During seed production, Brassica seed may become infected with Xanthomonas campestris pv. campestris after systemic colonization of plants upon leaf infection, or alternatively, after flower infection. Polytunnel experiments were conducted in 2007 and 2008 to study the relative importance of these colonization routes resulting in seed infection. Cauliflower plants (Brassica oleracea) were spray-inoculated at the 8-leaf stage, after formation of cauliflowers or during flowering, at which stage leaves or blossoms were inoculated. Inoculation at all stages resulted in a relatively high percentage of systemic infection; the average estimated infection incidences for stem base and peduncle infections were 16 % and 19 %, respectively. When seed samples were examined by dilution plating for deep-seated infection following hot water treatment, Xcc was detected in 61 % of the 23 seed samples harvested from plants with inoculated flowers. However, symptom development in seedlings raised from the seeds could not be confirmed in a grow-out test under favourable conditions for Xcc infection at a high RH (>95 %) and a relative high temperature (28 °C). Xcc was not detected in 59 seed samples harvested from leaf-inoculated plants with the exception of one sample from plants inoculated at peduncle formation. In a third polytunnel experiment carried out in 2009, the population dynamics of Xcc on inoculated flowers was investigated. Following spray-inoculation of flowers, 52 % of the flowers were infected with Xcc. During development of siliques, infection incidence decreased slowly and at 56 dpi, 20 % of the superficially disinfected siliques were infected with Xcc. It was estimated that 0.18 % of the seeds was infected and that 1–10 % of the infected siliques contained infected seeds. The implications of these results for control of Xcc in a seed production crop are discussed.  相似文献

Among energy crops, short‐rotation coppices (SRC) are recommended to provide renewable energy. Compared to annual crops, willows and poplars are regarded as plants with low requirements for nutrients, herbicides, pesticides, and soil maintenance. However, only little is known about N‐fertilizer effects on SRC and the few studies are even inconsistent. Therefore, we studied the effects of N on yields of willows and poplars in a field experiment. The effects of N fertilization on nitrate leaching and nitrous oxide emissions from the loamy‐sand soil were also measured. Cuttings of willows (Salix viminalis clone Inger) and poplars (Populus maximovizcii × P. nigra clone max 4) were planted on farmland in 2008. The experiment was arranged in a random block design with three levels of N fertilizer (0, 50, and 75 kg N ha–1 y–1). After 2 y, the trees were harvested for the first time. Fertilization did not affect the yields of willows or poplars. However, the application of 75 kg N ha–1 y–1 caused an average increase of N leaching in the willow and poplar plots of 25 kg N ha–1 y–1 and 40 kg N ha–1 y–1, respectively. Emissions of N2O were increased by a maximum of only 0.2 kg N ha–1 y–1. Further, the N fertilizer stimulated the growth of the weed biomass in case of the willow plots by 46% and of the weed N content by 52% (r = 0.53). In conclusion, in the first 2 y, SRC could be produced in a more effective and environmentally friendly manner without mineral fertilizer.  相似文献

Xanthomonas fragariae is the causative agent of angular leaf spot disease of strawberry. Greenhouse experiments were conducted using a X. fragariae isolate tagged with a green fluorescent protein (GFP) for detailed population dynamic studies in and on leaves after spray‐inoculation. The GFP‐tagged bacteria were monitored with dilution plating of leaf washings and leaf extracts, and analysis of intact leaves using a non‐invasive monitoring system called PathoScreen, based on laser radiation of fluorescent cells in plant tissues and signal recording with a sensitive camera. PathoScreen was also used to monitor bacteria grown on an agar medium after leaf printing. During the first 3 days after inoculation, bacterial populations washed off leaves rapidly decreased by at least a factor of 1000, after which populations remained stable until 14 days post‐inoculation (dpi), when symptoms first started to appear. Thereafter, populations increased to a level of 1012 colony‐forming units (CFU) g?1 of leaf material or higher. Similarly, densities in leaf extracts were low during the first 3 days after inoculation, at a level of 100–1000 CFU g?1 of leaf tissue. Gradually populations increased to a level of 109–1012 CFU g?1 at 28 dpi. Higher densities of epiphytic populations were found on the abaxial side than on the adaxial leaf side during the first 2 weeks after inoculation. After spray‐inoculation of leaves, bacterial populations released from infected plants remained low until symptoms appeared, after which plants became highly infectious, in particular under high humidity.  相似文献

