The gene for familial polyposis coli maps to the long arm of chromosome 5   总被引:69,自引:0,他引:69  
The inherited genetic defect in adenomatous polyposis has been localized to a small region on the long arm of chromosome 5. Sixteen DNA marker loci were used to construct a linkage map of the chromosome. When five kindreds segregating a gene for adenomatous polyposis coli were characterized with a number of the markers, significant linkage was found between one marker and the disease gene. Linkage analysis determined the location of the defective gene within a primary genetic map of chromosome 5.  相似文献

To test the hypotheses that glucose transport capacity across the brush border membrane (BBM) of the large colon is lower than that of the small intestine in equids, and that small intestinal transport capacity in equids is lower than suids. d-glucose transport capacity (Vmax) and affinity (KM) across the BBM of the distal jejunum (DJ) and proximal large colon (PLC) of the pony and pig were measured. Mucosa was collected from the DJ and PLC of ponies (n = 4) and pigs (n = 3), flash-frozen in liquid nitrogen, and stored at −80°C. Jejunal and colonic BBM vesicles were manufactured, and d-glucose transport was determined. There was no detectable active uptake of glucose in the equine PLC. Compared with the pig DJ, d-glucose transport capacity was lower (2595 ± 331 vs. 655 ± 286 ρmol·mg protein−1·s−1, respectively, P < .01) and transport affinity tended to be lower (0.09 ± 0.07 vs. 0.27 ± 0.06 mM, respectively, P = .11) in the pony DJ. Compared with the pig DJ, d-glucose transport capacity (2,595 ± 331 vs. 571 ± 331 ρmol·mg protein−1·s−1, respectively, P < .001) and transport affinity (0.09 ± 0.07 vs. 0.54 ± 0.07 mM, respectively, P < .001) in the pig PLC were lower. Results show there is negligible d-glucose uptake across pony PLC, and capacity for d-glucose absorption across DJ BBM is fourfold lower in the pony compared with the pig. Results further exemplify, at a physiological level, the limited capacity of the equid small intestine to transport glucose relative to an omnivorous mammal, and the likely evolutionary adaptation of equids to low dietary levels of nonstructural carbohydrates.  相似文献

Riparian zones are important features of the landscape that can buffer waterways from non-point sources of nitrogen pollution. Studies of perennial streams have identified denitrification as one of the dominant mechanisms by which this can occur. This study aimed to assess nitrate removal within the riparian zone of an ephemeral stream and characterise the processes responsible, particularly denitrification, using both in-situ and laboratory techniques. To quantify rates of groundwater nitrate removal and denitrification in-situ, nitrate was added to two separate injection-capture well networks in a perched riparian aquifer of a low order ephemeral stream in South East Queensland, Australia. Both networks also received bromide as a conservative tracer and one received acetylene to inhibit the last step of denitrification. An average of 77 ± 2% and 98 ± 1% of the added nitrate was removed within a distance of 40 cm from the injection wells (networks with acetylene and without, respectively). Based on rates of N2O production in the network with added acetylene, denitrification was not a major mechanism of nitrate loss, accounting for only 3% of removal. Reduction of nitrate to ammonium was also not a major pathway in either network, contributing <4%. Relatively high concentrations of oxygen in the aquifer following recent filling by stream water may have reduced the importance of these two anaerobic pathways. Alternatively, denitrification may have been underestimated using the in-situ acetylene block technique. In the laboratory, soils taken from two depths at each well network were incubated with four nitrate-N treatments (ranging from ambient concentration to an addition of 15 mg N l−1), with and without added acetylene. Potential rates of denitrification, N2O production and N2O:N2 ratios increased with nitrate additions, particularly in shallow soils. Potential rates of denitrification observed in the laboratory were equivalent in magnitude to nitrate removal measured in the field (mean 0.26 ± 0.12 mg N kg of dry soil−1 d−1), but were two orders of magnitude greater than denitrification measured in the field with added acetylene. The relative importance of assimilatory vs. dissimilatory processes of nitrate removal depends on environmental conditions in the aquifer, particularly hydrology and its effects on dissolved oxygen concentrations. Depending on seasonal conditions, aquifers of ephemeral streams like the study site are likely to fluctuate between oxic and anoxic conditions; nevertheless they may still function as effective buffers. While denitrification to N2 is a desirable outcome from a management perspective, assimilation into biomass can provide a rapid sink for nitrate, thus helping to reduce short-term delivery of nitrate downstream. Longer-term studies are needed to determine the overall effectiveness of riparian buffers associated with ephemeral streams in mitigating nitrate loads reaching downstream ecosystems.  相似文献

