Objective   We evaluated combinations of two commercial semen extenders and three concentrations of glycerol to determine the combination that yielded the highest post-thaw sperm motility.
Design   A randomised 2 × 3 block design was used.
Procedure   Semen was collected from four stallions (6 collections per stallion). The sample was diluted with either a dried skim-milk glucose extender (EZ Mixin Original Formula) or a chemically defined, milk-free diluent (INRA 96), and each was used in combination with 2%, 3% or 4% glycerol in standard commercial freezing medium. Sperm motility was assessed by microscopy in fresh and post-thaw semen.
Results   There was a significant difference between the two extenders in the motility of spermatozoa after cryopreservation (48.9% for INRA 96; 38.6% for EZ Mixin OF; P < 0.0001). Glycerol at 4% in freezing medium yielded the highest post-thaw motility, significantly better than 2% ( P < 0.05). Three of four stallions had significantly higher post-thaw motility using INRA 96 relative to EZ Mixin OF ( P < 0.01), and two of four stallions had significantly higher post-thaw motility using 4% glycerol ( P < 0.05). The combination of INRA 96 and 4% glycerol in freezing medium gave the highest average post-thaw motility of 51.5%.
Conclusion   In this study, INRA 96 combined with 4% glycerol yielded an average recovery of progressively motile sperm consistently above the 35% target.  相似文献

SUMMARY Three surgical techniques, grouped as intra-articular techniques, extra-capsular techniques and fibular head transposition, were used for repair of the cranial cruciate ligament (CCL)-deficient stifle in 113 dogs over a 4 year 4 month period. The clinical outcome of the techniques were compared using information provided by the owners and physical examination. Regardless of surgical technique, 85.7 to 91.0% of dogs showed clinical improvement after surgery. However, less than 50% of dogs became clinically sound on the operated leg and 9.0 to 14.3% of dogs remained persistently lame on the operated leg. No statistical association was found between result after surgery and age, body weight, sex, duration of injury before surgery, association with injury, tibial plateau angle, degree of radiographic osteoarthritis before surgery or the presence of concurrent medial meniscal injury. On physical examination, extra-capsular techniques appeared superior to the fibular head transposition in terms of joint stability and limb function. Concurrent medial meniscal injury necessitating meniscectomy existed in 48.0% of cases. Twenty-two percent of dogs ruptured their contralateral CCL at an average of 14 months after the first.  相似文献

OBJECTIVE: To evaluate postoperative morbidity in dogs after experimental cranial cruciate ligament transection and immediate stifle stabilization using an arthroscopically assisted or open arthrotomy technique. STUDY DESIGN: Experimental, prospective study. ANIMALS: Thirteen mature, healthy dogs. METHODS: Dogs were randomly assigned to 1 of 2 groups. Seven underwent open arthrotomy while 6 underwent arthroscopy of 1 stifle joint. Cranial cruciate ligaments were transected and debrided and all stifles were stabilized using 2 lateral extracapsular fabellar-tibial sutures. Minimally invasive suture placement was employed in the arthroscopy group. All animals were evaluated for 9 weeks using kinetic gait assessments, comfortable stifle range of motion measurements, thigh girth measurements, differential cell counts of synovial fluid, and subjective scores of behavior, limb use, and lameness. RESULTS: Significant differences in postoperative morbidity were observed during the 9-week postoperative period. Greater peak vertical force for 8 weeks (P=.015), vertical impulse for 6 weeks (P=.044), comfortable stifle range of motion for 9 weeks (P=.017), comfortable stifle flexion for 4 weeks (P=.005), and operative limb thigh circumference (P=.020) for 9 weeks were observed for the arthroscopy group. A trend towards a lower differential mean synovial polymorphonuclear cell count in the arthroscopy group was seen at 4 and 8 weeks postoperatively. No differences in subjective evaluation scores were noted. CONCLUSIONS: In this study population, significant differences were seen between the arthroscopy and arthrotomy groups for peak vertical force, vertical impulse, comfortable stifle range of motion, comfortable stifle flexion, and thigh circumference data. CLINICAL RELEVANCE: The results of this study suggest that short-term postoperative morbidity may be reduced in dogs receiving arthroscopic joint surgery with a limited approach for stifle stabilization as compared with a traditional open arthrotomy technique.  相似文献