Four Large Münsterländer cross‐bred dogs affected with black hair follicular dysplasia (BHFD) and one unaffected control littermate were observed, and skin was sampled weekly over the first 19 weeks of life. Affected dogs were born with silvery grey hair, a consequence of melanin clumping in the hair shafts. Hair bulb melanocytes were densely pigmented, and contained abundant stage IV melanosomes but adjacent matrix keratinocytes lacked melanosomes. Melanin clumping was not prominent in epidermal melanocytes in the haired skin but occurred in the foot pads. Follicular changes progressed from bulbar clumping, clumping in the isthmus/infundibulum and finally to dysplastic hair shafts. Alopecia developed progressively in pigmented areas. Silver‐grey hair, melanin clumping, accumulation of stage IV melanosomes within melanocytes and insufficient melanin transfer to adjacent keratinocytes are also classic features of human Griscelli syndrome. The underlying cause in Griscelli syndrome is a defect of melanocytic intracellular transport proteins leading to inadequate and disorganized melanosome transfer to keratinocytes with resultant melanin clumping. In view of the correlation in the phenotype, histology and ultrastructure between both disorders, a defect in intracellular melanosome transport is postulated as the pathogenic mechanism in BHFD.  相似文献

In seasonal climates the timing of logging operations can be adjusted to avoid the major growing periods of understory plants and seedlings. We evaluated the understory vegetation at five pairs of hardwood-dominated sites in northern Wisconsin, U.S.A. where one site in each pair was logged during winter and the other during summer. All sites were dominated by sugar maple (Acer saccharum) and basswood (Tilia americana), with varying amounts of other native tree species. Most (84%) of the 247 vascular plant species recorded during the study are indigenous to the region. Winter-logged sites supported significantly higher numbers and percent cover of ecologically vulnerable native plant species as defined by independently established coefficients of conservatism. These differences between winter-logged and summer-logged sites suggest that winter logging may have fewer negative impacts than summer logging on vulnerable plant species and, in the long run, may help maintain plant biodiversity in managed forests of this region. In both types of sites, understory plant richness and diversity were significantly higher near logging roads, due largely to higher numbers of alien species and early successional native plants. Away from roads, neither species richness nor diversity differed between winter-logged and summer-logged sites. Studies that fail to account for differences in species composition and studies that fail to include samples of access roads or skid trails are likely to ignore important impacts of logging activities.  相似文献

The survival in drain water of two strains of Ralstonia solanacearum and three strains of Ralstonia pseudosolanacearum, including two strains able to cause wilt in roses, was determined. Water draining from drip‐irrigated rock wool mats on which roses were grown was supplemented with the pathogen and survival was monitored at 4, 12, 20 and 28°C for up to 112 days. All strains were able to survive for at least 112 days in drain water at 12 and 20°C, but at 4°C maximum survival was 56 days. At 28°C, the survival period was strain dependent, but was at least 56 days. Populations declined gradually in non‐sterile drain water to a low level (maximum 100 cfu mL?1 after 112 days). In sterile drain water (autoclaved prior to addition of populations), no or only a limited decline in populations was found at 112 days, dependent on strain and temperature. Drain water that tested negative for Ralstonia in the dilution plating assay was tested for the presence of cells in a viable but non‐culturable state (VBNCs). Tomato plants were inoculated, but no symptoms developed, and plants sampled 22 days post‐inoculation were negative in a plating assay. Therefore, no indications were found that VBNCs were present.  相似文献

The present study investigated genetic diversity, structure and hybridization in a collection of the endangered wild pear species Pyrus pyraster (L.) Burgd. A total of 278 putative ‘true type’ P. pyraster trees originating from seven populations in the federal state of Saxony in Germany were analyzed along with 35 pear cultivars commonly cultivated in Saxony. The genetic analysis was performed using nine nuclear microsatellite markers (ncSSR) and two paternally inherited chloroplast marker (cpDNA) amplifying in the intergenic spacer region trnQrps16 and the intron region rps16. On basis of the ncSSR dataset after STRUCTURE analysis 80 % of the wild pear individuals were assigned as ‘true type’ P. pyraster genotypes. The cpDNA analysis showed shared haplotypes in P. pyraster and P. communis but with an unequal frequency in both species. The analysis of molecular variance resulted in a moderate (ncSSR) and great (cpDNA) variation among ‘true type’ P. pyraster and the pear cultivars. The genetic diversity in the ‘true type’ P. pyraster populations was still high and the genetic structure between the populations low (ncSSR and cpDNA) indicating a genetic exchange between the populations by pollen and seeds. The clear discrimination between the P. pyraster and P. communis confirms our expectation of the existence of ‘true type’ P. pyraster individuals in the study area. The existing genetic integrity and the high genetic diversity argue for the implementation of preservation measures in P. pyraster.  相似文献