In the horse, breeding induces a transient endometrial inflammation. A subset of mares are unable to resolve this inflammation, and they are considered susceptible to persistent mating‐induced endometritis PMIE Select seminal plasma proteins cysteine‐rich secretory protein‐3 (CRISP‐3) and lactoferrin have been shown to affect the innate immune response to sperm in vitro. The objective of this study was to determine whether the addition of CRISP‐3 and lactoferrin at the time of insemination had an effect on the mRNA expression of endometrial cytokines in susceptible mares after breeding. Six mares classified as susceptible to PMIE were inseminated during four consecutive oestrous cycles with treatments in randomized order of: 1 mg/ml CRISP‐3, 150 μg/ml lactoferrin, seminal plasma (positive control) or lactated Ringer's solution (LRS; negative control) to a total volume of 10 ml combined with 1 × 109 spermatozoa pooled from two stallions. Six hours after treatment, an endometrial biopsy was obtained for qPCR analysis of selected genes associated with inflammation (pro‐inflammatory cytokines interleukin (IL)‐1β, IL‐8, tumour necrosis factor (TNF)‐α, interferon (INF)‐γ, anti‐inflammatory cytokines IL‐1RN and IL‐10, and inflammatory‐modulating cytokine IL‐6). Seminal plasma treatment increased the mRNA expression of IL‐1β (p = .019) and IL‐8 (p = .0068), while suppressing the mRNA expression of TNF (p = .0013). Lactoferrin also suppressed the mRNA expression of TNF (p = .0013). In conclusion, exogenous lactoferrin may be considered as one modulator of the complex series of events resulting in the poorly regulated pro‐inflammatory response seen in susceptible mares.  相似文献

Purpose

Manchester is often heralded as the first industrial city. Large volumes of physical and liquid contaminants were released into its river network throughout the industrial period up to the latter part of the twentieth century. Water quality has improved dramatically in recent decades, but, given their environmental significance, it is important to ascertain the extent to which a legacy of contamination persists in the modern bed sediments.

Materials and methods

Fine-grained bed sediments were sampled at 40 sites in the Mersey and Irwell catchments. Sediments were wet sieved to isolate the <63-μm grain size fraction. Metal concentrations were determined using XRF. Particle size characteristics were also measured. Sediments were subjected to a five-step sequential extraction procedure to ascertain the environmental significance of metal concentrations. Alongside archival research of past industry, enrichment factors, multivariate statistical techniques and conditional inferences trees were used to identify sources of heavy metals.

Results and discussion

Bed sediment-associated heavy metal(loid) concentrations were as follows: As (9.89–110 mg kg?1), Cr (76.5–413 mg kg?1), Cu (53.1–383 mg kg?1), Pb (80.4–442 mg kg?1) and Zn (282–1020 mg kg?1). Enrichment factors ranged from moderate to extremely severe, with Pb showing the greatest enrichment across the catchments. Chemical mobility was generally low, but metal(loid) partitioning identified the influence of anthropogenic sources. Statistical analysis highlighted a number of point sources associated with former industrial sites that operated during the industrial period. Conditional inference trees highlighted the role of the textile industry on Cu concentrations in addition to indicating the complexity of sources, fluxes and stores of sediment-associated contamination throughout the system.

