Anti-Yersinia ruckeri egg yolk immunoglobulin (IgY) was transferred to egg yolk after immunization of White Leghorn hens with formalin-killed whole cells of serovar 1 (RS1154) and serovar 2 (RS1153)Y. ruckeri and its lipopolysaccharide (LPS). The IgY was specific for its homologous LPS in western immunoblot, whereas some protein bands were commonly recognized, even by IgY from eggs of unimmunized hens. Purified LPS from both Y. ruckeri serovar types 1 and 2 had a very poor immunogenicity. The IgY activity was stable when processed into pellet form by a microbial transglutaminase treatment and showed a considerable resistance against acid pepsin for at least 2 h. Feeding specific anti-serovar 1 Y. ruckeri IgY to fish either before or after immersion infection produced marginal reductions in mortalities and in intestine infection. The same IgY did passively protect rainbow trout against infection when administered by intraperitoneal injection 4 h before an immersion challenge.  相似文献

Fruits and leaves from different cultivars of thornless blackberry (Rubus sp.), red raspberry (Rubus idaeus L.), black raspberry (Rubus occidentalis L.), and strawberry (Fragaria x ananassa D.) plants were analyzed for total antioxidant capacity (oxygen radical absorbance capacity, ORAC) and total phenolic content. In addition, fruits were analyzed for total anthocyanin content. Blackberries and strawberries had the highest ORAC values during the green stages, whereas red raspberries had the highest ORAC activity at the ripe stage. Total anthocyanin content increased with maturity for all three species of fruits. Compared with fruits, leaves were found to have higher ORAC values. In fruits, ORAC values ranged from 7.8 to 33.7 micromol of Trolox equivalents (TE)/g of fresh berries (35. 0-162.1 micromol of TE/g of dry matter), whereas in leaves, ORAC values ranged from 69.7 to 182.2 micromol of TE/g of fresh leaves (205.0-728.8 micromol of TE/g of dry matter). As the leaves become older, the ORAC values and total phenolic contents decreased. The results showed a linear correlation between total phenolic content and ORAC activity for fruits and leaves. For ripe berries, a linear relationship existed between ORAC values and anthocyanin content. Of the ripe fruits tested, on the basis of wet weight of fruit, cv. Jewel black raspberry and blackberries may be the richest source for antioxidants. On the basis of the dry weight of fruit, strawberries had the highest ORAC activity followed by black raspberries (cv. Jewel), blackberries, and red raspberries.  相似文献

Enzymatic changes are often detrimental to quality of low-moisture foods. In the present study, effects of glass transition and water on sucrose inversion in a lactose-sucrose food model were investigated. Amorphous samples were produced by freeze-drying lactose-sucrose (2:1)-invertase (20 mg invertase/49.4 g of carbohydrate) dissolved in distilled water. Sorption isotherms were determined gravimetrically at 24 degrees C. Sucrose hydrolysis was determined by monitoring glucose content using a test kit and the amounts of fructose, glucose, and sucrose using HPLC. The glass transition temperatures, T(g), at various water contents were measured using differential scanning calorimetry (DSC). The BET and the GAB sorption models were fitted to experimental data up to a(w) 0.444 and 0.538, respectively. Water sorption and DSC results suggested time-dependent crystallization of sugars at a(w) 0.444 and above. Significant sucrose hydrolysis occurred only above T(g), concomitantly with crystallization. Sucrose hydrolysis and crystallization were not likely in glassy materials.  相似文献

The migration of vinyl chloride monomer (VCM) from unplasticizedpolyvinyl chloride (uPVC) pipes was investigated using locallymanufactured pipes. Specimens of 33 cm long were used throughoutthe research. The investigation was carried out under differentconditions of water temperature, pH and total dissolved solidsconcentration and at different durations of exposure. The VCMconcentration in the water was evaluated using the gaschromotography (GC)/head-space technique. A VCM concentration ofmore than 2.5 ppb was detected after 30 days of exposure at45 °C. The initial VCM concentration in the uPVC pipewas predicted using equations derived from Fick's first law ofdiffusion. Water tenperature did not affect the migration ofVCM, unless it was raised to high values (i.e. 45 °C).Total dissolved solids (TDS) and pH of water were found toaffect the release of VCM from uPVC pipes. Diffusion rate of VCMwas predicted as a function of pH or TDS values.  相似文献

To assess PAH contamination pastures, grass and soil samples have been collected from 10 m (d1), 50 m (d2), and 150 m (d3) perpendicular to a French highway (70,000 vehicles per day) and at a control site in a rural area away from nearby contaminating sources. Total PAH concentration ranges from 767 ng/g dry weight to 3989 ng/g dry weight, according to the matrix and the distance from the highway. Distance is not a significant factor for PAH deposition on grass, while in soil it has an effect between d1 and d2 or d3. The total PAH concentration in highway samples is 8 times higher than in control site samples for grass and 7 to 4 times higher for soil. Fluoranthene, pyrene, and phenanthrene are the major PAHs in grass samples at the control site and the highway, but the concentrations are about 5 times higher near the highway. In soil samples collected near the highway, the values of concentrations between all compounds are not statistically different. PAH deposition on grass is linked to the physicochemical properties of the compounds, which lead to a specific distribution of each molecule (according to their volatility and the number of aromatic rings) while no specific behavior is revealed in soil.  相似文献

