This gonad enhancement study investigates the effect of different fresh and formulated feeds and feeding regimes on the growth and gonad quality of wild‐collected adult sea urchin, Tripneustes gratilla, under farm conditions for over 18 weeks. In the first 12 weeks (phase 1), urchins were fed fresh Ulva rigida (U); a 50:50 mixture of fresh U. rigida and Gracilaria gracilis (UG); fresh G. gracilis (G) and a formulated diet 20U (containing 20% U. rigida), and in the final 6 weeks (phase 2) of the study, diet was changed to a formulated feed (20U diet). By the end of phase 1, urchins fed the 20U diet produced gonads (50.72 ± 5.4 g) that were significantly heavier (p < .001) than the gonads of urchins fed the fresh seaweed diets (U, UG & G). By the end of phase 2, gonad weight of urchins in treatment groups UG‐20U and G‐20U were similar to those fed the 20U‐20U diet. Gonad colour of urchins in the G‐20U treatment became significantly lighter (ANOVA, p = .029) and poorer quality, compared with urchins in the U‐20U group. This gonad enhancement study, conducted on wild‐collected adult T. gratilla, has shown that a formulated feed (20U diet) can enhance gonad growth and produce commercially acceptable gonads. This farm‐based study supports previous findings from aquarium‐based studies by our group and indicates that short‐term sea urchin gonad enhancement can be carried out under farm conditions in South Africa.  相似文献

2-Methylisoborneol (MIB) and structurally related terpenoid compounds are responsible for millions of dollars of lost revenue to catfish farmers. In an attempt to determine enzymatic pathways of biotransformation and elimination of MIB, the in vitro metabolism of MIB was examined in the Ulvade strain of channel catfish (Ictalurus punctatus). Although cytochrome P450 (CYP) activities were observed and correlated with expression of specific isoforms (i.e. steroid hydroxylation and CYP3A expression), no metabolites of MIB were observed. To determine whether extrahepatic biotransformation may be occurring the in vivo metabolism and disposition of 14C-MIB was examined in Uvalde, USDA-103 channel catfish, and a channel catfish X blue catfish (Ictalurus furcatus) hybrid species. Confirming in vitro hepatic studies, no metabolites were observed in plasma from animals treated with an intra-arterial dose of 14C-MIB. 14C-MIB elimination was predicted using a two compartment model in each strain of fish. There was no significant difference in terminal half-lives between strains but possible differences in total body clearance and apparent volumes of distribution which may be related to higher lipid content in the hybrids. Results of these studies indicate biotransformation has no involvement in MIB elimination and that other physiological processes may play a more significant role in MIB disposition within Ictalurid fish species.  相似文献

Coagulation profiles in 27 horses with large colon volvulus   总被引:3,自引:3,他引:0  
Objective: The objective of this study was to evaluate coagulation profiles in horses with surgical treatment of large colon volvulus (LCV), and determine if an association exists between hemostatic dysfunction and outcome. Design: Prospective clinical investigation from February to December 2000. Setting: Large animal intensive care unit in a veterinary teaching hospital. Interventions: Blood was collected from horses intra‐operatively, 24, and 48 hours following surgical treatment for LCV. Measurements: Coagulation profiles, thrombin–antithrombin (TAT) levels, and D‐dimer concentrations were determined for each time point. The number of tests abnormal in the standard coagulation profile, defined as the degree of hemostatic dysfunction, was determined for each horse for the duration of the study period. The association between each test and outcome, as well as the degree of hemostatic dysfunction for each horse and outcome, was determined using univariate analysis and logistic regression. TAT levels and D‐dimer concentrations were compared to the results of the standard coagulation profile and to patient outcome using univariate analysis and logistic regression. Main results: Seventy percent of horses evaluated with surgical treatment of LCV had evidence of hemostatic dysfunction (3/6 tests abnormal). Only 18% of those patients had clinical signs recognized by the attending clinician as a coagulopathy. There was an association between the development of a coagulopathy and outcome, with horses with 4/6 tests abnormal being more likely to be euthanized, and those with 3/6 tests abnormal having a prolonged hospital stay. Platelet count, prothrombin time, and TAT levels may be helpful in predicting outcome in horses with LCV. Conclusions: Hemostatic function should be evaluated in horses with surgical treatment of LCV to detect subclinical coagulopathies and direct subsequent intervention.  相似文献

Deladenus siricidicola is the primary biocontrol agent of the invasive woodwasp, Sirex noctilio. The nematode is mass-reared in culture on the S. noctilio symbiotic fungus, A. areolatum, but can also be induced to convert into its infective form in culture. The aim of this study was to explore the level of variation among a collection of D. siricidicola strains to convert into the infective form in culture. In addition, we measured variation in growth of the nematode on a strain of A. areolatum that is commonly used for laboratory rearing. Both traits could influence laboratory rearing and efficiency in the field. The 22 strains from three countries showed significant variation in the frequency of conversion into the infective form in culture. Even genetically uniform strains from South Africa displayed a range of variation in this trait. There was also significant variation in the growth of D. siricidicola on A. areolatum. These results emphasise the importance of considering variation in traits in D. siricidicola that could influence a biological control program. They also provide tools that could be used to screen larger collections of D. siricidicola for variation in traits linked to virulence and reproduction.  相似文献