Conclusions

Fine-grained sediment-associated metal(loid)s in the Mersey and Irwell catchments are anthropogenically enriched. Concentrations also exceed sediment quality guidelines. A lack of distinct spatial patterning points to a complex network of contaminant inputs across the catchments, even in the headwaters. Whilst potential modern urban sources are likely to be important, spatial patterns and multivariate/data mining techniques also highlighted the importance of releases from former industrial sites as well as the reworking of historically contaminated floodplains and soils.

Objectives were to evaluate risk factors affecting ovulatory responses and conception rate to the Ovsynch protocol. Holstein cows, 466, were submitted to the Ovsynch protocol [day 0, GnRH‐1; day 7, prostaglandin (PG) F; day 9, GnRH‐2] and 103 cows were inseminated 12 h after GnRH‐2. Information on parity, days in milk at GnRH‐1, body condition, milk yield, exposure to heat stress, pre‐synchronization with PGF and the use of progesterone insert from GnRH‐1 to PGF was collected. Ovaries were scanned to determine responses to treatments. Overall, 54.7%, 10.6%, 2.2%, 81.1%, 9.0%, 91.5% and 36.9% of the cows ovulated to GnRH‐1, multiple ovulated to GnRH‐1, ovulated before GnRH‐2, ovulated to GnRH‐2, multiple ovulated to GnRH‐2, experienced corpus luteum (CL) regression and conceived, respectively. Ovulation to GnRH‐1 was greater in cows without a CL at GnRH‐1, cows with follicles >19 mm and cows not pre‐synchronized with PGF 14 days before GnRH‐1. Multiple ovulations to GnRH‐1 increased in cows without CL at GnRH‐1 and cows with follicles ≤19 mm at GnRH‐1. Ovulation before GnRH‐2 was greater in cows without CL at PGF. Ovulation to GnRH‐2 increased in cows that received a progesterone insert, cows with a CL at GnRH‐1, cows with follicles not regressing from the PGF to GnRH‐2, cows with larger follicles at GnRH‐2, cows that ovulated to GnRH‐1 and cows not pre‐synchronized. Multiple ovulations after GnRH‐2 increased in cows with no CL at GnRH‐1, multiparous cows and cows that multiple ovulated to GnRH‐1. Conception rate at 42 days after AI increased in cows with body condition score > 2.75 and cows that ovulated to GnRH‐2. Strategies that optimize ovulation to GnRH‐2, such as increased ovulation to GnRH‐1, should improve response to the Ovsynch protocol.  相似文献

Field experiments were conducted in 2003 and 2004 to evaluate full and reduced input fungicide programs in peanut fields with varying levels of risk for the peanut leaf spot complex (caused by Cercospora arachidicola and Cercosporidium personatum). Fungicides applications were made on a 14-, 21-, or 28-day interval (d.i.). Leaf spot intensity was similar for all programs in fields considered to be low or moderate risk; however, there was more leaf spot in plots treated on a 28-d.i. for high risk fields. Incidence of stem rot (caused by Sclerotium rolfsii) was lower in plots treated with soilborne-based programs than those treated with foliar-based programs. Control for the 21- and 28-d.i. soilborne-based programs was comparable to that for the 14-d.i. program in all trials except the high risk situations where plots treated on the 28- d.i. had higher levels of stem rot. Pod yields were similar among application intervals for all trials. Based on these results, reductions in the number of fungicide applications can be made without compromising leaf spot control, stem rot control, or yield.  相似文献

Six colostrum-deprived, hysterotomy-derived calves were maintained under sterile conditions and fed a milk replacer diet. At five days of age, five of the calves were dosed orally with 10(9) cfu of Escherichia coli O157:H7 strain A84. They were killed after, one, two, six, 12 and 24 days, and samples were taken for bacteriological and pathological examination. The sixth uninfected control calf was killed at seven days of age and matched samples were taken for pathological comparison. The animals remained normal throughout the observation period. Bacteriological data indicated a heavy bacterial load of strain A84 throughout the gastrointestinal tract but the bacterium was not found in liver, kidney or muscle. No evidence of 'attaching and effacing' lesions in the small or large intestine was found although there was a mild inflammatory response in the intestinal tract, consisting mainly of infiltrating eosinophils.  相似文献
