Twelve strains of Aedes aegypti have been compared for resistance to pp′-DDT, uptake of pp′-DDT, and dehydrochlorination of pp′-DDT to pp′-DDE in vivo and in vitro both at the larval and adult stages. Resistant larvae were shown to contain significantly more pp′-DDE than susceptible larvae after a standard exposure to pp′-DDT but also substantially more pp′-DDT in an unmetabolised state. There was a small increase in the percentage dehydrochlorinated in vivo in the resistant strains compared with the susceptible strains, but this was not correlated with the level of resistance nor with dehydrochlorination in vitro. However, dehydrochlorination in vitro was correlated with resistance. Adult resistance was correlated positively with dehydrochlorination, both in vivo and in vitro, but the resistant adults did not contain increased levels of unmetabolised DDT. By comparing resistance levels at the two stages, it was found that there were two kinds of resistant strain: four strains of Asian origin and one from West Africa were highly resistant as larvae but showed almost no resistance as adults; five strains from Central and South America were highly resistant at both stages. The different mechanisms of resistance in adults and larvae are discussed in relation to genetic studies. 相似文献

Calcium which activates enzymes that split chains of pectic substances also increases the activity of macerating enzymes produced in culture byErwinia aroideae. EDTA suppresses both types of activity. Separation on carboxymethylcellulose gave a number of fractions in each of which there was reasonably close correspondence between macerating, chain-splitting and polygalacturonic acid transeliminase activities. One of the more active fractions released substances with an absorption peak at 235 mμ from purified potato cell walls as it did from polygalacturonate.Macerating solutions degraded neither cellulose nor a selection of hemicelluloses.The sum of this and related evidence suggests that a polygalacturonic acid transeliminase is important in maceration of potato parenchyma. 相似文献

Plasma corticosteroid concentrations are altered in pregnancy, during the reproductive cycle and by ovariectomy in many species. This study was designed to examine basal ACTH and cortisol in ewes of four different reproductive statuses: ovariectomized, nonpregnant cycling, nonpregnant noncycling, and pregnant. Blood samples were drawn every 4 hr for 48 hr from ewes during quiet, undisturbed conditions and analyzed for plasma ACTH, cortisol and progesterone concentrations. There were no significant changes in ACTH, cortisol or progesterone over time. Mean progesterone concentrations were significantly greater in the pregnant ewes than in all other ewes, and were greater in cycling ewes than noncycling or ovariectomized ewes. Mean ACTH was significantly greater in pregnant ewes than noncycling ewes, and mean cortisol was significantly greater in cycling ewes than in nonpregnant cycling or noncycling ewes. Ovariectomized ewes also had significantly greater mean cortisol concentrations than cycling ewes. Our objective was to examine whether mouse lines divergently selected for response to fescue toxicosis differed in the impact of increasing reproductive intensity on growth, final weight or first and second parity reproduction. Resistant (R) and susceptible (S) females were never mated (NR), mated only once (low reproduction, LR), mated after their first litter was weaned (moderate reproduction, MR) or paired continuously with a male (high reproduction, HR), allowing concurrent pregnancy and lactation. Final weight was significantly higher in mated than in not mated females (31.6 ± 0.6, 35.9 ± 0.6, 36.8 ± 0.6 and 37.2 ± 0.5 g for NR, LR, MR and HR females respectively), but the effect of increasing reproductive intensity was similar in both lines. Neither genetic line, reproductive treatment (LR and MR versus HR) nor their interaction affected first parity traits of dams or pups. In second parity, S dams produced larger litters at birth and weaning and heavier litters at birth than R dams, and MR dams produced more pups and heavier litters than HR dams. As with first litters, however, line by reproductive treatment interactions were not significant or important. Concurrent pregnancy and lactation (the HR treatment) had a similar dampening impact on the expected increase in litter size between first and second parity of both lines. Thus, lines divergently selected for toxicosis response did not differ significantly in the impact of increasing reproductive intensity on dam or pup growth, reproductive output through two parities or mature size. This conclusion should not be generalized. Selection responses in livestock should be monitored to ensure that improvement in one trait does not lead to deterioration in others. 相似文献