A new fluorescence‐based method for inbred haploid differentiation in maize kernels was developed by utilizing the R1‐nj colour marker in combination with fluorescence microspectroscopy and imaging. Seven inbred lines with varying R1‐nj expression were used in this study. The fluorescence response of the diploid kernels at the embryonic dye spot was shown to simultaneously exhibit lower intensity and occur at a higher wavelength than the fluorescence of the dye‐lacking haploid embryos. Intensity and area thresholds were applied to fluorescence images to sort the haploids from mixed sample populations, and sorting efficiencies of greater than 80% were achieved in all seven inbred lines (with values greater than 90% for five lines). The potential for high‐throughput sorting when fluorescence imaging is combined with existing technologies for seed handling as well as high sorting efficiency may make fluorescence a viable and promising alternative to current sorting methods for some inbred lines.  相似文献

This study investigated growth and gonad production of Tripneustes gratilla fed four protein‐rich artificial diets supplemented with varying amounts (0%, 5%, 15% and 20% weight/weight (w/w); designated 0, 5, 15 and 20U, respectively) of the macroalga Ulva over a 12‐week period. Gonad size, texture, colour and a number of production performance parameters were quantified and compared with urchins fed fresh Ulva (FU) and a combination of FU and artificial feed (FB). All artificial diets significantly ( 0.05) increased gonad somatic indices (GSI) compared with the FU treatment. The 20U treatment increased GSI by 205% by week 9, compared with a 57% increase in the GSI of urchins fed FU. Gonad colour was calculated using three colour parameters, namely L* (lightness), a* (redness) and b* (yellowness). Whereas L* and a* values did not differ significantly between treatments, b* values for all treatments, with the exception of the 20U and FB treatments, were significantly ( 0.05) lower or less yellow than the FU treatment by week 12. These results show that we have successfully formulated a feed (20U) which can produce commercially acceptable gonads in terms of both size and colour, indicating the potential for this artificial feed to support commercial echinoculture.  相似文献

Novel fire mitigation treatments that chip harvested biomass on site are increasingly prescribed to reduce the density of small-diameter trees, yet the ecological effects of these treatments are unknown. Our objective was to investigate the impacts of mechanical thinning and whole tree chipping on Pinus ponderosa (ponderosa pine) regeneration and understory plant communities to guide applications of these new fuel disposal methods. We sampled in three treatments: (1) unthinned forests (control), (2) thinned forests with harvested biomass removed (thin-only), and (3) thinned forests with harvested biomass chipped and broadcast on site (thin + chip). Plots were located in a ponderosa pine forest of Colorado and vegetation was sampled three to five growing seasons following treatment. Forest litter depth, augmented with chipped biomass, had a negative relationship with cover of understory plant species. In situ chipping often produces a mosaic of chipped patches tens of meters in size, creating a range of woodchip depths including areas lacking woodchip cover within thinned and chipped forest stands. Thin-only and thin + chip treatments had similar overall abundance and species richness of understory plants at the stand scale, but at smaller spatial scales, areas within thin + chip treatments that were free of woodchip cover had an increased abundance of understory vegetation compared to all other areas sampled. Relative cover of non-native plant species was significantly higher in the thin-only treatments compared to control and thin + chip areas. Thin + chip treated forests also had a significantly different understory plant community composition compared to control or thin-only treatments, including an increased richness of rhizomatous plant species. We suggest that thinning followed by either chipping or removing the harvested biomass could alter understory plant species composition in ponderosa pine forests of Colorado. When considering post-treatment responses, managers should be particularly aware of both the depth and the distribution of chipped biomass that is left in forested landscapes.  相似文献

Liver Protective Effects of Morinda citrifolia (Noni)   总被引:1,自引:0,他引:1  
This study evaluated the protective effects of Noni fruit juice on acute liver injury induced by carbon tetrachloride (CCl(4)) in female Sprague-Dawley (SD) rats. Liver damage (micro-centrilobular necrosis) was observed in animals pretreated with 20% placebo (drinking water) + CCl(4). However, pretreatment with 20% Noni juice in drinking water + CCl(4) resulted in markedly decreased hepatotoxic lesions. Furthermore, serum alanine aminotransferase and aspartate aminotransferase levels were significantly lower in the Noni group than the placebo group. In a correlative time-dependent study, one dose of CCl(4) (0.25 mL/kg in corn oil, p.o.) in female SD rats, pretreated with 10% placebo for 12 days, caused sequential progressive hepatotoxic lesions over a 24 h period, while a protective effect from 10% Noni juice pretreatment was observed. These results suggest that Noni juice is effective in protecting the liver from extrinsic toxin exposure.  相似文献
