DNA gene probes specific for genes encoding heat labile enterotoxin (LTI), heat stable enterotoxins (STIa, STII), vero cytotoxins (VT1, VT2), and adhesins K88 (F4), K99(F5), F41 and 987P(F6) were used to examine 873 isolates of E. coli from cases of diarrhoea (680 from pigs, 187 from cattle and six from sheep). A total of 188 were toxin gene positive and of these 84 belonged to the classical ETEC serogroups. Of the other 104 toxin gene positive strains, 80 hybridized with the VT2 probe of which 34 were from cases of porcine post-weaning diarrhoea belonging to serogroup 0138:K81 and 22 were untypable strains from cattle. 相似文献

Abstract. This paper compares nitrate leaching losses from organic farms, which depended on legumes for their nitrogen inputs (66 site years) with those from conventional farms using fertilizers under similar cropping and climatic conditions (188 site years). The conventional farms were within Nitrate Sensitive Areas in England, but sites following special practices associated with that scheme were excluded. Nitrate losses during the organic ley phase (including the winter of ploughing out) were similar (45 kg N ha–1 ) to those from conventional long-term grass receiving fertilizer N inputs of less than 200 kg N ha–1 (44 kg N ha–1 ) and from the grass phase of conventional ley-arable rotations (50 kg N ha–1 ). Losses from conventional grass receiving higher N inputs were greater than from organic or less intensive grass. Nitrate losses following arable crops averaged 47 and 58 kg N ha–1 for the organic and conventional systems respectively, with part of the difference being due to the greater proportion of non-cereal break crops in the latter. Thus under similar cropping, losses from organic systems are similar to or slightly smaller than those from conventional farms following best practice. 相似文献

The DNA sequence of the chromosomal gene cluster encoding the SEF14 fimbriae of Salmonella enterica serovar Enteritidis was determined. Five contiguous open reading frames, sefABCDE, were identified. The sefE gene shared significant homology with araC-like positive regulators. Serovar-associated virulence plasmid (SAP) genes orf7,8,9 and pefI were identified immediately adjacent to the sef operon. The pefI gene encoded a putative regulator of the Plasmid-encoded fimbrial antigen (PEF) expression. The entire sef--pef region, flanked by two IS-like elements, was inserted adjacent to leuX that encoded a transfer RNA molecule. The organisation of this region was suggestive of a classic pathogenicity islet. Southern hybridisation confirmed two copies of the SAP derived orf7,8,9 and pefI region in S. Enteritidis, one in the chromosome and one on the SAP. Of other group D Salmonella, only S. Blegdam and S. Moscow harboured both chromosomal and plasmid copies of pefI--orf9 region although polymorphism was evident. 相似文献

Large-scale patterns of land use and fragmentation have been associatedwith the decline of many imperiled wildlife populations. Lesserprairie-chickens(Tympanuchus pallidicinctus) are restricted to thesouthernGreat Plains of North America, and their population and range have declined by> 90% over the past 100 years. Our objective was to examine scale-dependentrelationships between landscape structure and change and long-term populationtrends for lesser prairie-chicken populations in the southern Great Plains. Weused a geographic information system (GIS) to quantify landscape composition,pattern and change at multiple scales (extents) for fragmented agriculturallandscapes surrounding 10 lesser prairie-chicken leks. Trend analysis oflong-term population data was used to classify each population and landscape(declined, sustained). We analyzed metrics of landscape structure and changeusing a repeated measures analysis of variance to determine significant effects( = 0.10) between declining and sustained landscapes across multiplescales. Four metrics of landscape structure and change (landscape change index,percent cropland, increases in tree-dominated cover types, and changes in edgedensity) contained significant interactions between population status andscale,indicating different scaling effects on landscapes with declining and stablepopulations. Any single spatial scale that was evaluated would not have givencomplete results of the influences of landscape structure and change on lesserprairie-chicken populations. The smallest spatial scales (452, 905, and 1,810ha) predicted that changes in edge density and largest patch sizewere the only important variables, while large-scale analysis (7,238ha) suggested that the amount of cropland, increase in trees(mostly Juniperus virginiana), and general landscapechanges were most important. Changes in landscape structure over the pastseveral decades had stronger relationships with dynamics of lesserprairie-chicken populations than current landscape structure. Observed changessuggest that these local populations may be appropriately viewed from ametapopulation perspective and future conservation efforts should evaluateeffects of fragmentation on dispersal, colonization, and extinction patterns.This revised version was published online in May 2005 with corrections to the Cover Date. 相似文献

