Tyrosine-amino transferase, an enzyme induced to the liver via glucocorticoids, caused clearly measurable activity rises in either sex, 24 hours after 1 and 2 applications. Increased levels continued to recordable from males on the 3rd day from application, whereas enzyme activities in females had already returned to normal by that time. Histological examination of liver sections, following 1 and 2 applications of parathion methyl (PM), revealed the tissue pattern of clearly manifest toxic hepatosis with detectable mesenchymal reactions. Histologically detectable damage reached its maximum on the 2nd and 3rd days from application. Sex-related differences were not recordable. Damage in response to the 2nd application was characterised by faster mesenchymal activation and higher frequency of cell necroses. The morphological picture of the liver was rated as having returned to normal, 14 days after 1 and 2 applications of PM. These results should not be interpreted as evidence to the involvement of specific, non-cholinergic mechanisms in the processes of damage observed. The alterations might rather be attributed to unspecific disorders in the energy balance or to the effect of "stress" during intoxication. 相似文献

OBJECTIVE: To determine the effect of maternally derived antibodies on induction of protective immune responses against bovine viral diarrhea virus (BVDV) type II in young calves vaccinated with a modified-live bovine viral diarrhea virus (BVDV) type I vaccine. DESIGN: Blinded controlled challenge study. ANIMALS: 24 neonatal Holstein and Holstein-cross calves that were deprived of maternal colostrum and fed pooled colostrum that contained a high concentration of (n = 6) or no (18) antibodies to BVDV. PROCEDURE: At 10 to 14 days of age, 6 seropositive and 6 seronegative calves were given a combination vaccine containing modified-live BVDV type I. All calves were kept in isolation for 4.5 months. Six calves of the remaining 12 untreated calves were vaccinated with the same combination vaccine at approximately 4 months of age. Three weeks later, all calves were challenged intranasally with a virulent BVDV type II. RESULTS: Seronegative unvaccinated calves and seropositive calves that were vaccinated at 2 weeks of age developed severe disease, and 4 calves in each of these groups required euthanasia. Seronegative calves that were vaccinated at 2 weeks or 4 months of age developed only mild or no clinical signs of disease. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicate that a single dose of a modified-live BVDV type-I vaccine given at 10 to 14 days of age can protect susceptible young calves from virulent BVDV type II infection for at least 4 months, but high concentrations of BVDV-specific maternally derived antibodies can block the induction of the response. 相似文献

The rise of atmospheric CO2 concentration ([CO2]) affects stomatal conductance and thus transpiration and leaf temperature. We evaluated the effect of elevated [CO2] levels under different water supply on daily sap flow and canopy microclimate (air temperature (Tc) and vapour pressure deficit (VPD)) of maize. The crop was cultivated in circular field plots under ambient (AMB, 378 μmol mol?1) and elevated [CO2] (FACE, 550 μmol mol?1) using free‐air CO2 enrichment with sufficient water in 2007, while in 2008 a DRY semicircle received only half as much water as compared to the WET semicircle from mid of July. In 2007, sap flow was measured in WET simultaneously under AMB and FACE conditions and was significantly decreased by elevated [CO2]. In 2008, sap flow was measured in all four treatments but not simultaneously. Therefore, data were correlated with potential evaporation and the slopes were used to determine treatment effects. Drought reduced whole‐plant transpiration by 50 % and 37 % as compared to WET conditions under AMB and FACE, respectively. Moreover, CO2 enrichment did not affect sap flow under drought but decreased it under WET by 20 % averaged over both years. The saving of water in the period before the drought treatment resulted in a displacement of dry soil conditions under FACE as compared to AMB. Under WET, CO2 enrichment always increased Tc and VPD during the day. Under DRY, FACE plots were warmer and drier most of the time in August, but cooler and damper short after the start of drought in July and from the end of August onwards. Thus, the CO2 effect on transpiration under drought was variable and detectable rather easy by measuring canopy microclimate. 相似文献

The N‐deposition in Germany is commonly calculated as values of about 20—30 kg/ha·yr. This range is based on the measurements of the nitrate and ammonium nitrogen bulk deposition, which does not include the gaseous N‐deposition and the direct N‐uptake by plants. The calculation of airbone N‐deposition from N‐balances of the Static Fertilization Experiment Bad Lauchstädt came to 50—58 kg/ha·yr. This is consistent with results from other European long‐term experiments. Using the newly developed 15N‐based ITNI‐system, the total airborne N‐deposition can be determined. For Bad Lauchstädt analogous to results of former measuring periods an annual N‐deposition of 65 kg/ha·yr was measured in 1998, a figure greater than the balanced values. The balanced and measured values show, that airborne N‐deposition is often underestimated and amounts to at least 50 kg/ha·yr, which is a significant burden on natural ecosystems. By taking this extra N‐input into account in calculations for fertilizer recommendations in agriculture a decrease of N‐losses can be achieved which, in turn can also induce a decrease in airborne N‐deposition. 相似文献

