Three major oat components, β-glucan, starch, and protein, and their interactions were evaluated for the impact on viscosity of heated oat slurries and in vitro bile acid binding. Oat flour from the experimental oat line "N979" (7.45% β-glucan) was mixed with water and heated to make oat slurry. Heated oat slurries were treated with α-amylase, lichenase, and/or proteinase to remove starch, β-glucan, and/or protein. Oat slurries treated with lichenase or lichenase combined with α-amylase and/or proteinase reduced the molecular weight of β-glucan. Heat and enzymatic treatment of oat slurries reduced the peak and final viscosities compared with the control. The control bound the least amount of bile acids (p < 0.05); heating of oat flour improved the binding. Heated oat slurries treated with lichenase or lichenase combined with α-amylase and/or proteinase bound the least amount of bile acid, indicating the contribution of β-glucan to binding. Oat slurries treated with proteinase or proteinase and α-amylase together improved the bile acid binding, indicating the possible contribution of protein to binding. These results illustrate that β-glucan was the major contributor to viscosity and in vitro bile acid binding in heated oat slurries; however, interactions with other components, such as protein and starch, indicate the importance of evaluating oat components as whole system.  相似文献

BACKGROUND: The liver has been suggested as a suitable target organ for gene therapy of Type 1 diabetes. However, the fundamental issue whether insulin-secreting hepatocytes in vivo will be destroyed by the autoimmune processes that kill pancreatic beta cells has not been fully addressed. It is possible that the insulin secreting liver cells will be destroyed by the immune system because hepatocytes express major histocompatibility complex (MHC) class I molecules and exhibit constitutive Fas expression; moreover the liver has antigen presenting activity. Together with previous reports that proinsulin is a possible autoantigen in the development of Type 1 diabetes, the autoimmune destruction of insulin producing liver cells is a distinct possibility. METHODS: To address this question, transgenic Non-Obese Diabetic (NOD) mice which express insulin in the liver were made using the Phosphoenolpyruvate Carboxykinase (PEPCK) promoter to drive the mouse insulin I gene (Ins). RESULTS: The liver cells were found to possess preproinsulin mRNA, translate (pro)insulin in vivo and release it when exposed to 100 nmol/l glucagon in vitro. The amount of insulin produced was however significantly lower than that produced by the pancreas. The transgenic PEPCK-Ins NOD mice became diabetic at 20-25 weeks of age, with blood glucose levels of 24.1 +/- 1.7 mmol/l. Haematoxylin and eosin staining of liver sections from these transgenic NOD PEPCK-Ins mice revealed the absence of an infiltrate of immune cells, a feature that characterised the pancreatic islets of these mice. CONCLUSIONS: These data show that hepatocytes induced to produce (pro)insulin in NOD mice are not destroyed by an ongoing autoimmune response; furthermore the expression of (pro)insulin in hepatocytes is insufficient to prevent development of diabetes in NOD mice. These results support the use of liver cells as a potential therapy for type 1 diabetes. However it is possible that a certain threshold level of (pro)insulin production might have to be reached to trigger the autoimmune response.  相似文献

Marine heatwaves are increasingly affecting marine ecosystems, with cascading impacts on coastal economies, communities, and food systems. Studies of heatwaves provide crucial insights into potential ecosystem shifts under future climate change and put fisheries social-ecological systems through “stress tests” that expose both vulnerabilities and resilience. The 2014–16 Northeast Pacific heatwave was the strongest and longest marine heatwave on record and resulted in profound ecological changes that impacted fisheries, fisheries management, and human livelihoods. Here, we synthesize the impacts of the 2014–2016 marine heatwave on US and Canada West Coast fisheries and extract key lessons for preparing global fisheries science, management, and industries for the future. We set the stage with a brief review of the impacts of the heatwave on marine ecosystems and the first systematic analysis of the economic impacts of these changes on commercial and recreational fisheries. We then examine ten key case studies that provide instructive examples of the complex and surprising challenges that heatwaves pose to fisheries social-ecological systems. These reveal important insights into improving the resilience of monitoring and management and increasing adaptive capacity to future stressors. Key recommendations include: (1) expanding monitoring to enhance mechanistic understanding, provide early warning signals, and improve predictions of impacts; (2) increasing the flexibility, adaptiveness, and inclusiveness of management where possible; (3) using simulation testing to help guide management decisions; and (4) enhancing the adaptive capacity of fishing communities by promoting engagement, flexibility, experimentation, and failsafes. These advancements are important as global fisheries prepare for a changing ocean.  相似文献