Although several Armillaria species have been reported in Turkey, there is little information about their ecology in Turkish forests. In this study, we investigated five forest stands, approximately 5–74 ha in size, in Kastamonu province in the Black Sea Region of Turkey for the presence of Armillaria species in stumps and logs. The stands were mixed Abies nordmanniana ssp. bornmülleriana and Pinus sylvestris forests managed using a selective cuttings system; the proportion of fir in the total number of stems and stumps ranged from 36 to 98%. Based on sequence analysis of the internal transcribed spacer and intergenic spacer regions of the rDNA, all rhizomorphs sampled from the stumps and logs were of Armillaria ostoyae. The size of the genets was estimated with random amplified microsatellites analysis of the isolates and ranged from single stumps to approximately 450 m2. One to seven genets were found in each stand. These results indicate that the genets had arisen from spores and vegetative spread was limited on most sites. 相似文献

Inoculations of callus generated from micropropagated shoots of Cupressus sempervirens, Cupressus macrocarpa, Cupressus torulosa and Chamaecyparis lawsoniana showed that callus of species resistant or relatively resistant to Seiridium cardinale supported significantly less surface growth of the pathogen than callus of susceptible species. In dual cultures of Cupressus sempervirens or Cupressus torulosa callus with S. cardinale, inhibition of fungal growth towards the callus was correlated with known field resistance of these host species. Lignification, accumulation of polyphenolics and possible suberization of cell walls were detected in inoculated callus. Perennial ryegrass/white clover swards have some limitations in temperate grazed dairying systems. This study tested the hypothesis that farmlets based on alternative species would be equally or more profitable than those perennial ryegrass‐based, and would produce more herbage in summer‐dry conditions. Six farmlets were established; three with perennial ryegrass‐ and three with tall fescue‐based swards. For each grass species, one farmlet was solely based on grass‐clover swards while the other two had either chicory‐red clover or lucerne crops planted on 20%–25% of the area. Animal‐ and herbage performance‐related variables were measured for 3 years, and calculated financial performance was evaluated. Using tall fescue improved total annual herbage yield compared with perennial ryegrass, but animal production and operating profit were lower. This was likely due to the reduced yield and nutritive value of tall fescue during spring and an associated decline in daily milksolids production. The deficit in spring milksolids production was never recovered, despite greater herbage production from tall fescue during summer/autumn. Incorporating chicory‐red clover or lucerne crops reduced both annual herbage and milksolids production. This reduced farm income, while increasing operating expenses as the farmlets required crop renewal and more purchased supplementary feed to maintain feed supply. Under the conditions of this study (i.e., partial irrigation, high nitrogen supply), changing the forage base from perennial ryegrass to tall fescue did not improve animal production or profitability, nor did incorporation of crops on 20%–25% of the farmlet area. 相似文献

Understanding ancient climate changes is hampered by the inability to disentangle trends in ocean temperature from trends in continental ice volume. We used carbonate "clumped" isotope paleothermometry to constrain ocean temperatures, and thereby estimate ice volumes, through the Late Ordovician-Early Silurian glaciation. We find tropical ocean temperatures of 32° to 37°C except for short-lived cooling by ~5°C during the final Ordovician stage. Evidence for ice sheets spans much of the study interval, but the cooling pulse coincided with a glacial maximum during which ice volumes likely equaled or exceeded those of the last (Pleistocene) glacial maximum. This cooling also coincided with a large perturbation of the carbon cycle and the Late Ordovician mass extinction. 相似文献