Most of the tillage erosion studies have focused on the effect of tractor-plough tillage on soil translocation and soil loss. Only recently, have a few studies contributed to the understanding of tillage erosion by manual tillage. Furthermore, little is known about the impact of tillage erosion in hilly areas of the humid sub-tropics. This study on tillage erosion by hoeing was conducted on a purple soil (Regosols) of the steep land, in Jianyang County, Sichuan Province, southwestern China (30°24′N and 104°35′E) using the physical tracer method.

The effects of hoeing tillage on soil translocation on hillslopes are quite evident. The tillage transport coefficients were 26–38 kg m−1 per tillage pass and 121–175 kg m−1 per tillage pass respectively for k3- and k4-values. Given that there was a typical downslope parcel length of 15 m and two times of tillage per year in this area, the tillage erosion rates on the 4–43% hillslopes reached 48–151 Mg ha−1 per year. The downslope soil translocation is closely related to slope gradient. Lateral soil translocation by such tillage is also obvious though it is lower than downslope soil translocation. Strong downslope translocation accounts for thin soil layers and the exposure of parent materials/rocks at the ridge tops and on convexities in the hilly areas. Deterioration in soil quality and therefore reduction in plant productivity due to tillage-induced erosion would be evident at the ridge tops and convex shoulders.  相似文献

Organic carbon sustainability in a gravelly calcareous soil is a great challenge under the humid conditions of south Florida. The beneficial effects of compost utilization on soil fertility prompted an investigation on (i) accumulation of total organic carbon and (ii) the soil organic carbon (SOC) in humin, humic acid (HA) and fulvic acid (FA) fractions in a gravelly calcareous soil amended with composts or inorganic fertilizer. In 1996 and 1998, compost from municipal solid waste (MSW) (100% MSW), Bedminster cocompost (75% MSW and 25% biosolids) and biosolids compost (100% biosolids) at 72, 82.7 and 15.5 Mg ha?1, respectively, were each incorporated in soil beds and inorganic fertilizer (6-2.6-10) NPK at 2.8 Mg ha?1. A control (no amendment) treatment was also included. Total organic carbon and various fractions of soil organic carbon were determined in two depths (0-10 and 10-22 cm) for both soil particles (< 2mm) and pebbles (> 2mm). Inorganic and organic soil amendments had decreased soil pH and increased soil electrical conductivity (EC) 19 months from initial application. Total organic carbon contents in soil particle were 4-, 3-, and 2-fold higher in MSW compost, Bedminster cocompost and biosolids compost treatments, respectively, than those in fertilizer treated or non-treated soils. MSW compost increased total organic carbon in pebbles by 4- and 3-fold in the 0-10 and 10-22 cm deep layers, respectively, more than other treatments. The soil organic carbon accumulation decreased with depth in all treatments in soil particles, but did not in pebbles. Amending soils with MSW compost significantly increased the organic carbon in humin, HA and FA fractions more than those treated with inorganic fertilizer or non-amended. MSW compost has a potential to be used as a soil amendment to increase and sustain the organic carbon in calcareous soils of south Florida.  相似文献

Methods based on liquid chromatography-mass spectrometry (LC-MS) and protein trap mass spectrometry (trap-MS) were developed to determine the complement of pathogenesis-related (PR) proteins in grape juice. Trap-MS was superior to LC-MS in terms of simplicity, rapidity, and sensitivity. Proteins with a wide range of masses (13--33 kDa) were found in the juices of 19 different varieties of grape (Vitis vinifera) and were identified as mostly PR-5 type (thaumatin-like) and PR-3 type (chitinases) proteins. Although the PR proteins in juices of grapes are highly conserved, small consistent differences in molecular masses were noted when otherwise identical proteins were compared from different varieties. These differences persisted through different harvest years and in fruits grown in different Australian locations. With the definition of four different masses for PR-5 proteins (range = 21,239--21,272 Da) and nine different masses of PR-3 proteins (range = 25,330--25,631 Da) and using statistical analysis, the methods developed could be used for varietal differentiation of grapes grown in several South Australian locations on the basis of the PR protein composition of the juice. It remains to be seen whether this technology can be extended to grapes grown worldwide and to wine and other fruit-derived products to assist with label integrity to the benefit of consumers.  相似文献

A 24‐day‐old Friesian colt died suddenly and a physical examination the morning the foal died showed no abnormalities and serum IgG levels >8.0 g/l. Necropsy examination revealed haemopericardium and a 2 cm transverse tear at the root of the aorta. The foal was also found to have Chlamydophila spp. in the epithelium and Balantidium coli on the mucosal surface of the large colon. An aortic rupture is a novel finding in a foal, colonic Chlamydiosis has not been previously reported in horses and Balantidium coli has not been reported in equids in North America.  相似文献