An adult Chinese red panda (Ailurus fulgens styani) transported by airplane from Florida to a North Dakota zoo died 1 week after arrival. Grossly, an interscapular abscess, subcutaneous inflammation, lymphadenitis, and pulmonary abscesses were observed. Microscopic findings included necrotizing inflammation in liver, lung, lymph node, and spleen. Chromobacterium violaceum was cultured from the interscapular abscess, liver, lung, and spleen and was injected into Swiss Webster mice. These mice died 18 hours postinoculation, and C. violaceum was cultured from liver, lung, and spleen. Chromobacterium violaceum is a sporadically reported but highly virulent pathogenic bacterium of both animals and humans typically found as a soil and water inhabitant of tropical and subtropical regions.  相似文献

Even after land is legally protected, ecological change continues to affect species and ecosystems. Changes vary from natural to human-caused, from beneficial to detremental, and from manageable to impossible to manage. Management can be approached on two levels: (1) an ecosystem-community (‘process-oriented’) level and (2) a species-population (‘species-oriented’) level. Management on these two levels sometimes conflicts. Key changes in reserves include: natural and human disturbance, manipulation of fire regimes, succession, imbalance of animal populations (e.g., elemination of large predators and population fluctuation of grazers), population and genetic change of rare species, introduction of exotics, pollution of air and water, interference with hydrologic regimes, and increased visitor pressure. Man-caused changes and poor management are still the greates threats to nature reserva systems.A preservationist ‘hands-off’ legacy, while idealistic and philosophically attractive, may contribute to lack of firm leadership in reserves by biologists and managers, Because the effects of human are now omnipresent, some form of management (if only to regulate visitor impact) usually is necessary. Dealing with ecological change requires statement of preservation goals, priorities, and clear policy on key issues; in addition, resource inventory, monitoring, and management-oriented research are required.  相似文献

A new method is described for hydroxymethylfurfural (HMF) in honey; accuracy and precision are improved over the most used optical and chemical methods. With a clarified honey solution containing 0.1% sodium bisulfite as reference and a similar solution without bisulfite as sample, a difference spectrum is obtained which represents only the HMF in the sample, without the interfering absorption of the honey. The average recovery was 97.5% for 24 additions to honey of 0.8--40 mg HMF/100 g. Forty honey samples ranging from 0 to 40 mg/100 g were analyzed by 3 methods with the following average results: Winkler optical method, 7.25; Winkler chemical method, 4.83; and new bisulfite method, 5.05 mg HMF/100 g honey. Values by the latter 2 methods did not differ at the P = 0.05 significance level.  相似文献

Flameless atomic absorption spectroscopy with a carbon rod atomizer was used to determine lead, cadmium, and chromium in whole-fish samples. Samples were dry-ashed, and the metals were separated by solvent extraction with ammonium pyrrolidine dithiocarbamate in methyl isobutyl ketone, and then back-partitioned into an aqueous acid solution for analysis. The back-partitioning step allows a direct comparison of sample solutions with aqueous solutions of the standard. Recoveries of the metals from fortified samples averaged 91% (+/-9.6) for lead and 100% (+/-5.6) for chromium at the 0.1-1 ppm level, and 100% (+/-13.3) for cadmium at the 0.01-0.1 ppm level.  相似文献

Potato is a major global crop that has an important role to play in food security, reducing poverty and improving human nutrition. Enhanced atmospheric CO2 concentrations provide an opportunity to increase potato yields in the future, but this will only be possible if the potato crop can cope with the other consequences of climate change caused by this rise in CO2. While climate change may impact biotic stress either positively or negatively, abiotic stresses are likely to be greatly increased and become a major threat to potato production. Increasing heat, drought and salinity stress will drive the need for greater understanding of genes, traits and management techniques that allow potato to cope with these stresses. In this review, we identify some of the key physiological and molecular adaptations of potato to these stresses and propose an ideotype which should include (1) optimal stomatal regulation to balance water loss and heat stress in leaves, (2) production of metabolites and transporters to scavenge reactive oxygen species and partition toxic elements, (3) enhanced root systems to maximise water capture, (4) maintenance of tuberisation under stress conditions and (5) stress avoidance by accelerating crop development and reducing time to yield. We discuss potential ways to achieve this ideotype, emphasising the need to benefit from genetic diversity in landrace and wild material by screening for traits in combined stress environments appropriate to future agroecosystems.  相似文献