The continuing increase in atmospheric CO 2 concentration is predicted to enhance biomass production and to alter biochemical composition of plant tissues. In the present study, winter wheat ( Triticum aestivum L. cv. 'Batis') was grown under ambient air (BLOW, CO 2 concentration: 385 muL L (-1)) and free-air CO 2 enrichment (FACE, CO 2 concentration: 550 muL l (-1)) and two different nitrogen (N) fertilization levels (normal N supply: N100, 50% of normal N supply: N50). Mature kernels were milled into white flour and analyzed for the contents of crude protein, Osborne fractions, single gluten protein types and glutenin macropolymer. Elevated CO 2 caused significant reductions in crude protein and all protein fractions and types ( p < 0.001) except albumins and globulins. Effects were more pronounced in wheat samples supplied with normal amounts of N fertilizer. Crude protein was reduced by 14% (N100) and 9% (N50), gliadins by 20% and 13%, glutenins by 15% and 15% and glutenin macropolymer by 19% and 16%, respectively. Within gliadins, omega5-gliadins (-35/-22%) and omega1,2-gliadins (-27/-14%) were more affected than alpha-gliadins (-21/-13%) and gamma-gliadins (-16/-12%). Within glutenins, HMW subunits (-23/-18%) were more affected than LMW subunits (-12/-15%). According to these results, flour from high CO 2 grown grain will have a diminished baking quality. To our knowledge, these are the first results of elevated CO 2 concentrations impacts on wheat grain protein composition gained under relevant growing conditions at least for Central Europe. 相似文献

Histological examination was performed on the cervical spinal cord from 13 horses with chronic cervical compressive myelopathy of 4 to 29 months duration. Structural alterations were correlated with clinical features. At the level of compression, the spinal cord was grossly deformed. Histological alterations included nerve fibre swelling and degeneration, occasional spheroids, astrocytic gliosis, increased macrophage activity and increased perivascular collagen. Myelin degeneration or loss at the level of the compressive lesion was greatest in the ventral and lateral funiculi and less consistently present in the dorsal funiculi. Asymmetry of lesions in the dorsal funiculi was associated with asymmetry of clinical signs in 5 horses. Histological alterations in areas of Wallerian degeneration were similar to that at the level of spinal cord compression, except that perivascular collagen was not increased. Wallerian degeneration was present cranial to the compressed site in the superficial portions of the lateral funiculi and in the middle of the dorsal funiculi. Caudal to the compressed site it was present in the ventral funiculi adjacent to the ventral median fissure and in the middle of the lateral funiculi. Deformation of the spinal cord did not correlate with the severity or duration of clinical signs but was positively correlated with the amount of perivascular collagen increase. The amount of nerve fibre swelling was not correlated with the severity of clinical signs but was negatively correlated with their duration. A rapid loss of nerve fibres apparently occurred early in the course of compression, since there was a marked decrease in the amount of nerve fibre swelling and Marchi stained degenerating myelin with increasing clinical duration.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

Twenty-four healthy, mixed-breed hound-type dogs were evenly and randomly assigned to a placebo control group, one of four dosages of deracoxib (0.3, 1, 3, or 10 mg/kg), or carprofen (2.2 mg/kg). Oral dosing of placebo, carprofen, or deracoxib was done 30 minutes before intraarticular injection of urate crystal suspension for induction of synovitis. Ground reaction forces, subjective clinical lameness scores, pain, joint effusion, and quantitative pain threshold responses were measured in a blinded fashion before induction of synovitis and 2, 4, 6, 8, 12, and 24 hours after injection. The medium and high dosages of deracoxib were effective in preventing lameness and pain associated with synovitis. Carprofen was also somewhat effective in attenuating the severity of urate-induced synovitis but to a lesser degree than the medium dose of deracoxib. Preemptive deracoxib treatment at dosages as low as 1 mg/kg reduced lameness and pain of synovitis associated with intraarticular administration of urate crystals. 相似文献

AIM: To determine the relationship between the concentrations of Cu in plasma and serum in red deer, and to compare this relationship with those previously reported in cattle and sheep. METHODS: Paired serum and heparinised plasma samples from 114 red deer from 10 herds (n=6-20 per herd) were analysed for concentrations of Cu. Samples were collected either at slaughter (n=84; eight herds) or by jugular venepuncture (n=30; two herds). Thirty-nine of the samples taken at slaughter were from adult hinds from four herds, while other samples were taken from 10–14-month-old males, except for one herd (10 samples) where an equal number of 8–9-month-old males and females were sampled. The effect of age, gender and herd on the relationship between concentrations of Cu in plasma and serum was assessed using univariate ANOVA. The individual results for concentrations of Cu in serum were compared with those in plasma, using limits-of-agreement plotting. RESULTS: The mean concentration of Cu in plasma was not significantly different from that of serum (0.048; 95% CI=-0.14 to 0.24 µmol/L). There was no effect of age, sex or herd on this relationship. CONCLUSIONS: In deer, there was no significant difference between concentrations of Cu in plasma and serum regardless of age, sex or herd of origin. CLINICAL RELEVANCE: In contrast to the situation in cattle and sheep, the concentration of Cu in serum can be used interchangeably with that in plasma for the estimation of concentration of Cu in blood of red deer. 相似文献

Pulsed field gel electrophoresis (PFGE) using 3 enzymes (Spe I, Xba I, Avr II) and repetitive sequence polymerase chain reaction (REP-PCR) with 3 primers (BOX, ERIC, REP) were compared with respect to their validity as a method for identifying transmission of Salmonella on swine farms. Sixty-eight isolates of Salmonella were obtained from feces of swine, cats, mice, and birds, insect body parts, water and floor samples, and boot scrapings collected on 9 swine farms in Illinois USA. Genetic distances between isolates were calculated using the Dice matching coefficient. Cluster analysis of distance matrices was conducted using the UPG-MA algorithm. There was no significant difference between PFGE and REP-PCR in the genetic diversity detected; however, REP-PCR differentiated between 14 pairs of isolates which PFGE identified as identical. There were no significant differences between PFGE and REP-PCR in identifying all or most close genetic links as isolates from the same farm, the same building, and from the same sampling visit, suggesting ecological validity for both methods. Thus, REP-PCR should be considered as an acceptable and perhaps preferable alternative to PFGE as a genotyping method for studies of Salmonella transmission. 相似文献