OBJECTIVES: To assess the association between goniodysgenesis, ocular measurements, and glaucoma in Great Danes. ANIMALS: 180 Great Danes. PROCEDURE: Eye examination and measurements were obtained from 180 Great Danes; for 30 of these dogs, depth of the anterior chamber, vitreal body length, and total depth of the globe were also measured. These data were merged with electronic pedigree information on 43,371 kennel club registered Great Danes. Relationships among goniodysgenesis, ocular measurements, and glaucoma and the heritability of goniodysgenesis were estimated. RESULTS: The degree of goniodysgenesis was significantly and positively associated with the likelihood of glaucoma. There was a significant association between the degree of goniodysgenesis in offspring and parents. The estimated heritability of the degree of goniodysgenesis was 0.52. The depth of the anterior chamber of the eye was also a good predictor of goniodysgenesis (ie, the dog was almost certain to have glaucoma if the depth was < 3.7 mm). If both parents had goniodysgenesis < 70%, then with 95% confidence, the occurrence of glaucoma in the ensuing offspring would be < 4/1000. This strategy translates to ensuring that the depth of the anterior chamber of the eye is > 3.7 mm for both parents. CONCLUSIONS AND CLINICAL RELEVANCE: The strong and significant correlation among goniodysgenesis, other eye measurements, and glaucoma and the significant heritability of goniodysgenesis suggests that glaucoma may be heritable in Great Danes. If so, glaucoma can be controlled by breeding only from sires and dams with a minimum degree of goniodysgenesis. 相似文献

Eighty-two dogs with lymphoma received a single 15-week course of chemotherapy, after which treatment was ceased until relapse. Fifty-six dogs (68%) achieved complete remission for a median 1st remission duration of 20 weeks. Forty-eight dogs relapsed, of which 30 repeated the induction cycle. In 22 of these dogs, 1st remission had been short, and they received maintenance chemotherapy; the other 8 dogs received 2 or 3 cycles of induction chemotherapy. Second remission rate for these 30 dogs was 87% (26 dogs). Overall disease control for the 38 dogs that remained on protocol was 44 weeks, which was not markedly shorter than for dogs treated with a previously reported protocol in which maintenance chemotherapy was instituted in all dogs after an identical 1st induction (VELCAP-L). Dogs that were febrile and dogs that were dyspneic were less likely to achieve a complete remission to induction chemotherapy. Of dogs that achieved a complete remission, those that were thrombocytopenic at entry had a shorter 1st remission, and dogs that were anorexic at entry had shorter overall disease control. There was a correlation between 1st remission duration and length of any subsequent remission obtained. The incidence of toxicity was high, particularly after the combination of doxorubicin and vincristine. Dose reductions because of toxicity did not markedly reduce remission duration. We conclude that discontinuous chemotherapy may reduce patient visits in a small number of patients because of long-term disease control. Three litters of Shetland Sheepdog pups born to the same bitch and 2 different sires were studied because of uncontrollable seizures or progressive neurologic dysfunction. Four pups from the 1st litter, 1 from the 2nd litter, and 4 from the 3rd litter had severe diffuse spongy degeneration of the white matter of the brain and spinal cord. An inherited basis for this syndrome was suspected. The purpose of this study was to evaluate the pups with currently available screening tests for the metabolic, biochemical, infectious, and toxicologic causes of leukodystrophy seen in humans and animals. Computed tomography scans revealed diffuse hypomyelination in the affected pup. Complete postmortem examination, including histopathology and electron microscopy, delineated a leukodystrophy resembling human Canavan's disease, but amino acid and organic acid metabolism abnormalities were not detected. No etiology for Shetland Sheepdog leukodystrophy has been found, but this condition represents another familial disease in the purebred dog population. 相似文献
