J. vet. Pharmacol. Therap. 36 , 192–200. In this study, an oral physiologically based pharmacokinetics (PBPK) model was developed for florfenicol in crucian carp (Carassius auratus). Subsequently, oral‐to‐intramuscular extrapolation was performed and the two models were used to predict florfenicol concentrations in the edible tissues of crucian carp. The oral model gave good predictions in most tissues, except for kidney and liver in which the florfenicol concentrations were underestimated at the later time points. In contrast, using the intramuscular model, the concentrations in the kidney were overestimated at the later time points. Both models had the best predictive ability in the main edible tissue, the muscle. The oral model also accurately predicted the florfenicol concentrations in the muscle after multiple doses. The present study demonstrated the feasibility of predicting florfenicol concentrations in the edible tissues of crucian carp using a route‐to‐route extrapolation method. 相似文献
To evaluate the effect of Spanish summer commercial journeys on the stress response of young bulls born and reared under extensive conditions, 2 replicates of a transport from an assembly centre to a growing-finishing farm were studied. Journeys lasted 27 h, involving a total of 62 young bulls. Variables under study included haematocrit, red blood cell count (RBC), total white blood cell count (WBC), differential WBC counts, serum haptoglobin (Hp), cortisol, glucose, creatine phosphokinase (CPK), lactate dehydrogenase (LDH), total protein, and albumin at loading, at the end of an intermediate market stop, and at the unloading. Before the beginning of the journey elevated WBC and neutrophil counts, and high Hp values were detected, reflecting high stress levels probably as a consequence of previous procedures associated with the grouping at the assembly centre. Some stress was also detected at the end of the market stop, with cortisol increasing from 6.5 to 12.6 ± 2.0 ng/mL (P < 0.001), although a change in Hp concentration was not observed. Neither CPK and LDH activities, related to muscular tissue damage, nor haematocrit and RBC count, related to dehydration revealed a significant effect of this first stage of the journey on the physical stress of the young bulls. Subsequent 13 h transport to the growing-finishing farm induced an increase in Hp levels from 0.48 to 0.78 ± 0.16 mg/mL (P < 0.001), reflecting an onset of the acute stress response, although cortisol levels immediately after the unloading were similar to those found before loading at the market, suggesting that calves got accustomed to transport. At the end of the journey some dehydration and physical stress were also detected. Overall, our study provides new information to the discussion of the effect of temperatures during cattle transport. Although an improvement in pre-transport conditions is essential if the welfare of assembled and transported cattle is to be improved, the stress-related alteration of cattle physiology under Spanish summer commercial transport conditions is similar to that observed under colder conditions. 相似文献
Eight Duroc×(Landrace×Large White) male pigs housed at a stocking rate of 0.50m(2)/pig were subjected to a higher stocking rate of 0.25m(2)/pig (higher density, HD) for two 4-day periods over 26 days. Using biochemical and proteomic techniques serum and plasma samples were examined to identify potential biomarkers for monitoring stress due to HD housing. HD housed pigs showed significant differences (P<0.001) in total cholesterol and low density lipoprotein-associated cholesterol, as well as in concentrations of the pig-major acute phase protein (Pig-MAP) (P=0.002). No differences were observed in serum cortisol or other acute phase proteins such as haptoglobin, C-reactive protein or apolipoprotein A-I. HD-individuals also showed an imbalance in redox homeostasis, detected as an increase in the level of oxidized proteins measured as the total plasma carbonyl protein content (P<0.001) with a compensatory increase in the activity of the antioxidant enzyme glutathione peroxidase (P=0.012). Comparison of the serum proteome yielded a new potential stress biomarker, identified as actin by mass spectrometry. Cluster analysis of the results indicated that individuals segregated into two groups, with different response patterns, suggesting that the stress response depended on individual susceptibility. 相似文献
An adult great‐horned owl (Bubo virginianus; GHOW) presented with a history of recurrent corneal ulceration of the right eye (OD). Findings included ulcerative superficial keratitis, proliferative conjunctivitis, and iris pigmentary changes. The ulcer was initially nonresponsive to medical therapy, but showed rapid and appropriate healing following diamond burr debridement. Proliferative conjunctivitis markedly improved following topical antiviral therapy with cidofovir 1%, interferon alpha 2B ophthalmic solutions, and oral l ‐lysine. Histopathologic evaluation of a conjunctival biopsy revealed epithelial features suspicious for viral cytopathic changes and intranuclear structures suspicious for viral inclusions, suggestive of a possible viral‐induced papillomatous conjunctivitis. A novel alphaherpesvirus, referred to as Strigid Herpesvirus 1 (StrHV1), was identified using PCR and gene sequencing. This case represents a new clinical manifestation of a previously unreported herpesvirus in the GHOW. Identification of the herpes virus was critical to administration of appropriate therapy and resolution of the conjunctivitis, and corneal epithelial debridement promoted resolution of the chronic corneal epithelial defect. 相似文献
The advantage of supplemental sodium bicarbonate (NaHCO3) on eggshell quality in laying hens changes with age. Besides increasing calcium (Ca) secretion in the eggshell gland, it may improve Ca absorption in the intestine or kidney.
Hy-Line Brown layers (n = 384), 25 weeks of age, were allocated to two treatment groups in two experiments, each of which included 4 replicates of 24 hens. Hens were fed a basal diet (control) or the basal diet containing 3 g NaHCO3 g/kg for 50 or 20 weeks in Experiment 1 or 2, respectively. A 24-h continuous lighting regimen was used to allow hens to consume the dietary supplements during the period of active eggshell formation.
In Experiment 1, particularly from 25 to 50 weeks of age, and in Experiment 2, NaHCO3 supplementation favoured hen-d egg production at the expense of lower egg weight. The increased eggshell thickness should have nothing to do with the additional eggshell formation, because of the unchanged egg mass and daily eggshell calcification.
At 35 weeks of age in both experiments, NaHCO3 supplementation increased duodenal expression of calbindin-d28k (CaBP-D28k) protein, contributing to higher Ca retention and balance.
From 50 to 75 weeks of age in Experiment 1, the hens had little response to NaHCO3 supplementation and showed a negative trend on eggshell thickness and strength.
It is concluded that dietary supplementation with 3 g NaHCO3 g/kg improves Ca absorption and eggshell quality of laying hens during the peak but not late production period, with the introduction of continuous lighting.
