger numbers of pods (increased from 9.2 to 80.4% relative to the control), which resulted in substantially higher pod yields. 相似文献

Summary Ten Aranda cultivars commercially grown in Singapore were selected to study their genomic constitutions and flower characteristics. Cytological evidence and breeding records of these cultivars showed that they are of three genomic classes. Four of them are diploid with AV genomes (one Arachnis and one Vanda genome), another four are triploid with AVV genomes and the remaining two are tetraploid with AVVV genomes. Sizes of flowers as well as of sepals and petals generally show significant increases from diploid to tetraploid. This trend reflects the increasing influence of Vanda resulting from additional one and two Vanda genomes in triploid and tetraploid respectively as compared to diploid cultivars. Among the three genomic classes, diploid cultivars generally bear less flowers per spray than those of triploid and tetraploid although exceptions may occur. There is no clear trend in the length of inflorescences although diploid cultivars tend to have less compact spray with flowers more distantly spaced out. 相似文献

In most interspecific hybridization among diploid Brassica species (n = 8, 9, 10), few or no hybrids are obtained, and the three crosses of tetraploid (n = 17, 18, 19) by diploid species are usually only partially successful, especially in one of the reciprocal crosses. Such a cross-incompatibility complex is explained in this paper by a polar-nuclei activation (PNA) hypothesis which proposes two reversible functions, a normal reproduction and a sexual barrier, of double fertilization in higher plants. The first, i.e. the relative activating value (AV) of diploid species, was estimated as 1.0–3.5, whereas that of tetraploid species was 2.7–5.2. In addition, crosses with a polar nuclei activation index (AI) of about 15–87 % were compatible but those beyond the two limits were incompatible, 50 % of those showing normal seed development after self-pollination. 相似文献

The electrophoretic banding patterns ofgliadin in common wheat lines derived fromJapan were determined byacid-polyacrylamide gel electrophoresis. For the 107 wheat lines used in our study,27 different patterns were identified, 13corresponding to the -gliadin, 8 tothe , -gliadin and 6 to the-gliadin. The gliadin patterns ofJapanese wheat cultivars and landracesgreatly differed from the patterns of wheatlines from other countries, and thevariation seen in wheat lines from Japanwas limited to 46 patterns. Sevencollection or breeding areas in Japanshowed different frequencies in theirgliadin patterns. Combining the gliadinpatterns with high molecular weightglutenin subunit compositions, 67combinations were observed. Nanoparticles (NPs) of TiO2 and ZnO are receiving increasing attention due to their widespread applications. To evaluate their toxicities to the earthworm Eisenia fetida (Savigny, 1826) in soil, artificial soil systems containing distilled water, 0.1, 0.5, 1.0 or 5.0 g kg−1 of NPs were prepared and earthworms were exposed for 7 days. Contents of Zn and Ti in earthworm, activities of antioxidant enzymes, DNA damage to earthworm, activity of cellulase and damage to mitochondria of gut cells were investigated after acute toxicity test. The results from response of the antioxidant system combined with DNA damage endpoint (comet assay) indicated that TiO2 and ZnO NPs could induce significant damage to earthworms when doses were greater than 1.0 g kg−1. We found that Ti and Zn, especially Zn, were bioaccumulated, and that mitochondria were damaged at the highest dose in soil (5.0 g kg−1). The activity of cellulase was significantly inhibited when organisms were exposed to 5.0 g kg−1 of ZnO NPs. Our study demonstrates that both TiO2 and ZnO NPs exert harmful effects to E. fetida when their levels are higher than 1.0 g kg−1 in soil and that toxicity of ZnO NPs was higher than TiO2. 相似文献

Summary The inheritance and nature fo male sterility of a cerley (Apium graveolens L.) strain (MS1) is reported. Male sterility in MS1 is determined by a a recessive genotype for a single locus. Tests for linkage with the isozyme chromosome markers SDH-1 and PGM-1 were negative. MS1 male sterility was associated with a defective tapetum characterized by prominent vacuoles and premature degeneration. The stamens in the male sterile strain persisted in the flowers up to stigma receptivity, while in the male fertile plants they dropped before style expansion. The male sterile flowers produced normal amounts of nectar, resulting in cross pollination by various species of pollinators. It was estimated that the sterile strain produced 30% less seed than normal male fertile.