SUMMARY Merino sheep, which were hand jetted with ivermectin jetting fluid, and untreated sheep were challenged with larval implants of Lucilia cuprina, Lucilia sericata, Calliphora noclva and Chrysomya ruflfacles at intervals of about 2 weeks from 6 to 16 weeks after treatment. Both Lucilia species produced strike rates of about 90% in untreated sheep; the respective rates were lower for Chr rufifacies (55%) and C nociva (60%). Strike rates for the treated group were about 17, 11, and 9% for L cuprina, L sericata, and Chr rufifacies, respectively. Only 1 implant site in the treated group was struck by C nociva. Treated sheep had significantly (P < 0.01) longer time to first strike than did untreated ones for each species of fly. L sericata, Chr rufifacies, and C nociva larvae implanted on treated animals had significantly (P < 0.05) longer time to first strike than did L cuprina larvae.  相似文献

AIMS: To assess the feasibility of applying animal-based welfare assessments developed for use in Europe on New Zealand dairy farms; in particular, to identify measures which could be evaluated during a single visit at milking time alongside whole herd locomotion scoring.

METHODS: A protocol for animal welfare assessment, developed in the United Kingdom (UK), was evaluated. Measures that were suitable for use on pasture-based dairy farms in New Zealand were then assessed for practicability on 59 farms across New Zealand, during and immediately after milking, alongside whole herd locomotion scoring. Where data were collected the results were compared to those from a UK study of 53 dairy farms.

RESULTS: Thirteen observations of the physical condition of cows were considered suitable for measurement, excluding observations related to hock lesions as they are rarely observed on pasture-based farms. Five of these measures were not assessed as there was not time to do so during milking alongside whole herd locomotion scoring. Thus, the prevalence of dirty flanks, hind limbs and udders, dull coat, thick hairy coat, significant hair loss, very fat cows (body condition score (BCS) ≥7 on 1–10 scale) and very thin cows (BCS≤3), were recorded. Three measures of behaviour were considered suitable for measurement on-farm, but only locomotion score was practicable and was measured. Farmer-estimates for the incidence of mastitis, lameness, sudden death, milk fever and other diseases were also obtained.Overall, dirty flanks, dirty udders and estimated milk fever incidence were more prevalent in this study than in the UK. The prevalence of thin and fat cows, lame cows and estimated mastitis incidence were much lower in the present study than on UK farms.

CONCLUSIONS AND CLINICAL RELEVANCE: Animal-based assessments can be used on dairy farms in New Zealand, but need to be modified from those developed for housed cows.Welfare on these farms was generally good compared to those in the UK, but these results need to be confirmed on more farms using a wider range of assessments than proved practicable in this study.  相似文献

The present study assesses the oxidative burst activity from polymorphonuclear leukocytes (PMNLs) from bovine leukemia virus (BLV)-infected cows. Fifteen clinically healthy cows were divided into serologically positive cows without any hematological alteration, serologically positive animals with persistent lymphocytosis (PL) and healthy serologically negative cows. The oxidative burst activity from the PMNLs was evaluated by flow cytometry using 2',7'-dichlorofluorescein diacetate as a probe. PMNLs from each cow were incubated with heat-killed Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) to stimulate oxidative burst activity. The results of the present work showed no significant difference in the oxidative burst activity without any stimulus and elicited by S. aureus. Conversely, a decrease in the oxidative burst index induced by E. coli in PMNLs was observed in BLV-infected cows.  相似文献

SUMMARY The decrease in the prevalence of Corynebacterium pseudotuberculosis after two generations of vaccination against the disease it causes, was used to estimate the rate of control of caseous lymphadenitis (CLA). Three groups of 150 sheep, of which 50 in each group were artificially infected with C pseudotuberculosis and 100 in each group were uninfected sheep, were run separately for 40 months and shorn 5 times to promote the spread of CLA. One lot of 50 infected sheep and 2 lots of 100 uninfected sheep were vaccinated against CLA. The rate of spread of CLA was recorded. Sheep vaccinated against CLA and naturally exposed to infection had a 74% lower infection rate than unvaccinated sheep. Sheep vaccinated against CLA and exposed to only vaccinated infected sheep had a 97% lower infection rate. Unvaccinated sheep had a 76% infection rate, with 77% of the transmission occurring at the 4th and 5th shearings, without any discharging CLA abscesses being observed. This study supports the view that in Australian wool producing flocks, CLA spreads mainly from sheep with discharging lung abscesses to sheep with shearing cuts. Vaccinated sheep infected with CLA have 96% fewer lung abscesses compared with unvaccinated infected sheep and are therefore less likely to spread this disease to other sheep .  相似文献
